RESUMEN

It is largely documented that neurodegenerative diseases can be effectively treated only if early diagnosed. In this context, the structural changes of some biomolecules such as Tau, seem to play a key role in neurodegeneration mechanism becoming eligible targets for an early diagnosis. Post-translational modifications are responsible to drive the Tau protein towards a transition phase from a native disorder conformation into a preaggregation state, which then straight recruits the final fibrillization process. Here, we show for the first time the detection of pre-aggregated Tau in artificial urine at femto-molar level, through the concentration effect of the pyro-electrohydrodynamic jet (p-jet) technique. An excellent linear calibration curve is demonstrated at the femto-molar level with a limit of detection (LOD) of 130 fM. Moreover, for the first time we show here the structure stability of the protein after p-jet application through a deep spectroscopic investigation. Thanks to the small volumes required and the relatively compact and cost-effective characteristics, this technique represents an innovative breakthrough in monitoring the early stage associated to neurodegeneration syndromes in different scenarios of point of care (POC) and such as for example in long-term human space exploration missions.

The electrohydrodynamic (EHD) jetting of fluids is used for several applications such as inkjet printing, atomization of analyte in mass spectrometry, liquid metal alloy ion sources, and electrospinning of polymer fibers. Historically, the bulk of research has focused on nonviscous, highly conductive fluids which are most suitable for EHD spray and printing, while there is relatively little experimental work on EHD jetting of highly viscous liquid dielectrics. We studied the dynamics of oscillation and pulsating jetting from a suspended drop of polydimethylsiloxane (PDMS) polymers in an electric field, with particular attention to the viscosity dependence of the oscillation period and meniscus elongation and contraction time over a wide viscosity range (102-105 cSt). The reported results could help the appropriate design of EHD processes and may open new possibilities for the rheological characterization of liquid polymers using small volumes at the scale of nanoliters.

OBJECTIVE: The aim: To increase the efficiency of surgical treatment of patients with chronic lung abscesses by developing mini-invasive methods of surgical treatment using electrosurgical technologies. PATIENTS AND METHODS: Materials and methods: Conducted study of the results of surgical treatment of 78 patients with chronic lung abscesses operated from 2011 to 2021. Patients were divided into two groups: the main group (37 patients who were treated using developed technologies) and a comparison group (41 patients, treated using traditional tactics). RESULTS: Results: Transthoracic and endobronchial sanitation of the purulent cavity in the lung at the first stage of treatment contributed to the rapid elimination of inflammation and significantly accelerated the regeneration of lung tissue. Clinical effectiveness in the main group was expressed in reducing the phenomena of intoxication, decrease in Leukocyte intoxication index (LII) (early as on day 5 after surgery), on the 10th day, a significant reduction in patients bacterial excretion was noted (in the main group by 18.9%, in the comparison group - by 14.6%), the average time of reducing the abscess cavity by 1/4 of the volume 6 days less, the healing time of the cavity of the AL which is on average 13 and 16 days, respectively, less. CONCLUSION: Conclusions: The developed methods of surgical interventions made it possible to significantly positively influence the level of endogenous intoxication indicators, avoid resection surgical interventions, reduce the number of postoperative complications, avoid damage to neighboring organs, reduce the time of patients with achieving a stable positive effect.

To meet the growing demand for early fatal disease screening among large populations, current fluorescence detection instruments aiming at point-of-care diagnosis have the tendency to be low cost and high sensitivity, with a high potential for the analysis of low-volume, multiplex analytes with easy operation. In this work, we present the development of a miniaturized, high numerical aperture confocal fluorescence scanner for sub-micro-liter fluid diagnosis. It is enhanced with high-rate analyte accumulation using a pyroelectro-hydrodynamic dispensing system for generating tiny, stable sample droplets. The simplified confocal fluorescence scanner (numerical aperture 0.79, working distance 7.3 mm) uses merely off-the-shelf mass-production optical components. Experimental results show that it can achieve a high-sensitive, cost-efficient detection for sub-micro-liter, low-abundant (0.04 µL, 0.67 attomoles) fluid diagnosis, promising for point-of-care diagnosis.

HYPOTHESIS: Thin liquid films are important in many scientific fields. In particular, films with both the surface layers exposed to a different fluid phase, known as freestanding films, are relevant in the ambit of foams and emulsions. Hence, there is a great interest in developing novel techniques allowing to form large and stable freestanding liquid films and to follow their dynamics. EXPERIMENTS: We develop a novel opto-mechanical tool allowing to perform and study the preparation and the capillary leveling flow of axisymmetric bare freestanding liquid films. The tool is composed by a customized motorized iris diaphragm and by an innovative joint imaging setup combining digital holography and white light color interferometry that enables real-time measurement of film thickness over a large field of view. The dynamics of films made of a model Newtonian fluid, i.e., high-viscosity silicone oil, is studied. Direct numerical simulations and a hydrodynamic model based on the lubrication theory are used to support the experimental results. FINDINGS: Iris opening induces the formation of large circular freestanding films with a stepped profile. Once iris opening is stopped, the films undergo a capillary leveling flow tending to flatten their profile. The leveling flow follows the theoretical scaling given by Ilton et al. [1]. We prove through numerical simulations that an equi-biaxial extensional flow occurs at the film center. Furthermore, we observe the formation and dynamics of dimples in bare freestanding films for the first time.

The demand for sensors capable of measuring low-abundant collagen in human fluids has highly increased in recent years. Indeed, collagen is expected to be a biomarker for chronic diseases and could monitor their progression. Here we show detection of highly diluted samples of collagen at picogram level thanks to an innovative pyro-electrohydrodynamic jet (p-jet) system. Through the intense electric fields generated by the pyroelectric effect in a ferroelectric crystal, the collagen solution was concentrated on a small area of a slide that was appropriately functionalized to bind proteins. The collagen molecules were labeled by an appropriate fluorophore to show how the number of tiny droplets influences the limit of detection of the technique. The results show that the p-jet is extremely promising for overcoming the current detection limits of collagen-based products in human fluids, performing 10 times better than the enzyme-linked immunosorbent assay (ELISA) and thus paving the way for the early diagnosis of related chronic diseases.

We investigate a hybrid re-configurable three dimensional metamaterial based on liquid crystal as tuning element in order to build novel devices operating in the terahertz range. The proposed metadevice is an array of meta-atoms consisting of split ring resonators having suspended conducting cantilevers in the gap region. Adding a "third dimension" to a standard planar device plays a dual role: (i) enhance the tunability of the overall structure, exploiting the birefringence of the liquid crystal at its best, and (ii) improve the field confinement and therefore the ability of the metadevice to efficiently steer the THz signal. We describe the design, electromagnetic simulation, fabrication and experimental characterization of this new class of tunable metamaterials under an externally applied small voltage. By infiltrating tiny quantities of a nematic liquid crystal in the structure, we induce a frequency shift in the resonant response of the order of 7-8% in terms of bandwidth and about two orders of magnitude change in the signal absorption. We discuss how such a hybrid structure can be exploited for the development of a THz spatial light modulator.

Monitoring of multiple sclerosis (MS) requires additional molecular markers. Recently, we used original TCA-precipitation/extraction approach in combination with MALDI TOF/TOF mass-spectrometry and identified earlier unknown 48 kDa form of the unconventional myosin IC isoform b (Myo1C) in blood serum of the MS patients. Further examination of TCA-extracted fraction of blood serum of these patients by means of thin-layer chromatography and HPLC gel-filtration allowed detecting 300-500 Da peptides. MALDI TOF/TOF massspectrometry of these peptides showed that they contain Ser-Pro-Cys amino acid sequence. We discussed potential mechanisms of a release of these peptides that were earlier unknown in blood serum of the MS patients.

RESUMEN

Control of liquid crystal (LC) orientation using a proper SiO2 alignment layer is essential for the optimization of vertically aligned nematic (VAN) displays. With this aim, we studied the optical anisotropy of thin SiO2 films by generalized ellipsometry as a function of deposition angle. The columnar SiO2 structure orientation measured by a noninvasive ellipsometry technique is reported for the first time, and its morphology influence on the LC alignment is demonstrated for large deposition angles.

The switchable three-component reactions of 5-amino-3-methylisoxazole, salicylaldehyde and N-aryl-3-oxobutanamides under different conditions were studied and discussed. The unexpected influence of the aryl substituent in N-aryl-3-oxobutanamides on the behavior of the reaction was discovered. The key influence of ultrasonication and Lewis acid catalysts led to an established protocol to selectively obtain two or three types of heterocyclic scaffolds depending on the substituent in the N-aryl moiety.

We study the presence of dark and bright modes in a planar metamaterial with a double rod unit cell introducing geometric asymmetry in rod lengths. The dark mode displays a Fano-type resonance with a sharp asymmetric profile, rendering it far more sensitive than the bright mode to slight variations of the dielectric environment. This peculiar feature may envisage the possible application of the asymmetric dimer metamaterial as an optical sensor for chemical or biological analysis, provided that the effect of material losses on the dark mode quality factor is properly taken into account.

Recently important efforts have been dedicated to the realization of a new kind of photonic crystals, known as photonic quasicrystals, in which the lack of the translational symmetry is compensated by rotational symmetries not achievable by the conventional periodic crystals. Here we show a novel approach to their fabrication based on the use of a programmable Spatial Light Modulator encoding Computer-Generated Holograms. Using this single beam technique we fabricated Penrose-tiled structures possessing rotational symmetry up to 23-fold, and a two-dimensional Thue-Morse structure, which is an aperiodic structure not achievable by multiple beam holography.

We have studied the electro-optical and angular behavior of holographic-polymer-dispersed liquid crystal gratings at different wavelengths, in the visible and in the near-infrared range. As usual in these kinds of materials, a strong polarization dependent behavior was observed. Our samples showed very high diffraction efficiency for p-polarized radiation at 1.55 microm, which is very interesting for many possible applications in the telecom field. However, we also observed a very unusual behavior for visible p-polarized light and we try to suggest some explanation for that. By analyzing the angular dependence of the diffraction efficiency, we could measure the components of the permittivity modulation tensor and infer important information about the main parameters involved in the grating structure: the degree of phase separation and the anisotropy in the liquid crystal droplet distribution. In our opinion, this simple and nondestructive methodology can be very useful for studying these kinds of materials and getting information on their morphology, in view of optimizing their properties. Finally, we discuss the role of the refractive index optical dispersion in order to describe the behavior of these materials at different wavelengths. These remarks are especially important when properties in the infrared range are extrapolated from measurements in the visible.
