This essay aims to evaluate the composition of fiber in neutral detergent, digestibility of plant components, as well as ruminal disappearance rate of fodder dry matter from five ryegrass fields submitted to different cut systems for the production of haylage. The experimental design was the randomized blocks one, at a 5 x 3 factorial scheme, with five cultivars (cv.) of ryegrass Lolium multiflorum (cv. Barjumbo, cv. BarHQ, cv. Ribeye, cv. Potro and cv. Willians) associated with three harvesting management systems: 1- single harvesting system at a pre-floral stage; 2 - double harvesting system, with a cut at vegetative stage and another at pre-floral stage; and 3 - triple harvesting system, with a cut at vegetative stage, a cut at full vegetative stage and a cut at pre-floral stage, with three repetitions each. Similarities were noticed on the results of fodder digestibility of different fields, however, cv. Potro presented a higher ruminal disappearance rate after 48 hours, with 69.27%. Generally, the harvesting management system with the increase of ryegrass cut frequency determined increments on the percentage of lignin and the reduction in the concentration of total nutrients and dry matter digestibility of the resulting hylage.(AU)


O objetivo do presente trabalho foi avaliar a composição da fibra em detergente neutro, digestibilidade dos componentes da planta, bem como a taxa de desaparecimento ruminal da matéria seca da forragem de cinco cultivares de azevém submetidos a diferentes sistemas de cortes para produção de silagem pré-secada. O delineamento experimental foi em blocos ao acaso,, em um fatorial 5 x 3, sendo cinco cultivares de azevém Lolium multiflorum (cv. Barjumbo, cv. BarHQ, cv. Ribeye, cv. Potro e cv. Willians) associados a três sistemas de manejo de colheita: 1 - sistema com única colheita no estádio de pré-florescimento; 2 - sistema com uma colheita no estádio vegetativo e outra no estádio de pré-florescimento; e 3 - sistema com uma colheita no estádio vegetativo, uma no estádio pleno vegetativo e outra no estádio de pré-florescimento, com três repetições cada. Notou-se similaridade entre os resultados de digestibilidade da forragem entre os cultivares, porém o cv. Potro apresentou a maior taxa de desaparecimento ruminal após 48 horas, com 69,27%. De maneira geral, o sistema de manejo de colheita mostrou que o aumento do número de cortes do azevém determinou incrementos nos teores de lignina e redução na concentração de nutrientes digestíveis totais e da digestibilidade da matéria seca nas silagens pré-secadas resultantes.(AU)

O objetivo do presente trabalho foi avaliar a produção de fitomassa fresca e de fitomassa seca, assim como a composição física e bromatológica do feno do trigo cultivar BRS Umbu sob efeitos de dois níveis de adubação nitrogenada (120kg ha-1 e 180kg ha-1) e dois estádios fenológicos de colheita (pré-florescimento e grão farináceo). O delineamento experimental foi blocos ao acaso, em esquema fatorial 2x2, composto por quatro tratamentos com quatro repetições. Não houve interação (P>0,05) entre os níveis de adubação nitrogenada e os estádios fenológicos de colheita para todas as variáveis. A produção de fitomassa seca (P<0,05) nos estádios de grão farináceo e pré-florescimento foi de 10.171 e 4.982kg ha-1, respectivamente. A maior dose de N incrementou a produção de fitomassa seca em 775kg ha-1. Houve aumento da participação de espigas com o avanço do ciclo, apresentando-se 43,0% e 16,2% nos estádios grão farináceo e pré-florescimento, respectivamente. No estádio fenológico de pré-florescimento, a participação de folha verde foi superior (37,1% contra 9,8% da MS total). O feno colhido em estádio de grão farináceo apresentou menores teores de proteína bruta e fibra em detergente neutro. Os fenos produzidos apresentaram características distintas, o que permite seu uso em diferentes estratégias de alimentação de ruminantes.(AU)


The objective was to evaluate the production of the fresh and dry weight, physical and chemical composition of wheat hay, cv. BRS UMBU, under effects of two levels of nitrogen fertilization (120kg ha-1 and 180kg ha-1) and two harvest stages (pre-flowering and grain dough). The experimental design was randomized blocks in factorial 2X2, composed of four treatments with four replications. There was no interaction (p>0.05) between the levels of nitrogen fertilization and growth stages of harvest for all variables. The production of dry matter in the dough stage and pre-flowering were 10.171 and 4.982kg ha-1, respectively. The higher N rates increased the production of dry matter of 775kg ha-1. There was increased participation of spikes with the advancement of the cycle, presenting 43.0% and 16.2% in the dough stage and pre-flowering, respectively. In the growth stage of pre-flowering, the share of green leaf was higher (37.1% against 9.8% of the total MS). The hay harvested at dough stage had lower NDF and CP levels. Thus, each treatment presented favorable characteristics allowing its use in different strategies in ruminant nutrition.(AU)

O objetivo do trabalho foi avaliar a produtividade e as características bromatológicas dos estratos superior e inferior de gramíneas perenes em sistema contínuo de cortes. As cultivares avaliadas foram: Cynodon nlemfuensis Vanderyst cv. Estrela-roxa; Cynodon dactilon (L.) Pears cv. Coast-cross 1; Cynodon nlemfuensis Vanderyst cv. Tifton 68; Cynodon ssp. cv. Tifton 85; Cynodon dactilon sp. cv. Jiggs; Hemárthria altissima cv. Roxinha e Penissetum clandestinum cv. Quicuio. Todos os cortes foram realizados no momento em que cada cultivar alcançava 95% de interceptação luminosa. As cutivares Jiggs e Tifton 68, com 21.348 e 21.016kg ha-1, respectivamente, tiveram as maiores produções (P<0,05) de fitomassa seca acumulada. Na média geral dos cortes, o quarto corte apresentou a menor relação estrato superior:inferior em relação aos demais (P<0,05). Em relação ao estrato superior, a Tifton 85 apresentou o maior teor de PB, sendo superior estatisticamente (P<0,05) no primeiro, segundo e quarto cortes, enquanto no terceiro corte a Coast-cross apresentou o maior teor proteico. No geral, as cv. Jiggs e Tifton 68 apresentaram a maior produção de fitomassa acumulada. Já a cv. Tifton 85 foi responsável pelos maiores níveis de PB, enquanto os teores de carboidratos estruturais mantiveram padrão constante, tanto no estrato superior quanto inferior, em sistema de quatro cortes sucessivos.(AU)


The aim of this study was to evaluate the productivity and the bromatologic characteristics of the upper and lower strata of perennial grasses in four cut systems. The cultivars evaluated were: Cynodon nlemfuensis Vanderyst cv. Estrela Roxa; Cynodon dactilon (L.) Pears cv. Coast-cross 1; Cynodon nlemfuensis Vanderyst cv. Tifton-68; Cynodon ssp. cv. Tifton-85; Cynodon dactilon sp. cv. Jiggs; Hemárthria altissima cv. Roxinha, and Penissetum clandestinum cv. Quicuio. All cuts were made when each cv. reached 95% light interception. The cv. Jiggs and Tifton 68, with 21348 and 21016kg ha-1, respectively, were responsible for the higher yields (P<0.05) of accumulated dry biomass. The overall average of the cuts, the fourth cut had the lowest ratio upper stratum: lower compared to the others (P<0.05). Regarding the upper stratum, the cv. Tifton 85 had the highest CP content, statistically higher (P<0.05) in the first, second and fourth cut, while in the third cut the cv. Coast-cross had the highest protein content. In general, cv. Jiggs and Tifton 68 had the highest number of accumulated biomass. The cv. Tifton 85 was responsible for the higher levels of CP, whereas the levels of structural carbohydrates maintained a constant pattern, in both the upper and lower strata in four cut systems.(AU)

The isolation of single biological cells and their further cultivation in dedicated arrayed chambers are key to the collection of statistically reliable temporal data in cell-based biological experiments. In this work, we present a hydrodynamic single cell trapping and culturing platform that facilitates cell observation and experimentation using standard bio-lab equipment. The proposed design leverages the stochastic position of the cells as they flow into the structured microfluidic channels, where hundreds of single cells are then arrayed in nanoliter chambers for simultaneous cell specific data collection. Numerical simulation tools are used to devise and implement a hydrodynamic cell trapping mechanism that is minimally detrimental to the cell cycle and retains high overall trapping efficiency (∼70%) with the capability of reaching high fill factors (>90%) in short loading times (1-4 min) in a 400-trap device. A Monte Carlo model is developed using the design parameters to estimate the system trapping efficiencies, which show strong agreement with the experimentally acquired data. As proof of concept, arrayed mammalian tissue cells (MIA PaCa-2) are cultured in the microfluidic chambers for two days without viability problems.

BACKGROUND: Irritable bowel syndrome with constipation (IBS-C) significantly decreases quality of life and the ability to perform daily living activities. AIM: To demonstrate the long-term safety, tolerability and patient outcomes of lubiprostone in patients with IBS-C. METHODS: This extension study enrolled 522 IBS-C patients who had completed one of two randomised phase 3 studies. All enrolled patients received open-label lubiprostone orally for 36-weeks (8 mcg, twice daily). The primary objective was the assessment of long-term safety and tolerability, monitored via adverse events (AEs), laboratory parameters and vital signs. Additional outcome endpoints included monthly responder rates and patient evaluations of IBS-C symptom severity and impact on quality of life. RESULTS: The evaluable safety population comprised of 520 patients; 476 of which had patient reported outcome data available. The overall safety profile of lubiprostone during this study was similar to that observed in the preceding phase 3 studies. The most common AEs were diarrhoea (11.0%), nausea (11.0%), urinary tract infection (9.0%), sinusitis (9.0%) and abdominal distention (5.8%). Diarrhoea and nausea were the most common treatment-related AEs. No serious AEs were considered treatment-related. Seventeen patients discontinued due to a treatment-related AE, of which diarrhoea and nausea accounted for six (1.2%) and three (0.6%) respectively. For responder rates and patient-evaluated parameters (n = 476), all groups experienced significant improvements from baseline, with initial improvements maintained throughout the study. CONCLUSION: In patients with irritable bowel syndrome with constipation, lubiprostone 8 mcg twice daily was found to be safe and well tolerated over 9-13 months of treatment.

BACKGROUND: Lubiprostone is a prostone analog with a novel mechanism of action involving type-2 chloride channel activation. The aim of this work was to perform a dose-finding study for lubiprostone for the treatment of constipation with or without irritable bowel syndrome (IBS) in Japan. METHODS: A total of 170 patients (128 without IBS and 42 with IBS) with chronic idiopathic constipation (CIC) randomly received a placebo (n=42) or 16µg (n=41), 32µg (n=43), or 48µg (n=44) of lubiprostone daily for 2weeks. KEY RESULTS: There was a statistically significant and dose-dependent increase in change from baseline in the weekly average number of spontaneous bowel movements at week 1 (placebo: 1.5±0.4; 16µg: 2.3±0.4, 32µg: 3.5±0.5; and 48µg: 6.8±1.1, per week, mean±SE; P<0.0001). These primary endpoint results were significant on stratified analysis when patients were limited to those without IBS (P<0.0001). The primary endpoint in patients with IBS treated with 48µg of lubiprostone was significantly better than those given placebo (P=0.0086). Dose dependency was also seen for the secondary efficacy endpoints. Lubiprostone produced no serious side effects. CONCLUSIONS & INFERENCES: Our results suggest that lubiprostone produced a steady and effective improvement in the symptoms of CIC with or without IBS in a dose-dependent manner with a good safety profile and tolerability in a Japanese population.

BACKGROUND: Effective treatments for irritable bowel syndrome with constipation (IBS-C) are lacking. AIM: To assess the efficacy and safety of lubiprostone in IBS-C. METHODS: A combined analysis was performed among 1171 patients with a Rome II diagnosis of IBS-C in two phase-3 randomized trials of lubiprostone 8 mcg vs. placebo twice daily for 12 weeks. Using a balanced seven-point Likert scale ranging from significantly relieved (+3), to significantly worse (-3), patients responded on their electronic diary to the question: 'How would you rate your relief of IBS symptoms over the past week compared to how you felt before you entered the study?'. The primary efficacy endpoint was the percentage of overall responders. RESULTS: Using an intent-to-treat analysis with last observation carried forward, a significantly higher percentage of lubiprostone-treated patients were considered overall responders compared with those treated with placebo (17.9% vs. 10.1%, P=0.001). Patients treated with lubiprostone reported a similar incidence of adverse events to those treated with placebo. CONCLUSIONS: The percentage of overall responders based on patient-rated assessments of IBS-C symptoms was significantly improved in patients treated with lubiprostone 8 mcg twice daily compared to those treated with placebo. Lubiprostone was well tolerated with a favourable safety profile.

Proteins are expected to exhibit collective vibrational modes at terahertz frequencies. We have developed a promising approach to measure these motions by using a membrane device to hold samples. Samples of bovine serum albumin (BSA) in native and thermally denatured conformations were measured using terahertz time-domain spectroscopy. Clear differences were observed in transmittance and phase between native-conformation BSA samples and thermally denatured BSA samples. Time-domain data shows that samples exhibited relative time shifts when compared with a standard. Results suggest that there were differences in dielectric responses in the BSA samples, and these are probably associated with molecular conformational changes in the membrane device.

BACKGROUND: Analyses of a trial in constipated patients indicated that lubiprostone may be an effective treatment for irritable bowel syndrome with constipation. AIM: To assess the efficacy and safety of three lubiprostone doses for irritable bowel syndrome with constipation. METHODS: 195 irritable bowel syndrome with constipation patients received daily doses of 16 [8 microg twice daily (b.d.)], 32 (16 microg b.d.) or 48 microg (24 microg b.d.) lubiprostone or placebo b.d. for 3 months. Gastrointestinal parameters were recorded in diaries daily by patients. RESULTS: After 1 month, lubiprostone showed significantly greater improvements in mean abdominal discomfort/pain scores vs. placebo (P = 0.023). After 2 months, all lubiprostone groups showed significantly greater improvements in mean abdominal discomfort/pain scores (P < or = 0.039). After 3 months of treatment, the improvement in each lubiprostone arm was greater than placebo, but the test for trend was no longer significant. Treatment with lubiprostone showed significantly higher rates of gastrointestinal adverse events (P = 0.020), especially diarrhoea and nausea. CONCLUSION: Lubiprostone significantly improved gastrointestinal symptoms of irritable bowel syndrome with constipation at all doses. Higher doses of lubiprostone, especially the 48 microg/day group, were associated with more gastrointestinal adverse events. From these data, the 16 microg/day dose demonstrated the optimal combination of efficacy and safety. These results warrant further study of lubiprostone for treatment of irritable bowel syndrome with constipation patients.

BACKGROUND: Lubiprostone, a locally acting type-2 chloride channel activator, induces intestinal fluid secretion. AIM: To assess efficacy and safety of oral lubiprostone at multiple doses for the treatment of chronic constipation. METHODS: A total of 129 patients with chronic constipation were randomized to receive lubiprostone (24, 48 or 72 mcg/day) or placebo for 3 weeks. Spontaneous bowel movement (SBM) frequency, rescue medication use, symptom assessments and adverse events (AEs) were tracked. RESULTS: Over the double-blinded period, mean SBM frequencies were higher for lubiprostone groups (5.1-6.1) vs. placebo (3.8) and the overall difference was statistically significant (P = 0.046). SBM frequencies at week 1 were significantly higher in patients taking lubiprostone 48 or 72 mcg/day (P < or = 0.003) and, at week 2, all three lubiprostone doses yielded significantly higher SBM rates vs. placebo (P < or = 0.020). Significantly larger proportions of patients taking lubiprostone 48 and 72 mcg/day also experienced a SBM on the first treatment day (P < or = 0.009). The most common AEs were nausea, headache and diarrhoea. CONCLUSIONS: Lubiprostone improved SBM rates in a dose-dependent manner. AEs were tolerable for most patients. Increased AE severity at 72 mcg/day did not provide a clear risk-to-benefit advantage compared with lubiprostone 48 mcg/day, the dose chosen for subsequent Phase 3 studies.

AIM: To determine the efficacy of using albumin tear supplements in the treatment of ocular surface disorders as a substitute for serum eye drops. METHODS: The effects of albumin on the viability of serum deprived conjunctival cell were observed in vitro. The ability for albumin to compensate for serum was demonstrated by measuring the activity of the apoptosis related enzyme, caspase-3. In an animal study, corneal erosions were inflicted in 40 Japanese white rabbits. Rabbits were treated with 5% or 10% solutions of human albumin, and the decrease in epithelial defect size was compared with saline control and 0.3% sodium hyaluronate. A clinical case series trial of 5% albumin drops was conducted in nine patients with Sjögren's syndrome with severe dry eye. RESULTS: The addition of albumin to serum deprived conjunctival cells inhibited caspase activity and increased cell viability, showing that albumin can compensate for some of the physiological properties of serum. Corneal erosions in rabbits healed significantly faster (p<0.05) in eyes treated with 10% albumin compared with control and sodium hyaluronate. Patients with Sjögren's syndrome used albumin drops showed statistically significant improvement in fluorescein and rose bengal scores, but not in tear break up time and subjective symptoms. No adverse effects of albumin were observed during the study. CONCLUSIONS: The use of albumin as a protein supplement in artificial tear solutions is a viable approach in the treatment of ocular surface disorders associated with tear deficiency.

RESUMEN

Pauwels' femoral valgus osteotomy was performed on two osteoarthritic hips that developed as a sequela of infectious coxitis in the childhood. One of the hips was diagnosed as osteomyelitis of the ipsilateral femur before the operation and was treated with curettage and implantation of antibiotic-impregnated cement beads. The results of both hips were excellent and maintained for 9-11 years.

A 54-year-old woman visited our hospital because of gingival bleeding on May 31, 1998. After hematological and bone marrow examinations, she was diagnosed as having acute promyelocytic leukemia (APL) and given all-trans retinoic acid (ATRA) therapy starting on June 1. Anti-cancer drugs were administered for 5 days from June 12 because of an increase in the number of APL cells. The patient developed fever on June 20, and nodular erythematous eruptions appeared on June 23. Sweet's syndrome was diagnosed from biopsy samples of the eruption. ATRA was therefore discontinued, and prednisolone was started on June 29. The fever and skin eruptions improved rapidly, and complete remission was obtained on July 13. Sweet's syndrome due to ATRA may be a partial form of retinoic acid syndrome, in which the differentiated leukemic neutrophils increase and invade various organs. However, Sweet's syndrome must be considered regardless of the WBC count because in this case the syndrome occurred even when the WBC count was not high.

Yeast mitochondria were found to affect the zeta-potential of protoplasts, resulting in electrofusion of membrane behavior. For modeling purposes, two fusion systems were investigated: (1) parent yeasts of Saccharomyces cerevisiae G706 (rho(+)) x O708-11-16A (rho(+)), with zeta-potentials of -10 to -27 mV and -28 to -45 mV, depending on MgCl(2) concentration in the medium, respectively; and (2) parent yeasts of S. cerevisiae G706-1 (rho(-)) x O708-11-16A (rho(+)), with zeta-potentials of -30 to -60 mV, depending on MgCl(2). Yields of the hybrids in system (2) were over 100-fold higher than those in system (1). Thus, regulation of yeast electrofusion was found to be possible by mitochondrial mutations.

Between November 1983 and December 1992, 136 hips (119 patients) with coxarthritis were operated on using joint-preserving techniques based on the rationale of Pauwels' osteotomy. The criterion for selection was a patient in whom the height of the joint space in the weight-bearing area of the hip was less than 1 mm. The mean age at operation was 48 years and the mean follow-up 109 months (60 to 171). Hips were categorised using Bombelli's classification of osteoarthritis, into atrophic and non-atrophic types. The endpoint was defined as that at which the height of the joint space became less than 1 mm again. The Kaplan-Meier curve showed that the rate of survival of the non-atrophic group was significantly better than that of the atrophic group. Cox's proportional hazard model indicated that the factors influencing the results of joint-preserving operations included Bombelli's classification, postoperative incongruence of the joint and the height of the joint space.

We report the results of cementless, anatomic, medullary locking hip prosthesis application in our first consecutive series. We used the so-called Asian size of prosthesis with proportionally smaller stem sizes in both diameter and length. Forty-seven stems and sockets were analyzed with a mean follow-up of 69 months. The mean Merle d'Aubigné hip scores were 8 points preoperatively and 16 points at the final follow-up. Radiologically, the stems showed excellent stability without loosening. Stress shielding around the stems did occur in most cases but did not progress. Preoperative bone quality influenced the extent of stress shielding evaluated at the final follow-up: higher stress shielding was noted in poorer quality bones at the time of operation. There were problems with the sockets. The shallow socket and impingement at the protruded rim seemed to cause a high incidence of dislocation (13%). Massive polyethylene wear occurred in 5 sockets. These sockets were 48 and 46 mm in diameter with 26 mm heads. In conclusion, the stems of the anatomic medullary locking hip prostheses used in Japan showed satisfactory stability even in poor quality bones, but there were problems with the polyethylene liners. Our solution was to use larger sockets with 22 mm heads.

Patients with osteoarthritis of the hip were treated with a conservative therapy of heel lifting. Orthoses were applied on 35 hips in 33 subjects and the cases were followed for 23 months on average. Dramatic pain relief was reported, but the time required to reduce or completely relieve pain increased according to the stage of osteoarthritis. The radiological results were not satisfactory. During the follow-up, only two hips showed improvement, 22 showed no change, and 11 deteriorated. The mechanism of heel lifting in relation to the hip joint was analysed, showing that pelvic obliquity was achieved and the trunk stabilized. In conclusion this simple orthosis was effective as a palliative therapy for osteoarthritis of the hip.

Human skin has various distributions and arrangements of elastic fiber (EF). Previous reports did not clearly show the distribution of EF in the face skin because of various contents during aging. In this study, a color image analyzer indicated distribution of elastic, oxytalan, and muscle fibers in human face skin. During aging the muscle fiber size and the content of the EF decreased in the modiolus and inferior labial regions of the human skin, and the ratio of the EF was lower than that of oxytalan fiber measured areas. That is, the dimension of oxytalan fiber may reflect the content of EF, and muscle has a role in the distribution of the EF in human face skin. In the deeper regions, small and large EF bundles were found near the sheath of gland and muscles. Therefore, face movement might be an important aspect to maintaining the EF content of human face skin.
