RESUMEN

AIM: To develop and evaluate a new fundus image optimization software based on red, green, blue channels (RGB) for the evaluation of age-related macular degeneration (AMD) in the Chinese population. METHODS: Fundus images that were diagnosed as AMD from the Shanghai Changfeng Study database were analyzed to develop a standardized optimization procedure. Image brightness, contrast, and color balance were measured. Differences between central lesion area and normal retinal area under different image brightness, contrast, and color balance were observed. The optimal optimization parameters were determined based on the visual system to avoid image distortion. A paired-sample diagnostic test was used to evaluate the enhancement software. Fundus optical coherence tomography (OCT) was used as the gold standard. Diagnostic performances were compared between original images and optimized images using McNemar's test. RESULTS: A fundus image optimization procedure was developed using 86 fundus images of 74 subjects diagnosed with AMD. By observing gray-scale images, choroid can be best displayed in red channel and retina in green channel was found. There was limited information in blue channel. Totally 104 participants were included in the paired sample diagnostic test to assess the performance of the optimization software. After the image enhancement, sensitivity increased from 74% to 88% (P=0.008), specificity decreased slightly from 88% to 84% (P=0.500), and Youden index increased by 0.11. CONCLUSION: The standardized image optimization software increases diagnostic sensitivity and may help ophthalmologists in AMD diagnosis and screening.

Wetland is rich in biodiversity and cultural diversity, possessing higher tourism value and environmental education and community participation functions. Wetland ecotourism reflects the sustainable development of tourism economy and wetland protection, having received great concern from governments and scholars at home and abroad. This paper summarized the related theories and practices, discussed the research advances in wetland ecotourism from the aspects of significance, progress, contents, methods and results, and pointed out the important research fields in the future, aimed to accelerate the development of wetland ecotourism research and to provide reference about the resources exploitation, environment protection, and scientific administration of wetland and related scenic areas.

Pot-culture experiments were used to examine the individual and combined effects of Cu and Cd pollutants on Trifolium repens L. seedlings, both on their growth and their active oxygen metabolism system, mainly superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) activities. The results showed that the negative action took place at low concentrations of Cu (less than 500 ppm) and Cd (less than 0.5 ppm), which had no obvious effects on the seedlings' growth. However, as the concentrations of Cu and Cd increased (500-3000 ppm and 0.5-50 ppm respectively), synergistic activities was observed, showing obvious negative effects (P less than 0.05). Compared with the control samples, the seedlings affected by Cu and Cd pollutants were shorter and smaller, their fresh/dry weight and content of soluble protein decreased drastically, their leaf electric conductivity increased, and the contents of their leaf pigments decreased. Chlorophyll a was more sensitive than chlorophyll b to Cu and Cd pollutants, and chlorophyll b was more sensitive than carotenoid. It was also shown that the active oxygen metabolism of T. repens seedlings was destroyed by high amounts of Cu and Cd, the balance of the anti-oxidase system was broken, and the CAT and SOD activities noticeably decreased while POD activity evidently increased. Cd had a more noticeable effect on seedling growth than Cu.

AIM: To identify specifically expressed genes in the adult and fetal testes. METHODS: A human testis cDNA microarray was established. Then the mRNA of adult and fetal testis was purified and probes were prepared by a reverse transcription reaction with the testis mRNA as template. The microarray was hybridized with probes of adult and fetal testes. The nucleic sequences of differentially expressed genes were determined and homologies were searched in the databases of the GenBank. RESULTS: When hybridized with adult or fetal testis probes, the positive clones were 96.8 % and 95.4 %, respectively. Among these genes, one was a new testis-specific gene, which was named TSP1. TSP1 was highly expressed in human adult testis. The cDNA of TSP1 was 1,484 bp in length. The cDNA sequence of this clone was deposited in the Genbank (AF333098). TSP1 was also determined as Interim Gen Symbol (Unigene, No. Hs.98266). Protein analysis showed that TSP1 contained two functional domains: an N-terminal basic helix-loop-helix (bHLH) and a C-terminal leucine zipper (Zip). Homologous analysis showed that the 430 amino acid sequences deduced from the 1293 bp open reading frame (ORF) had a homology with the human gene FLJ2509 (AK098575). TSP1 had also a sequence homology with Spz 1 protein of mouse. Expression profiles showed that TSP1 was specifically and strongly expressed in the testis. CONCLUSION: TSP1 is a gene highly expressed in adult testis. It may play an important role in spermatogenesis in the humans.
