RESUMEN

The diagnosis of solid pseudopapillary neoplasm of the pancreas (SPN) can be challenging due to potential confusion with other pancreatic neoplasms, particularly pancreatic neuroendocrine tumors (NETs), using current pathological diagnostic markers. We conducted a comprehensive analysis of bulk RNA sequencing data from SPNs, NETs, and normal pancreas, followed by experimental validation. This analysis revealed an increased accumulation of peroxisomes in SPNs. Moreover, we observed significant upregulation of the peroxisome marker ABCD1 in both primary and metastatic SPN samples compared with normal pancreas and NETs. To further investigate the potential utility of ABCD1 as a diagnostic marker for SPN via immunohistochemistry staining, we conducted verification in a large-scale patient cohort with pancreatic tumors, including 127 SPN (111 primary, 16 metastatic samples), 108 NET (98 nonfunctional pancreatic neuroendocrine tumor, NF-NET, and 10 functional pancreatic neuroendocrine tumor, F-NET), 9 acinar cell carcinoma (ACC), 3 pancreatoblastoma (PB), 54 pancreatic ductal adenocarcinoma (PDAC), 20 pancreatic serous cystadenoma (SCA), 19 pancreatic mucinous cystadenoma (MCA), 12 pancreatic ductal intraepithelial neoplasia (PanIN) and 5 intraductal papillary mucinous neoplasm (IPMN) samples. Our results indicate that ABCD1 holds promise as an easily applicable diagnostic marker with exceptional efficacy (AUC=0.999, sensitivity=99.10%, specificity=100%) for differentiating SPN from NET and other pancreatic neoplasms through immunohistochemical staining.

The Carbon Fiber Reinforced Polymer (CFRP)-wrapped steel pipeline system is emerging as an alternative for the repair and long-distance transportation of oil and gas. This system is recognized as a composite material. Under internal pressure, the pipe predominantly undergoes circumferential stretching. This study derives the tensile constitutive model of the steel-CFRP composite material using uniaxial tensile tests and the rule of mixtures. Subsequently, utilizing this newly calibrated constitutive model, a computational model to assess the internal pressure capacity of the steel-carbon fiber composite pipe was established. Finally, a comparison between the theoretical outcomes of the computational model and the actual internal pressure test results revealed a high degree of correlation. This holds substantial significance for the design and practical implementation of this novel composite pipeline system.

RESUMEN

Neurosyphilis (NS) is a central nervous system (CNS) infection caused by Treponema pallidum (T. pallidum). NS can occur at any stage of syphilis and manifests as a broad spectrum of clinical symptoms. Often referred to as "the great imitator," NS can be easily overlooked or misdiagnosed due to the absence of standard diagnostic tests, potentially leading to severe and irreversible organ dysfunction. In this study, proteomic and machine learning model techniques are used to characterize 223 cerebrospinal fluid (CSF) samples to identify diagnostic markers of NS and provide insights into the underlying mechanisms of the associated inflammatory responses. Three biomarkers (SEMA7A, SERPINA3, and ITIH4) are validated as contributors to NS diagnosis through multicenter verification of an additional 115 CSF samples. We anticipate that the identified biomarkers will become effective tools for assisting in diagnosis of NS. Our insights into NS pathogenesis in brain tissue may inform therapeutic strategies and drug discoveries for NS patients.

Prostate cancer detected by autopsy is named latent prostate cancer. As the repertoire of clinical prostate cancer, latent cancer may better reflect the disease burden. Unlike clinical prostate specimens, which are obtained exclusively from biopsy-positive cases, prostate specimens obtained through autopsy provide information on biopsy-negative cases, helping calculate the true sensitivity of prostate biopsy. From 2014 to 2021, we collected autopsy specimens of the prostate from body donors in China and performed transperineal and transrectal biopsies on specimens before step-sectioning and pathological measurements. We found that the crude prevalence of latent prostate cancer in middle-aged and elderly men was 35.1% (81/231), which was higher than previous estimates for Chinese populations. The overall per-patient sensitivities of transperineal and transrectal biopsies were not significantly different (33.3% vs. 32.1%, p = 0.82), but the two approaches differed in preferential sampling area along the proximal-distal axis of the prostate. Transperineal biopsy had a higher sensitivity for detecting clinically significant lesions in the distal third (34.7% vs. 16.3%, p = 0.02) and distal half (30.6% vs. 18.1%, p = 0.04), while transrectal biopsy had a higher sensitivity for lesions in the proximal half (25.0% vs. 13.9%, p = 0.046). Both transperineal and transrectal methods of biopsy missed most small lesions (<0.1 mL) and 35.3% (6/17) of large lesions (>0.5 mL). In conclusion, the prevalence of latent prostate cancer in China has increased over the past 2 decades. Systematic transperineal and transrectal methods of biopsy had comparable sensitivities but had different preferential sampling areas. Both approaches miss one-third of large lesions.

To tackle microbial contamination and spoilage during pork storage, transportation, and sales, a novel packaging film with exogenously driven controlled release behavior was designed through the demand-directed preparation of the emulsion of shellac-encapsulated cinnamaldehyde nano-capsules (SNCs) and the ingenious integration of chitosan (CS) film matrix. Among them, the SNCs were synthesized using the solvent exchange method, allowing controlled release of cinnamaldehyde (CA) in response to volatile alkaline substances present in the meat. The electrostatic interaction between SNCs and CS molecules improved the thermal stability, water resistance, tensile strength, and viscosity of the film-forming solution, while the CA in SNCs enhanced UV resistance and antimicrobial properties of the film. Notably, the CS film with SNC-100 loaded (CSNCs-100 film) achieved 99% inhibition against both E. coli and S. aureus, regulated environmental moisture, and prolonged the freshness duration of pork by an impressive six days.

BACKGROUND: Therapeutic approaches for extrahepatic cholangiocarcinoma (EHCC) are limited, due to insufficient understanding to biomarkers related to prognosis and drug response. Here, we comprehensively assess the molecular characterization of EHCC with clinical implications. METHODS: Whole-exome sequencing (WES) on 37 tissue samples of EHCC were performed to evaluate genomic alterations, tumor mutational burden (TMB) and microsatellite instability (MSI). RESULTS: Mutation of KRAS (16%) was significantly correlated to poor OS. ERBB2 mutation was associated with improved OS. ERBB2, KRAS, and ARID1A were three potentially actionable targets. TMB ≥10 mutations per megabase was detected in 13 (35.1%) cases. Six patients (16.2%) with MSIsensor scores ≥10 were found. In multivariate Cox analysis, patients with MSIsensor sore exceed a certain threshold (MSIsensor score ≥0.36, value approximately above the 20th percentile as thresholds) showed a significant association with the improved OS (HR = 0.16; 95% CI: 0.056-0.46, p < 0.001), as well as patients with both TMB ≥3.47 mutations per megabase (value approximately above the 20th percentile) and MSIsensor score ≥0.36. CONCLUSIONS: TMB and MSI are potential biomarkers associated with better prognosis for EHCC patients. Furthermore, our study highlights important genetic alteration and potential therapeutic targets in EHCC.

Pancreatic cancer is a highly lethal form of malignancy that continues to pose a significant and unresolved health challenge. Doublecortin-like kinase 1 (DCLK1), a serine/threonine kinase, is found to be overexpressed in pancreatic cancer and holds promise as a potential therapeutic target for this disease. However, few potent inhibitors have been reported currently. Herein, a series of novel purine, pyrrolo [2,3-d]pyrimidine, and pyrazolo [3,4-d] pyrimidine derivatives were designed, synthesized, and evaluated their biological activities in vitro. Among them, compound I-5 stood out as the most potent compound with strong inhibitory activity against DCLK1 (IC50 = 171.3 nM) and remarkable antiproliferative effects on SW1990 cell lines (IC50 = 0.6 µM). Notably, I-5 exhibited higher in vivo antitumor potency (Tumor growth inhibition value (TGI): 68.6 %) than DCLK1-IN-1 (TGI: 24.82 %) in the SW1990 xenograft model. The preliminary mechanism study demonstrated that I-5 not only inhibited SW1990 cell invasion and migration, but also decreased the expression of prominin-1 (CD133) and cluster of differentiation 44 (CD44), which are considered as differentiation markers for SW1990 stem cells. All the results indicated that I-5, a novel DCLK1 inhibitor, shows promise for further investigation in the treatment of pancreatic cancer.

Niobate Li+ -storage anode materials with shear ReO3 crystal structures have attracted intensive attention due to their inherent safety and large capacities. However, they generally suffer from limited rate performance, cyclic stability, and temperature adaptability, which are rooted in their insufficient interlayer spacings. Here, sodium niobate (NaNb13 O33 ) micron-sized particles are developed as a new anode material owning the largest interlayer spacing among the known shear ReO3 -type niobates. The large interlayer spacing of NaNb13 O33 enables very fast Li+ diffusivity, remarkably contributing to its superior rate performance with a 2500 to 125 mA g-1 capacity percentage of 63.2%. Moreover, its large interlayer spacing increases the volume-accommodation capability during lithiation, allowing small unit-cell-volume variations (maximum 6.02%), which leads to its outstanding cyclic stability with 87.9% capacity retention after as long as 5000 cycles at 2500 mA g-1 . Its cyclic stability is the best in the research field of niobate micron-sized particles, and comparable to that of "zero-strain" Li4 Ti5 O12 . At a low temperature of -10 °C, it also exhibits high rate performance with a 1250 to 125 mA g-1 capacity percentage of 65.6%, and even better cyclic stability with 105.4% capacity retention after 5000 cycles at 1250 mA g-1 . These comprehensively good electrochemical results pave the way for the practical application of NaNb13 O33 in high-performance Li+ storage.

RATIONALE: Myositis ossificans circumscripta (MOC) is a rare disorder that causes heterotopic bone formation in soft tissues. It usually occurs after trauma and affects large muscles of the extremities. MOC of the pectineus muscle is extremely rare and has not been reported to be treated surgically. PATIENT CONCERNS: A 52-year-old woman presented with left hip pain and dysfunction 4 months after a traffic accident that caused pelvic and humeral fractures and cerebral hemorrhage. DIAGNOSES: Radiological imaging revealed isolated ossification of the left pectineus muscle. The patient was diagnosed with MOC. INTERVENTIONS: The patient underwent surgical resection of the ossified pectineus muscle followed by local radiation and medical therapy. OUTCOMES: At 12 months postoperatively, she was asymptomatic and had normal hip function. No recurrence was observed on radiography. LESSONS: MOC of the pectineus muscle is a rare condition that can cause severe hip dysfunction. Surgical resection combined with radiation and anti-inflammatory drugs can be an effective treatment option for patients who do not respond to conservative management.

Niobates are very promising anode materials for Li+-storage rooted in their good safety and high capacities. However, the exploration of niobate anode materials is still insufficient. In this work, we explore ~1 wt% carbon-coated CuNb13O33 microparticles (C-CuNb13O33) with a stable shear ReO3 structure as a new anode material to store Li+. C-CuNb13O33 delivers a safe operation potential (~1.54 V), high reversible capacity of 244 mAh g-1, and high initial-cycle Coulombic efficiency of 90.4% at 0.1C. Its fast Li+ transport is systematically confirmed through galvanostatic intermittent titration technique and cyclic voltammetry, which reveal an ultra-high average Li+ diffusion coefficient (~5 × 10-11 cm2 s-1), significantly contributing to its excellent rate capability with capacity retention of 69.4%/59.9% at 10C/20C relative to 0.5C. An in-situ XRD test is performed to analyze crystal-structural evolutions of C-CuNb13O33 during lithiation/delithiation, demonstrating its intercalation-type Li+-storage mechanism with small unit-cell-volume variations, which results in its capacity retention of 86.2%/92.3% at 10C/20C after 3000 cycles. These comprehensively good electrochemical properties indicate that C-CuNb13O33 is a practical anode material for high-performance energy-storage applications.

Ectopic secretion of parathyroid hormone (PTH) is a rare cause of hypercalcemia in malignancy patients. A 56-year-old woman with life-threatening hypercalcemia was caused by poorly-differentiated endometrial carcinoma secreting PTH with concomitant nodular goiter mimic parathyroid tumors. The elevated level of PTH and calcium decreased immediately after cytoreductive surgery (CRS). The pathology confirmed mismatch repair (MMR)-deficient endometrial carcinoma with PTH expression. The patient received four-course chemotherapy and one-course immunotherapy after CRS. The disease progression led to multiple organ failure and death about five months after CRS. To our knowledge, this is the first case of hypercalcemia caused by MMR-deficient endometrial carcinoma with ectopic PTH secreting and the first report of malignancy associated hypercalcemia complicated with nodular goiter.

Objective: The study aimed to explore the correlation of serum chemokine (C-C motif) ligand 21 (CCL21) and heat shock protein 90 (Hsp90) with preeclampsia (PE). Methods: Between June 2021 and June 2022, 50 pregnant women with PE were included in the PE group, and 50 healthy pregnant women were included in the control group. The serum levels of CCL21 and Hsp90 were compared between the two groups. Results: PE patients showed significantly higher levels of CCL21 and Hsp90 than healthy pregnant women (P < 0.05). Correlation analysis showed a positive correlation between CCL21 and Hsp90 levels (r > 0, (P < 0.05)). Binary logistic regression analysis suggested that high expression of CCL21 and Hsp90 were influencing factors for PE (OR >1, (P < 0.05)). The area under the receiver operating characteristic (AUC) curves of Hsp90 and CCL21 levels for predicting PE were 0.895 and 0.864, respectively, suggesting a good predictive value. Conclusion: Serum CCL21 and Hsp90 show great potential as disease markers for PE prediction. Further trials are, however, required prior to clinical promotion.

Objective: The purpose of the study was to determine the significance of heat shock protein 90 (HSP 90) and serum chemokine ligand 21 (CCL-21) in pregnant women with preeclampsia (PE). Methods: From June 2021 to June 2022, the study enrolled 100 women undergoing obstetric examinations and delivering in our hospital; 50 PE patients undergoing routine obstetric examinations and delivering during the same period were enrolled in the research group; according to the severity, they were divided into mild PE and severe PE groups, while 50 healthy pregnant women undergoing obstetric examinations and delivering in our hospital during the same period were enrolled in the control group. In a subsequent analysis, serum levels of CCL-21 and HSP90 were compared between the two groups, and the correlation among CCL-21, HSP 90, and PE severity was analyzed. Results: An overall total of 50 patients with PE were enrolled in the study, which included 32 patients with mild PE and 18 patients with severe PE. Patients with severe PE had lower mean arterial pressure (MAP), HSP 90, and CCL21 index levels than those with mild PE; MAP, HSP 90, and CCL21 in the severe PE group were higher than those in the mild PE group, but the difference was not statistically significant; In the research group, MAP was weakly correlated with HSP90 concentration and CCL21 concentration, with correlation coefficients of 0.33 and 0.30, respectively, and the correlation analysis was significant. Conclusion: Patients with PE showed significantly increased serum concentrations of HSP90 and CCL-21, but a significant difference did not exist between mild and severe PE. In addition, there was a weak relationship between HSP90 and CCL-21 concentrations in PE patients and MAP, suggesting that HSP90 and CCL-21 play an instrumental role in the pathogenesis of PE, although more studies are needed to clarify the exact mechanisms.

In the present study, the PLC characteristic parameters and DSA mechanism of Al-(2.86~9.41) Mg alloy sheets were investigated during tensile testing at room temperature with a tensile rate of 1 × 10-3 s-1. On the basis of the solution Mg concentrations in the α-Al matrix, the initial vacancy concentration, the second-phase particle configuration and the recrystallized grain configuration are almost the same by quenching treatment. The results show that the type of room-temperature tensile stress-strain curves of quenched Al-(2.86~9.41) Mg alloy sheets varied according to the Mg content. The type of stress-strain curve of the Al-2.86 Mg alloy sheet was B + C, while the type of stress-strain curve of the Al-(4.23~9.41) Mg alloy sheets was C. When the quenched Al-(2.86~9.41) Mg alloy sheets were stretched at room temperature, the strain cycle of the rectangular waves corresponding to the high stress flow ΔεTmax and stress drop amplitude Δσ on the zigzag stress-strain curve of alloy sheets increased with increasing the Mg content. Moreover, the strain cycle of ΔεTmax and Δσ on the stress-strain curve of alloy sheets increased gradually with increasing tensile deformation. The yield stress of quenched Al-(2.86~9.41) Mg alloy sheets increased gradually with increasing the Mg content. Moreover, the critical strain corresponding to yield stress εσ and the critical strain corresponding to the occurrence of the PLC shearing band εc of alloy sheets both increased with increasing the Mg content. However, the difference in flow strain value Δεc-σ between εc and εσ of alloy sheets decreased gradually with increasing the Mg content.

Parathyroid carcinomas are difficult to distinguish from adenomas according to the current diagnostic criteria. The judgment of local infiltration is subjective and inconsistent. Existing studies have found that the CDC73 gene encoding parafibromin is related to the occurrence of parathyroid carcinomas. This study is aimed at investigating whether the immunohistochemistry of parafibromin is helpful in distinguishing malignant from benign parathyroid tumors. A total of 53 patients with parathyroid carcinoma from Peking Union Medical College Hospital were included. Metastasis was found in 17 of 53 patients. In addition, another 53 patients with parathyroid adenomas were included as controls. Appropriate sections were stained with an immunohistochemical autostainer. Three senior pathologists evaluated the sections and analyzed their clinicopathological features independently. The loss of parafibromin expression only occurred in malignant tumors, including all carcinomas with metastasis (17/17) and 14 of 36 carcinomas with only local infiltration. All staining results of adenomas (53/53) were positive. Considering invasion as the gold standard of malignancy, the sensitivity of parafibromin staining is 58%, and the specificity is 100%. If the gold standard is changed to metastasis, the sensitivity becomes 100%, and the specificity becomes 84%. By analyzing clinicopathological features with metastasis and parafibromin staining, it is found that local-infiltrative carcinomas with positive staining results have better biological behaviors than carcinomas that lack parafibromin expression. Parafibromin staining is highly recommended as an auxiliary method in the diagnosis of parathyroid carcinoma.

In the present study, the fatigue life and fatigue fracture characteristics of annealed 7005 aluminum alloy plates subjected to different pre-tensile deformations were investigated. The results obtained upon increasing the pre-tensile deformation of the alloy plate to 20% revealed that the second-phase particles did not show any obvious changes, and that the thickness of the thin strip grain slightly decreased. The dislocation distribution in the alloy matrix varied significantly among the grains or within each grain as the dislocation density gradually increased with increasing pre-tensile deformation. Moreover, the fatigue performance of the annealed 7005 aluminum alloy plate was significantly improved by the pre-tensile deformation, and the alloy plate subjected to 20% pre-tensile deformation exhibited an optimal fatigue life of ~1.06 × 106 cycles, which was 5.7 times and 5.3 times that of the undeformed and 3% pre-stretched alloy plates, respectively. Two fatigue life plateaus were observed in the pre-tensile deformation ranges of 3-5% and 8-12%, which corresponded to heterogeneous dislocation distribution among various grains and within each grain, respectively. Moreover, two large leaps in the plot of the fatigue-life-pre-tensile-deformation curve were observed, corresponding to the pre-tensile deformation ranges of 5-8% and 16-20%, respectively.

Inspired by the classic ion-exchange reaction, a single phase material of Mg0.66Al0.34(OH)2(Mo3S13)0.03(NO3)0.14(CO3)0.07·H2O (Mo3S13-LDH) was masterly constructed by intercalating Mo3S132- into the MgAl-LDH gallery. Prepared Mo3S13-LDH displays excellent binding affinity and high selectivity for Ag(I) and Hg(II) in a mixed solution, in which an apparent selectivity order of Hg(II) > Ag(I) ≫ Pb(II), Cu(II), Ni(II), Co(II), Cd(II), and Mn(II) is observed. Enormous capture capacities (qmAg = 446.4 mg/g, qmHg = 354.6 mg/g) and fast equilibration time (within 60 min) place Mo3S13-LDH in the upper ranks of materials for such removal. For oxoanions, As(III) (HAsO32-) and Cr(VI) (CrO42-) can be specifically trapped by Mo3S13-LDH with comparable loading ability (qmAs = 61.8 mg/g, qmCr = 90.6 mg/g) in the coexistence of multiple interfering anions. Notably, high Hg(II) and Cr(VI) concentrations are finally reduced below the safe limit of drinking water. The excellent capture capacity of Mo3S13-LDH benefits from the rational design by following two aspects: (i) the multiple sulfur ligands in Mo3S132- groups give place to various capture modes and different affinity orders for target ions, and (ii) large-sized Mo3S132- groups widen the interlayer spacing of LDH, thereby accelerating the mass transfer process. Furthermore, the satisfactory structural stability of Mo3S13-LDH is also reflected through the unchanged hexagonal prismatic shape after adsorption. All of these highlight the great potential of Mo3S13-LDH for the application in water remediation.

Purpose: Xanthomatous hypophysitis (XHP) is an extremely rare form of primary hypophysitis for which there is a lack of clinical experience. A comprehensive understanding of its clinical characteristics, diagnosis and treatment is needed. Methods: Here, we report a case study and conduct a systematic review of XHP. Thirty-six cases were included, and their clinical manifestations, endocrine assessment, imaging features, treatment and follow-up data were collected and analyzed. Results: The mean age at diagnosis was 39.1 years, and females were predominant (75.0%). The most common symptom was headache (68.6%), and 66.7% of female patients presented menstrual disorders. The most common pituitary dysfunction was growth hormone (GH) deficiency. More than half of patients exhibited central diabetes insipidus (CDI). The majority of patients had an imaging presentation of a cystic lesion with peripheral enhancement. Pituitary stalk thickening was observed in half of the patients. Total lesion resection was achieved in 57.1% of cases. The recurrence rate after partial resection and biopsy was significantly higher than that after total lesion resection (57.1% vs. 0.0%, P = 0.0147). The most common pituitary hormone abnormalities to resolve after surgery were hyperprolactinemia (100.0%) and GH deficiency (91.7%). The typical pathological feature was inflammatory infiltration of foamy histiocytes, which showed positivity for CD68. Conclusion: Diagnosis of XHP is difficult when relying on clinical symptoms and imaging features. Therefore, surgical histopathology is necessary. Based on the available evidence, total lesion resection is recommended for treatment. However, the long-term prognosis for this rare disease remains unclear.

BACKGROUND: Ectopic adrenocortical tissue is a lesion usually found incidentally during autopsy or inguinal surgery. Here, we demonstrate an extremely unusual case of intrahepatic adrenocortical adenoma which highly mimicks hepatocellular carcinoma (HCC) and brings challenges for clinicians and pathologists. The diagnostic pitfalls have been discussed in detail to provide clues for guiding differential diagnosis and future treatment. CASE PRESENTATION: A 44-year-old man was admitted into our hospital for evaluation of a hepatic mass identified during routine examination. Enhanced CT revealed its margin displayed apparent enhancement in arterial phase, but hypointensity in portal and delayed phase. HCC was suspected and partial hepatectomy was performed. Microscopically, cells were arranged in solid sheets. Most of the tumor cells were large, polygonal, had prominent nucleoli and were rich in eosinophilic cytoplasm. Pleomorphic nucleus was frequently found. Focally, smaller cells were found with small nuclei and granular cytoplasm. Immunohistochemically, tumor cells were negative for Arg-1, glypican-3 (GPC3), hepatocyte specific antigen (HSA), and positive for synaptophysin (Syn), α-inhibin, and Melan A. The Ki-67 index was 1 %. The final diagnosis was ectopic adrenocortical oncocytic adenoma and the patient was uneventful after the surgery. CONCLUSION: Intrahepatic adrenocortical adenoma in the liver can hardly be diagnosed through radiology and little experience in pathology has been reported. In the present case, massive oncocytic changes and huge pleomorphism add greatly to the difficulties of making correct diagnosis. This lesion should be carefully kept in mind and a combination of markers is suggested for differentiating from HCC.
