RESUMEN

Fushi-tarazu factor 1 (FTZ-F1) is a class of transcription factors belonging to the nuclear receptor superfamily and an important molting regulator in insects; however, its detailed function in the molting process of Locusta migratoria is still unclear. This study identified two FTZ-F1 transcripts (LmFTZ-F1-X1 and LmFTZ-F1-X2) in L. migratoria. The classical domains of FTZ-F1 were present in their protein sequences and distinguished based on their variable N-terminal domains. Reverse-transcription quantitative polymerase chain reaction analysis revealed that LmFTZ-F1-X1 and LmFTZ-F1-X2 were highly expressed in the integument. RNA interference (RNAi) was used to explore the function of LmFTZ-F1s in the molting of the third-instar nymph. Separate LmFTZ-F1-X1 or LmFTZ-F1-X2 silencing did not affect the normal development of third-instar nymphs; however, the simultaneous RNAi of LmFTZ-F1-X1 and LmFTZ-F1-X2 caused the nymphs to be trapped in the third instar stage and finally die. Furthermore, the hematoxylin-eosin and chitin staining of the cuticle showed that the new cuticles were thickened after silencing the LmFTZ-F1s compared to the controls. RNA-seq analysis showed that genes encoding four cuticle proteins, two chitin synthesis enzymes, and cytochrome P450 303a1 were differentially expressed between dsGFP- and dsLmFTZ-F1s-injected groups. Taken together, LmFTZ-F1-X1 and LmFTZ-F1-X2 are involved in the ecdysis of locusts, possibly by regulating the expression of genes involved in cuticle formation, chitin synthesis, and other key molting processes.

LiPF6-based carbonate electrolytes have been extensively employed in commercial Li-ion batteries, but they face numerous interfacial stability challenges while applicating in high-energy-density lithium-metal batteries (LMBs). Herein, this work proposes N-succinimidyl trifluoroacetate (NST) as a multifunctional electrolyte additive to address these challenges. NST additive could optimize Li+ solvation structure and eliminate HF/H2O in the electrolyte, and preferentially be decomposed on the Ni-rich cathode (LiNi0.8Co0.1Mn0.1O2, NCM811) to generate LiF/Li3N-rich cathode-electrolyte interphase (CEI) with high conductivity. The synergistic effect reduces the electrolyte decomposition and inhibits the transition metal (TM) dissolution. Meanwhile, NST promotes the creation of solid electrolyte interphase (SEI) rich in inorganics on the Li metal anode (LMA), which restrains the growth of Li dendrites, minimizes parasitic reactions, and fosters rapid Li+ transport. As a result, compared with the reference, the Li/LiNi0.8Co0.1Mn0.1O2 cell with 1.0 wt.% NST exhibits higher capacity retention after 200 cycles at 1C (86.4% vs. 64.8%) and better rate performance, even at 9C. In the presence of NST, the Li/Li symmetrical cell shows a super-stable cyclic performance beyond 500 h at 0.5 mA cm-2/0.5 mAh cm-2. These unique features of NST are a promising solution for addressing the interfacial deterioration issue of high-capacity Ni-rich cathodes paired with LMA.

Background: The endoscopic nasojejunal (NJ) placement plays a pivotal role in the nutritional support of critically ill patients. However, the conventional endoscopy-guided tube insertion method presents issues of excessive procedural duration. We have enhanced the traditional endoscopy-guided catheter placement method, enabling a faster and more convenient catheter insertion. Methods: We improved the traditional endoscopically guided technique by incorporating an extra silk thread knot at the 25 cm mark on the jejunal segment of the NJ tube to assist endoscopists in accurate tube placement. We conducted the improved NJ tube placement on critically ill patients in need of enteral nutrition (EN). Laboratory data were retrospectively collected before and after the 7-day period of NJ tube placement and EN treatment to evaluate the effectiveness and safety of the improved method. Results: A total of 88 critically ill patients, with an average age of 59.6±15.5 years, and a male ratio of 86.4%, who underwent the improved NJ tube placement method were enrolled into analysis finally, achieving a 100% success rate of NJ tube insertion. The average time for tube insertion was 5.9±2.2 min, with a mean insertion depth of 108.8±12.5 cm. The EN tolerance score was 0.79±0.98. Following 7 days of EN therapy, the patients showed significant improvement in serum albumin levels compared to baseline (36.42 vs. 33.66 g/L, P<0.001). Conclusions: The improved endoscopically guided NJ tube placement technique is a rapid and safe procedure with excellent patient tolerance. It significantly improves the nutritional status of critically ill patients and facilitates the administration of EN, which requires further validation through randomized controlled trials.

Cytochrome P450 monooxygenases (P450s) are a superfamily of multifunctional heme-containing proteins and could function as odorant-degrading enzymes (ODEs) in insect olfactory systems. In our previous study, we identified a P450 gene from the antennal transcriptome of Locusta migratoria, LmCYP6MU1, which could be induced by a variety of volatiles. However, the regulatory mechanisms of this gene in response to volatiles remain unknown. In current study, we investigated the tissues and development stages expression patterns of LmCYP6MU1 and determined its olfactory function in the recognition of the main host plant volatiles which induced LmCYP6MU1 expression. The results showed that LmCYP6MU1 was antenna-rich and highly expressed throughout the antennal developmental stages of locusts. LmCYP6MU1 played important roles in the recognition of trans-2-hexen-1-al and nonanal. Insect CncC regulates the expression of P450 genes. We tested whether LmCncC regulates LmCYP6MU1 expression. It was found that LmCncC knockdown in the antennae resulted in the downregulation of LmCYP6MU1 and repressed the volatiles-mediated induction of LmCYP6MU1. LmCncC knockdown reduced the electroantennogram (EAG) and behavioral responses of locusts to volatiles. These results suggested that LmCncC could regulate the basal and volatiles-mediated inducible expression of LmCYP6MU1 responsible for the recognition of trans-2-hexen-1-al and nonanal. These findings provide an original basis for understanding the regulation mechanisms of LmCncC on LmCYP6MU1 expression and help us better understand the LmCncC-mediated olfactory plasticity.

BACKGROUND: The cap 'n' collar (Cnc) belongs to the Basic Leucine Zipper (bZIP) transcription factor super family. Cap 'n' collar isoform C (CncC) is highly conserved in the animal kingdom. CncC contributes to the regulation of growth, development, and aging and takes part in the maintenance of homeostasis and the defense against endogenous and environmental stress. Insect CncC participates in the regulation of various kinds of stress-responsive genes and is involved in the development of insecticide resistance. RESULTS: In this study, one full-length CncC sequence of Locusta migratoria was identified and characterized. Upon RNAi silencing of LmCncC, insecticide bioassays showed that LmCncC played an essential role in deltamethrin and imidacloprid susceptibility. To fully investigate the downstream genes regulated by LmCncC and further identify the LmCncC-regulated genes involved in deltamethrin and imidacloprid susceptibility, a comparative transcriptome was constructed. Thirty-five up-regulated genes and 73 down-regulated genes were screened from dsLmCncC-knockdown individuals. We selected 22 LmCncC-regulated genes and verified their gene expression levels using RT-qPCR. Finally, six LmCYP450 genes belonging to the CYP6 family were selected as candidate detoxification genes, and LmCYP6FD1 and LmCYP6FE1 were further validated as detoxification genes of insecticides via RNAi, insecticide bioassays, and metabolite identification. CONCLUSIONS: Our data suggest that the locust CncC gene is associated with deltamethrin and imidacloprid susceptibility via the regulation of LmCYP6FD1 and LmCYP6FE1, respectively.

Commercial Li-ion batteries use LiPF6-based carbonate electrolytes extensively, but there are many challenges associated with them, like dendritic Li growth and electrolyte decomposition, while supporting the aggressive chemical and electrochemical reactivity of lithium metal batteries (LMBs). This work proposes 1,1,1,3,3,3-hexafluoroisopropyl methacrylate (HFM) as a multifunctional electrolyte additive, constructing protective solid-/cathode-electrolyte interphases (SEI/CEI) on the surfaces for both lithium metal anode (LMA) and Ni-rich cathode to solve these challenges simultaneously. The highly fluorinated group (-CF3) of the HFM molecule contributes to the construction of SEI/CEI films rich in LiF that offer excellent electronic insulation, high mechanical strength, and surface energy. Accordingly, the HFM-derived LiF-rich interphases can minimize the electrolyte-electrode parasitic reactions and promote uniform Li deposition. Also, the problems of LiNi0.8Co0.1Mn0.1O2 particles' inner microcrack evolution and the growth of dendritic Li are adequately addressed. Consequently, the HFM additive enables a Li/LiNi0.8Co0.1Mn0.1O2 cell with higher capacity retention after 200 cycles at 1 C than the cell with no additive (74.7 vs 52.8%), as well as a better rate performance, especially at 9 C. Furthermore, at 0.5/0.5 mAh cm-2, the Li/Li symmetrical battery displays supersteadfast cyclic performance beyond 500 h when HFM is present. For high-performance LMBs, the HFM additive offers a straightforward, cost-effective route.

The systematical understanding of metal-dependent activity in electrocatalyzing oxygen reduction reaction (ORR), a vital reaction with sluggish kinetics for zinc-air batteries, remains quite unclear. An atomic and spatial engineering modulating ORR activity over hollow carbon quasi-sphere (HCS) confined in a series of single M-N (M = Cu, Mn, Ni) sites is reported here. Based on the theoretical prediction and experimental validation, Cu-N4 site with the lowest overpotential shows a better ORR kinetics than Mn-N4 and Ni-N4 . The ORR activity of single-atom Cu center can be further improved by decreasing the coordination number of N to two, namely Cu-N2 , due to the enhancement of electrons with lower coordination structure. Benefitting from the unique spatial confinement effect of the HCS structure in modulating electronic feature of active sites, the Cu-N2 site confined in HCS also delivers highly improved ORR kinetics and activity relative to that on planner graphene. Additionally, the best catalyst holds excellent promise in the application of zinc-air batteries. The findings will pave a new way to atomically and electronically tune active sites with high efficiency for other single-atom catalysts.

Precocene I is a juvenile hormone antagonist that needs to be activated via oxidative biotransformation catalyzed by cytochrome P450 (CYP). NADPH-cytochrome P450 reductase (CPR) supplies CYP with electrons in the oxidation-reduction process; however, its functional role in the activation of precocene I remains unexplored. Here, the representative characteristics of CPRs were analyzed in the CPR gene of Locusta migratoria (LmCPR), the result of model docking indicated that the hydrogen bonds were formed between reduced nicotinamide adenine dinucleotide phosphate (NADPH), flavin adenine dinucleotide (FAD), flavin mononucleotide (FMN) and NADPH-, FAD-, FMN-domains of LmCPR, respectively. Treating the fourth-instar nymphs with precocene I decreased the juvenile hormone titers of nymphs to 0.55-fold of that in acetone-treated controls, and extended the interval time between fourth- and fifth-instar nymphs. 68.75% of the treated fourth-instar nymphs developed into precocious adults in the fifth-instar. LmCPR knockdown decreased the response to precocene I in the nymphs, the occurrence rate of precocious adults induced by precocene I treatment reduced by 23.11%. Therefore, LmCPR may be involved in the activation of precocene I in L. migratoria. In addition, we generated an active recombinant LmCPR protein using a prokaryotic expression system, its activity in reducing cytochrome c was 33.13 ± 11.50 nmol CytCred/min/µg protein. This study lays the foundation for further research on the role of LmCPR in precocene I activation.

The energy density of solid-state lithium batteries (SSLBs) has been primarily limited by the low ionic conductivity of solid electrolyte and poor interface compatibility between electrolyte and electrodes. Herein, a multifunctional composite solid polymer electrolyte (CSPE) based on polyethylene oxide (PEO) embedded with zeolitic imidazolate framework-8 deposited on carboxymethyl cellulose (ZIF@CMC) is reported. The ZIF@CMC interpenetrated in PEO matrix creates a continuous Li+ conductive network by combining Zn2+ in ZIF with the unsaturated group in PEO to boost the Li+ transport through the PEO chain segment. On the other hand, Zn2+ can bond with bis(trifluoromethane)sulfonimide (TFSI-) anion, thus promoting the dissolution of lithium salt and releasing more lithium ions. This CSPE demonstrates brilliant electrochemical properties, including a high ionic conductivity of 1.8 × 10-4 S cm-1 at room temperature and a wide electrochemical window of 5 V. The integrated LiFePO4/CSPE/Li batteries using 20 wt.% ZIF-8@CMC show excellent reversible capacity of 145.6 mAh g-1 with a capacity retention of 88.95 % after 200 cycles at a high current density of 0.5C. Our study proposed a novel and effective strategy to construct high-performance solid-state lithium batteries.

Cytochrome P450 monooxygenases (P450s) are a large superfamily of heme-thiolate proteins and play a vital role in the biosynthesis and inactivation of endogenous substances as well as the detoxification of exogenous substances. They also function as odor-degrading enzymes (ODEs) in insect olfactory sensory systems. In the present study, a P450 gene was obtained from the antennae of Locusta migratoria and named as CYP6FD5. Multiple alignment of P450 proteins revealed that LmCYP6FD5 contained five conserved motifs, including the helix C motif, an oxygen-binding site, helix K motif, a meander region, and the haem-binding motif. The expression of LmCYP6FD5 in various tissues and antennal development stages was determined by using RT-qPCR. Our results showed that LmCYP6FD5 was antenna-specific and highly expressed throughout the antennal developmental stages of female and male locusts. Furthermore, the role of LmCYP6FD5 in the perception of host plant volatiles was assessed using RNAi in combination with electroantennogram (EAG) and behavioral responses. Our findings showed that after silencing LmCYP6FD5, the EAG responses of female and male locusts to the main volatiles of gramineous plants, including trans-2-Hexen-1-al, cis-3-Hexenyl acetate, and decanal, were significantly diminished. Moreover, a significant decrease in EAG response of male antennae to benzaldehyde was also observed. In addition, behavioral assay showed that the locust response to single volatile from host plant or wheat remained unchanged after the silencing of LmCYP6FD5. Antenna-specific expression and EAG responses of locusts to host plant volatiles still suggested that LmCYP6FD5 was potentially involved in host plant recognition, although no behavioral changes were observed.

OBJECTIVE: To explore the influence of predictive nursing combined with emotional therapy on the rehabilitation effect and psychological state of patients with brain injury after the operation and to analyze its application value to patients with brain injury after the operation. Methods. 70 postoperative patients with brain injury who went to our hospital for diagnosis and treatment were selected as the key figures of this experiment, including 31 female patients and 39 male patients. In order to ensure the uniformity of data, medical staff need to divide patients into an experimental group and a control group through the double-blind method, with 35 cases in each group. The patients in the two groups need to take the relevant diagnosis and surgical treatment measures after admission. The patients in the control group take routine nursing, providing patients with a clean and tidy ward is convenient for patients to manage nursing, and for patients to bring comfortable treatment relief. The patients in the experimental group take predictive nursing combined with emotional therapy, which is to master the basic situation of patients, according to the quality of patients, and develop comprehensive nursing measures for patients. The psychological status, complication incidence rate, treatment effectiveness, and nursing satisfaction of the two groups were compared and analyzed. Results. The psychological status, complication incidence rate, nursing efficiency, and nursing satisfaction of patients in the experimental group were significantly better than those in the control group (P < 0.05). Conclusion. Predictive nursing combined with emotional therapy for patients with brain injury after the operation can effectively alleviate bad mood of patients, reduce the incidence rate of complications, improve the nursing satisfaction of patients, as well as provide patients with high-quality treatment and nursing services. It has a certain clinical application value and is worth popularizing.

To evaluate the correlation of neutrophil-lymphocyte ratio (NLR), platelet-lymphocyte ratio (PLR), and mean platelet volume (MPV) with parameters related to gout activity. The general data of the patients and healthy controls (HCs), including complete blood count, C-reactive protein (CRP), erythrocyte sedimentation rate (ESR), serum uric acid (SUA), and the presence of tophi were retrospectively analyzed. NLR, MPV, and PLR were calculated in patients with intercritical gout and gout flares. Correlation of the 3 markers with clinical features, like ESR, CRP, SUA, and the presence of tophi, were analyzed. The results revealed that NLR and PLR were elevated and MPV was markedly decreased in patients with gout compared with HCs (all P < .05). In patients with gout flares, NLR, and PLR were higher and MPV was lower than in intercritical gout patients (all P < .05). NLR and PLR were positively correlated with ESR and CRP, whereas MPV was negatively correlated with ESR. NLR, PLR, and MPV showed no obvious correlation with SUA and the presence of tophi. The receiver operating characteristic curve showed that NLR was more valuable in assessing gout disease activity. NLR, PLR, and MPV were correlated with inflammatory parameters in gout; they may be used as complementary tools to evaluate gout activity.

Objective: To analyze the prognostic impact of staged nursing interventions on the treatment of patients with chronic obstructive pulmonary diseases (COPD) combined with type II respiratory failure. Methods: 120 patients with COPD combined with type II respiratory failure admitted to our hospital between January 2021 and January 2022 were divided into a control group and a study group, with 60 patients in each group. The control group received conventional strategy interventions, and the study group received staged nursing interventions. Pulmonary function, blood gases, health impairment, knowledge, mood, hope level, and quality of survival were evaluated before and after patient care, and satisfaction and the impact on patient prognosis were assessed. Results: The improvement of pulmonary function and blood gas in the study group was better than that in the control group aftercare, and the difference was statistically significant (P < 0.05). Health impairment and mood scores were lower in the study group compared to the control group aftercare, and the difference was statistically significant (P < 0.05). Knowledge awareness, hope, and quality of survival scores were higher in the study group compared to the control group aftercare, with statistically significant differences (P < 0.05). The rate of excellent prognosis and satisfaction was higher in the study group compared with the control group, and the difference was statistically significant (P < 0.05). Conclusion: The implementation of staged nursing interventions during the treatment of patients with COPD combined with type II respiratory failure can significantly improve patient prognosis and has a high application value.

Rechargeable lithium metal batteries (LMBs) with high energy densities can be achieved by coupling a lithium metal anode (LMA) and a LiNi0.8Co0.1Mn0.1O2 (NCM811) cathode. Nevertheless, Li dendrite growth on the LMA surface and structural collapse of the NCM811 material, closely tied with the fragile cathode-/solid-electrolyte interphases (CEI/SEI) and corrosive hydrogen fluoride (HF), seriously deteriorate their performances. Herein, trimethylsilyl trifluoroacetate (TMSTFA) as a multifunctional electrolyte additive is proposed for regulation of the CEI/SEI films and elimination of HF. For one thing, the TMSTFA-derived CEI film rich in C-O species is conductive to Li+ transport and structural stability of NCM811 materials, and the TMSTFA-derived SEI film mainly consisting of inorganics (Li2CO3 and LiF) and organics (C-O and O-C═O species) can significantly promote Li+ homogeneous deposition and impede the Li dendrite growth. For another thing, the undesired reactions of the solvents and LiPF6 salt are effectively retarded by the TMSTFA additive. Consequently, in the presence of TMSTFA, the capacity retention of Li/NCM811 cell is increased by 17% after 200 cycles at 1C, and the lifespan of symmetrical Li/Li cells is prolonged beyond 600 h at 0.5 mA cm-2.

Globally eruptive harmful algal blooms (HABs) have caused numerous negative effects on aquatic ecosystem and human health. Conversion of HABs into biohythane via dark fermentation (DF) is a promising approach to simultaneously cope with environmental and energy issues, but low HABs harvesting efficiency and biohythane productivity severely hinder its application. Here we designed a gradient electro-processing strategy for efficient HABs harvesting and disruption, which had intrinsic advantages of no secondary pollution and high economic feasibility. Firstly, low current density (0.888-4.444 mA/cm2) was supplied to HABs suspension to harvest biomass via electro-flocculation, which achieved 98.59% harvesting efficiency. A mathematic model considering coupling effects of multi-influencing factors on HABs harvesting was constructed to guide large-scale application. Then, the harvested HABs biomass was disrupted via electro-oxidation under higher current density (44.44 mA/cm2) to improve bioavailability for DF. As results, hydrogen and methane yields of 64.46 mL/ (g VS) and 171.82 mL/(g VS) were obtained under 6 min electro-oxidation, along with the highest energy yield (50.1 kJ/L) and energy conversion efficiency (44.87%). Mechanisms of HABs harvesting and disruption under gradient electro-processing were revealed, along with the conversion pathways from HABs to biohythane. Together, this work provides a promising strategy for efficient disposal of HABs with extra benefit of biohythane production.

RESUMEN

Harmful algal blooms (HABs) are growing threats that cause tens of billion dollars economic loss annually. Aiming at efficient disposal of HABs, a cheap and eco-friendly cationic straw was developed by etherification of wheat straw, which replaced hydroxyl groups on cellulose by quaternary ammonium groups. It endowed the cationic straw with high positive charge and achieved 93.92% of harvesting efficiency by enhancing HABs cells aggregation via charge neutralization. Different from inorganic salts-based flocculants, HABs harvesting by the cationic straw is a spontaneous and exothermic process with negative ΔG° and ΔH° under all adsorption conditions. Thermodynamics and kinetics analysis elucidated that HABs adsorption process by cationic straw were mainly driven by physical forces. Together, cationic straw preparation and HABs harvesting processes were comprehensively optimized with orthogonal experiments. The work may inspire cost-effective HABs disposal and fill knowledge gaps of process nature for HABs harvesting.

Wave-absorbing materials are developing in the direction of "light weight, wide frequency band, thin layer and high strength", and it is difficult to achieve the synergy between wave-absorbing performance and mechanical properties when graphene absorbent is compounded with a single resin matrix. In this paper, based on the preparation of a new composite absorbing wire with a graphene (GR)/spherical graphite (SG) double absorbent and polylactic acid (PLA)/thermoplastic polyurethane (TPU) double matrix, we proposed a new method to prepare samples for testing the electromagnetic parameters and tensile strength by fused deposition modeling (FDM). Furthermore, the effect of SG/GR ratio on the microwave absorbing properties and mechanical properties of PLA/TPU composites was specifically studied. It was found that when the ratio of SG/GR was small (0:5, 1:4), the dielectric loss (interfacial polarization loss, dipole polarization loss, conductivity loss) and attenuation ability of the composites were stronger, and the impedance matching was better. When the SG/GR ratio was large (5:0, 4:1), the composites had high strength and toughness. When the ratio of SG/GR was moderate (2:3, 3:2), it could retain the absorbing and mechanical properties of the absorbing materials. On the one hand, the SG and PLA/TPU matrix formed an "island structure", which improves the dispersion of GR; on the other hand, the GR and PLA/TPU matrix formed a "core-shell structure", which promotes polarization and multiple scattering.

Halloween genes are involved in the biosynthesis of the molting hormone, which plays a key role in insect ecdysis, development, metamorphosis, and reproduction. Our previous work identified five Halloween genes from Locusta migratoria, but their functions are currently unknown. In this study, the sequences of these five Halloween genes were analyzed and characterized. LmCYP307a2, LmCYP306a1, LmCYP302a1, and LmCYP315a1 were primarily expressed in the prothoracic glands, while LmCYP314a1 was universally expressed in peripheral tissues, especially in the ovaries and Malpighian tubules. All five Halloween genes were mainly expressed from the 5th to the 7th d in 5th-instar nymphs. RNA interference (RNAi) silencing of LmCYP307a2 resulted in severe molting delays and molting failure, which could be rescued by supplementary 20-hydroxyecdysone. A hematoxylin and eosin staining analysis suggested that the RNAi of LmCYP307a2 inhibited the ecdysis process by inhibiting the apolysis and degradation of the old cuticle, and by promoting the synthesis of a new cuticle. Quantitative reverse transcription polymerase chain reaction results showed that the expressions of LmE74, LmCht5, and LmCht10 were dramatically down-regulated, while that of LmChsI was substantially up-regulated, after knockdown of LmCYP307a2. The results suggest that LmCYP307a2 is related to the molt process via regulation of chitin synthesis and degradation.

In this study, we obtained four alternative splicing transcripts of intracellular copper/zinc superoxide dismutase 1 (icCuZnSOD1) in Oxya chinensis. OcicCuZnSOD1a has all common characteristics of CuZnSOD family and is a canonical CuZnSOD. OcicCuZnSOD1b is missing a Zn binding site. OcicCuZnSOD1c lacks Zn ion and is a Cu-only SOD. OcicCuZnSOD1d is missing a CuZnSOD conserved sequence and lacks the E-loop, a conserved disulfide bond, and an active site arginine. OcicCuZnSOD1a was the most heat-resistant and OcicCuZnSOD1c was the most unstable at high temperatures above 55 °C. They were stable at a wide pH range, especially in alkaline conditions. The four variants expressed at the throughout developmental stages and had various tissue expression patterns. OcicCuZnSOD1a and OcicCuZnSOD1d were significantly induced by 8.79 mM CuCl2 and OcicCuZnSOD1b was significantly up-regulated by 14.67 mM CuCl2. OcicCuZnSOD1a was significantly inhibited by 19.13 mM ZnSO4 while OcicCuZnSOD1d were significantly induced by 22.61 mM ZnSO4. Disc diffusion assay showed that the four isoforms of OcicCuZnSOD1 made the killing zones smaller surrounding the CdCl2-soaked filter discs. However, the reduction ratios of OcicCuZnSOD1a were the highest. These results implied that the four transcripts played roles in defense against CdCl2-induced oxidative stress while OcicCuZnSOD1a had stronger antioxidant capacity.

BACKGROUND: Balloon dilation is a commonly used minimally invasive endourological treatment of ureteral stricture, but the postoperative recurrence rate is relatively high. And factors contributing to recurrence after treatment are poorly understood. Herein, we sought to develop a novel clinical nomogram to predict ureteral stricture-free survival in patients suffering from ureter stricture and performed balloon dilation. METHODS: The nomogram was established based on a retrospective analysis of 321 patients who received endoscopic balloon dilation alone for ureter strictures from January 2016 to January 2020 in Sun Yat-sen Memorial Hospital using the Cox regression model. Perioperative clinical data and disease outcomes were analyzed. The primary endpoint was the onset of ureteral re-stricture after ureter balloon dilation. Discrimination of the nomogram was assessed by the concordance index (C-index) and the calibration curve. The results were internally validated using bootstrap resampling. RESULTS: Overall, 321 patients with a median follow-up of 590 days were enrolled in the study, among which 97 patients (30.2%) developed recurrence of ureteral stricture during follow-up. Five variables remained significant predictors of ureteral re-stricture after multivariable analyses: stricture nature (P < 0.001), urinary nitrite (P = 0.041), CKD (P = 0.005), stent retention time (P < 0.001), and balloon size (P = 0.029). The calibration craves for the probability of 1-, 3-, and 5-years stricture-free survival (SFS) presented satisfied with the consistency of nomogram prediction and actual observation. The C-index of the model was 0.74 (95% CI 0.70-0.79). CONCLUSIONS: Our study developed the first nomogram to effectively predict stricture-free survival in patients suffering from ureter stricture after balloon dilation. It is helpful to identify the optimal patients with ureter stricture for balloon dilation and improve treatment outcomes. However, further external validation of the nomogram is warranted.
