RESUMEN

FMRFamide, a member of the neuropeptide family, is involved in numerous physiological processes. FMRFamide-activated sodium channels (FaNaCs) are a family of non-voltage-gated, amiloride-sensitive, Na+-selective channels triggered by the neuropeptide FMRFamide. In the present study, the full-length cDNA of the FaNaC receptor of Sepiella japonica (SjFaNaC) was cloned. The cDNA of SjFaNaC was 3004 bp long with an open reading frame (ORF) of 1812 bp, encoding 603 amino acid residues with no signal peptide at the N-terminus. Sequence analysis indicated that SjFaNaC shared a high identity with other cephalopods FaNaCs and formed a sister clade with bivalves. The protein structure was predicted using SWISS-MODEL with AcFaNaC as the template. Quantitative real-time PCR (qRT-PCR) revealed that SjFaNaC transcripts were highly expressed in both female and male reproductive organs, as well as in the optic lobe and brain of the central nervous system (CNS). Results of in situ hybridisation (ISH) showed that SjFaNaC mRNA was mainly distributed in the medulla and deep retina of the optic lobe and in both the supraesophageal and subesophageal masses of the brain. Subcellular localisation indicated that the SjFaNaC protein was localised intracellularly and on the cell surface of HEK293T cells. In summary, these findings may lay the foundation for future exploration of the functions of SjFaNaC in cephalopods.

The flathead gray mullet (Mugil cephalus L.) is a cosmopolitan fish that lives in warm and temperate zones over 42°N-42°S. It is a key fish species for industrial fishing off coastal Taiwan. Gray mullets enter the coastal waters of the southeastern Taiwan Strait (22°N-25°N) to spawn in winter and feed in the coastal and tidal waters of China (25°N-30°N). From 1986 to 2010, the annual catch of gray mullet decreased substantially and remained low. Although the Pacific Decadal Oscillation and El Niño-Southern Oscillation are recognized to affect gray mullet migration, the increase in sea surface temperature may be the main cause of the aforementioned decrease. We explored how weather changes affect fishing conditions and patterns at the gray mullet fishing grounds in Taiwan's coastal areas. Because of the decrease in gray mullet catches, the most common method for catching gray mullet in Taiwan's coastal areas between 1990 and 2010 was the use of drift or trawl nets instead of two-boat purse-seiner fleets. Since 2012, purse-seiner fleets have become the most common method for catching gray mullet. This trend indicates that the local fishing industry is adapting to changing environmental conditions.

RESUMEN

Forced normalization (FN) is a unique phenomenon that is often seen in the treatment of epilepsy. FN is characterized by abnormal mental behavior and disordered emotions in epilepsy patients despite a significantly improved electroencephalogram and successful seizure control; the occurrence of FN seriously affects patients' quality of life. The causes of FN include antiseizure medications (ASMs), epilepsy surgery and vagus nerve stimulation, with ASMs being the most common cause. However, with the timely reduction or discontinuation of ASMs and the use of antipsychotic drugs, the overall prognosis is good. Here, we perform an extensive review of the literature pertaining to FN, including its epidemiology, possible mechanisms, clinical features, treatment and prognosis.

The Long Short-Term Memory neural network (LSTM) has excellent learning ability for the time series of the nuclear pulse signal. It can accurately estimate the parameters (such as amplitude, time constant, etc.) of the digitally shaped nuclear pulse signal (especially the overlapping pulse signal). However, due to the large number of pulse sequences, the direct use of these sequences as samples to train the LSTM increases the complexity of the network, resulting in a lower training efficiency of the model. The convolution neural network (CNN) can effectively extract the sequence samples by using its unique convolution kernel structure, thus greatly reducing the number of sequence samples. Therefore, the CNN-LSTM deep neural network is used to estimate the parameters of overlapping pulse signals after digital trapezoidal shaping of exponential signals. Firstly, the estimation of the trapezoidal overlapping nuclear pulse is considered to be obtained after the superposition of multiple exponential nuclear pulses followed by trapezoidal shaping. Then, a data set containing multiple samples is set up; each sample is composed of the sequence of sampling values of the trapezoidal overlapping nuclear pulse and the set of shaping parameters of the exponential pulse before digital shaping. Secondly, the CNN is used to extract the abstract features of the training set in these samples, and then these abstract features are applied to the training of the LSTM model. In the training process, the pulse parameter set estimated by the present neural network is calculated by forward propagation. Thirdly, the loss function is used to calculate the loss value between the estimated pulse parameter set and the actual pulse parameter set. Finally, a gradient-based optimization algorithm is applied to update the weight by getting back the loss value together with the gradient of the loss function to the network, so as to realize the purpose of training the network. After model training was completed, the sampled values of the trapezoidal overlapping nuclear pulse were used as input to the CNN-LSTM model to obtain the required parameter set from the output of the CNN-LSTM model. The experimental results show that this method can effectively overcome the shortcomings of local convergence of traditional methods and greatly save the time of model training. At the same time, it can accurately estimate multiple trapezoidal overlapping pulses due to the wide width of the flat top, thus realizing the optimal estimation of nuclear pulse parameters in a global sense, which is a good pulse parameter estimation method.

Neuropeptides are released by neurons that are involved in a wide range of brain functions, such as food intake, metabolism, reproduction, and learning and memory. A full-length cDNA sequence of an FMRFamide gene isolated from the cuttlefish Sepia pharaonis (designated as SpFMRFamide) was cloned. The predicted precursor protein contains one putative signal peptide and four FMRFamide-related peptides. Multiple amino acid and nucleotide sequence alignments showed that it shares 97% similarity with the precursor FMRFamides of Sepiella japonica and Sepia officinalis and shares 93% and 92% similarity with the SpFMRFamide gene of the two cuttlefish species, respectively. Moreover, the phylogenetic analysis also suggested that SpFMRFamide and FMRFamides from S. japonica and S. officinalis belong to the same sub-branch. Tissue expression analysis confirmed that SpFMRFamide was widely distributed among tissues and predominantly expressed in the brain at the three development stages. The combined effects of SpFMRFamide+SpGnRH and SpFLRFamide+SpGnRH showed a marked decrease in the level of the total proteins released in the CHO-K1 cells. This is the first report of SpFMRFamide in S. pharaonis and the results may contribute to future studies of neuropeptide evolution or may prove useful for the development of aquaculture methods for this cuttlefish species.

Objective: To investigate the in vivo pharmacokinetic characteristics of 17 bioactive components including ginsenoside Rg1, Rb1, Rd, berberine, epiberberine, jatrorrhizine, palmatine, columbamine, coptisine, evodiamine, dehydroevodiamine, rutaecarpine, limonin, hyperin, curcumin, demethoxycurcumin and bisdemethoxycurcumin in rat plasma after oral administration of Xintiantai I extract powder (XI) and Xintiantai I without guide drug borneol extract powder (XI without borneol), and study the compatibility effects of guide drug borneol on the pharmacokinetics. Methods: A UHPLC-MS/MS method was established and fully validated for the comparative pharmacokinetics of 17 bioactive components. The pharmacokinetics parameters of 17 bioactive components after oral administration of XI and XI without borneol were calculated by the software of DAS 3.0 and intercompared. Results: The specificity, linearity, lower limit of quantification (LLOQ), precision, accuracy, extraction recovery rates, matrix effects, and stability of the UHPLC-MS/MS assay were good within the acceptance criteria from FDA guidelines. Guide drug borneol can significantly increase AUC of G-Rd, palmatine, hyperin, curcumin, demethoxycurcumin, bisdemethoxycurcumin and C max of 16 bioactive components except for dehydroevodiamine (P < 0.05), decrease T max of G-Rd, berberine, columbamin, coptisine, limonin and MRT of 17 bioactive components in XI group (P < 0.05). Conclusion: Guide drug borneol enhanced the absorption of G-Rd, palmatine, hyperin, curcumin, demethoxycurcumin and bisdemethoxycurcumin.

BACKGROUND: Thermally significant blood flows into locally cooled diseased tissues and warm them during cryosurgery so that the iceball is often hard to cover the whole diseased volume. This paper is aimed at investigating the effects of large arterial bifurcation on the temperature distribution during cryosurgery through simulation method. METHODS: A parametric geometry model is introduced to construct a close-to-real arterial bifurcation. The three-dimensional transient conjugate heat transfer between bifurcated artery and solid tissues with phase change during cryosurgery is performed by finite volume method. RESULTS: The discussion was then made on the effects of the relative position between cryoprobe and artery bifurcation, the inlet velocity of root artery and the layout of multiple cryoprobes on the temperature distribution and iceball evolution. The results show that the thermal interaction between blood flow and iceball growth near bifurcation is considerable complex. The thermal effects of bifurcation could modulate the iceball morphology, severely weaken its freezing volume and prevent the blood vessel from being frozen. CONCLUSION: The present work is expected to be valuable in optimizing cryosurgery scheme of the situation that the bifurcated artery is embedded into the disease tissue.

The present paper reviews the development courses of acupuncture and moxibustion and gives an introduction to the present situation, education and legislation of acupuncture and moxibustion in United Kingdom. Acupuncture and moxibustion have been developed in United Kingdom since 1960s, the London College of TCM was established by Beijing University of Chinese Medicine join forced with Acu-medic Foundation in 1993, and so far, acupuncture has been taught as an undergraduate program in four United Kingdom universities. Since 2002, the British government and Ministry of Health have begun on the legislation of Chinese Medicine and acupuncture and moxibustion, the lawmaking group of Ministry of Health in British submitted a bill about the legislation of acupuncture and herbal medicine as well as Chinese Medicine to the British government in 2008. Clinical levels of acupuncture workers are quite different and the therapy of acupuncture and moxibustion for nearly 20 diseases displays certain effect. The government or one's own can pay the expenses for acupuncture and moxibustion treatment.

<p><b>OBJECTIVE</b>To evaluate the accordance of tidal volume (TV) with thoracic inflating volume (TIV) measured by pressure whole-body or double-chamber plethysmography in mice.</p><p><b>METHODS</b>TV and TIV by double-chamber plethysmography were simultaneously measured in 6 mice with spontaneous respiratory frequencies 90 - 120/min; TV and TIV by whole-body plethysmography were measured in 8 paralyzed mice ventilated by fixed frequency.</p><p><b>RESULT</b>TIV value by double-chamber plethysmography was significantly higher than TV [(0.369 +/- 0.014) ml vs (0.356 +/- 0.012) ml, P < 0.01)] in 6 spontaneously breathing mice. TIV value by whole-body plethysmography was significantly lower than TV[(0.233 +/- 0.003) ml vs 0.3 ml, P < 0.001] in 8 paralyzed mice.</p><p><b>CONCLUSION</b>The TV value assessed by TIV measurement can not be accurate, because of the humidifying and heating of inhaled air and negative thoracic pressure during the measurement.</p>

RESUMEN

<p><b>OBJECTIVE</b>Acute pulmonary thromboembolism (PTE) is a serious high mortality pulmonary vascular disease whose effective treatment decreases morbidity and mortality. To determine if low-molecular-weight-heparin (LMWH) is clinically as efficient and safe as unfractionated heparin (UH) in patients with diagnosis of acute non-massive PTE, our study compares the efficacy, adverse effects and costs of LMWH and UH.</p><p><b>METHODS</b>One hundred and fourteen patients with non-massive acute PTE were randomly divided into LMWH (nadroparin calcium) and UH groups. Oxygenation index, D-dimer, fibrinogen (FG), lung ventilation/perfusion (V/Q) scan and computed tomography pulmonary angiography (CTPA) were observed before anticoagulation and on day 14 after anticoagulation.</p><p><b>RESULTS</b>In both groups, the ABG (arterial blood gas) analysis showed PaO(2) and PaCO(2) were elevated, P(A-a)O(2) was decreased and oxygenation index (PaO(2)/FIO(2)) was elevated, D-dimer and fibrinogen were decreased, lung V/Q and CTPA showed embolized segments reduced (P<0.05). Hemorrhage and thrombocytopenia occurred in 3.5% of the LMWH group. Hemorrhage occurred in 5.3% and thrombocytopenia occurred in 7.0% of the UH group. The average cost in the LMWH group was RMB 1218.60 Yuan and RMB 1541.40 Yuan in the UH group.</p><p><b>CONCLUSION</b>LMWH and UH are equally effective for treatment of non-massive acute PTE, but LMWH may have a lower prevalence of complications and is less expensive.</p>
