RESUMEN

The pathogenesis of psoriasis, an immune-mediated chronic inflammatory skin disease, remains unclear. Studies have shown an association between psoriasis and intestinal inflammation; in this context, the influence of the gut microbiota on the immune response of psoriasis has become a focus of recent research. The present research evaluated the composition and diversity of the gut microbiota of 21 participants with psoriasis from a Brazilian referral dermatology service compared to 24 healthy controls. A stool sample was collected from each participant at the time of inclusion in the study, and the samples were analysed by sequencing the 16S rRNA gene. The recruitment of research participants involved matching between groups by sex, age, body mass index, comorbidities and smoking and the exclusion of several criteria that could potentially influence the gut microbiota and the interpretation of the data. There was an increase in the Dialister genus and Prevotella copri species in patients with psoriasis compared to the control group. A reduction in the Ruminococcus, Lachnospira and Blautia genera, as well as in the Akkermansia muciniphila species, was also verified in the psoriasis group compared to the control group. Furthermore, patients with psoriasis exhibited less gut microbiota diversity than controls.

RESUMEN

Viruses belonging to the genus Begomovirus (family Geminiviridae) have circular single-strand DNA genomes encapsidated into quasi-icosahedral particles, and are transmitted by whiteflies of the Bemisia tabaci complex. Biological and molecular properties of begomoviruses have been studied efficiently with infectious clones containing dimeric genomic components. However, current approaches employing enzymatic digestion and ligation to binary vectors are laborious, mostly due to many cloning steps or partial digestion by restriction enzyme. Here, an infectious clone of the bipartite begomovirus Bean golden mosaic virus (BGMV) was obtained using PCR and Gibson Assembly (GA). Common bean (Phaseolus vulgaris) seedlings displayed severe yellow mosaic and stunt symptoms 15 days after agroinoculation with DNA-A and DNA-B of BGMV. The approach based on PCR-GA protocol is a fast and useful tool to obtain infectious clones of a circular DNA plant virus.

BACKGROUND: Experts in medical invasive evaluations, like colonoscopy, could be exposed to ergonomic risks during their work. Little attention has been given to these spectrum of occupational health. Its aimed to analyze possible clinical problems related to upper limb of physicians who perform those exams. METHODS: Cross-sectional study in a health service in Sao Paulo, Brazil. For evaluation of the workplace of the colonoscopist physician were applied two tools: Couto's check list and Sue Rodgers Method. The results direct the potential risk to upper limb injuries. RESULTS: The procedure is done and the final report is issued. There are no mandatory pauses during workday. The dominant hand holds the tube insertion with pincer movement during the exam, digital prehension being necessary at times. For this activity the employee has to use some strength. Couto's check list indicates a significant biomechanical factor. Sue Rodgers's Method states high ergonomic risk for dominant hand/wrist/fingers. CONCLUSION: The main risk for the development of health disorders are associated with the use of force and repetitive movements of hands. The adoption of regular breaks to recover most of the structures required is recommended.

In order to investigate an alternative way to correct diaphysial bone defects, granulated decalcified homologous cortical bone was used as a graft. Because of the suitable anatomic arrangement, the forearm of rabbits was chosen as an experimental model of bone defect. A 2 cm long bone cylinder was removed from the diaphysis of both radii preserving the periosteum. The artificial bone defect was filled with granular decalcified bone on the right side. The left side was used as control and kept empty or filled with undecalcified granular bone. The 18 animals were sacrificed in batches, 3,6, and 9 weeks after the operation. New bone formation was followed by X-rays, routine histology and incorporation of calcein blue, xylenol orange and tetracycline. In the decalcified granular bone grafts new bone formation was already detected at the first week and 9 weeks after the graft operation there was a well developed cylindric ossicle, in 89% of the cases. In some cases a medullary canal was present. No bone formation was found neither in the empty defects nor in the ones filled with undecalcified granular bone grafts.

RESUMEN

No presente trabalho, foi investigada uma forma alternativa de correcao de falhas osseas diafisarias com enxerto homologo de osso cortical descalcificado, na forma de granulos. Tendo o coelho como animal de experimentacao, foram retirados cilindros de 2 cm de comprimentos da regiao medio-diafisaria de ambos os radios, preservando-se o periosteo. A falha criada foi sempre preenchida por enxerto osseo nao descalcificado a esquerda, que serviu como controle. Os animais foram estudados ate 9 semanas apos a cirurgia. Os resultados foram avaliados radiologicamente, atraves de incorporacao de corantes fluorescentes indicadores de osso neoformado, tendo como marcadores calceina azul, xilenol laranja e tetraciclina, possibilitando um estudo dinamico da osteogenese. Nas falhas preenchidas com osso descalcificado, ja havia neoformacao ossea na primeira semana, e apos 9 semanas, houve correcao completa da falha ossea em 89% dos casos, formando-se um ossiculo cilindrico, perfeitamente solidario com o restante do radio e apresentando canal medular em alguns casos

RESUMEN

Os resultados do tratamento cirurgico de 29 pacientes portadores de lesoes traumaticas da coluna cervical sao apresentados e comentados. Na maioria dos casos as etapas seguidas foram as seguintes: 1a. etapa - Reducao: manipulacao sob anestesia seguida de tracao; 2a. etapa - Manutencao: artrodese da coluna cervical por via anterior. As radiografias de controle realizadas entre o 2o. e 3o. mes de pos-operatorio mostraram a integracao do enxerto e boa estabilidade da coluna. Nos casos apropriados o retorno do paciente as suas atividades deu-se em torno do 4o.mes

RESUMEN

Os efeitos da isquemia sobre o musculo esqueletico foram estudados experimentalmente em caes adultos. Os animais foram subdivididos em dois grupos: grupo I - 35 caes - submetidos unicamente a isquemia de 1 a 10 horas. O grupo R - 20 caes - era constituido pelos animais submetidos a isquemia de 1 a 10 horas, seguido de revascularizacao ate o maximo de 10 horas. O estudo morfologico do material foi realizado visando a analise das alteracoes isquemicas, juntamente com a citometria das fibras, no membro isquemiado, no revascularizado e no controle. Os diametros das fibras musculares variam entre si, tanto na fase isquemica como pos-isquemica. Observou-se recuperacao parcial dos diametros quando a isquemia era de ate 2 horas. Os resultados obtidos atraves da correlacao entre o coeficiente de variabilidade dos diametros e as demais alteracoes teciduais revelaram que a fase isquemica de ate 2 horas pode ser suportada pelo tecido muscular. A partir deste tempo, as lesoes tornam-se irreversiveis

RESUMEN

The integration of immature cartilage allografts to knee joints of both normal rabbits and those submitted to arthritis through papain was studied for a period of up to 9 months. Two types of grafts were compared: chondral--constituted only by cartilage, and osteochondral--cartilage plus a thin layer of subchondral bone. The integration of the grafts was analyzed through inspection of the articular surface and histologic sections. The viability of the grafts was checked through the incorporation of 35SO4 on the chondral matrix. The following conclusions were drawn: (1) A large member of the grafted cartilages were well accepted without histological evidence of immunological rejection, and metabolically active 9 months after transplantation. (2) The chondral graft was found to be superior to the osteochondral one regarding the integration, but both showed tendency to degeneration with time. (3) The presence of arthritis previously induced by papain affected the integration of the grafts causing a higher precentage of degeneration on the grafts causing a higher precentage of degeneration on the grafted cartilage.
