RESUMEN

Spastic paraplegia type 4 (SPG4) is the most common type of autosomally inherited spastic paraplegia. Its main clinical features include typical simple hereditary spastic paraplegia, with neurological impairments limited to lower limb spasticity, hypertonic bladder dysfunction, and mild weakening of lower limb vibration sensation, without accompanying features such as nerve atrophy, ataxia, cognitive impairment, seizures, and muscle tone disorders. SPAST is the main pathogenic gene underlying SPG4, and various pathogenic SPAST variants have been discovered. This disease has featured a high degree of clinical heterogeneity, and the same pathogenic variant can have different age of onset and severity among different patients and even within the same family. There is a lack of systematic research on the correlation between the genotype and phenotype of SPG4, and the pathogenic mechanism has remained controversial. This article has provided a review for the clinical characteristics, pathogenic gene characteristics, correlation between the genotype and phenotype, and pathogenic mechanism of this disease, with an aim to provide reference for its clinical diagnosis and treatment.

RESUMEN

@#To analyze the clinicial features magnetic resonance imaging manifestations of contrast transthoracic echocardiography(cTTE)on patients cryptogenic stroke,in order to discover the specific clinical characteristics of patients with cTTE+ and contribute to the etiological diagnosis of CS.Methods The clinical and imaging data of 108 patients who were diagnosed with CS in Ningbo Huamei Hospital from August 2020 to May 2022 were retrospectively collected. According to the cTTE results, they were classified intotwo groups:cTTE+ group and cTTEgroup. Then the age of onset, gender, risk factors of cerebrovascular disease(hypertension, diabetes, heart disease, smoking history, drinking history, etc. ),symptoms,NIHSS score,mRS score, left atrium size, infarct types(number, location, size)of cerebral infarction on diffusion weighted imaging(DWI)of the two groups were compared and analyzed. The differences of infarct lesions on DWI with different grades of right-to-left shunt were analyzed.Results(1)The cTTE+ positive rate among 108 ptients with CS was 39.8%,including 30 males(69.8%)and 13 females(30.2%),aged(52.8±17.5)years. Right-to-left shunt was found in 21 patients with grade 1,12 with grade 2,and 10 with grade 3.In the positive group,4 cases of extracardiac shunt were found, and 2 cases of pulmonary arteriovenous fistula were confirmed by operation.(2)Among the two groups of patients, the cTTE+ group had a higher ratio of young adults(P=0.021)and a higher incidence of non-specific symptoms at onset(P=0.008),while the negative group had a higher ratio of core symptoms and personal history of smoking(P=0.012),with statistically significant differences.(3)The proportion of multi-vessel area involved in the cTTE+ group was significantly higher than that in the cTTE-group(P=0.004),Meanwhile,more cerebral infarction lesions were distributed in the vertebrobasilar artery system(P=0.021),Less in the right internal carotid artery system(P=0.045);and the maximum diameter of stroke infarct lesions in the cTTE-group was more distributed in [1cm,3cm],with statistical significance(P<0.05).With the increase of right to left shunt grade, the proportion of lesions<1 cm showed an increasing trend, but there was no significant statistical difference.Conclusion(1)Non-specific symptom onset of CS mainly occurs in multi-vessel distribution areas under 50 years old,and the pathogenesis should be considered to be related to right-to-left shunt.(2)The detection of right-to-left shunt by cTTE is of supplementary value to the etiological diagnosis of CS,which can detect some hidden and undetectable extracardial shunt, suggesting that CS patients should actively undergo cTTE screening, which is helpful to define the etiology and guide the selection of stroke prevention and treatment plan.

RESUMEN

Photoperiod plays an important role in transformation from vegetative growth to reproductive growth in plants. CONSTANS (CO), as a unique gene in the photoperiod pathway, responds to changes of day length to initiate flowering in the plant. In this study, the expression level of FaCONSTANS (FaCO) gene under long-day, short-day, continuous light and continuous darkness conditions was analyzed by real-time quantitative PCR. We constructed the over-expression vector p1300-FaCO and infected into Arabidopsis thaliana by Agrobacterium-mediated method. We constructed the silencing vector p1300-FaCO-RNAi and infected into Festuca arundinacea by Agrobacterium-mediated method. The expression of FaCO gene was regulated by photoperiod. The over-expression of FaCO promoted flowering in wild type of Arabidopsis thaliana under long day condition and rescued the late flowering phenotype in co-2 mutant of Arabidopsis thaliana. Silencing FaCO gene in Festuca arundinacea by RNAi showed late-flowering phenotype or always kept in the vegetative growth stage. Our understanding the function of FaCO in flowering regulation will help further understand biological function of this gene in Festuca arundinacea.

RESUMEN

Objective:To investigate the relationship among happiness, resilience, optimistic intelligence quotient and perceived social support in patients with advanced gynecological cancer. Methods:A total of 269 patients with advanced gynecological cancer from January 2018 to January 2019 in three top three hospitals in Shanghai were selected as subjects by convenient sampling. The basic information questionnaire, Memorial University of New-foundland scale of happiness, Connor-Davidson Resilience Scale simplified Chinese version, Optimistic Intelligence Quotient Questionnaire and Perceived Social Support Scale were used to investigate. Results:The score of happiness was (32.45±6.37), the score of resilience was (30.64±8.78),the score of optimistic intelligence quotient was (62.74±12.49) and the score of perceived social support was (68.14±4.25) in patients with advanced gynecological cancer. Linear regression equation analysis showed that happiness and optimistic intelligence quotient × happiness had significant predictive effect on resilience ( β values were 0.547, 0.016, P < 0.05), optimistic intelligence quotient had no significant predictive effect on resilience ( P>0.05), and optimistic intelligence quotient, happiness, resilience, optimistic intelligence quotient×happiness had significant predictive effect on perceived social support ( β values were 0.015-0.251, P<0.01). Happiness could regulate the direct path and the first half of the mediating effect of resilience between perceived social support and optimistic intelligence quotient. According to Johnson-Neyman's analysis, the boundary value of the moderating effect of happiness between optimistic intelligence quotient and resilience was 26.19, and the boundary value of the moderating effect of happiness between optimistic intelligence quotient and perceived social support was 22.08. Conclusions:There is a positive correlation between optimistic intelligence quotient and perceived social support in patients with advanced gynecological cancer. Resilience can play a significant mediating role between optimistic intelligence quotient and perceived social support under the regulation of happiness, which can provide corresponding psychological intervention for patients with advanced gynecological cancer.

RESUMEN

Autophagy is essential for the maintenance of cellular homeostasis and its dysfunction has been linked to various diseases. Autophagy is a membrane driven process and tightly regulated by membrane-associated proteins. Here, we summarized membrane lipid composition, and membrane-associated proteins relevant to autophagy from a spatiotemporal perspective. In particular, we focused on three important membrane remodeling processes in autophagy, lipid transfer for phagophore elongation, membrane scission for phagophore closure, and autophagosome-lysosome membrane fusion. We discussed the significance of the discoveries in this field and possible avenues to follow for future studies. Finally, we summarized the membrane-associated biochemical techniques and assays used to study membrane properties, with a discussion of their applications in autophagy.

RESUMEN

OBJECTIVE: To investigate the relationship between Interleukin-1beta (IL-1beta) and diabetic peripheral neuropathy (DPN) using animal models. METHODS: The rat model of diabetic neuropathy was induced by intraperitoneal injection of a single dose of streptozotocin (STZ) at 65mg/kg. Diabetic rats were randomly divided into two groups (10 each), one treated with 0.9% saline (DMS group) and the other with interleukin-1 receptor antagonist (IL-1RA) at 50mg/kg (DMI group) twice a day for 5 weeks. Ten normal rats matched for weight, age and sex served as normal controls (Con group) and were treated with saline. Morphologic studies of sciatic nerves were achieved using light and transmission electron microscopy. RESULTS: Transmission electron microscopy of the sciatic nerve showed the ultrastructure of myelin and the axon in the IL-1RA group was highly protected compared to diabetic controls. CONCLUSION: High levels of circulating IL-1beta may be associated with the risk of DPN and anti-IL-1 treatment may provide a potential strategy for the prevention of diabetic neuropathy.

RESUMEN

Light signaling and cortical microtubule (MT) arrays are essential to the anisotropic growth of plant cells. Microtubule-associated proteins (MAPs) function as regulators that mediate plant cell expansion or elongation by altering the arrangements of the MT arrays. However, current understanding of the molecular mechanism of MAPs in relation to light to regulate cell expansion or elongation is limited. Here, we show that the MPS SPR1 is involved in light-regulated directional cell expansion by modulating microtubule arrangement. Overexpression of SmSPR1 in Arabidopsis results in right-handed helical orientation of hypocotyls in dark-grown etiolated seedlings, whereas the phenotype of transgenic plants was indistinguishable from those of wild-type plants under light conditions. Phenotypic characterization of the transgenic plants showed reduced anisotropic growth and left-handed helical MT arrays in etiolated hypocotyl cells. Protein interaction assays revealed that SPR1, CSN5A (subunits of COP9 signalosome, a negative regulator of photomorphogenesis), and ELONGATED HYPOCOTYL 5 (HY5, a transcription factor that promotes photomorphogenesis) interacted with each other in vivo. The phenotype of Arabidopsis AtSPR1-overexpressing transgenic lines was similar to that of SmSPR1-overexpressing transgenic plants, and overexpression of Salix SmSPR1 can rescue the spr1 mutant phenotype, thereby revealing the function of SPR1 in plants.
