RESUMEN

Objective:To investigate the influences of circular RNA(circRNA)BICD2 on the proliferation,apoptosis and epitheli-al-mesenchymal transition(EMT)of oral squamous cell carcinoma(OSCC)cells by regulating the miR-218-5p/Ras homolog gene family member A(RhoA)axis.Methods:qRT-PCR and Western blot were applied to detect CircBICD2,miR-218-5p gene expres-sion and RhoA protein expression in OSCC tissue,paracancerous tissue,human oral epithelial cell line HOEC and OSCC cell line HSC-4,CAL-27 and SCC-15,respectively.SCC-15 cells were divided into 7 groups of Ct(control,A),si-NC(B),si-CircBICD2(C),miR-NC(D),miR-218-5p(E),si-CircBICD2+anti-NC(F)and si-CircBICD2+anti-miR-218-5p(G).qRT-PCR was ap-plied to detect the expression of CircBICD2 and miR-218-5p in the cells;CCK-8 assay and plate cloning assay were applied to detect the cell proliferation;flow cytometry was applied to detect cell apoptosis;Western blot was applied to detect the protein expressions of RhoA,E-cadherin,Vimentin and N-cadherin;dual luciferase was applied to verify the relationship between CircBICD2 and miR-218-5p,miR-218-5p and RhoA,respectively.Results:In OSCC tissues and cells,CircBICD2 and RhoA proteins were highly ex-pressed,miR-218-5p was lowly expressed.In SCC-15 cells,the expression of CircBICD2 and RhoA protein was the highest,and the expression of miR-218-5p was the lowest(P<0.05),SCC-15 cells in the 7 groups showed that,compared with group B,the expres-sion of CircBICD2 and RhoA in group C was decreased,and the expression of miR-218-5p was increased(P<0.05);compared with group D,in group E RhoA protein expression level was decreased,and the miR-218-5p expression level was increased(P<0.05);compared with group C and group F,the expression of miR-218-5p decreased in group G,and the expression of RhoA protein in-creased(P<0.05).Down-regulation of CircBICD2 or over-expression of miR-218-5p inhibited SCC-15 cell proliferation and EMT,and induced cell apoptosis;down-regulation of miR-218-5p attenuated the inhibition of SCC-15 cell proliferation and EMT,the cell apoptosis was promoted by silencing CircBICD2.Cir-cBICD2 targeted the miR-218-5p/RhoA axis.Conclusion:Silencing CircBICD2 may inhibit RhoA expression by up-regulating miR-218-5p,inhibits SCC-15 cell proliferation and EMT,and induces cell apoptosis.

RESUMEN

Objective:To comprehensively retrieve and summarize the best evidence on the prevention of position-related nerve injury in adult patients undergoing general anesthesia, in order to provide evidence-based guidance for standardized position management during general anesthesia surgery in adults.Methods:Clinical decision-making, guidelines, evidence summaries, best practice, practice advisories, systematic reviews, expert consensuses were systematically search in UpToDate, BMJ Best Practice, Guidelines International Network (GIN), Canadian Medical Association: Clinical Practice Guideline(CMA Infobase), National Guideline Clearinghouse (NGC), National Institute for Health and Care Excellence(NICE), Cochrane Library, PubMed, CNKI, Wanfang, and related websites. The literature retrieval period was from the database construction to September 30,2022. The guidelines were independently assessed by 4 researchers, and the remaining literature was independently evaluated by 2 researchers. The literature that met the criteria was extracted. Finally, the expert meeting integrated the evidence and summarized the evidence topics.Results:A total of 17 articles were included, including 6 clinical decision-making, 3 guidelines, 2 practice advisories,5 systematic reviews, and 1 expert consensus. A total of 32 pieces of best evidence and 5 evidence topics were formed: personnel placement, perioperative evaluation, points of surgical position, key points in special surgery or situation, other general principles.Conclusion:This study summarized the best evidence for the prevention and management of surgical position related nerve injuries, and provides a scientific theoretical reference for postural management of adult patients undergoing operation with general anesthesia, to reduce the incidence of nerve injuries related position.

RESUMEN

Objective:To investigate the correlation of PELP1/MNAR expression with expressions of estrogen receptor (ER), Ki-67, human epidermal growth factor receptor 2 (HER2) and PIK3CA gene mutation.Methods:A total of 80 paraffin-embedded tissue specimens of primary invasive breast cancer patients in the Fifth People's Hospital of Datong in Shanxi Province from January 2008 to December 2018 were collected. The expression of PELP1/MNAR was examined by immunohistochemistry EliVision tow-step method. The polymerase chain reaction (PCR)-Sanger sequencing method was used to detect the mutation of PIK3CA gene. The expressions of PELP1/MNAR among patients with different expression status of ER, Ki-67, HER2 and with or without PIK3CA gene mutation were compared, and the correlations between each index and the expression of PELP1/MNAR were analyzed.Results:The high expression rates of PELP1/MNAR protein in patients with ER-positive [86.1% (31/36) vs. 59.1% (26/44)], Ki-67 high expression [100.0% (13/13) vs. 65.7% (44/67)], HER2-positive [81.0% (34/42) vs. 60.5% (23/38)] were high, and the differences were statistically significant (all P<0.05). There was no significant difference in the high expression rate of PELP1/MNAR protein between patients with mutant and wild-type PIK3CA [60.0% (12/20) vs. 75.0% (45/60), P = 0.199]. The expression of PELP1/MNAR was negatively correlated with the expression level of ER ( r = -0.195, P < 0.05), positively correlated with the expression level of Ki-67 ( r = 0.198, P < 0.05), positively correlated with the expression level of HER2 ( r = 0.225, P < 0.05), and negatively correlated with lymph node metastasis ( r = -0.269, P < 0.05). Conclusions:The expression of PELP1/MNAR in invasive breast cancer is negatively correlated with the expression level of ER, and positively correlated with the expression level of Ki-67 and HER2. There is no correlation between PELP1/MNAR expression and PIK3CA gene mutation, and the two may play their own role in the PI3K-AKT-mTOR regulatory pathway of breast cancer.

RESUMEN

The construction of biologic functional surfaces of materials, from the visual angle of material science, is aimed to make the biomaterials adapted by tissues, and to endow them with dynamic conformity; moreover, from the view-point of clinical applications, it is the functional surface to join the environmental tissues with the implanted material, playing the role of artificial extracellular matrix (ECM). The architecture of biologic functional surface is very important in tissue engineering science. Here the primary concepts of biological surface science and the construction and application of biofunctional surfaces in tissue engineering are reviewed.
