RESUMEN

Asymmetric transmission in a passive vortex system is highly desirable, as it enables the development of compact vortex-based devices. However, breaking the mirror symmetry of transmission via a single metasurface poses challenges due to the inherent symmetric transmission properties in reciprocity. Here, we theoretically propose and experimentally demonstrate a novel transmission-reflection phase coupling mechanism to achieve the broken mirror symmetry of sound vortex transmission. This mechanism establishes a special coupling link between transmission and reflection waves, superimposing asymmetric reflection phases on the transmission phases. By utilizing a single passive phase gradient metasurface with asymmetric reflection phase twists, distinct transmission phase twists for mirror-symmetric incident vortices can be achieved within a cylindrical waveguide. This is typically difficult to imple-ment in a reciprocal system. Numerical and experimental results both demonstrate the broken mirror symmetry of vortex transmission and reflection. Our findings offer a new strategy for controlling vortex wave propagation, which may inspire new directional applications and extend to the field of photonics.

RESUMEN

Precipitation nowcasting refers to the use of specific meteorological elements to predict precipitation in the next 0-2 h. Existing methods use radar echo maps and the Z-R relationship to directly predict future rainfall rates through deep learning methods, which are not physically constrained, but suffer from severe loss of predicted image details. This paper proposes a new model framework to effectively solve this problem, namely LSTMAtU-Net. It is based on the U-Net architecture, equipped with a Convolutional LSTM (ConvLSTM) unit with the vertical flow direction and depthwise-separable convolution, and we propose a new component, the Efficient Channel and Space Attention (ECSA) module. The ConvLSTM unit with the vertical flow direction memorizes temporal changes by extracting features from different levels of the convolutional layers, while the ECSA module innovatively integrates different structural information of each layer of U-Net into the channelwise attention mechanism to learn channel and spatial information, thereby enhancing attention to the details of precipitation images. The experimental results showed that the performance of the model on the test dataset was better than other examined models and improved the accuracy of medium- and high-intensity precipitation nowcasting.

The essential COPI coat mediates retrieval of transmembrane proteins at the Golgi and endosomes following recruitment by the small GTPase, Arf1. ArfGAP proteins regulate COPI coats, but molecular details for COPI recognition by ArfGAPs remain elusive. Biochemical and biophysical data reveal how ß'-COP propeller domains directly engage the yeast ArfGAP, Glo3, with a low micromolar binding affinity. Calorimetry data demonstrate that both ß'-COP propeller domains are required to bind Glo3. An acidic patch on ß'-COP (D437/D450) interacts with Glo3 lysine residues located within the BoCCS (binding of coatomer, cargo, and SNAREs) region. Targeted point mutations in either Glo3 BoCCS or ß'-COP abrogate the interaction in vitro, and loss of the ß'-COP/Glo3 interaction drives Ste2 missorting to the vacuole and aberrant Golgi morphology in budding yeast. These data suggest that cells require the ß'-COP/Glo3 interaction for cargo recycling via endosomes and the TGN, where ß'-COP serves as a molecular platform to coordinate binding to multiple proteins, including Glo3, Arf1, and the COPI F-subcomplex.

RESUMEN

Reproducing ion channel-based neural functions with artificial fluidic systems has long been an aspirational goal for both neuromorphic computing and biomedical applications. In this study, neuromorphic functions were successfully accomplished with a polyelectrolyte-confined fluidic memristor (PFM), in which confined polyelectrolyte-ion interactions contributed to hysteretic ion transport, resulting in ion memory effects. Various electric pulse patterns were emulated by PFM with ultralow energy consumption. The fluidic property of PFM enabled the mimicking of chemical-regulated electric pulses. More importantly, chemical-electric signal transduction was implemented with a single PFM. With its structural similarity to ion channels, PFM is versatile and easily interfaces with biological systems, paving a way to building neuromorphic devices with advanced functions by introducing rich chemical designs.

RESUMEN

Retromer (VPS26/VPS35/VPS29 subunits) assembles with multiple sorting nexin proteins on membranes to mediate endosomal recycling of transmembrane protein cargoes. Retromer has been implicated in other cellular processes, including mitochondrial homeostasis, nutrient sensing, autophagy, and fission events. Mechanisms for mammalian retromer assembly remain undefined, and retromer engages multiple sorting nexin proteins to sort cargoes to different destinations. Published structures demonstrate mammalian retromer forms oligomers in vitro, but several structures were poorly resolved. We report here improved retromer oligomer structures using single-particle cryo-EM by combining data collected from tilted specimens with multiple advancements in data processing, including using a 3D starting model for enhanced automated particle picking in RELION. We used a retromer mutant (3KE retromer) that breaks VPS35-mediated interfaces to determine a structure of a new assembly interface formed by the VPS26A and VPS35 N-termini. The interface reveals how an N-terminal VPS26A arrestin saddle can link retromer chains by engaging a neighboring VPS35 N- terminus, on the opposite side from the well-characterized C-VPS26/N-VPS35 interaction observed within heterotrimers. The new interaction interface exhibits substantial buried surface area (∼7000 Å2) and further suggests that metazoan retromer may serve as an adaptable scaffold.

RESUMEN

Nanofluidic memristors are memory resistors based on nanoconfined fluidic systems exhibiting history-dependent ion conductivity. Toward establishing powerful computing systems beyond the Harvard architecture, these ion-based neuromorphic devices attracted enormous research attention owing to the unique characteristics of ion-based conductors. However, the design of nanofluidic memristor is still at a primary state and a systematic guidance on the rational design of nanofluidic system is desperately required for the development of nanofluidic-based neuromorphic devices. Herein, we proposed a systematic review on the history, main mechanism and potential application of nanofluidic memristors in order to give a prospective view on the design principle of memristors based on nanofluidic systems. Furthermore, based on the present status of these devices, some fundamental challenges for this promising area were further discussed to show the possible application of these ion-based devices.

Deciphering mechanisms controlling SNARE localization within the Golgi complex is crucial to understanding protein trafficking patterns within the secretory pathway. SNAREs are also thought to prime coatomer protein I (COPI) assembly to ensure incorporation of these essential cargoes into vesicles, but the regulation of these events is poorly understood. Here, we report roles for ubiquitin recognition by COPI in SNARE trafficking and in stabilizing interactions between Arf, COPI, and Golgi SNAREs in Saccharomyces cerevisiae. The ability of COPI to bind ubiquitin, but not the dilysine motif, through its N-terminal WD repeat domain of ß'-COP or through an unrelated ubiquitin-binding domain is essential for the proper localization of Golgi SNAREs Bet1 and Gos1. We find that COPI, the ArfGAP Glo3, and multiple Golgi SNAREs are ubiquitinated. Notably, the binding of Arf and COPI to Gos1 is markedly enhanced by ubiquitination of these components. Glo3 is proposed to prime COPI-SNARE interactions; however, Glo3 is not enriched in the ubiquitin-stabilized SNARE-Arf-COPI complex but is instead enriched with COPI complexes that lack SNAREs. These results support a new model for how posttranslational modifications drive COPI priming events crucial for Golgi SNARE localization.

The efficiency of warehouses is vital to e-commerce. Fast order processing at the warehouses ensures timely deliveries and improves customer satisfaction. However, monitoring, analyzing, and manipulating order processing in the warehouses in real time are challenging for traditional methods due to the sheer volume of incoming orders, the fuzzy definition of delayed order patterns, and the complex decision-making of order handling priorities. In this paper, we adopt a data-driven approach and propose OrderMonitor, a visual analytics system that assists warehouse managers in analyzing and improving order processing efficiency in real time based on streaming warehouse event data. Specifically, the order processing pipeline is visualized with a novel pipeline design based on the sedimentation metaphor to facilitate real-time order monitoring and suggest potentially abnormal orders. We also design a novel visualization that depicts order timelines based on the Gantt charts and Marey's graphs. Such a visualization helps the managers gain insights into the performance of order processing and find major blockers for delayed orders. Furthermore, an evaluating view is provided to assist users in inspecting order details and assigning priorities to improve the processing performance. The effectiveness of OrderMonitor is evaluated with two case studies on a real-world warehouse dataset.

The retromer complex (Vps35-Vps26-Vps29) is essential for endosomal membrane trafficking and signaling. Mutation of the retromer subunit Vps35 causes late-onset Parkinson's disease, while viral and bacterial pathogens can hijack the complex during cellular infection. To modulate and probe its function, we have created a novel series of macrocyclic peptides that bind retromer with high affinity and specificity. Crystal structures show that most of the cyclic peptides bind to Vps29 via a Pro-Leu­containing sequence, structurally mimicking known interactors such as TBC1D5 and blocking their interaction with retromer in vitro and in cells. By contrast, macrocyclic peptide RT-L4 binds retromer at the Vps35-Vps26 interface and is a more effective molecular chaperone than reported small molecules, suggesting a new therapeutic avenue for targeting retromer. Last, tagged peptides can be used to probe the cellular localization of retromer and its functional interactions in cells, providing novel tools for studying retromer function.

Perfect absorbers with high quality factors (Q-factors) are of great practical significance for optical filtering and sensing. Moreover, tunable multiwavelength absorbers provide a multitude of possibilities for realizing multispectral light intensity manipulation and optical switches. In this study, we demonstrate the use of vanadium dioxide (VO2)-assisted metasurfaces for tunable dual-band and high-quality-factor perfect absorption in the mid-infrared region. In addition, we discuss the potential applications of these metasurfaces in reflective intensity manipulation and optical switching. The Q-factors of the dual-band perfect absorption in the proposed metasurfaces are greater than 1000, which can be attributed to the low radiative loss induced by the guided-mode resonances and low intrinsic loss from the constituent materials. By utilizing the insulator-metal transition in VO2, we further proved that a continuous tuning of the reflectance with a large modulation depth (31.8 dB) can be realized in the designed metasurface accompanied by a dual-channel switching effect. The proposed VO2-assisted metasurfaces have potential applications in dynamic and multifunctional optical devices, such as tunable multiband filters, mid-infrared biochemical sensors, optical switches, and optical modulators.

Arf GTPase activating (ArfGAP) proteins are critical regulatory and effector proteins in membrane trafficking pathways. Budding yeast contain two ArfGAP proteins (Gcs1 and Glo3) implicated in COPI coat function at the Golgi, and yeast require Glo3 catalytic function for viability. A new X-ray crystal structure of the Glo3 GAP domain was determined at 2.1 Å resolution using molecular replacement methods. The structure reveals a Cys4-family zinc finger motif with an invariant residue (R59) positioned to act as an "arginine finger" during catalysis. Comparisons among eukaryotic GAP domains show a key difference between ArfGAP1 and ArfGAP2/3 family members in the final helix located within the domain. Conservation at both the sequence and structural levels suggest the Glo3 GAP domain interacts with yeast Arf1 switch I and II regions to promote catalysis. Together, the structural data presented here provide additional evidence for placing Glo3 near Arf1 triads within membrane-assembled COPI coats and further support the molecular niche model for COPI coat regulation by ArfGAPs.

A four-fold-degenerate three-dimensional (3D) Dirac point, represents a degenerate pair of Weyl points carrying opposite chiralities. Moreover, 3D Dirac crystals have shown many exotic features different from those of Weyl crystals. How these features evolve from 3D Dirac to Weyl crystals is important in research on 3D topological matter. Here, we realized a pair of 3D acoustic Dirac points from band inversion in a hexagonal sonic crystal and observed the surface states and helical interface states connecting the Dirac points. Furthermore, each Dirac point can transition into a pair of Weyl points with the introduction of chiral hopping. The exotic features of the surface states and interface states are inherited by the resulting Weyl crystal. Our work may serve as an ideal platform for exploring exotic physical phenomena in 3D topological semimetals.

Metazoan retromer (VPS26/VPS35/VPS29) associates with sorting nexins on endosomal tubules to sort proteins to the trans-Golgi network or plasma membrane. Mechanisms of metazoan retromer assembly remain undefined. We combine single-particle cryoelectron microscopy with biophysical methods to uncover multiple oligomer structures. 2D class averages reveal mammalian heterotrimers; dimers of trimers; tetramers of trimers; and flat chains. These species are further supported by biophysical solution studies. We provide reconstructions of all species, including key sub-structures (∼5 Å resolution). Local resolution variation suggests that heterotrimers and dimers adopt multiple conformations. Our structures identify a flexible, highly conserved electrostatic dimeric interface formed by VPS35 subunits. We generate structure-based mutants to disrupt this interface in vitro. Equivalent mutations in yeast demonstrate a mild cargo-sorting defect. Our data suggest the metazoan retromer is an adaptable and plastic scaffold that accommodates interactions with different sorting nexins to sort multiple cargoes from endosomes their final destinations.

Weyl points (WPs), as the doubly degenerate points in three-dimensional momentum band structures, carry quantized topological charges and give rise to a variety of extraordinary properties, such as robust surface wave and chiral anomaly. Type-II Weyl semimetals, which have conical dispersions in Fermi surfaces and a strongly tilted dispersion with respect to type I, have recently been proposed in condensed-matter systems and photonics. Although the type-II WPs have been theoretically predicted in acoustics, the experimental realization in phononic crystals has not been reported so far. Here, we experimentally realize a type-II Weyl phononic crystal. We demonstrate the topological transitions observed at the WP frequencies and the topological surface acoustic waves between the Weyl frequencies. The experiment results are in good accordance with our theoretical analyses. Due to the violation of the Lorentz symmetry, the type-II WPs only exist in low energy systems. As the analog counterpart in classical waves, the phononic crystal brings a platform for the research of type-II WPs in macroscopic systems.

Coding acoustic metasurfaces can combine simple logical bits to acquire sophisticated functions in wave control. The acoustic logical bits can achieve a phase difference of exactly π and a perfect match of the amplitudes for the transmitted waves. By programming the coding sequences, acoustic metasurfaces with various functions, including creating peculiar antenna patterns and waves focusing, have been demonstrated.

A novel method is proposed to generate vector beams with arbitrary spatial variation of phase and linear polarization at the nanoscale using compact plasmonic metasurfaces with rectangular nanoapertures. The physical mechanism underlying the simultaneous control of light polarization and phase is explained. Vector beams with different spiral phasefronts are obtained by manipulating the local orientation and geometric parameters of the metasurfaces. In addition, radially and azimuthally polarized vector beams and double-mode vector beams are achieved through completely compensating for the Berry phase, which provides additional degrees of freedom for beam manipulation.
