BACKGROUND: Overexpression of solute carrier family 7 member 8 (SLC7A8) has been shown to relate to the survival time and tumor progression in cancer patients. However, the role of SLC7A8 in lung adenocarcinoma (LUAD) is still obscure. METHOD: The relationships between SLC7A8 expression in LUAD tissues and clinical values as well as immune infiltration were explored through bioinformatics. The functions and pathways of SLC7A8 in LUAD were investigated using Kyoto Encyclopedia of Genes and Genomes enrichment analysis, Gene Set Enrichment Analysis, Western blotting, and other methods. RESULTS: We found that the expression of SLC7A8 was decreased significantly in LUAD tissues compared with normal tissues, which was related to the dismal survival time and disease progression. Moreover, it carried diagnostic value in LUAD and was a risk factor for dismal prognosis. Receiver operating characteristic curve analysis indicated that the expression level of SLC7A8 carried significant diagnostic value in LUAD. Overexpression of SLC7A8 inhibited the proliferation, invasion, and migration of LUAD cells, likely through a mechanism involving the cell cycle. SLC7A8 expression in LUAD was significantly correlated with the infiltration of immune cells, especially B cells, interstitial dendritic cells, mast cells, CD56 bright cells, natural killer cells, plasmacytoid dendritic cells, T follicular helper cells, T helper 2 and 17 cells, and immune factors. CONCLUSION: The downregulation of SLC7A8 was related to a dismal prognosis and immune cell infiltration in LUAD. Increasing the expression of SLC7A8 inhibited the growth and migration of LUAD cells, thereby improving the prognosis of patients.

OBJECTIVE: The roles of MTFR1 in the drug resistance of lung adenocarcinoma (LAC) to cisplatin remain unexplored. In this study, the expression, clinical values and mechanisms of MTFR1 were explored, and the relationship between MTFR1 expression and immune microenvironment was investigated in LAC using bioinformatics analysis, cell experiments, and meta-analysis. METHODS: MTFR1 expression and clinical values, and the relationship between MTFR1 expression and immunity were explored, through bioinformatics analysis. The effects of MTFR1 on the growth, migration and cisplatin sensitivity of LAC cells were identified using cell counting kit-8, wound healing and Transwell experiments. Additionally, the mechanisms of drug resistance of LAC cells involving MTFR1 were investigated using western blotting. RESULTS: MTFR1 was elevated in LAC tissues. MTFR1 overexpression was associated with sex, age, primary therapy outcome, smoking, T stage, unfavourable prognosis and diagnostic value and considered an independent risk factor for an unfavourable prognosis in patients with LAC. MTFR1 co-expressed genes involved in the cell cycle, oocyte meiosis, DNA replication and others. Moreover, interfering with MTFR1 expression inhibited the proliferation, migration and invasion of A549 and A549/DDP cells and promoted cell sensitivity to cisplatin, which was related to the inhibition of p-AKT, p-P38 and p-ERK protein expression. MTFR1 overexpression was associated with stromal, immune and estimate scores along with natural killer cells, pDC, iDC and others in LAC. CONCLUSIONS: MTFR1 overexpression was related to the unfavourable prognosis, diagnostic value and immunity in LAC. MTFR1 also participated in cell growth and migration and promoted the drug resistance of LAC cells to cisplatin via the p-AKT and p-ERK/P38 signalling pathways.

Esophageal cancer (ESCA) is one of the common malignant tumors. The roles and signaling mechanisms of spindle apparatus coiled-coil protein 1 (SPDL1) in ESCA progression have not been reported previously. Therefore, the expression levels and potential clinical roles of SPDL1 were investigated using data from multiple databases and tissue samples of 53 ESCA patients who underwent 18F-FDG positron emission tomography (PET)/computed tomography (CT) before therapy. The signaling mechanisms of SPDL1 involved in ESCA progression were investigated via bioinformatics analysis. The effects of SPDL1 on the growth and migration of ESCA cells were investigated using CCK-8, Edu, and transwell assays. SPDL1 was upregulated in ESCA tissues. Increased SPDL1 expression was associated with age, grade, drinking history, cancer stage, lymph node metastasis, TP53 mutation, and poor prognosis in patients with ESCA. SPDL1 overexpression was significantly correlated with SUVmax, SUVmean, and TLG of PET/CT. SPDL1 silencing inhibited cell proliferation, migration, and invasion. SPDL1 was significantly enriched in cell cycle, spliceosome, DNA replication, and other processes. The hub genes of a constructed protein-protein interaction network included CDK1, BUB1, CCNB1, BUB1B, CCNA2, CDC20, MAD2L1, AURKB, NDC80, and PLK1, which were related to SPDL1 expression. The findings of this study suggest that SPDL1 may serve as a biomarker of ESCA prognosis.

High-performance lithium ion batteries (LIBs) juggling high reversible capacity, excellent rate capability and ultralong cycle stability are urgently needed for all electronic devices. Here we report employing a vesicle-like porous N-doped carbon material (abbr. N/C-900) as a highly active anode for LIBs to balance high capacity, high rate and long life. The N/C-900 material was fabricated by pyrolysis of a designed crystal MOF LCU-104, which exhibits a graceful two-fold interpenetrating structural feature of N-rich nanocages {Zn6(dttz)4} linked through an N-donor ligand bpp (H3dttz = 4,5-di(1H-tetrazol-5-yl)-2H-1,2,3-triazole, bpp = 1,3-bis(4-pyridyl)propane). The features of LCU-104 combine high N content (35.1%), interpenetration, and explosive characteristics, which endow the derived N/C material with optimized N-doping for tuning its chemical and electronic structure, a suitably thicker wall to enhance its stability, and a vesicle-like structure to improve its porosity. As an anode material for LIBs, N/C-900 delivers a highly reversible capacity of ca. 734 mA h g-1 at a large current density of 1 A g-1 until the 2000th cycle, revealing its ultralong cycle stability and excellent rate capability. The unique structure and preferential interaction between abundant pyridinic N active sites and Li atoms are responsible for the improved excellent lithium storage capacity and durability performances of the anode according to analysis of the results of computational modeling.

Reproductive traits are important factors in sheep production. The Booroola fecundity (FecB) gene-the first major gene for prolificacy identified in sheep-has a positive effect on ovulation rates and litter size under natural reproductive conditions. However, the effect of the FecB gene on reproductive performance under assisted reproduction, which uses many artificial hormones, remains unclear. In the present study, we evaluated the effect of FecB (BMPR-1B mutation) on reproductive performance under assisted reproduction, and examined offspring body weight at birth and weaning and survival rate at weaning. There were no differences among three genotype groups (homozygous carrier, BB; heterozygous carrier, B+; non-carrier, ++) in terms of estrus detection rate, time to estrus onset, or estrus duration following estrus synchronization (P > 0.05). The pregnancy rates at 60 d were similar among three genotype groups after artificial insemination (P > 0.05). However, the B allele had an additive effect on litter size (one copy resulted in an increase of 0.88 lambs and two copies produced an additional 0.41 lambs; P < 0.01), and increased lambing and fecundity rates (P < 0.01). After multiple ovulation, the average numbers of recovered embryos per ewe were 9.16 ±â€¯0.79, 8.20 ±â€¯0.77, and 8.44 ±â€¯0.61 in the BB, B+, and ++ ewes, respectively (P > 0.05). There were no differences in the fertilization rate or numbers of grade 1-2 embryos among different groups (P > 0.05). The birth and weaning weights of lambs from BB and B+ ewes were lower than those of lambs born from ++ ewes (P < 0.01) owing to the high fecundity. The survival rate of lambs at weaning did not differ among groups (P > 0.05). Our results indicated that the presence of the B allele had an additive effect on litter size after artificial insemination, but it did not influence the parameters of estrus synchronization and multiple ovulation. Furthermore, the higher prolificacy in ewes carrying the B allele was associated with a reduction in offspring body weight at birth and weaning.

The type 1 insulin-like growth factor receptor (IGF-1R) and its downstream signaling components have been increasingly recognized to drive the development of malignancies, including non-small cell lung cancer (NSCLC). This study aimed to investigate the effects of IGF-1R and its inhibitor, AG1024, on the progression of lung cancer. Tissue microarray and immunohistochemistry were employed to detect the expressions of IGF-1 and IGF-1R in NSCLC tissues (n=198). Western blotting was used to determine the expressions of IGF-1 and phosphorylated IGF-1R (p-IGF-1R) in A549 human lung carcinoma cells, and MTT assay to measure cell proliferation. Additionally, the expressions of IGF-1, p-IGF-1R and IGF-1R in a mouse model of lung cancer were detected by Western blotting and real-time fluorescence quantitative polymerase chain reaction (FQ-PCR), respectively. The results showed that IGF-1 and IGF-1R were overexpressed in NSCLC tissues. The expression levels of IGF-1 and p-IGF-1R were significantly increased in A549 cells treated with IGF-1 as compared to those treated with IGF-1+AG1024 or untreated cells. In the presence of IGF-1, the proliferation of A549 cells was significantly increased. The progression of lung cancer in mice treated with IGF-1 was significantly increased as compared to the group treated with IGF-1+AG1024 or the control group, with the same trend mirrored in IGF-1/p-IGF-1R/IGF-1R at the protein and/or mRNA levels. It was concluded that IGF-1 and IGF inhibitor AG1024 promotes lung cancer progression.

Ancient eggshells over the past 700 years were extracted from an ornithogenic sediment profile on Guangjin Island, South China Sea. Based on SEM and nitrogen isotope analyses, we determined that neither post-depositional processes nor seabirds' dietary changes had a large influence on eggshell Hg levels. The historical change of Hg in these eggshells was reconstructed. Eggshell Hg was a marker for past Hg deposition in marine environment. The eggshell Hg showed three small peaks at around 1300AD, 1600 AD and 1700-1750AD and rapid increase since 1800 AD. Before 1970 AD the Hg deposition in the Xisha area had global distribution characteristics, with increased Hg emissions due to global anthropogenic activities in industrial times. However, after 1970 AD, a further sharp increase up to present day occurred, implying that the Hg production center had gradually shifted from Europe and America to Asia.

PURPOSE: This study was to explore the chroma of a custom shade guide with metal substrate. METHODS: A custom shade guide sample was fabricated using the central maxillary incisor as a model,which was corresponding to the colour of VITAPAN shade guide.The colour of each shade tab was measured with PR-650 spectrophotometer when every porcelain was added. RESULTS: From the opaque to glaze, the lightness was decreased and the chroma was increased distinctively; When the body porcelain were baked over their corresponding opaque, the lightness was decreased and the chroma was increased; When the enamel porcelain and the translucent porcelain were added on, the lightness and chroma were decreased slightly; the glaze could increase the lightness. CONCLUSION: The "add on" porcelain can affect the final color of the restoration, and the effects of each porcelain layer were different from each other.

Technical framework for water environment simulation of contaminations is established based on visualization and a spatial environmental model is built. The main two contaminations, namely NH: -N and TP, are simulated on the platform of MapInfo and Delft3D in the Qiantang River at the low water period, to analyze its space-time diversity. For NH4+ -N, the measured values are 0.19 mg/L and 0.66 mg/L larger than simulated values at the Lanjiang River mouth and the Yanlingwu, 0.16 mg/L, 0.54 mg/L and 0.49 mg/L smaller at the Zhaixi, the Yushan and the Yuanpu. For TP, the measured values are 0.13 mg/L and 0.14 mg/L higher than simulated values at the Meicheng Water facility and Yanlingwu. However, the measure values are slightly lower than simulated ones at Zhaixi, Yushan, Puyang River mouth and Yuanpu, the trend of which accords with actual situation. The results indicate that the contaminations of the Qiantang Reach mostly come from the Lanjiang River, the Fuchun River and the Puyang River on the upstream, among which the Lanjiang River and the Puyang River have a very high concentration of polluted materials, which means bad water quality, and influence the water downstream. The Lanjiang River becomes the chief contaminative source in the Fuchun River. When the discharge from the Xin'an River Dam is small, the recirculation region may be formed and makes part of the Xin'an Reach contaminated. Otherwise, when the discharge is large, the water quality in the Fuchun River is apparently improved. And the Puyang River, which brings the contaminations from the upstream, along with the polluted water let into it from the industries along the reach, has significant impacts on the water quality in Qiantang Reach.

OBJECTIVE: To compare the color character of VITA shade guide and the custom shade guide with metal substrate. METHODS: Color of 9 sites of 5 series of VITA shade guides and custom shade guides with metal substrate were measured by means of PR-650 spectrophotometer. RESULTS: The color range of VITA shade guide were that L* was 56.86-73.86, a* was-1.29-3.69, b* was 7.09-21.94, and the transparence was 3.20-7.59. The color range of the custom shade guide was that L* was 60.59-78.54, a* was -1.09-4.99, b* was 7.60-22.35, and the transparence was 0.23-10.98. L*, a* and b* of the custom shade guide with metal substrate were higher than VITA shade guide, and the transparence was differet. The color difference of homonymy shade tab between custom shade guide and VITA shade guide was larger than 1.5 NBS. CONCLUSION: There is color difference between VITA shade guide and custom shade guide. Metal substrate has effects on the color of cero-metal prosthesis, and color selection in clinic should be carefully considered.
