RESUMEN

Chloroplast capture, a phenomenon that can occur through interspecific hybridization and introgression, is frequently invoked to explain cytonuclear discordance in plants. However, relatively few studies have documented the mechanisms of cytonuclear coevolution and its potential for driving species differentiation and possible functional differences in the context of chloroplast capture. To address this crucial question, we chose the Aquilegia genus, which is known for having minimal sterility among species, and inferred that A. amurensis captured the plastome of A. parviflora based on cytonuclear discordance and gene flow between the two species. We focused on the introgression region and its differentiation from corresponding regions in closely related species, especially its composition in a chloroplast capture scenario. We found that nuclear genes encoding cytonuclear enzyme complexes (CECs; i.e., organelle-targeted genes) of chloroplast donor species were selectively retained and displaced the original CEC genes in chloroplast-receiving species due to cytonuclear interactions during introgression. Notably, the intrinsic correlation of CEC introgression was a greater degree of evolutionary distance for these CECs between A. amurensis and A. parviflora. Terpene synthase activity genes (GO: 0010333) were overrepresented among the introgressed genes, and more than 30% of these genes were CEC genes. These findings support our observations that floral terpene release pattern is similar between A. amurensis and A. parviflora compared with A. japonica. Our study clarifies the mechanisms of cytonuclear coevolution, species differentiation and functional differences in the context of chloroplast capture and highlights the potential role of chloroplast capture in adaptation.

Water ice and other volatiles that accumulated in the Moon's polar regions are among the top priority targets for lunar exploration, due to their significances in both lunar geology and extraterrestrial resource utilization. Locating suitable landing sites and determining the provenance of sampled/measured surface materials are critical for future landed missions. Here, we map over 800 sites of plains terrains in the Moon's south polar region, with a total surface area of ~46,000 km2. Orbital measurements and analog studies show that most of these plains have apparently higher albedo and lower iron content than volcanic mare plains, suggesting an origin of ejecta-induced debris flows from distant impact craters, especially from the Schrödinger basin. Our findings suggest that the entire lunar south polar region probably have experienced contributions from distant basin materials. We recommend these plains as priority landing sites for future exploration of lunar polar volatiles and early bombardment history.

BACKGROUND: Aquilegia is a model system for studying the evolution of adaptive radiation. However, very few studies have been conducted on the Aquilegia mitochondrial genome. Since mitochondria play a key role in plant adaptation to abiotic stress, analyzing the mitochondrial genome may provide a new perspective for understanding adaptive evolution. RESULTS: The Aquilegia amurensis mitochondrial genome was characterized by a circular chromosome and two linear chromosomes, with a total length of 538,736 bp; the genes included 33 protein-coding genes, 24 transfer RNA (tRNA) genes and 3 ribosomal RNA (rRNA) genes. We subsequently conducted a phylogenetic analysis based on single nucleotide polymorphisms (SNPs) in the mitochondrial genomes of 18 Aquilegia species, which were roughly divided into two clades: the European-Asian clade and the North American clade. Moreover, the genes mttB and rpl5 were shown to be positively selected in European-Asian species, and they may help European and Asian species adapt to environmental changes. CONCLUSIONS: In this study, we assembled and annotated the first mitochondrial genome of the adaptive evolution model plant Aquilegia. The subsequent analysis provided us with a basis for further molecular studies on Aquilegia mitochondrial genomes and valuable information on adaptive evolution in Aquilegia.

The chiral pesticide enantiomers often show selective efficacy and non-target toxicity. In this study, the enantioselective degradation characteristics of the chiral organophosphorus insecticide isocarbophos (ICP) by Cupriavidus nantongensis X1T were investigated systematically. Strain X1T preferentially degraded the ICP R isomer (R-ICP) over the S isomer (S-ICP). The degradation rate constant of R-ICP was 42-fold greater than S-ICP, while the former is less bioactive against pest insects but more toxic to humans than the latter. The concentration ratio of S-ICP to R-ICP determines whether S-ICP can be degraded by strain X1T. S-ICP started to degrade only when the ratio (CS-ICP/CR-ICP) was greater than 62. Divalent metal cations could improve the degradation ability of strain X1T. The detected metabolites that were identified suggested a novel hydrolysis pathway, while the hydrolytic metabolites were less toxic to fish and green algae than those from P-O bond breakage. The crude enzyme degraded both R-ICP and S-ICP in a similar rate, indicating that enantioselective degradation was due to the transportation of strain X1T. The strain X1T also enantioselectively degraded the chiral organophosphorus insecticides isofenphos-methyl and profenofos. The enantioselective degradation characteristics of strain X1T make it suitable for remediation of chiral organophosphorus insecticide contaminated soil and water.

Chlorpyrifos is an important organophosphorus insecticide. It is highly toxic to mammals and can pollute the environment. Cupriavidus nantongensis X1T can efficiently degrade chlorpyrifos. Immobilization technology can also improve the viability, stability and catalytic ability of bacteria. In this study, strain X1T was, therefore, captured on various composite immobilized carriers, sodium alginate (SA), diatomite (KLG), chitosan (CTS) and polyvinyl alcohol (PVA). The four types of immobilized beads (SA, SA + KLG, SA + CTS and SA + PVA) could form a slice and honeycomb structure to capture strain X1T. The results showed that SA + CTS (SC) was an optimal material combination for the immobilization of strain X1T to degrade chlorpyrifos. Compared with SA-X1T, after adding CTS, the specific surface area and adsorption capacity for chlorpyrifos were increased 3.4 and 1.7 fold, respectively. SC-X1T could degrade 96.6% of chlorpyrifos at 20 mg/L within 24 h and the degradation rate constant was 4.8 fold greater than immobilized strain LLBD2, a well-studied chlorpyrifos-degrading strain. The immobilized beads SC-X1T also showed a more stable and greater degradation ability than X1T free cells for chlorpyrifos in industrial wastewater. The synergy of adsorption and degradation of immobilized strain X1T is suitable for in-situ remediation of chlorpyrifos contaminated environment.

The unequal distribution of volcanic products between the Earth-facing lunar side and the farside is the result of a complex thermal history. To help unravel the dichotomy, for the first time a lunar landing mission (Chang'e-4, CE-4) has targeted the Moon's farside landing on the floor of Von Kármán crater (VK) inside the South Pole-Aitken (SPA). We present the first deep subsurface stratigraphic structure based on data collected by the ground-penetrating radar (GPR) onboard the Yutu-2 rover during the initial nine months exploration phase. The radargram reveals several strata interfaces beneath the surveying path: buried ejecta is overlaid by at least four layers of distinct lava flows that probably occurred during the Imbrium Epoch, with thicknesses ranging from 12 m up to about 100 m, providing direct evidence of multiple lava-infilling events that occurred within the VK crater. The average loss tangent of mare basalts is estimated at 0.0040-0.0061.

Organophosphorus insecticides account for approximately 28% of the global commercial insecticide market, while 40% of them are chiral enantiomers. Chiral enantiomers differ largely in their toxicities. Enantiomers that are less active or inactive do not offer the needed efficacy but pollute the environment and cause toxicities to non-target species. Cupriavidus nantongensis X1T, a recently isolated bacterial strain, could degrade S-profenofos 2.3-fold faster than R-profenofos, while the latter is the active enantiomer potently against pest insects and has greater mammalian safety. The degradation enzyme encoded by opdB was expressed via Escherichia coli and purified. The degradation kinetics of R- and S-profenofos showed that both the purified OpdB and crude enzyme extracts had no enantiomer degradation selectivity, which strongly indicated that the degradation selectivity occurred in the uptake process. Metabolite analyses suggested a novel dealkylation pathway. This is the first report of bacterial selective uptake of organophosphates. Selective degradation of S-profenofos over R-profenofos by the strain X1T suggests a concept of co-application of racemic pesticides and degradation-selective bacteria to minimize contamination and non-target toxicity problems.
