RESUMEN

To investigate the effect analysis and preventive effects of nucleic acid testing combined with enzyme-linked immunosorbent assay (ELISA) on blood-borne diseases. This study included 72335 blood samples that were collected in our hospital from March 2019 to March 2020. All the samples were tested for anti HIV (AIDS antibody), anti HCV (hepatitis C antibody), HBsAg (hepatitis B surface antigen) and anti TP (syphilis antibody) in blood respectively with two manufacturers' reagents. The results of anti HIV, anti HCV, HBsAg and anti TP of all samples were analyzed, and blood samples with 0.7< sample test value / critical value (s / CO) < 3.0 were tested by ELISA and negative blood samples were tested by ELISA and nucleic acid testing. Then we analyzed the results of nucleic acid testing. 610 blood samples failed to pass the test of anti HIV, anti HCV, HBsAg and anti TP ELISA, accounting for 0.84% of the total numbers, including 100 blood samples with 1.0 3.0, 338 blood samples with 0.7< s/CO <1.0 and 71725 blood samples were qualified. We used nucleic acid testing to test 71725 qualified samples tested by ELISA and then there were 50 samples with positive HBV-DNA , accounting for 0.07% (50 / 71725), no one with positive HIV-RNA and positive HCV-RNA, accounting for 0.00% (0/71725). In this study, we describe the development and evaluation of a novel multiple-antigen ELISA for rapid diagnosis and screening of active tuberculosis (TB). The humoral immune responses of 136 active TB patients and 57 healthy subjects against antigens Rv3425, 38kDa and lipoarabinomannan (LAM) from Mycobacterium tuberculosis H37Rv were examined by ELISA. Three essential results were obtained. (i) Rv3425 antigen is a potential candidate for serodiagnosis of active TB. Of 136 active TB patients, Rv3425 antigen provided a sensitivity of 31.6%, lower than that of LAM antigen, but higher than that of 38kDa antigen, with an overall specificity of 100%. (ii) For 62 smear-negative pulmonary TB patients and 15 extra-pulmonary TB patients, the multiple-antigen test provided a sensitivity of 43.5% and 26.7%, respectively, representing an improvement over acid-fast bacilli (AFB) smear-based diagnosis. (iii) Compared with the single-antigen ELISA and the two available commercial kits, the multiple-antigen test offered the highest accuracy (71.0%). In conclusion, the multiple-antigen ELSIA test based on Rv3425, 38kDa, and LAM antigens is a potentially useful tool for the serodiagnosis and screening of active TB. Combinations of Rv3425 with other mycobacterial antigens may also be worthy of further investigation.

OBJECTIVE: To construct protein fingerprints in serum and to early diagnose chronic allograft nephropathy (CAN) using surface enhanced laser ionization/desorption-time of flight-mass spectrometry (SELDI-TOF-MS). METHODS: Serum protein spectra were detected by SELDI-TOF-MS and weak cation exchange (CM10), which were from long-term survival patients with well-functioning kidney allograft (LS) (n = 24) and CAN (n = 15), and then biomarkers were screened through the analysis of database with Biomarker Wizard and Biomarker Pattern softwares. RESULTS: 78 protein peaks of interest were generated in each sample, 18 protein spectra statistically show difference between LS and CAN (P < 0.05), 6 of which (m/z 2476.0, 3078.7, 3190.5, 4076.5, 4506.0, 6178.4) statistically show significant difference (P < 0.01), a single biomarker (m/z 3078.7) differentiated LTS and CAN (P = 0.0001), it was upregulated in the former and downregulated in the latter, its diagnostic sensitivity and specificity were 87.5% and 81.8% respectively. Its cost-effective value is 0.307. CONCLUSION: Establishment of protein fingerprint signature in serum by SELDI-TOF-MS is clinically predictive of CAN.
