RESUMEN

An obligately anaerobic, Gram-positive, rod-shaped bacterium (1.8-5.5 µm long, 0.6-0.9 µm wide), designated ZC22-4T, was isolated from a pickle-processing wastewater treatment plant in Zhejiang province, P.R. China. Strain ZC22-4T grows optimally at 37-40 °C and pH 7.0 in the presence of 1% (w/v) NaCl or 2.0% (w/v) sea salts. It contained C16:0 (25.9%), C14:0 (13.6%), and C16:1 cis 9 (10.6%) as the dominant cellular fatty acid (> 10%). Polar lipids include phosphatidylglycerol (PG), diphosphatidylglycerol (DPG), phosphatidylethanolamine (PE), one unidentified phospholipid (PL), two unidentified glycolipids (GL), three unidentified amino phosphoglycolipids (APGL1-3), one unidentified aminoglycolipid (AGL), and one unidentified lipid (L). The genomic DNA G + C content of ZC22-4T was 28.7%. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain ZC22-4T belonged to the genus Clostridium and formed a clade with the most closely related Clostridium aestuarii HY-45-18T (96.3%), Clostridium ganghwense HY-42-06T (95.9%). The average nucleotide identity and DNA-DNA hybridization values among the genomes of strain ZC22-4T and C. aestuarii HY-45-18T and C. ganghwense HY-42-06T were 75.7% and 77.3%, 21.7% and 23.0%, respectively. Based on the phenotypic, phylogenetic, and genetic data, strain ZC22-4T represents a novel species in the Clostridium cluster I, for which the name Clostridium brassicae sp. nov. is proposed. The type strain is ZC22-4T (= MCCC 1K07510T = JCM 35370T).

Although most children conceived by assisted reproductive technology (ART) are healthy, there are concerns regarding the potential long-term health implications of ART. It has been reported that alterations in insulin-induced gene (INSIG), sterol regulatory element binding protein (SREBP), and SREBP cleavage-activating protein (SCAP) are involved in cardiometabolic changes. Thus, ART mouse models were established via in vitro fertilization (IVF), intracytoplasmic injection (ICSI), and in vitro oocyte maturation (IVM). A significantly higher systolic blood pressure was identified in the IVM aged female mice. In addition, abnormalities in the blood lipids and liver function were identified in the IVM- or ICSI-conceived elderly mice. Furthermore, ICSI or IVM significantly affected the hepatic expression and methylation of INSIG-SCAP-SREBP from a young to old age. Our animal data indicated that ICSI or IVM result in a higher risk of cholesterol metabolism dysfunction in older mice, which may be associated with long-term alterations of INSIG-SCAP-SREBP.

RESUMEN

PURPOSE: Recent clinical trials showed that expression of excision repair cross-complementation group 1 (ERCC1), ribonucleotide reductase M1 (RRM1), and thymidylate synthase (TS) proteins was able to predict the effects of non-small cell lung cancer (NSCLC) to chemotherapy. However, it remains unknown whether the adjuvant chemotherapy based on expression of the three proteins has survival significance in Chinese NSCLC patients. METHODS: We investigated 128 Chinese patients receiving chemotherapy after tumor resection for expression of these proteins using immunohistochemistry. Based on protein expression, patients were assigned to two groups for different adjuvant chemotherapy regimes. The disease-free survival (DFS) data were collected and analyzed using Kaplan-Meier curves and Cox models. RESULTS: We found that DFS of these patients with carboplatin and a third-generation agent (gemcitabine or pemetrexed) stratified by protein expression showed no statistical difference between individual treatment versus non-individuation treatment analyzed using Kaplan-Meier method (P = 0.143, median 23.9 vs. 30.8 months). Furthermore, the multivariate analysis showed that histology and tumor stages were independent predictors for DFS in these patients. CONCLUSIONS: The results suggest that chemotherapy based on ERCC1, RRM1, and TS expression did not have significant impact on DFS of patients with resection of NSCLC.

AIMS: Indoleamine 2,3-dioxygenase (IDO) inhibits T-cell proliferation by catalyzing the conversion of l-tryptophan to l-kynurenine. IDO-induced immune tolerance weakens the clinical outcomes of immunotherapies. Sodium butyrate (NaB), one of the histone deacetylase inhibitors (HDACIs), has potential anti-tumor effects. Our previous studies revealed that NaB could inhibit IFN-γ induced IDO expression in nasopharyngeal carcinoma cells, CNE2. In the present study, we aim to investigate to the mechanism of NaB interfering with the interferon-gamma (IFN-γ)-mediated IDO expression signaling transduction. MAIN METHODS: IDO expression and STAT1 phosphorylation in CNE2 cells were analyzed by western blotting and STAT1 acetylation was evaluated by immunoprecipitation. STAT1 nuclear translocation and NF-κB activity were detected by transient transfection and reporter gene assay. KEY FINDINGS: We found that NaB inhibited IFN-γ-induced IDO expression in CNE2 cells via decreasing phosphorylation and nuclear translocation of STAT1, but not via down-regulation of IFN-γ-receptor (IFNGR). Immunoprecipitation assays revealed that NaB increased STAT1 acetylation. Furthermore, NaB elevated the activity of NF-κB in CNE2 cells, and blocking the NF-κB activity had no effect on the IFN-γ-induced IDO expression. SIGNIFICANCE: These results suggest that NaB inhibited IFN-γ-induced IDO expression via STAT1 increased acetylation, decreased phosphorylation, and reduced nuclear translocation. These provided new evidence for the anti-tumor action of NaB and potential drug targets to reduce the IDO-induced immune tolerance.

OBJECTIVE: To assess the correlation between expression of proliferation antigen Ki-67, early apoptotic protein M30 (M30CytoDEATH, CK18) and biologic characteristics in endometrial carcinoma. METHODS: SP immunohistochemical technique was used to detect the expression of Ki-67 and M30 in 79 cases of endometrial carcinoma respectively. RESULTS: The mean Ki-67 indices varied with histological grading and clinical stage of the tumor, being 20.48 +/- 14.86 in grade 1, 24.12 +/- 14.42 in grade 2 and 38.84 +/- 11.88 in grade 3; 20.65 +/- 13.56 in stage I; 26.92 +/- 14.71 in stage II; and: 35.14 +/- 14.70 in stage III. The mean M30 indices varied with the grading and stage of the tumor, being 1.03 +/- 1.42 in grade 1, 1.03 +/- 1.64 in grade 2 and 1.94 +/- 1.20 in grade 3; 0.30 +/- 0.58 in stage I; 1.66 +/- 1.74 in stage II; and 2.07 +/- 1.62 in stage III. CONCLUSIONS: Ki-67 and M30 indexes are significantly correlated with biologic behavior and prognosis in endometrial carcinoma. There is a positive relationship between Ki-67 and M30 indexes.
