RESUMEN

Meiotic resumption in mammalian oocytes involves nucleus and organelle structural changes, notably chromatin configuration transitioning from non-surrounding nucleolus (NSN) to surrounding nucleolus (SN) in germinal vesicle (GV) oocytes. Our study found that nuclear speckles, a subnuclear structure mainly composed of serine-arginine (SR) proteins, changed from a diffuse spotted distribution in mouse NSN oocytes to an aggregation pattern in SN oocytes. We further discovered that SRPK1, an enzyme phosphorylating SR proteins, co-localized with NS at SN stage and NSN oocytes failed to convert into SN oocytes after inhibiting the activity of SRPK1. Furthermore, the typical structure of chromatin ring around the nucleolus in SN oocytes collapsed after inhibitor treatment. To explore the underlying mechanism, phosphorylated SR proteins were confirmed to be associated with chromatin by salt extraction experiment, and in situ DNase I assay showed that the accessibility of chromatin enhanced in SN oocytes with SRPK1 inhibited, accompanied by decreased repressive modification on histone and abnormal recurrence of transcriptional signal. In conclusion, our results indicated that SRPK1-regulated phosphorylation on SR proteins was involved in the NSN to SN transition and played an important role in maintaining the condensation nucleus of SN oocytes via interacting with chromatin.

Serving as a vital linkage between surface water and groundwater, the hyporheic zone (HZ) plays a fundamental role in improving water quality and maintaining ecological security. In arid or semi-arid areas, effluent discharge from wastewater treatment facilities could occupy a predominant proportion of the total base flow of receiving rivers. Nonetheless the relationship between microbial activity, abundance and environmental factors in the HZ of effluent-receiving rivers appear to be rarely addressed. In this study, a spatiotemporal field study was performed in two representative effluent-dominated receiving rivers in Xi'an, China. Land use data, physical and chemical water quality parameters of surface and subsurface water were used as predictive variables, while the microbial respiratory electron transport system activity (ETSA), the Chao1 and Shannon index of total microbial community, as well as the Chao1 and Shannon index of denitrifying bacteria community were used as response variables, while ETSA was used as response variables indicating ecological processes and Shannon and Chao1 were utilized as parameters indicating microbial diversity. Two machine learning models were utilized to provide evidence-based information on how environmental factors interact and drive microbial activity and abundance in the HZ at variable depths. The models with Chao1 and Shannon as response variables exhibited excellent predictive performances (R2: 0.754-0.81 and 0.783-0.839). Dissolved organic nitrogen (DON) was the most important factor affecting the microbial functions, and an obvious threshold value of ∼2 mg/L was observed. Credible predictions of models with Chao1 and Shannon index of denitrifying bacteria community as response variables were detected (R2: 0.484-0.624 and 0.567-0.638), with soluble reactive phosphorus (SRP) being the key influencing factor. Fe (Ⅱ) was favorable in predicting denitrifying bacteria community. The ESTA model highlighted the importance of total nitrogen in the ecological health monitoring in HZ. These findings provide novel insights in predicting microbial activity and abundance in highly-impacted areas such as the HZ of effluent-dominated receiving rivers.

BACKGROUND: Anthocyanins are the most important compounds for nutritional quality and economic values of blood orange. However, there are few reports on the pre-harvest treatment accelerating the accumulation of anthocyanins in postharvest blood orange fruit. Here, we performed a comparative transcriptome and metabolomics analysis to elucidate the underlying mechanism involved in seasonal drought (SD) treatment during the fruit expansion stage on anthocyanin accumulation in postharvest 'Tarocco' blood orange fruit. RESULTS: Our results showed that SD treatment slowed down the fruit enlargement and increased the sugar accumulation during the fruit development and maturation period. Obviously, under SD treatment, the accumulation of anthocyanin in blood orange fruit during postharvest storage was significantly accelerated and markedly higher than that in CK. Meanwhile, the total flavonoids and phenols content and antioxidant activity in SD treatment fruits were also sensibly increased during postharvest storage. Based on metabolome analysis, we found that substrates required for anthocyanin biosynthesis, such as amino acids and their derivatives, and phenolic acids, had significantly accumulated and were higher in SD treated mature fruits compared with that of CK. Furthermore, according to the results of the transcriptome data and weighted gene coexpression correlation network analysis (WGCNA) analysis, phenylalanine ammonia-lyase (PAL3) was considered a key structural gene. The qRT-PCR analysis verified that the PAL3 was highly expressed in SD treated postharvest stored fruits, and was significantly positively correlated with the anthocyanin content. Moreover, we found that other structural genes in the anthocyanin biosynthesis pathway were also upregulated under SD treatment, as evidenced by transcriptome data and qRT-PCR analysis. CONCLUSIONS: The findings suggest that SD treatment promotes the accumulation of substrates necessary for anthocyanin biosynthesis during the fruit ripening process, and activates the expression of anthocyanin biosynthesis pathway genes during the postharvest storage period. This is especially true for PAL3, which co-contributed to the rapid accumulation of anthocyanin. The present study provides a theoretical basis for the postharvest quality control and water-saving utilization of blood orange fruit.

BACKGROUND: Adverse childhood experiences (ACEs) are one of the causes of depression in middle-aged and older adults, but the combined effects of ACEs, cognitive function and ability to perform activities of daily living (ADL) on depression have not been fully explored. METHODS: This study was based on data from 4 waves (2013, 2014, 2015 and 2018) of data from the China Health and Retirement Longitudinal Study, including 10,995 middle-aged and older adults. ACEs were derived from the 2014 self-report life history module. A latent variable growth curve model was used to assess the mediating effect of cognitive function and ability to perform ADL in the relationship between ACEs and depression. RESULTS: ACEs were significantly associated with lower initial cognitive status (ß = -0.156, P < .001), worse ability to perform ADL (ß = 0.051, P < .001) and higher severity of depression (ß = 0.228, P < .001). The results of mediation analysis indicated that the association between ACEs and the intercept of depression was partly mediated by the initial level of cognitive function and ADL, and the association between ACEs and the slope of depression was total mediated by cognitive (intercept and slope) and ADL (intercept and slope). CONCLUSIONS: ACEs were associated with higher severity of depression in part due to lower cognitive function and worse ability to perform ADL. Interventions that focus on reducing ACEs and improving cognitive level and ability to perform ADL may effectively reduce the incidence of depression among middle-aged and older individuals.

Aluminum alloy adhesive bonding joint widely appears in many industrial products. Improving the mechanical performances of aluminum alloy bonding joints has been attracting much effort. To acquire more excellent bonding strength, this paper focused on the effects of different surface treatments, including laser ablation and milling superposed by phosphoric acid anodizing (PAA). The treated surfaces were characterized by roughness and contact angle, and the effects of the geometric parameters of microstructures on wettability, failure mode, and shear strength were examined. The results indicate that those surfaces where the spacing is smaller than the diameter present a hydrophilic property and the corresponding specimens are mainly subject to cohesive failure, and vice versa. Additionally, laser ablation with a properly designed dimple pattern can greatly improve the bonding strength, and the maximum average shear strength of specimens with a thickness of 50 µm reaches 32.82 MPa, which is an increase of 28.15% compared with the original milling specimen. Moreover, fabricating groove or grid patterns on the surfaces and applying PAA treatment can also significantly enhance the bonding strength, reaching up to 36.28 MPa.

To further enrich the side-filling structure system of goaf-retaining roadways and explore the compression reaction mechanism of the composite in the support environment of underground mine roadways, this paper introduces a double-wall hollow composite pier structure (FPRSC structure) that is composed of the fiber-reinforced polymer (FRP) composite and polyvinyl chloride plastic (PVC) as restraint materials and the infill material featured with a high water-to-powder ratio. A total of 16 circular specimens with a diameter and height of 100 mm were tested to explore the axial performance of the combined support structure. The main control variables in the present research included the water-to-cement ratio of the high-water material (e.g., 2:1, 3:1, and 4:1), the thickness of the FRP pipe (i.e., 6 mm and 3 mm), the inner diameter of the PVC pipe (i.e., 29 mm and 22 mm), as well as the thickness of the PVC pipe (1.5 mm and 5 mm). Test results showed that the high-water material was under triaxial stress due to the double-wall tube binding, and the bearing capacity of the composite was higher than that of the single material. Meanwhile, the FPRSC structure exhibited obvious strain-hardening characteristics when the infill material is under the combined constraints of double-wall hollow tubes. Moreover, the ratio of PVC-c, FRP-A, and high-water material with a water-cement ratio of 3:1 shows the best axial mechanical properties. The new composite pier structure with high toughness and strength has wide application prospects in the field of goaf retention in deep underground mines.

In the context of increasing aridity due to climate changes, effluent from wastewater treatment plants (WWTPs) became dominant in some rivers. However, the prevalence of antibiotic resistance genes (ARGs) and virulence factors (VFs) in effluent-dominated rivers was rarely investigated. In this study, the profiles of ARGs and VFs in the sediment of two effluent-dominated rivers were revealed through the metagenomic sequencing technique. In each river, samples from the effluent discharge point (P site) and approximately 500 m downstream (D site) were collected. Results showed that the abundances of ARGs and VFs were both higher in D sites than those in P sites, indicating higher risks in the downstream areas. The compositions of ARGs were similar in the P sites of two rivers while being distinct in the D sites. The same was true for changes in the VFs compositions. Microbial community structure variations were the main driver for the changes in ARGs and VFs. Network analysis revealed that the interaction of ARGs and VF genes (VFGs) in sediment was intense. Two VFGs and eleven ARGs were identified to play important roles in the network. Metagenome-assembled genomes (MAGs) were generated to evaluate the coexistence of ARGs and VFGs at the single genome level. It was found that 38.4 % of the MAGs contained both ARGs and VFGs, and two MAGs were from pathogenic genera. These results suggested that high microbiological risks existed in effluent-dominated rivers, and necessary measures should be taken to prevent the potential threat to public health.

The China Society of Explosives and Blasting required a larger than 20% annual increase in the national use of digital electronic detonators since 2018. So, this article conducted a large number of on-site tests and then used the Hilbert-Huang Transform method to analyze and compare the vibration signals of digital electronic and nonel detonators during the excavation process of minor cross-sectional rock roadways from the perspective of time, frequency, and energy. Then, through vibration energy analysis, identification of actual delay time, and formula derivation, it was proved that the delay time error of the detonator can control vibration wave random interference and reduce vibration. The analysis results showed that when using a segmented simultaneous blasting network for excavation in small-sectioned rock tunnels, nonel detonators may provide more excellent protection to structures than digital electronic detonators. In the same segment, the timing error of nonel detonators produces a vibration wave with a random superposition damping effect, resulting in an average vibration reduction of 19.4% per segment compared to digital electronic detonators. However, digital electronic detonators are superior to nonel detonators for the fragmentation effect on rock. The research conducted in this paper has the potential to facilitate a more rational and comprehensive promotion of digital electronic detonators in China.

Background: Although a significant amount of literature has examined the association between childhood adversity and adverse health outcomes, which may be affected by sleep duration and life satisfaction. However, this relationship has not been researched in the Chinese population. This study aimed to assess the association between childhood adversity and health outcomes, with sleep duration and life satisfaction as mediators. Methods: A total of 14,693 subjects aged 45 and over from the 2018 China Health and Retirement Longitudinal Study (CHARLS) were included. Taking childhood adversity as the independent variable, the health level of middle-aged and older individuals as the dependent variable, and sleep time and satisfaction as the mediating factors, Mplus 8.0 software was used to establish a structural equation model (SEM) to analyze the link between childhood adversity and health level and to explore the mediating effect of target mediators between childhood adversity and health level. Results: In this study, childhood adversity was positively associated with depression symptoms, activities of daily living (ADL), and the number of chronic diseases (r = 0.116, 0.026 and 0.050, respectively, P < 0.001). Associations between adverse childhood experiences (ACEs) and depressive symptoms, ADL, and number of chronic diseases were mediated by sleep duration and life satisfaction, respectively. Conclusion: Adversity experienced in childhood can affect the health status of middle-aged and older people. By ensuring adequate sleep and improving life satisfaction, health outcomes can be improved, and the negative effects of childhood adversity can be reduced.

Serum exosomal microRNAs (miRNAs) are potential biomarkers for tumor diagnosis. Clinically, reverse transcription-quantitative polymerase chain reaction (RT-qPCR) can be used to determine the expression of exosomal miRNAs in the serum of breast cancer patients. The prerequisites for obtaining meaningful serum exosomal miRNA data of breast cancer patients include a suitable extraction method for exosomes and RT-qPCR data standardized by internal reference genes. However, the appropriate methods for the extraction of exosomes and the applicability of reference genes for analyzing exosomal miRNAs in breast cancer patients remain to be studied. This study compared the effects of three exosome extraction methods as well as the expression of exosomal miRNA in different initial serum amounts and at different serum states to identify the selection of the best method for serum exosome extraction. Five candidate reference genes including miR-16, miR-484, miR-1228, miR-191 and miR-423 for standardizing serum exosomal miRNAs were screened using five algorithms and were used for the quantification of serum exosomal miR-940. Significant downregulation of serum exosomal miR-940 expression in breast cancer was detected using miR-191 and miR-1228, whereas no significant down or up regulation was observed with miR-484, miR-423 and miR-16. Previous studies have shown that the expression level of miR-940 is downregulated in breast cancer tissues. The absolute quantitative results showed that miR-940 was significantly downregulated in breast cancer serum exosomes, which was consistent with the results from the analysis using miR-191 or miR-1228 as reference genes. Therefore, miR-191 and miR-1228 could serve as reference genes for the relative quantification of serum exosomal miRNAs. This finding indicated the importance of rigorously evaluating the stability of reference genes and standardization for serum exosomal miRNA expression. Moreover, the level of serum exosomal miR-940 in breast cancer could reflect the presence of lymph node metastasis and the status of HER2/neu, which indicates its potential as a biomarker for breast cancer metastasis. In summary, an optimized protocol for the detection of serum exosomal miR-940 as a breast cancer marker was preliminarily established.

Freshwater biofilms play an important role in the migration and transformation of organic pollutants, especially under illumination conditions. Nonetheless, the roles of variable fractions in freshwater biofilms, e.g., extracellular polymeric substances (EPS), microbial cells and original biofilms, in promoting the photodegradation of trace organic pollutants remain largely unclear. In this study, two contaminants, i.e., methyl orange (MO) and bisphenol A (BPA), were selected, and the roles of different fractions in freshwater biofilms in their photodegradation performances were investigated. After dosing 696 mg/L SS biofilm harvested from an effluent-receiving river, the direct photodegradation rate of MO and BPA was increased 8.7 times and 5.6 times, respectively. River biofilm EPS contained more aromatic fractions, chromogenic groups and conjugated structures than biofilm harvested from a less eutrophic pond, which might be responsible for the enhanced photodegradation process. The quenching experiments suggested that when EPS fractions derived from river biofilm were dosed, 3EPS* was the major reactive oxygen species during the photodegradation of MO and BPA. Meanwhile, for EPS derived from the pond biofilm, ·OH/1O2 was predominantly responsible for the enhanced photodegradation. Batch experimental results suggested that the cells and EPS in river biofilms could collaboratively interact with each other to enhance the preservation of reactive species and protection of microbes, thus facilitating the photoactivity of biofilms. Our results might suggest that biofilms generated from eutrophic waterbodies, such as effluent-receiving rivers, could play a more important role in the photodegradation processes of contaminants.

A novel composite biochar (NCB) was produced from the pyrolysis of co-fermentation products of sewage sludge, food wastes and rice straw, and exhibited higher nitrogen and phosphorus adsorption capacity due to the larger surface area (14.7 m2 g-1) and higher Ca content (51753.7 mg kg-1) than single rice straw biochar. The addition of NCB was then investigated in a non-aerated vertical baffled flow constructed wetland (VBFCW) for contaminated water treatment. The VBFCW with NCB addition significantly improved CODMn, NH4+-N, TN and TP removal efficiencies of 83.3 ±â€¯5.3%, 95.9 ±â€¯3.4%, 28.0 ±â€¯4.0% and 59.5 ±â€¯11.8%, respectively, at a hydraulic retention time (HRT) of 3 d. In addition, the TN and TP removal rates at a decreased HRT of 2 d were much higher than those at an HRT of 3 d without NCB addition. The presence of NCB in the VBFCW system enhanced nutrient adsorption and improved the enrichment of bacteria for organic and nitrogen removal mainly including the genera Bacillus and Lactococcus.
