RESUMEN

Acute kidney injury is a common problem in dogs and is associated with significant morbidity and mortality. So, the present study aimed to evaluate symmetric dimethylarginine (SDMA) and Doppler ultrasonography including resistive index (RI) in the diagnosis of acute kidney injury in dogs. Ten healthy mongrel dogs were injected with gentamicin sulfate 10% at the dose of 30 mg/kg body weight daily for 10 days for induction of acute kidney injury. Clinical, biochemical, ultrasonographic, and Doppler ultrasonographic examinations and urinalysis were performed for all dogs on 0 day before induction, on the 5th day, and on the 10th day of induction. The results of the current study showed significant increase in plasma level of SDMA, serum urea, creatinine, phosphorus, and potassium and a significant decrease in serum sodium, calcium, and chloride on the 5th day and 10th day of induction, and there was an increase in renal cortical echogenicity of the right and left kidney compared to adjacent liver and spleen, respectively. RI value showed a significant increase on the 5th day and 10th day of induction. The present study showed that SDMA is a sensitive and promising biomarker for diagnosis of acute kidney injury in dogs compared to routine biomarkers; also, the RI of Doppler ultrasonography is useful for early identifying acute kidney injury when the only observable change is an increase in cortical echogenicity.

Because of the increasingly emerging praziquantel resistance, there is a crucial need to develop new anti-schistosomal agents. This work was conducted to assess the therapeutic efficacy of a new benzimidazole compound (BTP-OH) in mice experimentally infected with Schistosoma mansoni. A total of 40 Swiss albino female mice were divided into an infected untreated group and three infected treated groups (using praziquantel and BTP-OH). The compound activity was evaluated through parasitological, histopathological and scanning electron microscopy studies. Praziquantel and BTP-OH at both doses significantly reduced male (75%, 42.67% and 61.08%, respectively), female (71.45%, 48.94% and 68.13%, respectively) and total worm burden (75.21%, 42.42% and 62.28%, respectively), as well as tissue egg load in the liver (71.22%, 42.12% and 66.04%, respectively). In oogram, praziquantel significantly increased the percentage of dead eggs (65.89%), while BTP-OH significantly reduced the percentage of immature eggs (30.43% and 19.64%). BTP-OH significantly diminished granuloma count (33.87% and 44.77%) and diameter (39.23% and 49.40%), and caused ultrastructural changes in the tegument of adult schistosomes. This study provides evidence for the schistosomicidal efficacy of BTP-OH. However, future studies are needed to elucidate the full mechanisms of action and effects of BTP-OH on other human schistosomes.

Schistosomiasis represents a public health problem and praziquantel is the only drug used for treatment of all forms of the disease. Thus, the development of new anti-schistosomal agents is of utmost importance to increase the effectiveness, reduce side effects and delay the emergence of resistance. The present study was conducted to report the therapeutic efficacy of PPQ-8, a new synthetic quinoline-based compound against Schistosoma mansoni. Mice were treated with PPQ-8 at day 49 post infection using two treatment regimens (20 and 40 mg/kg). Significant reductions were recorded in hepatic (62.9% and 83.6%) and intestinal tissue egg load (57.4% and 73.5%), granuloma count (75.4% and 89.1%) and diameter (26.2% and 47.3%), in response to the drug regimens, respectively. In addition, both treatment regimens induced significant decrease in liver (23.3% and 32.8%) and spleen (37.5% and 45.3%) indices. Also, there were significant reductions in mature ova, total worm and female count, which were more prominent with the higher dose. The reduction in the level of nitric oxide in the liver by both therapeutic regimens to 22.5% and 47.2% indicates the anti-oxidant activity of PPQ-8. Bright field microscopic examination of worms recovered from infected and PPQ-8-treated mice showed nearly empty intestinal caeca with no observable changes in the tegument. Our findings hold promise for the development of a novel anti-schistosomal drug using PPQ-8, but further in vitro and in vivo studies are needed to elucidate the possible mechanism/s of action and to study the effect of PPQ-8 on other human schistosomes.

OBJECTIVE: To report 3 patients who presented with effort-induced thrombosis of the upper limbs. CASE PRESENTATIONS AND INTERVENTION: The 1st patient presented with a 2-week history of fever, shortness of breath, and increasing swelling of the neck after strenuous manual work. The 2nd patient presented with a 2-day history of pain, swelling, and discolouration of the right upper limb following a session of intense weight lifting. The 3rd patient presented with a 2-day history of swelling, pain, and discolouration of the left upper limb due to repeated rotatory movements of the arm at work. In all 3 cases, the diagnosis of effort-induced thrombosis of subclavian and axillary veins was made. In the 1st case, the lesion was bilateral and complicated by superior vena cava obstruction. In the 2nd and in the 3rd case, right and left subclavian veins were thrombosed, respectively. After consulting the vascular team, anticoagulation therapy was initiated in all 3 cases. The patients described showed gradual recovery with eventual recanalization of the thrombosed veins on follow-up. CONCLUSIONS: These cases indicate the need to consider thrombosis of the subclavian vein as a part of the differential diagnosis in a patient with a history of strenuous manual work. Therapeutic options include immediate anticoagulation or thrombolysis, while vascular surgery should remain an option for cases with underlying anatomical anomalies.

RESUMEN

Cryptosporidium paruum has become one of the most important contaminant found in drinking water and is associated with high risk of waterborne diseases particularly for the immunocompromised. In the present study, Cryptosporidium was found in uncovered water tanks, nearby the shore in El Noubareya and El Mahmoudeya canals and in one swimming pool. Direct flourescence monoclonal antibody was more sensitive in detecting the organism. Ziehl-Neelsen although less sensitive, yet it has the advantage that it could recognize other protozoa causing diarrhoea as G. lamblia, B hominis and Cyclospora sp.

RESUMEN

Three Leishmania strains of the old world were the subject of this study. Both the amastigote and promastigote stages were subjected to staining by Feulgen procedure and examined by computerized image analyser system to determine their nuclear DNA content. Results of this study showed that, in the promastigote stage there is a single cell population, whereas in the amastigote stage there are two population of parasite nuclei. One with low level of DNA resembling that of the promastigote and a second population with double DNA level. This latter formed when apparently 2 nuclei fused and form one with double DNA level. The data obtained in the present study further indicate that Leishmania of the old world can undergo nuclear fusion which is a strong evidence for the presence of sexual reproduction in the parasite.

RESUMEN

Ivermectin, a broad spectrum antiparasitic agent has become a promising drug for treating parasitic infection and infestation. This study was carried out to investigate its activity against two common protozoal infection, giardiasis and cryptsporidiosis in a rat model. The results of this study showed that ivermectin therapy is effective against both parasites at a dose of 200 micrograms/kg.

Swiss strain albino mice were inoculated intradermally in a shaved area above the tail with Leishmania tropica strain. The dissemination of the parasites to various organs and distant cutaneous sites was followed up to 16 weeks. The infection was detected in the metastatic foci by examination of impression smears, inoculated culture tubes containing Tanabe's medium with specimen obtained from lesions and by histopathological changes. It was found that the parasite disseminated from the site of inoculation to visceral organs namely: the liver, spleen, kidney and lung. However, no invasion of the testis was observed throughout the experiment. Metastatic cutaneous lesions occurred late in the infection and was observed only in 2 animals. Cutaneous metastatic lesions developed exclusively on the tail and the face.

Ivermectin is used in veterinary practice against many ectoparasites and endoparasites and is the drug of choice for treatment of human onchocerciasis. This study was carried out to investigate the effect of topical application of this drug against human ectoparasites (Sarcoptes scabiei and Pediculus humanus capitis). Ivernectin was found to have a curative effect on head lice after a single topical application. In patients with scabies, the drug was also found to be effective after a single application. However, in 50% of the cases, another application was needed five days later.

In the present study, a trial was made to evaluate the monoclonal antibody produced as a tool in the detection of circulating antigen. For the preparation of monoclonal antibodies, four Balb/C mice were immunized with Trichinella spiralis larvae. Immunized spleen cells were prepared and a suitable mutant NS1 myeloma cell line was used for fusion. Eighty white swiss albino mice were orally infected with 500 L1 T. spiralis larvae. Their serum was collected at different periods i.e. 5, 11, 18 and 28 days post infection for the detection of circulating antigen which was done by the ELISA technique. Circulating antigen could not be detected on day 5 post infection, while on day 11 it was clearly identified; on day 18 it could be detected but at a lesser O.D. reading however the antigen disappeared completely on the 28th day. This study confirmed that monoclonal antibodies may be a valuable tool in the early diagnosis of trichinosis by the detection of specific antigens, even in small amounts whenever present in the circulation.
