RESUMEN

Along with mounting evidence that gut microbiota and their metabolites migrate endogenously to distal organs, the 'gut-lung axis,' 'gut-brain axis,' 'gut-liver axis' and 'gut-renal axis' have been established. Multiple animal recent studies have demonstrated gut microbiota may also be a key susceptibility factor for neurological disorders such as Alzheimer's disease, Parkinson's disease and autism. The gastrointestinal tract is innervated by the extrinsic sympathetic and vagal nerves and the intrinsic enteric nervous system, and the gut microbiota interacts with the nervous system to maintain homeostatic balance in the host gut. A total of 1507 publications on the interactions between the gut microbiota, the gut-brain axis and neurological disorders are retrieved from the Web of Science to investigate the interactions between the gut microbiota and the nervous system and the underlying mechanisms involved in normal and disease states. We provide a comprehensive overview of the effects of the gut microbiota and its metabolites on nervous system function and neurotransmitter secretion, as well as alterations in the gut microbiota in neurological disorders, to provide a basis for the possibility of targeting the gut microbiota as a therapeutic agent for neurological disorders.

Previous studies have demonstrated that gut microbiota dysbiosis promotes the development of mastitis. The interaction of the vagus nerve and gut microbiota endows host homeostasis and regulates disease development, but whether the vagus nerve participates in the pathogenesis of mastitis is unclear. Here, vagotomized mice exhibit disruption of the blood-milk barrier and mammary gland inflammation. Notably, mastitis and barrier damage caused by vagotomy are dependent on the gut microbiota, as evidenced by antibiotic treatment and fecal microbiota transplantation. Vagotomy significantly alters the gut microbial composition and tryptophan metabolism and reduces the 5-hydroxyindole acetic acid (5-HIAA) level. Supplementation with 5-HIAA alleviates vagotomy-induced mastitis, which is associated with the activation of the aryl hydrocarbon receptor (AhR) and subsequent inhibition of the NF-κB pathway. Collectively, our findings indicate the important role of the vagus-mediated gut-mammary axis in the pathogenesis of mastitis and imply a potential strategy for the treatment of mastitis by targeting the vagus-gut microbiota interaction.

Introduction: The drawbacks of using antibiotics as feed additives for blue foxes have gradually become apparent; moreover, thymol has wide-spectrum antimicrobial activity and has the potential to replace antibiotics in various animals. However, there are few reports on the effects of thymol on blue foxes. Methods: This study aimed to investigate the effects of different concentrations of thymol on the growth performance, apparent nutrient digestibility, serum biochemical indicators, intestinal morphology, and gut microbiota of blue foxes. Twenty-four male blue foxes (120 ± 5 d) of similar weight (6.05 ± 0.16 kg) were randomly divided into 4 groups. 0, 100, 200, and 300 mg/kg thymol were added to the basal diets of groups C, L, M, and H, respectively. Results: Compared with those in the C group, the addition of 100 mg/kg thymol to the diet significantly increased organic matter (OM) digestibility, crude protein (CP) digestibility, immunoglobulin (Ig) A, IgM, the VH of the duodenum, the CD of the jejunum, the VH of the ileum, and the VH/CD of the ileum (P < 0.05) and strongly significantly increased IgG (P < 0.01). The addition of 200 mg/kg thymol to the diet increased the VH/CD of the duodenum (P < 0.05). The addition of 300 mg/kg thymol to the diet significantly increased the VH and CD of the jejunum (P < 0.05). The addition of 200 mg/kg and 300 mg/kg thymol to the diets increased the final weight (FW) (P < 0.05). Adding 100 mg/kg thymol significantly increased the levels of interleukin-4 (IL-4) and catalase (CAT) compared with those in the other groups (P < 0.05). 16S rRNA gene detection revealed that thymol can change the abundances of Bifidobacterium, Fusobacterium, Allobaculum, Streptococcus, Megasphaera, and Lactobacillus in the gut. Conclusion: The addition of thymol to diets can increase the abundance of Bifidobacterium, Fusobacterium, and Allobaculum, which may contribute to improving the growth performance of blue foxes.

Endometritis is one of the important causes of infertility. Puerarin (PU) can inhibit oxidative stress and reduce inflammation; however, it is unclear whether PU has a protective effect on the endometritis. In our study, we used Staphylococcus aureus to induce mouse endometritis. The PU group (100 mg/kg PU) and the S. aureus + PU group received daily intraperitoneal injection of PU (25, 50 or 100 mg/kg PU). The results showed that S. aureus significantly increased the levels of MPO, TNF-α, IL-1ß and IL-6 in uterine tissue, and increased the expression of p-p65 and p-IκBα proteins in uterine tissue to induce endometritis in mice (p < 0.05). Furthermore, it has been found that S. aureus promotes the occurrence of ferroptosis by reducing GSH and ATP content, increasing MDA and iron content and reducing GPX4 and SLC7A11 protein expression levels (p < 0.05). S. aureus significantly increase the expression of NLRP3, ASC, caspase-1 and P2X7 proteins in uterine tissue (p < 0.05). However, PU obviously reduced the inflammatory response and reversed the changes of ferroptosis and the expression of P2X7 receptor/NLRP3 pathway associated proteins of the uterus induced by S. aureus (p < 0.05). Taken together, these findings emphasize the protective effect of PU on endometritis by regulating the P2X7 receptor/NLRP3 signalling pathway and inhibiting ferroptosis.

Refreezing the remaining genetic resources after in vitro fertilization (IVF) can conserve genetic materials. However, the precise damage inflicted by repeated freezing and thawing on bovine sperm and its underlying mechanism remain largely unexplored. Thus, this study investigates the impact of repeated freeze-thaw cycles on sperm. Our findings indicate that such cycles significantly reduce sperm viability and motility. Furthermore, the integrity of the sperm plasma membrane and acrosome is compromised during this process, exacerbating the advanced apoptosis triggered by oxidative stress. Additionally, transmission electron microscopy exposed severe damage to the plasma membranes of both the sperm head and tail. Notably, the "9 + 2" structure of the tail was disrupted, along with a significant decrease in the level of the axonemal protein DNAH10, leading to reduced sperm motility. IVF outcomes revealed that repeated freeze-thaw cycles considerably impair sperm fertilization capability, ultimately reducing the blastocyst rate. In summary, our research demonstrates that repeated freeze-thaw cycles lead to a decline in sperm viability and motility, attributed to oxidative stress-induced apoptosis and DNAH10-related dynamic deficiency. As a result, the utility of semen is compromised after repeated freezing.

The uterine environment provides necessary conditions for the existence of endometrial microbiota, which in turn plays an important role in maintaining the homeostasis of the uterine environment. The endometrial microbiome is highly susceptible to external factors such as age, hormones, menstrual, pregnancy, etc. When the microbiota is imbalanced, it will further promote the occurrence of uterine diseases such as endometritis and endometrial cancer. Regulating the microbiome of the endometrium is of positive significance for promoting uterine health. Among them, antibiotics, probiotics, prebiotics, and microbial transplantation may be important pathways for regulating endometrial microbiota in the future. However, there is currently no unified plan for evaluating the endometrial microbiota. In addition, due to the small sample size, it is easy to be contaminated by exogenous bacterial DNA, which poses great challenges for studying the mechanism of microbial community regulating uterine health. Therefore, there are still many areas worth exploring for the future of endometrial microbiome.

Mastitis is the most frequent disease of cows and has well-recognized detrimental effects on animal wellbeing and dairy farm profitability. With the advent of the postantibiotic era, alternative antibiotic agents, especially probiotics, have received increasing attention in the treatment of mastitis. Based on research showing that Lactobacillus reuteri (L. reuteri) has anti-inflammatory effects, this study explored the protective effects and mechanisms of L. reuteri against mastitis induced by Staphylococcus aureus (S. aureus) in mice. First, mice with S. aureus-induced mastitis were orally administered L. reuteri, and the inflammatory response in the mammary gland was observed. The results showed that L. reuteri significantly inhibited S. aureus-induced mastitis. Moreover, the concentration of oxytocin (OT) and protein expression of oxytocin receptor (OTR) were measured, and inhibition of OTR or vagotomy reversed the protective effect of L. reuteri or its culture supernatant (LCS) on S. aureus-induced mastitis. In addition, in mouse mammary epithelial cells (MMECs), OT inhibited the inflammation induced by S. aureus by inhibiting the protein expression of OTR. It was suggested that L. reuteri protected against S. aureus-induced mastitis by releasing OT. Furthermore, microbiological analysis showed that the composition of the microbiota was altered, and the relative abundance of Lactobacillus was significantly increased in gut and mammary gland after treatment with L. reuteri or LCS. In conclusion, our study found the L. reuteri inhibited the mastitis-induced by S. aureus via promoting the release of OT, and treatment with L. reuteri increased the abundance of Lactobacillus in both gut and mammary gland.

The microbiota gut brain (MGB) axis has been shown to play a significant role in the regulation of inflammatory and infective diseases. Exploring the structure and communication mode of MGB axis is crucial for understanding its role in diseases, and studying the signaling pathways and regulatory methods of MGB axis regulation in diseases is also of profound significance for future clinical research. This article reviews the composition, communication mechanism of MGB axis and its role in inflammatory and infective diseases, including Parkinson's disease (PD), Alzheimer's disease (AD), multiple sclerosis (MS), autism spectrum disorder (ASD), depression, psoriasis, irritable bowel syndrome (IBS), and inflammatory bowel diseases (IBD). In addition, our investigation delved into the regulatory functions of the inflammasome, IFN-I, NF-κB, and PARK7/DJ-1 innate immune signaling pathway in the context of inflammatory and infective diseases. Ultimately, we discussed the efficacy of various interventions, including fecal microbiota transplantation (FMT), antibiotics, probiotics, prebiotics, synbiotics, and postbiotics, in the management of inflammatory and infective diseases. Understanding the role and mechanism of the MGB axis might make positive effects in the treatment of inflammatory and infective diseases.

Background: Ketosis is one of the most frequent and costly metabolic disorders in high-producing dairy cows, and negatively associated with the health and reproductive performance of bovine. Ketosis is mainly caused by the accumulation of ketone body ß-hydroxybutyric acid and its diagnosis is based on ß-hydroxybutyrate (ßHB) concentration in blood. Methods: In this study, we investigated the effects of ßHB on bovine oocyte maturation in the concentration of subclinical (1.2 mM) ßHB and clinical (3.6 mM). Results: The results showed ßHB disrupted bovine oocyte maturation and development capacity. Further analysis showed that ßHB induced oxidative stress and mitochondrial dysfunction, as indicated by the increased level of reactive oxygen species (ROS), disrupted mitochondrial structure and distribution, and depolarized membrane potential. Furthermore, oxidative stress triggered early apoptosis, as shown by the enhanced levels of Caspase-3 and Annexin-V. Moreover, 3.6 mM ßHB induced the disruption of the pyruvate dehydrogenase (PDH) activity, showing with the decrease of the global acetylation modification and the increase of the abnormal spindle rate. Conclusion: Our study showed that ßHB in subclinical/clinical concentration had toxic effects on mitochondrial function and PDH activity, which might affect energy metabolism and epigenetic modification of bovine oocytes and embryos.

Fatty acids (FAs) are classified into different types according to the degree of hydrocarbon chain saturation, including saturated fatty acids (SFAs), monounsaturated fatty acids (MUFAs), omega-3 polyunsaturated fatty acids (omega-3 PUFAs) and omega-6 polyunsaturated fatty acids (omega-6 PUFAs), which play an important role in maintaining semen quality. This review focuses on the regulation of FAs in semen, diet and extender on semen quality, and expounds its effects on sperm motility, plasma membrane integrity, DNA integrity, hormone content, and antioxidant capacity. It can be concluded that there are species differences in the FAs profile and requirements in sperm, and their ability to regulate semen quality is also affected by the addition methods or dosages. Future research directions should focus on analyzing the FAs profiles of different species or different periods of the same species and exploring suitable addition methods, doses and mechanism of regulating semen quality.

Introduction: Flaxseed oil (FO) and vitamin E (VE) both have antioxidant effects on sperm. The present study investigated the effects of dietary supplementation with FO and/or VE on semen quality. Methods: 16 fertile Simmental bulls were selected and randomly divided into 4 groups (n = 4): the control group (control diet), FO group (control diet containing 24 g/kg FO), VE group (control diet containing 150 mg/kg VE) and FOVE group (control diet containing 150 mg/kg VE and 24 g/kg FO), and the trial lasted 10 weeks. Results: The results showed that the addition of FO independently can increase sperm motion parameters, the levels of catalase (CAT), glutathione peroxidase (GSH-Px), testosterone (T) and estradiol (E2), while reduce oxidative stress in seminal plasma (P < 0.05). Supplement of VE independently can increased the motility, motility parameters, CAT and superoxide dismutase (SOD) levels, and reduce oxidative stress in seminal plasma (P < 0.05). There was an interaction effect of FO × VE on motility and reactive oxygen species (ROS), while GSH-Px and ROS were affected by week × VE 2-way interaction, levels of T and E2 were also affected by the dietary FO × week interaction (P < 0.05). The triple interaction effects of FO, VE and week were significant for malondialdehyde (MDA) (P < 0.05). Compared with the control group, sperm from the FOVE group had a significantly higher in vitro fertilization (IVF) rate, and subsequent embryos had increased developmental ability with reduced ROS levels at the eight-cell stage, then increased adenosine triphosphate (ATP) content and gene expression levels of CAT, CDX2, Nanog, and SOD at the blastocyst stage (P < 0.05). Metabolomic and transcriptomic results indicated that dietary supplementation of FO and VE increased the expression of the metabolite aconitic acid, as well as the expression of ABAT and AHDHA genes. Conclusion: With in-silico analysis, it can be concluded that the effects of dietary FO and VE on improving semen quality and embryo development may be related to increased aconitic acid via the ABAT and AHDHA genes involved in the propionic acid metabolism pathway.

Introduction: Adding antibiotics to animal basal diets can improve growth and production performance. However, the use of antibiotics poses a potential threat to public health safety. Methods: The study was conducted to investigate the effects of different levels of mannan oligosaccharides (MOS) on the fur quality, nutrient apparent digestibility, serum immunity, antioxidant status, intestinal morphology, and gut microbiota of fur-growing raccoon dogs. Divide 24 male raccoon dogs (120 ± 5 d) of similar weight (5.01 ± 0.52 kg) into 4 groups randomly. Add 0, 0.05, 0.1, and 0.2% MOS to the basal diets of groups C, L, M, and H, respectively. Results: Compared to the C group, the addition of 0.05% and 0.1% MOS in the diet increased the apparent digestibility of crude protein (CP), Underfur length (UL), Guard hair length (GL), immunoglobulin A (IgA), immunoglobulin G (IgG), and immunoglobulin M (IgM) levels in the serum (p < 0.05); Under the dosage of 0.05 % MOS, the activities of Superoxide Dismutase (SOD) and catalase (CAT) increased (p < 0.05). Compared to the C group, adding 0.05% MOS significantly increased the VH/CD of the duodenum and ileum, while also increasing the VH and CD of the jejunum (p < 0.05). Through Spearman correlation analysis of the gut microbiota, it was found that MOS can improve fur quality by reducing the abundance of Dorea while improving the immune response of raccoon dogs by reducing the abundance of Blautia and Gemmiger. Discussion: In conclusion, MOS can improve the fur quality, serum immunity, antioxidant capacity, and gut microbiota of raccoon dogs. Therefore, MOS has the potential to replace antibiotics.

Sika deer velvet antler is the most important animal nutraceutic in traditional Chinese medicine. Reducing the breeding cost of sika deer by looking for a low-cost diet is the main research direction at present. The purpose of this experiment was to find an alternative diet for sika deer and reduce the cost of the diet by using spent mushroom substrate (SMS) as a concentrate supplement. The apparent digestibility for sika doe and the hematological changes of sika doe and suckling fawn were measured by replacing 10% of the concentrate supplement with SMS of Pleurotus ostreatus (SMS-MP). Compared with the control group, the digestibility of dry matter (DM), total protein (TP), globulin (GLO), and cholesterol (CHOL) of sika doe were significantly decreased (p < 0.05), and glucose (GLU), alanine (Ala), phenylalanine (Phe), and proline (Pro) of sika doe were significantly increased (p < 0.05) after the replacement of SMS-MP. Compared with the control group, the serum GLU of suckling fawn was significantly decreased (p < 0.05) and the phosphatase (ALP) was significantly increased after the replacement of SMS-MP (p < 0.05). There were no significant differences in the immune globulin and amino acid of suckling fawns between the two groups (p > 0.05). The present findings confirm the applicability of SMS-MP as a sika doe concentrate supplement. At the same time, using SMS, a waste resource, can not only reduce the breeding cost of sika doe, but also make full use of SMS to reduce environmental pollution.

At present, spent mushroom substrate (SMS) is a waste resource that is producing a pollution problem in China, and which has some use as animal feed or fertilizer, has not been assessed as a feed for deer. The purpose of this study is to expand the feed of male sika deer and reduce the feeding cost by using the waste resource of SMS. The 10% concentrated supplement was replaced with SMS and the feed intake, apparent digestibility, blood index and velvet production of male sika deer were measured. As the results showed, compared to the control group, the substitution of SMS for 10% of the concentrate supplement decreased the concentration of IgA (p < 0.01), replacing 10% concentrated supplement with SMS of Pleurotus ostreatus (SMS-MP) reduced the intake of organic matter (OMI) and improved the digestibility of ether extract (EE), while replacing 10% concentrated supplement with SMS of Flammulina velutipes (SMS-MF) had no effect on apparent nutrient digestibility, feed intake, velvet antler production, and biochemical indexes. In conclusion, SMS had no effect on serum biochemical indexes and the ratio of the feed weight of the deer supplement to the weight of velvet antler (p > 0.05). At the same time, SMS could reduce the feed consumption and improve the economy by using SMS as a waste resource.

The purpose of this experiment is to expand the feed of growing sika deer and to explore the effects on growing sika deer of the spent mushroom substrate of Pleurotus ostreatus (SMS-MP). Twelve immature female growing sika deer were randomly assigned to four groups. The ratios of SMS-MP to replace concentration supplements were 0%, 10%, 20%, and 30%, respectively, and the growth performance, feed intake and apparent digestibility, serum biochemical indexes, blood physiological indexes, serum immune globulin and plasma amino acid of growing sika deer were measured. The results of the current study confirmed the applicability of SMS-MP as a feed ingredient in growing sika deer diets. There was no significant change in growth performance and hematology of growing sika deer when the concentrate supplement was replaced with 10-20% SMS-MP. However, replacing 30% of concentrate supplements with SMS-MP in the growing sika deer diet resulted in significantly decreased Hb and HCT levels. It can be concluded that, as a waste resource, adding a small amount of SMS-MP has no significant effect on the growth of sika deer, and at the same time can reduce the consumption of concentrate supplements, thereby improving the economic benefits of sika deer breeding.

It is important to inhibit oxidative stress to maintain sperm motility during cryopreservation. The present study was performed to investigate the effects of supplementing oligomeric proanthocyanidins (OPC) and bamboo leaf flavonoids (BLF) or their combination as an extender for Simmental bull semen freezing. OPC, BLF, or their combination were added to the frozen diluent of bovine semen. Afterwards, computer-assisted semen analysis (CASA), detection of membrane functionality, acrosome integrity, mitochondrial integrity, CAT, SOD, GSH-PX, MDA, and ROS were conducted. The results showed that adding 50 mg/L OPC or 4 mg/L BLF could improve the quality of frozen sperm. Compared with 50 mg/L OPC alone, the combination of 50mg/L OPC and 2 mg/L BLF significantly increased the kinematic parameters of sperm, and sperm CAT, GSH-PX and SOD levels (p < 0.05), whereas the MDA of sperm was decreased (p < 0.05). These results indicated that compared to the addition of 50 mg/L OPC alone, a combination of 50 mg/L OPC and 2 mg/L BLF could further improve the quality of frozen semen. The results could provide theoretical data support for the development of a new protective agent and are significant for the cryopreservation of bovine semen in the future.

To determine the effects of epigallocatechin-3-gallate (EGCG) on the cryopreservation of bovine semen, epigallocatechin-3-gallate dissolved with double distilled water to 0.2, 0.4, 0.6 and 0.8 mg/ml were added to the cryopreservation diluent of the bull semen. Then, we used computer-assisted analysis of semen kinematic parameters, staining method to detect membrane function, acrosome integrity, enzyme-linked immunosorbent assay to detect catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), malondialdehydes (MDA) and reactive oxygen levels. The results showed that adding 0.6 mg/L of epigallocatechin-3-gallate could improve the cryopreserved sperm quality, which significantly increased the total motility, distance average path, distance straight line, distance curved line, average path velocity, curvilinear-velocity, straight-line velocity, amplitude of lateral head displacement and beat/cross frequency, as well as sperm CAT, GSH-Px and SOD levels (p < 0.05), whilst reducing the reactive oxygen species and MDA levels (p < 0.05). Hence, these results indicate that the addition of 0.6 mg/ml of EGCG has a protective effect on the cryopreservation of the bovine semen.

The present study was performed to investigate the effects of supplementing flaxseed oil (FO) or vitamin E (VE) or their combination to an extender for Simmental bull semen cryopreservation. In experiment 1, different concentrations of FO (0, 10, 100, and 1000 ng/mL) and VE (0.05, 0.1, and 0.2 mg/mL) were added to the extenders. In experiment 2, FO, VE, and FO + VE were added and a control group was included. Sperm viability, motility, motion parameters, acrosome integrity and membrane integrity, endogenous antioxidant indices, reactive oxygen species, and malondialdehyde levels were evaluated after semen thawing. A higher percentage of viability, motion parameters, endogenous antioxidant indices, and membrane integrity was observed after supplementation with 10 ng/mL FO or 0.1 mg/mL VE compared with the control group (p < 0.05). Also, combined supplementation of 10 ng/mL FO +0.1 mg/mL VE further improved the quality of frozen-thawed sperm by regulating viability, motion parameters, membrane integrity, and endogenous antioxidant indices compared with the FO or VE alone (p < 0.05). These results indicated that FO (10 ng/mL) + VE (0.1 mg/mL) could further improve the protective effects on bull sperm post-thaw.

The purpose of this study was to determine the effects of apigenin and astragalus polysaccharides on the cryopreservation of bovine semen. Apigenin, astragalus polysaccharides, or their combination were added to a frozen diluent of bovine semen. Afterwards, Computer Assisted Semen Analysis (CASA), membrane functionality, acrosome integrity, mitochondrial integrity, CAT, SOD, GSH-Px, MDA, and ROS detection were conducted. The results showed that adding 0.2 mmol/L AP or 0.5 mg/mL APS could improve the quality of frozen sperm. Compared to 0.2 mmol/L AP alone, the combination of 0.2 mmol/L AP and 0.3 mg/mL APS significantly increased the total motility (TM), average path distance (DAP), straight line distance (DSL), average path velocity (VAP), curvilinear velocity (VCL), wobble (WOB), and sperm CAT and SOD levels (p < 0.05), while reducing the ROS and MDA levels (p < 0.05). These results indicated that the addition of 0.2 mmol/L AP or 0.5 mg/mL APS alone has a protective effect on the freezing of bovine semen. Compared to the addition of 0.2 mmol/L AP, a combination of 0.2 mmol/L AP and 0.3 mg/mL APS could further improve the quality of frozen semen.
