RESUMEN

Maximizing the production of heterologous biomolecules is a complex problem that can be addressed with a systems-level understanding of cellular metabolism and regulation. Specifically, growth-coupling approaches can increase product titers and yields and also enhance production rates. However, implementing these methods for non-canonical carbon streams is challenging due to gaps in metabolic models. Over four design-build-test-learn cycles, we rewire Pseudomonas putida KT2440 for growth-coupled production of indigoidine from para-coumarate. We explore 4,114 potential growth-coupling solutions and refine one design through laboratory evolution and ensemble data-driven methods. The final growth-coupled strain produces 7.3 g/L indigoidine at 77% maximum theoretical yield in para-coumarate minimal medium. The iterative use of growth-coupling designs and functional genomics with experimental validation was highly effective and agnostic to specific hosts, carbon streams, and final products and thus generalizable across many systems.

RESUMEN

Physiological circuits differ across increasing isometric force levels during unilateral contraction. Therefore, we first explored the possibility of predicting the force level based on electroencephalogram (EEG) activity recorded during a single trial of unilateral 5% or 40% of maximal isometric voluntary contraction (MVC) in right-hand grip imagination. Nine healthy subjects were involved in this study. The subjects were required to randomly perform 20 trials for each force level while imagining a right-hand grip. We proposed the use of common spatial patterns (CSPs) and coherence between EEG signals as features in a support vector machine for force level prediction. The results showed that the force levels could be predicted through single-trial EEGs while imagining the grip (mean accuracy = 81.4 ± 13.29%). Additionally, we tested the possibility of online control of a ball game using the above paradigm through unilateral grip imagination at different force levels (i.e., 5% of MVC imagination and 40% of MVC imagination for right-hand movement control). Subjects played the ball games effectively by controlling direction with our novel BCI system (n = 9, mean accuracy = 76.67 ± 9.35%). Data analysis validated the use of our BCI system in the online control of a ball game. This information may provide additional commands for the control of robots by users through combinations with other traditional brain-computer interfaces, e.g., different limb imaginations.

RESUMEN

By reverse transcription-polymerase chain reaction (RT-PCR), an open reading frame of pathogenesis-related protein 1 (PR1) was isolated from Panax notoginseng and named as PnPR1. Molecular and bioinformatic analyses of PnPR1 revealed that an open reading frame of 501 bp was predicted to encode a 166-amino acid protein with a deduced molecular mass of 18.1 kD. Homology analysis showed that the deduced amino acid sequence of PR1 protein of Panax notoginseng had a high similarity with other higher plants had the same conservative structure domain of cysteine-rich secretory protein (CAP). The recombinant expressed plasmid pET28a(+)-PnPR1 was expressed in Escherichia coli BL21. The expression conditions were optimized by induction at different times, different temperatures, different IPTG concentrations and different giving times. The optimum expression condition was 0.4 mmol.L-1 IPTG at 28 degrees C for 20 h. The successful expression of PnPR1 provides some basis for protein purification and preparation of the monoclonal antibody.

RESUMEN

Objective To discuss the effect and treatment methods of ureteral obstruction caused by primary or metastatic pelvic malignancy. Methods Clinical data of 51 cases with ureteral obstruction caused By pelvic malignancy were reviewed retrospectively. The treatment included traditional open operation in 17 cases in which 3 cases with uretero-uwtero, 9 cases with uretero-bladder anastomotic stoma and 5 cases with cutaneous ureterostomy, retrograde stenting in 19 cases (11 cases with unilateral ureter, 8 cases with bilateral ureter), pereutaneous nephrostomy in 15 cases. Results The median follow-up time was 21 months (range 6 to 72 months ). Three months after operation, uhrasonograph or intravenous urography(IVU)showed that 39 (76.5%) cases had nomal renal function, 12 (23.5%) cases had hydronephrosis relief and renal function improvement. No stricture in the uretero-uretero or uretero-bladder anastomotic stoma was recorded. Conclusions Appropriate treatment is dependent upon the accurate identification of the underlying pathological process and it can relieve ureteral obstruction and increase the quality of life for ureteral obstruction caused by pelvic malignancy.

RESUMEN

Objective To explore the relationship of bone marrow micrometastases(BMM) with nm23 expression of breast cancer(BC) in patients with stage Ⅰ BC. Methods BMM and nm23 expression of carcinoma tissue in 52 cases of stage Ⅰ BC were examined by immunohistochemical technique with monoclonal anti epithelial membrane antigen(anti EMA) and nm23 H1. Results BMM was observed in 10 of 52 patients(19.2%). In the group of poor differentiated cancer, the positive rate of BMM was significantly higher than that in well differentiated cancer(P
