BACKGROUND: Pharmacogenomics is a facet of personalized medicine that explores how genetic variants affect drug metabolism and adverse drug reactions. Therefore, this study aims to detect distinct pharmacogenomic variations among the Jingpo population and explore their clinical correlation with drug metabolism and toxicity. METHODS: Agena MassARRAY Assay was used to genotype 57 VIP variants in 28 genes from 159 unrelated Jingpo participants. Subsequently, the chi-squared test and Bonferroni's statistical tests were utilized to conduct a comparative analysis of genotypes and allele frequencies between the Jingpo population and the other 26 populations from the 1000 Genome Project. RESULTS: We discovered that the KHV (Kinh in Ho ChiMinh City, Vietnam), CHS (Southern Han Chi-nese, China) and JPT (Japanese in Tokyo, Japan) exhibited the smallest differences from the Jingpo with only 4 variants, while ESN (Esan in Nigeria) exhibited the largest differences with 30 variants. Besides, a total of six considerably different loci (rs4291 in ACE, rs20417 in PTGS2, rs1801280 and rs1799929 in NAT2, rs2115819 in ALOX5, rs1065852 in CYP2D6, p < 3.37 × 10-5) were identified in this study. According to PharmGKB, rs20417 (PTGS2), rs4291 (ACE), rs2115819 (ALOX5) and rs1065852 (CYP2D6) were found to be associated with the metabolism efficiency of non-steroidal anti-inflammatory drugs (NSAIDs), aspirin, montelukast and tamoxifen, respectively. Meanwhile, rs1801280 and rs1799929 (NAT2) were found to be related to drug poisoning with slow acetylation. CONCLUSION: Our study unveils distinct pharmacogenomic variants in the Jingpo population and discovers their association with the metabolic efficiency of NSAIDs, montelukast, and tamoxifen.

Herein, a self-enhanced molecularly imprinted polymer luminescence (MIP-ECL) sensing platform based on gold-copper doped Tb-MOFs (Au@Cu:Tb-MOFs) was constructed for ultra-sensitive detection of chlorpyrifos (CPF). In this work, Au@Cu:Tb-MOFs as co-reaction promoters greatly improve the ECL emission signal, while Au@Cu:Tb-MOFs were used as cathode emitters. And chlorpyrifos and 4,7-bis(thiophene-2-yl)benzo [c][1,2,5] thiadiazole were electropolymerized on electrode surface to form MIP, where this films with thiophene derivatives could greatly improve ECL signal. Notably, the introduction of MIP as recognition elements enabled specific identification of target analytes, in which molecular docking technique validated target analyte and functional monomers are tightly bound through Pi-alkyl interaction. As the concentration of CPF increases, the ECL signal gradually decreases, showing a good linear relationship in the range of 0.1-106 pg/mL with a low detection limit (LOD) of 0.029 pg/mL. Moreover, actual sample testing experiment of this method displayed a special correlation in organophosphorus detection and development potential in actual sample analysis.

OBJECTIVES: This study aimed to detect the correlation between SOWAHB polymorphisms and Thyroid cancer (TC) risk in the Chinese Han population. METHODS: We genotyped SOWAHB variants in 510 TC patients and 509 controls using Agena MassARRAY. We assessed the association between SOWAHB polymorphisms and TC susceptibility, with the significant results evaluated through FPRP analysis. We predicted TC risk by the SNP-SNP interaction, analyzed by MDR. RESULTS: Carriers with rs2703129 CC had a lower probability of TC (codominant, recessive: p = 0.002), while subjects with rs1874564 AG had an increased risk of developing TC (codominant, recessive: p = 0.000, log-additive: p = 0.028). In subjects aged > 45 years, rs2703129 may reduce TC predisposition (codominant: p = 0.011, recessive: p = 0.007), but there was an increased association between rs1874564 and TC risk (codominant: p = 0.030, dominant: p = 0.047). Also, rs2703129 was associated with a lower risk of TC among males (codominant: p = 0.018, recessive: p = 0.013). Conversely, rs1874564 was associated with an increased risk of TC in females (codominant: p = 0.001, dominant: p = 0.003). CONCLUSION: SOWAHB SNPs were related to the occurrence of TC, and rs2703129 may be a protective site for TC.

BACKGROUND: Stroke is an important cause of death and disability worldwide, ranking second in the cause of death, and it is thought to be related to genetic factors. The purpose of our study is to investigate the association between CASZ1, WNT2B and PTPRG single nucleotide polymorphisms (SNPs) and stroke risk in the Chinese population. METHODS: We recruited 1418 volunteers, comprised of 710 stroke cases and 708 controls in this study. We used MassARRAY iPLEX GOLD method to genotype the three SNPs on CASZ1, WNT2B and PTPRG. Logistic regression was used to analyse the association between these SNPs and stroke, and odds ratios (ORs) and 95% confidence intervals (CIs) were then calculated. What's more, the interactions among SNPs were predicted by multi-factor dimensionality reduction (MDR) analysis. RESULTS: This research demonstrated that CASZ1 rs880315 and PTPRG rs704341 were associated with reduced stroke susceptibility. More precisely, CASZ1 rs880315 was associated with reduced stroke susceptibility in people aged ≤64 years and women. PTPRG rs704341 was associated with reduced stroke susceptibility in people aged >64 years, women, non-smokers and non-drinkers. Conversely, WNT2B rs12037987 was related to elevated stroke susceptibility in people aged >64 years, women and non-smokers. In addition, CASZ1 rs880315, WNT2B rs12037987 and PTPRG rs704341 had a strong redundancy relationship. CONCLUSION: Our study concludes that CASZ1 rs880315, WNT2B rs12037987 and PTPRG rs704341 are associated with stroke, and the study provides a basis for assessing genetic variants associated with stroke risk in the Han Chinese population.

BACKGROUND: Our study aimed to elucidate the association between single nucleotide polymorphisms (SNPs) in CYP2B6 gene and susceptibility to lung cancer (LC). METHODS: Five SNPs in CYP2B6 were genotyped in Chinese Han population (507 cases and 505 controls) utilizing Agena MassARRAY. The relationship between these SNPs and LC susceptibility was assessed using odds ratios, 95% confidence intervals, and χ2 tests. Additionally, multifactor dimensionality reduction was employed to analyze SNP-SNP interactions. Bioinformatics methods were applied to investigate the function of these SNPs. RESULTS: We found that rs2099361 was associated with an increased susceptibility to LC in the codominant model (OR = 1.31, p = 0.045). Stratification analysis revealed the allele G at rs4803418 and the allele T at rs4803420 of CYP2B6 (BMI >24 kg/m2) were significantly linked to decreased susceptibility of LC. Conversely, the allele C at rs12979270 (BMI >24 kg/m2) showed increased susceptibility to LC. Moreover, a robust redundant relationship between rs12979270 and rs4803420 was identified in the study. According to the VannoPortal database, we found that rs4803420, rs12979270 and rs2099361 may modulate the binding affinity of LMNB1, SP1 and HDAC2, respectively. CONCLUSIONS: Our results suggest that SNPs in the CYP2B6 gene play crucial roles in LC susceptibility.

As an important element in organism, the lack and excess of ferric ions (Fe3+) may lead to an extensive range of diseases presenting with distinct clinical manifestations. In our design, a multi-channel probe with reversible enol-to-keto-to-enol tautomerization for the specific recognition and high sensitivity detection of Fe3+ was prepared. This paper reported a novel Cop-NC probe, Tris (4-formylphenyl) amine bearing 1,4-cyclohexanedione groups, which provides binding site for Fe3+ and also contributes both fluorescent and electrochemical signals. The as-synthesized Cop-NC exhibit intense fluorescence under an excitation wavelength at 378 nm with a quantum yield of 26%. Results of spectroscopic measurement show that Fe3+ can significantly cause a "Switch-off" fluorescence intensity effect. Simultaneously, the addition of Fe3+ can cause a "Switch-on" effect in electrochemical channel. It has realized the detection of Fe3+ with concentration as low as 0.4 µM and 1.0 nM in the fluorescence channel and redox channel, respectively. The development of the joint probe with multi-channel signals provides a more convenient and rapid detection method for food, medical treatment, environmental monitoring and other fields.

Eighteen derivatives of pentacyclic triterpene carboxylic acids (Maslinic acid, Corosolic acid and Asiatic acid) have been prepared by coupling the piperazine complex of l-amino acids at the C-28 site of the parent compounds. The α-glucosidase inhibitory activities of the pristine derivatives were evaluated in vitro. The results indicated that the inhibitory activity of some compounds (15e IC50 = 591 µM, 16e IC50 = 423 µM) was closed to that of the reference acarbose (IC50 = 347 µM) in ethanol-water system. In addition, compound 16e (IC50 = 380 µM) showed superior inhibitory activity than acarbose (IC50 = 493 µM) in the measurement system with DMSO as solvent. The comparison of two different solvent systems showed that the derivatives had better α-glucosidase inhibitory activity in the DMSO system than that of in ethanol-water system. Regrettably, all of the as-synthesized derivatives exhibited inferior α-glucosidase inhibitory activities than those of the parent compounds in both test solvent systems. Furthermore, the result of enzyme kinetics demonstrated that the inhibition mechanism of compound 16e was noncompetitive inhibition with the inhibition constant Ki = 552 µM.

A novel symmetric bianthracene derivative (D2) containing one benzene ring and two ethyl thioglycolates connecting to the benzene ring on both sides of the bianthracene unit was designed and synthesized. D2 can detect Fe3+ and Hg2+ in acetonitrile/water (6:4, v/v) solution via different changes of absorbance and fluorescence in the pH range from 3 to 10. D2 exhibits high colorimetric sensitivity for Fe3+ with low detection limit (1.87 × 10-5 mol/L). The absorbance intensity of D2 in acetonitrile/water solution show a linear response to Fe3+ in the wide concentration range of 0 -1000 µM, which is beneficial for quantitative analysis. D2 also displays highly selective fluorescent sensing for Hg2+ with a low detection limit of 1.07 µM over other metal ions and can accurately detect the existence of Hg2+ in water samples.

Prior to the maturation of next-generation energy storage devices, the actual lithium-ion batteries for commercial purposes are still expected to fulfill some critical requirements, among which the high energy density, wide operating temperature range, and related long-term cycling stability are the most challenging issues. Herein a multiple additives strategy is employed to simultaneously optimize the solid electrolyte interphase on the large-area anode and cathode in a 2 Ah artificial graphite (AGr)/LiNi0.5Co0.2Mn0.3O2 (NCM523) pouch cell with high gravimetric (>260 Wh kg-1) and volumetric (>630 Wh L-1) energy density. By introducing a rational mixture of electrolyte additives, a highly sulfurized surface layer and a uniform and thin passivation layer are separately formed on the anode and cathode of the AGr/NCM523 pouch cell, exhibiting high storage stability at 60 °C, much improved discharge capacity at -10 and -20 °C, high anodic stability at high voltage of 4.4 V, and stable cyclic performance with a capacity retention of 85.5% after 500 cycles, significantly outperforming the value of 75.7% after only 200 cycles of the cell without additional additives. These results demonstrate the critical effect of simultaneous optimizations of anode and cathode interphase layers to construct stable high-energy-density lithium-ion pouch cells.

A new indolo[2,3-a]carbazole based colorimetric chemosensor C1 was designed and synthesized. The optical properties of C1 were investigated by UV-vis spectroscopy, UV-vis titrations, and fluorescence spectroscopy. The HOMO and LUMO values of C1 were obtained using density functional theory (DFT) calculations. The results indicate that C1 has specific selectivity and high sensitivity for detection of Cu2+ and Fe3+ ions compared with metal ions (K+, Na+, Mg2+, Zn2+, Cd2+, Pb2+, Hg2+, Ag+, Co2+, Cr2+, Ni2+, Mn2+, Ba2+). The presence of Cu2+ ions leads to visible color change from yellow to colorless in an aqueous-acetonitrile solution, especially. Moreover, the detection limit for the analysis of Cu2+ and Fe3+ ions is found to be as low as 2.93 × 10-7 M and 4.10 × 10-7 M, respectively. Therefore, C1 should have potential applications as a practical colorimetric chemosensor for simultaneous detection of Cu2+ and Fe3+ in the environment and biological systems.

Combining two bioactive moieties by covalent bond into a novel single hybrid biological entity in view of the principle of active splicing, twenty-two C28-modified derivatives of pentacyclic dihydroxytriterpene carboxylic acids with saturated nitrogen heterocycle segments (i.e. 1-deoxynojirimycin or piperazines) have been synthesized. The inhibitory activities of all final target compounds on α-glucosidase were evaluated in vitro. The results of α-glucosidase inhibition assay indicate that some derivatives (e.g. 4b: IC50 = 1468.4 µM; 12b: IC50 = 499.6 µM 12c: IC50 = 768.5 µM, 13c: IC50 = 819.2 µM) show superior inhibitory activity in α-glucosidase than that of the precursor maslinic acid (IC50 = 2540.6 µM) or corosolic acid (IC50 = 1363.7 µM), in which compound 12b (IC50 = 499.6 µM) possesses stronger inhibitory activity than that of acarbose (IC50 = 606 µM). In addition, the result of enzyme kinetics study reveals that the inhibitory mechanism of the compound 12b is non-competitive inhibition and the inhibition constant Ki is 570 µM. The binding interaction between compounds with α-glucosidase are predicted by molecular docking simulation.

9,10-Diphenylanthracene cubic nanoparticles (DPA CNPs) with aggregation-induced emission characteristic (AIEgens) were synthesized through a facile reprecipitation method; then, a bright and stable electrochemiluminescence (ECL) signal can be observed when the DPA CNPs were modified at the glassy carbon electrode (GCE) in the presence of Tri-n-propylamine (TPA). This phenomenon is ascribed to the molecules with restricted movement that greatly blocked the energy leakage during the relaxation of the excited state, which facilitated the emission of energy in the form of photons. In addition, the size confinement effect of DPA CNPs in the aggregated state effectively enhanced the ECL emission. The application of DPA CNPs with AIE characteristics in an electrochemiluminescence immunosensor has not been reported. In this contribution, a free-label aggregation-induced electrochemiluminescence immunosensor based on DPA CNPs was fabricated and a simple strategy for ultrasensitive detection of aflatoxin B1 (AFB1) was proposed. The ECL signal is quenched linearly in the range of 0.01 pg/mL to 100 ng/mL for AFB1, and the detection limit is 3 fg/mL. In summary, the prepared sensor exhibits high sensitivity, acceptable accuracy, good anti-interference ability and stability, and satisfactory detection toward AFB1 in walnut samples. Therefore, the fabricated immunosensor will have significant applications in the fields of food, medicine, and so on.

Five novel metal-free organic dyes DQ1-5 containing a dipentyldithieno[3,2-f:2',3'-h]quinoxaline (DPQ) unit were synthesized and applied in dye-sensitized solar cells (DSSCs), where DPQ was employed as a π-spacer for the first time. Their photophysical, electrochemical, and theoretical calculations and photovoltaic properties were systematically investigated. All the five dyes show broad photoresponse. Especially the absorption edges of DQ3-5 extend to 800 nm on the TiO2 films. The inserted electron-rich unit 3,4-ethylenedioxythiophene or electron-withdrawing group benzothiadiazole (BTD) in DPQ-based dyes can greatly influence the optoelectronic properties of the dyes. In addition, the different electron donors also significantly affect the performance of the DSSCs. Under standard global AM 1.5 solar light conditions, the DQ5 sensitized solar cell obtained a power conversion efficiency of 7.12%. The result indicates that the rigid DPQ-based organic dye is a promising candidate for efficient DSSCs.

A novel efficient metal free sensitizer containing asymmetric double donor-π-acceptor chains (DC) was synthesized for dye-sensitized solar cells (DSSCs). Comparing to 3.80%, 4.40% and 4.64% for the DSSCs based on the dyes with single chain (SC1, SC2) and cosensitizers (SC1 + SC2), the overall conversion efficiency reaches 6.06% for DC-sensitized solar cells as a result of its longer electron lifetime and higher incident monochromatic photon-to-current conversion efficiency.
