BACKGROUND AND AIMS: Most pediatric IBD studies are performed after medications are approved in adults and the majority of participants in these studies are adolescents. We hypothesized that adolescent-onset IBD is not fundamentally different than adult-onset IBD. If this is correct, the value of delaying access to novel drugs in adolescents becomes questioned. METHODS: Data from 11 randomized, double-blind, placebo-controlled adult phase 2 and 3 trials of 4 biologics were analyzed. Participants were categorized as having adolescent- or adult-onset disease (diagnosed 12 to <18, or ≥18 years). Multivariable modelling explored the association between age at diagnosis and response to treatment after adjustment for disease duration, extent, and severity at baseline. Data from dose arms were pooled to evaluate similarity of therapeutic response between adolescent- and adult-onset IBD within the same trial (not between doses or across trials). Ratios of odds ratios between the two groups were evaluated. RESULTS: Data from 6,283 study participants (2,575 with Crohn's disease [CD], 3,708 with ulcerative colitis [UC]) were evaluated. Of 2,575 study participants with CD, 325 were 12-<18 years old at diagnosis; 836 participants (32.4%) received placebo. Of 3,708 participants with UC, 221 were 12-<18 years old at diagnosis; 1,212 (33%) were receiving placebo. The majority of the ratios of ORs were within two-fold, suggesting that responses in adolescent and adult-onset participants are generally similar. CONCLUSION: Data presented lend support for extrapolating efficacy of biologics from adults to adolescents with IBD, which would facilitate earlier labeling and patient access.

Aboveground Biomass (AGB) in the grassland senescence period is a key indicator for assessing grassland fire risk and autumnal pasture carrying capacity. Despite the advancement of remote sensing in rapid monitoring of AGB on a regional scale, accurately monitoring AGB during the senescence period in vast arid areas remains a major challenge. Using remote sensing, environmental data, and 356 samples of grassland senescence period AGB data, this study utilizes the Gram-Schmidt Pan Sharpening (GS) method, multivariate selection methods, and machine learning algorithms (RF, SVM, and BP_ANN) to construct a model for AGB during senescence grassland, and applies the optimal model to analyze spatio-temporal pattern changes in AGB from 2000 to 2021 in arid regions. The results indicate that the GS method effectively enhances the correlation between measured AGB and vegetation indices, reducing model error to some extent; The accuracy of grassland AGB inversion models based on a single vegetation index is low (0.03 ≤ |R| ≤ 0.63), while the RF model constructed with multiple variables selected by the Boruta algorithm is the optimal model for estimating AGB in arid regions during the senescence period (R2 = 0.71, RMSE = 519.74 kg/ha); In the span of 22 years, the annual average AGB in the senescence period of arid regions was 1413.85 kg/ha, with regions of higher AGB primarily located in the northeast and southwest of the study area. The area experiencing an increase in AGB during the senescence period (79.97 %) was significantly larger than that with decreased AGB (20.03 %).

Plant species diversity is one of the critical factors for maintaining multi-function and stability of terrestrial ecosystem. We reviewed the traditional methods for measuring plant species diversity of grassland (PSDG), and then introduced the new ideas and methods used for PSDG monitoring. Traditionally, PSDG monitoring depended heavily on ground-based investigation, which usually required large amounts of time, labor, and cost, and therefore was only suitable for small scale investigation. Grassland plant species were typically small in size and highly mixed. It was difficult to identify and measure by remote sensing due to the limitation of resolution. Consequently, most studies on PSDG were based on remote-sensing retrieval or habitat simulation. Characterized with high spatial-temporal resolution, flexible and low cost, the unmanned aerial vehicle (UAV) technology was regarded as the bridge between ground-based investigation and satellite remote sensing. It could be the breakthrough for monitoring PSDG accurately at large scales. In the future, we should establish PSDG monitoring network by combining the fixed monitoring sites and dynamic monitoring sites of UAV and satellite remote sensing, and integrating UAV and automatic target recognition organically.

Determining the relationship between net primary productivity (NPP) and grassland phenology is important for an in-depth understanding of the impact of climate change on ecosystems. In this study, the NPP of grassland in Xinjiang, China, was simulated using the Carnegie-Ames-Stanford approach (CASA) model with Moderate Resolution Imaging Spectroradiometer (MODIS) grassland phenological (MCD12Q2) data to study trends in phenological metrics, grassland NPP, and the relations between these factors from 2001-2014. The results revealed advancement of the start of the growing season (SOS) for grassland in most regions (55.2%) in Xinjiang. The percentage of grassland area in which the end of the growing season (EOS) was delayed (50.9%) was generally the same as that in which the EOS was advanced (49.1%). The percentage of grassland area with an increase in the length of the growing season (LOS) for the grassland area (54.6%) was greater than that with a decrease in the LOS (45.4%). The percentage of grassland area with an increase in NPP (61.6%) was greater than that with a decrease in NPP (38.4%). Warmer regions featured an earlier SOS and a later EOS and thus a longer LOS. Regions with higher precipitation exhibited a later SOS and an earlier EOS and thus a shorter LOS. In most regions, the SOS was earlier, and spring NPP was higher. A linear statistical analysis showed that at various humidity (K) levels, grassland NPP in all regions initially increased but then decreased with increasing LOS. At higher levels of K, when NPP gradually increased, the LOS gradually decreased.

To investigate whether microcystin-LR (MC-LR) influences children's cognitive function and memory ability, we measured serum MC-LR and whole blood lead levels in 697 primary students, and collected their academic and neurobehavioral test scores. The median of serum MC-LR levels was 0.80 µg/L (the value below the limit of detection to 1.67 µg/L). The shapes of the associations of serum MC-LR levels (cut-point: 0.95 µg/L) with scores on academic achievements, digit symbol substitution test and long-term memory test were parabolic curves. Logistic regression analysis showed that MC-LR at concentrations of 0.80-0.95 µg/L was associated with the increased probability of higher achievements on academic achievements [odds ratio (OR) = 2.20, 95% confidence interval (CI): 1.28-3.79], and also with scores on digit symbol substitution test (OR = 1.73, 95% CI: 1.05-2.86), overall memory quotient (OR = 2.27, 95% CI: 1.21-4.26), long-term memory (OR = 1.85, 95% CI: 1.01-3.38) and short-term memory (OR = 2.13, 95% CI: 1.14-3.98) after adjustment for confounding factors. Antagonism of MC-LR and lead on long-term memory was observed (synergism index = 0.15, 95% CI: 0.03-0.74). In conclusion, serum MC-LR at concentrations of 0.80-0.95 µg/L was positively associated with higher scores on cognitive and neurobehavioral tests, and antagonism between MC-LR at concentrations of 0.80-1.67 µg/L and lead exposure was obviously observed on long-term memory in children. Concerning that MC-LR is a neurotoxin at high doses, our observation is interesting and need further investigation.

Net primary productivity (NPP) is an index of the increase in plant biomass. Plant biomass is an important component of the global carbon cycle that indicates the health of an ecosystem. Environmental restoration has recently received much attention in Xinjiang, and it is thus important to quantify the dynamic effects of the drivers of NPP in the region. NPP was calculated for the annual growing season from 1982 to 2013 using the Carnegie-Ames-Stanford Approach (CASA) model. The effects of climate factors on NPP were analyzed, and the relationships between NPP and climate factors as well as human activity were quantified. Additionally, an innovative method based on partial derivatives and residual error was proposed to calculate the contributions of climate factors and human activities. The results show that average annual NPP in Xinjiang was 57.45 g C m-2 from 1982 to 2013 and that the average increase in annual NPP was 0.23 g C m-2 year-1. The average increases in annual NPP due to temperature, precipitation, and solar radiation were 0.0095, 0.2679, and 0.2541 g C m-2 year-1; the average decreases were respectively - 0.0133, - 0.0521, and - 0.0725 g C m-2 year-1. Precipitation and solar radiation influence NPP more than temperature. Precipitation had the greatest effect on NPP in the first 19 years, but solar radiation became more influential after 2000. Climate conditions were favorable for increase in NPP before 2000. The environmental restoration also occurred in Xinjiang during that period, and human activity slightly decreased NPP. Human activity increased and had a greater effect on NPP after 2000.

Climate change and human activities are two key factors that affect grassland ecosystem. Accurately estimating the effects of these two factors on grassland dynamics and understanding the driving forces of the dynamics are important in controlling grassland degradation. In this study, the potential Net Primary productivity (NPPP) and the difference between NPPP and actual NPP (NPPA) are used as indicators of climate change and human activities on grassland ecosystem in Xinjiang. An overall grassland NPPA increase than decrease (69.7% vs 30.3%) is found over the study period of 2000 to 2014. While human activities played a dominant role for such a NPPA increase, both human activities and climate change contributed almost equally to the grassland NPPA decrease. Within the three types of grasslands in Xinjiang, the desert grassland showed the greatest NPPA increasing trend that mostly attributed to human activities; the meadow showed an overall NPPA decreasing trend that was mainly caused by human activities; the steppe showed similar NPPA decreasing and increasing trend in terms of area percentage. Based on this study, our recommendations are (1) to continue the grazing prohibition policy in desert grassland and (2) to extensively implement the rest grazing policy in steppe and meadow grasslands.

AFB1 and MC-LR are two major environmental risk factors for liver damage worldwide, especially in warm and humid areas, but there are individual differences in health response of the toxin-exposed populations. Therefore, we intended to identify the susceptible genes in transport and metabolic process of AFB1 and MC-LR and find their effects on liver damage. We selected eight related SNPs that may affect liver damage outcomes in AFB1 and MC-LR exposed persons, and enrolled 475 cases with liver damage and 475 controls of healthy people in rural areas of China. The eight SNPs were genotyped by PCR and restriction fragment length polymorphism. We found that SLCO1B1 (T521C) is a risk factor for liver damage among people exposed to high AFB1 levels alone or combined with MC-LR, and that GSTP1 (A1578G) could indicate the risk of liver damage among those exposed to high MC-LR levels alone or combined with high AFB1 levels. However, GSTP1 (A1578G) could reduce the risk of liver damage in populations exposed to low MC-LR levels alone or combined with high AFB1 levels. In conclusion, SLCO1B1 (T521C) and GSTP1 (A1578G) are susceptible genes for liver damage in humans exposed to AFB1 and/or MC-LR in rural areas of China.

Abnormal liver function (ALF) plays a key role in metabolic syndrome (MetS), but only few data on the relationship between MetS and the risk factors for ALF (e.g., biotoxins) are available. We aimed to provide the prevalence of MetS and its association with the risk factors for ALF in rural area of Southwest China. A cross-sectional study within the hepatocellular carcinoma cohort was conducted, and included 5493 people with age from 30 to 85 years old. MetS was defined according to the Joint Scientific Statement. We observed that the prevalence of MetS was 31.8% (39.0% in women and 19.8% in men). Logistic regression analysis showed that significantly increased risk of MetS was found in those showing ALF (OR = 3.00, 95% CI: 2.43-3.71). Significantly decreased risk of MetS was found in those with higher HBV DNA titers (OR = 0.49, 95% CI: 0.33-0.74), and in those with higher aflatoxin B1 exposure (estimated daily intake, EDI) (OR = 0.60, 95% CI: 0.53-0.67). No significant change was found in those with higher microcystin-LR exposure (EDI). Therefore, the different risk factors for ALF might exert different effects on MetS. However, there should be an interaction effect existing that might decide the severity of MetS.

Three liver hazards, two confirmed-hepatitis B virus (HBV) and aflatoxin (AFB), and one rarely studied in populations-microcystin (MC), simultaneously exist in tropical and humid areas; however, there are no epidemiological data on their risks in the same population. We conducted a community-based cross-sectional survey among 5493 adults in two rural towns and statistically analyzed the comparative and combinative effects of the three factors after detecting HBsAg and HBV DNA titers, determining estimated daily intakes (EDIs) of AFB1 and MC-LR and testing serum AST and ALT as liver injury markers for each participant. We observed a HBsAg(+) rate of 7.6%, a relatively high AFB1 exposure level (mean EDIAFB1 = 471.30 ng/d), and a relatively low MC-LR exposure level (mean EDIMC-LR = 228.25 ng/d). ORs for abnormal AST (2.42, 95%CI = 1.69-3.45) and ALT (2.87, 95%CI = 1.91-4.29) increased in HBV infections compared with HBV-unexposed participants but did not increase in participants with separate or combined exposure to AFB1 and MC-LR (EDIs ≥ mean). Meanwhile, after adjustment for confounding factors, means of AST and ALT and ORs of abnormal AST and ALT were successively elevated after exposure to HBV, HBV&AFB1 (or HBV&MC-LR), and HBV&AFB1&MC-LR, especially in the group with detectable HBV DNA (AST: OR = 11.38, 95%CI = 3.91-33.17; ALT: OR = 17.09, 95%CI = 5.36-54.53). Notably, ORs for abnormal AST and ALT in the HBV exposed group were not significantly different from those in HBV&AFB1 or in the HBV&MC-LR exposed group but were significantly higher in the HBV&AFB1&MC-LR exposed group (P = 0.029 and P = 0.037, respectively). Our study indicated that microcystin may have the potential to increase the risk of liver injury induced by combined exposure to HBV and aflatoxin. However, in consideration of the uncertainties in the detection of the toxins and evaluation of the EDIs, more epidemiological data are expected to determine the increasing toxic effects of microcystins.

Although the nephrotoxicity of microcystin and aflatoxin has been observed in animal and clinical cases, few population data are available. We conducted a cross-sectional study in Southwest China to investigate the association of renal function indicators (RFIs, including BUN, SCr, and eGFR) with exposure to microcystin and aflatoxin in 5493 members of the general population. Microcystin-LR levels in water and aquatic products and aflatoxin B1 levels in daily foods were measured by ELISA, and individual estimated daily intake (EDI) was assessed on the basis of the measurement and questionnaire. We found that participants with abnormal RFIs had a much higher mean level of microcystin-LR EDI than those with normal RFIs and that there was a significant increasing trend for abnormal rates and odds ratios of RFIs with increasing microcystin-LR EDI quartiles (p for trend = 0.000). Compared with the lowest quartile of microcystin-LR exposure, those in the highest quartile had significantly higher risks of abnormal BUN (OR = 1.80, 95% CI = 1.34-2.42), SCr (OR = 4.58, 95% CI = 2.92-7.21), and eGFR (OR = 4.41, 95% CI = 2.55-7.63), respectively, but no higher risk was found in subjects with higher AFB1 exposure. After adjustment for confounding factors, risk associations with microcystin-LR persisted. Consequently, our results suggest that microcystin, rather than aflatoxin, might be one important risk of renal-function impairment.

BACKGROUND: Microcystin-producing Microcystis bloom is a severe water problem in the world. Some reports indicate that chronic exposure to microcystin may result in liver damage in adults, but information on effects in children is limited. OBJECTIVE: We investigated the relationship between microcystin exposure and liver damage in children. METHODS: We measured microcystin concentrations in drinking water and aquatic food (carp and duck) from two lakes and four wells. Participants were 1,322 children 7-15 years of age who obtained drinking water from one of the tested sources, completed questionnaires, and provided blood samples for serum liver enzymes [alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and γ-glutamyltransferase (GGT)] and serum microcystin analysis. Multivariable logistic regression was used to identify risk factors associated with liver damage (two or more abnormal serum enzyme levels in ALT, AST, ALP, or GGT). RESULTS: Microcystin was detected in most samples of water and aquatic food from two lakes. Children who drank water from the lake with the highest microcystin concentrations had a total estimated daily microcystin intake of 2.03 µg, a value much higher than the tolerable daily intake (0.40 µg) proposed by the World Health Organization for children. Hepatitis B virus (HBV) infection, use of hepatotoxic medicines, and microcystin exposure were associated with liver damage. AST and ALP levels were significantly higher in high-microcystin-exposed children than in low-exposed children and unexposed children when participants who were HBV-positive or hepatotoxic medicine users were excluded from the analysis. CONCLUSION: These results suggest that chronic exposure to microcystin may be associated with liver damage in children in the Three Gorges Reservoir Region.

In this paper, bulk polymeric composites with staggered orientation of carbon nanotubes (CNTs) in polymer matrix were prepared by means of a macro layer-by-layer (MLBL) method, while an alternating current (AC) electric field was applied for inducing alignment of the CNTs. Test results verified that there existed a relationship between conductive capacity of the composites and orientation of the CNTs in matrix. Conductivity of the composites containing aligned CNTs represented a dependency on time and testing history on the composite specimens. Among the composites with different orientation types of CNTs in the matrices, the composite specimens including staggered orientation of CNTs in polymer matrix demonstrated the most outstanding electric conductivity and showed similar conductive properties in the two directions of the CNT alignment.

An ideal carbon nanotube (CNT) network structure in poly(methyl methacrylate) (PMMA) can be attained by adjusting simultaneously both evolution of the CNTs alignment in methyl methacrylate (MMA) monomer under alternating current (AC) electric field and polymerization of MMA. Alignment of the CNTs in liquid media is time dependent and can be enhanced by increasing suitably the voltage and frequency of the electric field. There is an optimal time window in which the ideal alignment is achieved after which the alignment degrades and the CNTs start to agglomerate due to the transverse interactions. Viscosity of the MMA increases with its polymerization, and the macro motion of CNTs stops when the viscosity is high enough. Polymerization of the MMA can be accelerated or slowed down by changing polymerization temperature and initiator concentration. If the two courses, the alignment of CNTs and the polymerization of MMA, intersect at a proper point, preferred CNTs alignment will be preserved and the prepared CNT/PMMA composites will be endowed with novel properties.
