RESUMEN

OBJECTIVE: To determine the effective and safe intravenous doses of mesenchymal stem cells (MSCs)-derived microvesicles (MVs) and to elucidate the possible causes of death in mice receiving high-dose MVs. METHODS: MVs were isolated from human MSCs by gradient centrifugation. Mice with collagen-induced arthritis were treated with different doses of intravenous MVs or MSCs. Arthritis severity, white blood cell count, and serum C-reactive protein levels were measured. To assess the safety profile of MSCs and MVs, mice were treated with different doses of MSCs and MVs, and LD50 was calculated. Mouse lungs and heart were assessed by live fluorescence imaging, histopathological measurements, and immunohistochemistry to explore the possible causes of death. Serum concentrations of cTnT, cTnI, and CK-MB were determined by ELISA. With the H9C2 cardiomyocyte cell line,  cellular uptake of MVs was observed using confocal microscopy and cell toxicity was assessed by CCK-8 and flow cytometry. RESULTS: Intravenous treatment with MSCs and MVs alleviated inflammatory arthritis, while high doses of MSCs and MVs were lethal. Mice receiving a maximum dose of MSCs (0.1 mL of MSCs at 109/mL) died immediately, while mice receiving a maximum dose of MVs (0.1 mL of MVs at 1012/mL) exhibited tears, drooling, tachycardia, shortness of breath, unbalanced rollover, bouncing, circular crawling, mania, and death. Some mice died after exhibiting convulsions and other symptoms. All mice died shortly after injecting the maximum dose of MSCs. Histologically, mice receiving high doses of MSCs frequently developed pulmonary embolism, while those receiving high doses of MVs died of myocardial infarction. Consistently, the serum levels of cTnT, cTnI, and CK-MB were significantly increased in the MVs-treated group (P < 0.05). The LD50 of intravenous MVs was 1.60 × 1012/kg. Further, MVs could enter the cell. High doses of MVs induced cell apoptosis, though low concentrations of MVs induced cell proliferation. CONCLUSIONS: Appropriate dosages of MVs and MSCs are effective treatments for inflammatory arthritis while MVs and MSCs overdose is unsafe by causing cardiopulmonary complications.

RESUMEN

Heat is almost everywhere. Unlike electricity, which can be easily manipulated, the current ability to control heat is still highly limited owing to spontaneous thermal dissipation imposed by the second law of thermodynamics. Optical illumination and pressure have been used to switch endothermic/exothermic responses of materials via phase transitions; however, these strategies are less cost-effective and unscalable. Here, we spectroscopically demonstrate the glassy crystal state of 2-amino-2-methyl-1,3-propanediol (AMP) to realize an affordable, easily manageable approach for thermal energy recycling. The supercooled state of AMP is so sensitive to pressure that even several megapascals can induce crystallization to the ordered crystal, resulting in a substantial temperature increase of 48 K within 20 s. Furthermore, we demonstrate a proof-of-concept device capable of programable heating with an extremely high work-to-heat conversion efficiency of ∼383. Such delicate and efficient tuning of heat may remarkably facilitate rational utilization of waste heat.

RESUMEN

Effects of fermentation by Lactobacillus Plantarum NCU116 on the antihypertensive potential of black sesame seed (BSS) and structure characteristics of fermented black sesame seed protein (FBSSP) were investigated. Angiotensin-I-converting enzyme (ACE) inhibition and zinc chelating ability of fermented black sesame seed hydrolysate (FBSSH) reached the highest of 60.78 ± 3.67 % and 2.93 ± 0.04 mg/mL at 48 h and 60 h of fermentation, respectively. Additionally, the antioxidant activities of FBSSH and surface hydrophobicity of FBSSP were increased noticeably by fermentation. The α-helix and ß-rotation of FBSSP tended to decrease and increase, respectively, during fermentation. Correlation analysis indicated strong positive relationships between ß-turn and ACE inhibition activity as well as zinc chelating ability with correlation coefficients r of 0.8976 and 0.8932. Importantly, novel ACE inhibitory peptides LLLPYY (IC50 = 12.20 µM) and ALIPSF (IC50 = 558.99 µM) were screened from FBSSH at 48 h using in silico method. Both peptides showed high antioxidant activities in vitro. Molecular docking analysis demonstrated that the hydrogen bond connected with zinc ions of ACE mainly attributed to the potent ACE inhibitory activity of LLLPYY. The findings indicated that fermentation by Lactobacillus Plantarum NCU116 is an effective method to enhance the antihypertensive potential of BSS.

RESUMEN

Covalent organic frameworks (COFs) with tunable pore sizes and ordered structures are ideal materials for engineering nanofiltration (NF) membranes. However, most of the COFs prepared by solvothermal synthesis are unprocessable powders and fail to form well-structured membranes, which seriously hinders the development of COF NF membranes. Herein, colloidal 2D-COFs with processable membrane formation ability were synthesized by oil-in-water emulsion interfacial polymerization technology. COF NF membranes with tailored thickness and surface charge were fabricated via a layer-by-layer (LBL) assembly strategy. The prepared COF NF membrane achieved precise sieving of dye molecules with high permeance (85 L·m-2·h-1·bar-1). In this work, the strategy of prepared COF NF membranes based on colloid 2D-COF LBL assembly is proposed for the first time, which provides a new idea for the on-demand design and preparation of COF membranes for precise molecular sieving.

RESUMEN

It is not enough to develop an ideal hydrogen evolution reaction (HER) electrocatalysts by single strategy. Here, the HER performances are significantly improved by the combined strategies of P and Se binary vacancies and heterostructure engineering, which is rarely explored and remain unclear. As a result, the overpotentials of MoP/MoSe2 -H heterostructures rich in P and Se binary vacancies are 47 and 110 mV at 10 mA cm-2 in 1 m KOH and 0.5 m H2 SO4 electrolytes, respectively. Especially, in 1 m KOH, the overpotential of MoP/MoSe2 -H is very close to commercial Pt/C at the beginning and even better than Pt/C when current density is over 70 mA cm-2 . The strong interactions between MoSe2 and MoP facilitate electrons transfer from P to Se. Thus, MoP/MoSe2 -H possesses more electrochemically active sites and faster charge transfer capability, which are all in favor of high HER activities. Additionally, Zn-H2 O battery with MoP/MoSe2 -H as cathode is fabricated for simultaneous generation of hydrogen and electricity, which displays the maximum power density of up to 28.1 mW cm-2 and stable discharging performance for 125 h. Overall, this work validates a vigorous strategy and provides guidance for the development of efficient HER electrocatalysts.

RESUMEN

In this study, we characterized a WRKY family member gene, SsWRKY1, which is located in the nucleus and contains multiple stress-related cis-acting elements. In addition, constructed SsWRKY1-overexpressing Arabidopsis thaliana had higher antioxidant enzyme activity and proline content under drought stress conditions, with lower malondialdehyde content and reactive oxygen species (ROS) accumulation, and the expression levels of six stress-related genes were significantly upregulated. This indicates that the overexpression of SsWRKY1 in Arabidopsis thaliana improves resistance to drought stress. SsWRKY1 does not have transcriptional autoactivation activity in yeast cells. The yeast two-hybrid (Y2H) system and the S. spontaneum cDNA library were used to screen 21 potential proteins that interact with SsWRKY1, and the interaction between SsWRKY1 and ATAF2 was verified by GST pull-down assay. In summary, our results indicate that SsWRKY1 plays an important role in the response to drought stress and provide initial insights into the molecular mechanism of SsWRKY1 in response to drought stress.

RESUMEN

Strict carbon emission regulations are set with respect to countries' territorial seas or shipping activities in exclusive economic zones to meet their climate change commitment under the Paris Agreement. However, no shipping policies on carbon mitigation are proposed for the world's high seas regions, which results in carbon intensive shipping activities. In this paper, we propose a Geographic-based Emission Estimation Model (GEEM) to estimate shipping GHG emission patterns on high seas regions. The results indicate that annual emissions of carbon dioxide equivalent (CO2-e) in shipping on the high seas reached 211.60 million metric tonnes in 2019, accounting for about one-third of all shipping emissions globally and exceeding annual GHG emissions of countries such as Spain. The average emission from shipping activities on the high seas is growing at approximately 7.26% per year, which far surpasses the growth rate of global shipping emission at 2.23%. We propose implementation of policies on each high seas region with respect to the main emission driver identified from our results. Our policy evaluation results show that carbon mitigation policies could reduce emissons by 25.46 and 54.36 million tonnes CO2-e in the primary intervention stage and overall intervention stage, respectively, with 12.09% and 25.81% reduction rates in comparison to the 2019 annual GHG emissions in high seas shipping.

RESUMEN

Depression is a common psychiatric symptom in patients with Sjögren's syndrome (SS). Prevalence of depression in SS patients is significantly higher than that in the general population. Increasing evidence suggests a crucial role of gut-microbiota-brain axis in depression. In this study, we investigated whether non-obese diabetic (NOD) mice, a widely used animal model of SS, exhibit depression-like phenotypes and abnormal composition of gut microbiota. Eleven-week-old NOD mice spontaneously exhibited SS-related symptoms without pancreatic destruction. NOD mice displayed depression-like behaviors, decreased expression of synaptic proteins in the prefrontal cortex (PFC), and abnormal composition of gut microbiota. Interestingly, SS-related proinflammatory factors in the submandibular gland (SMG) and autoantibodies (anti-SSA and anti-SSB) in the plasma were correlated with the expression of synaptic proteins in the PFC or depression-like behaviors. In addition, there were correlations between the relative abundance of microbiota and SS-related symptoms (or depression-related phenotypes). These data suggest that SS-related symptoms and abnormal composition of gut microbiota may play a role in depression-like phenotypes in NOD mice through gut-microbiota-brain axis.

RESUMEN

The NAC (NAM, ATAF1/2, and CUC2) transcription factor family is one of the largest families unique to plants and is involved in plant growth and development, organs, morphogenesis, and stress responses. The NAC family has been identified in many plants. As the main source of resistance genes for sugarcane breeding, the NAC gene family in the wild species Saccharum spontaneum has not been systematically studied. In this study, 115 SsNAC genes were identified in the S. spontaneum genome, and these genes were heterogeneously distributed on 25 chromosomes. Phylogenetic analysis divided the SsNAC family members into 18 subgroups, and the gene structure and conserved motif analysis further supported the phylogenetic classification. Four groups of tandemly duplicated genes and nine pairs of segmentally duplicated genes were detected. The SsNAC gene has different expression patterns at different developmental stages of stems and leaves. Further qRT-PCR analysis showed that drought, low-temperature, salinity, pathogenic fungi, and other stresses as well as abscisic acid (ABA) and methyl jasmonate (MeJA) treatments significantly induced the expression of 12 SsNAC genes, indicating that these genes may play a key role in the resistance of S. spontaneum to biotic and abiotic stresses. In summary, the results from this study provide comprehensive information on the NAC transcription factor family, providing a reference for further functional studies of the SsNAC gene.

RESUMEN

Liquid crystal devices using organic molecules are nowadays widely used to modulate transmitted light, but this technology still suffers from relatively weak response, high cost, toxicity and environmental concerns, and cannot fully meet the demand of future sustainable society. Here, an alternative approach to color-tunable optical devices, which is based on sustainable inorganic liquid crystals derived from 2D mineral materials abundant in nature, is described. The prototypical 2D mineral of vermiculite is massively produced by a green method, possessing size-to-thickness aspect ratios of >103 , in-plane magnetization of >10 emu g-1 , and an optical bandgap of >3 eV. These characteristics endow 2D vermiculite with sensitive magneto-birefringence response, been several orders of magnitude larger than organic counterparts, as well as capability of broad-spectrum modulation. The finding consequently permits the fabrication of various magnetochromic or mechanochromic devices with low or even zero-energy consumption during operation. This work creates opportunities for the application of sustainable materials in advanced optics.

RESUMEN

Interfacial solar-driven evaporation provides one of the most promising green and sustainable technologies to deal with the knotty water crisis by extracting vapor from a variety of water sources powered by solar energy. Advanced photothermal materials play critical roles in interfacial solar-driven evaporation by photothermal conversion and heat localization. Herein, inspired by the unique hierarchical structure and light-harvesting function of diatoms, we propose a novel photothermal material with a diatom-like hierarchical nanostructure derived from TiO2-PANi-decorated bilayer melamine foam (TiO2-PANi@MF) for solar-driven clean water generation. The diatom-like hierarchical nanostructured TiO2-PANi@MF can realize full-spectrum light absorption and photothermal conversion by enhancing multiple light reflection and light scattering. Thanks to the diatom-like hierarchical nanostructure, TiO2-PANi@MF not only impressively achieves an evaporation rate of 2.12 kg m-2 h-1 under 1 sun irradiation but also shows a high solar steam conversion efficiency up to 88.9%. Notably, the TiO2-PANi composite also endows TiO2-PANi@MF with photocatalytic degradation capability. Apart from the excellent steam generation capability, optimized TiO2-PANi@MF also provides the high photocatalytic efficiency of dye degradation and maintains a high evaporation rate of more than 2 kg m-2 h-1. We believe that the proposed photothermal material with a diatom-like hierarchical nanostructure can envision promising practical applications in seawater desalination and sewage purification.

RESUMEN

The aim of this study was to investigate the lactic acid bacteria (LAB) and yeast community from home-made sauerkraut collected from Southwest China through culture-dependent and culture-independent technology. Forty-eight samples of home-made sauerkraut were collected from households at three different locations in Southwest China. The pH, total acidity and salt contents among these fermented vegetables were 3.69 ± 0.42, 0.86 ± 0.43 g/100 ml, and 3.86 ± 2.55 g/100 ml, respectively. The number of lactic acid bacteria (LAB) and yeasts were 7.25 ± 1.05 log10 colony-forming units (CFU)/ml and 3.74 ± 1.01 log CFU/ml, respectively. A total of 182 LAB and 81 yeast isolates were identified. The dominant isolates were Lactobacillus plantarum, L. brevis, Pediococcus ethanolidurans, Pichia membranifaciens, P. fermentans and Kazachstania bulderi. Denaturing gradient gel electrophoresis (DGGE) showed that L. plantarum, uncultured Lactobacillus sp, P. ethanolidurans, and K. exigua were the predominant microflora. Our studies demonstrated that the DGGE technique combined with a culture-dependent method is very effective for studying the LAB and yeast community in Chinese traditional fermentation vegetables. The results will give us an understanding of LAB and yeast community of Chinese sauerkraut and improve the knowledge of LAB and yeast community of Chinese sauerkraut.

RESUMEN

Superwetting membranes with opposite wettability to oil and water have drawn intense attention in recent years for oil/water separation. Superhydrophilic and underwater superoleophobic membranes have shown unique advantages in the efficient treatment of oily wastewater containing oil-in-water emulsions. Facile interfacial engineering and microstructural design of the hierarchical architectures and the hydrophilic chemistry is of significance but still challenging. In this study, a hydrophilic hierarchical hybrid layer derived from metal-phenolic network (MPN)/metal-organic framework (MOF) synergy is constructed on the membrane surface via a proposed coordination-directed alternating assembly strategy. The assembly of MPN multilayers provides a hydrophilic chemical basis, and the assembly of MOF nanocrystals provides a hierarchical structural basis. Notably, the coordination interfacial interaction enables the formation of well-defined hydrophilic hierarchical architectures. The obtained membrane is thus endowed with robust superhydrophilicity, underwater superoleophobicity, and anti-oil-adhesion capability, which make it capable of highly efficient oil-water separation with high water permeance (above 6300 L/m2 h), high oil rejection (above 99.4%), and recyclable antifouling property. The high performance of the developed superwetting membrane makes it a competitive candidate for oil/water separation. Additionally, the demonstrated MPN/MOF assembly strategy may offer new prospects for the facile and versatile design of other superwetting materials.

RESUMEN

The development of antifouling membranes plays a vital role in the widespread application of membrane technology, and the hybridization strategy has attracted a significant amount of attention for antifouling applications. In this work, TA/PEI@TiO2 hierarchical hybrid nanoparticles (TPTi HHNs) are first synthesized through a simple strategy combining the multiple catechol chemistries of phenolic tannic acid (TA) with the biomimetic mineralization chemistry of titania. The TPTi HHNs are used as nanofillers to prepare PVDF/TPTi hybrid membranes. The TPTi HHNs endow the membrane with higher porosity, hierarchical roughness, greater hydrophilicity, and underwater superoleophobicity. Upon TPTi HHN loading, the PVDF/TPTi hybrid membranes exhibit enhanced antifouling performance. The flux recovery ratio can reach 92% when utilized to separate oil-in-water emulsion. Even being applied to the three-cycle filtration of oil-in-water emulsion with much higher concentration, the PVDF/TPTi membrane can still maintain a high flux recovery ratio about 85%. This study will provide a facial polyphenol-based platform to fabricate antifouling hybrid nanofillers and antifouling hybrid membranes with promising applications in oil/water separation.

RESUMEN

Auxiliary fields have been used to enhance the performance of needle electrospinning. However, much less has been reported on how auxiliary fields affect needleless electrospinning. Herein, we report a novel needleless electrospinning technique that consists of an aerodynamic field and a second electric field. The second electric field is generated by setting two grounded inductive electrodes near the spinneret. The two auxiliary fields have to be applied simultaneously to ensure working of the electrospinning process. A synergistic effect was observed between inductive electrode and airflow. The aerodynamic-electric auxiliary field was found to significantly increase fiber production rate (4.5 g h-1), by 350% in comparison to the setup without auxiliary field (1.0 g h-1), whereas it had little effect on fiber diameter. The auxiliary fields allow running needleless electrospinning at an applied voltage equivalent to that in needle electrospinning (e.g. 10-30 kV). The finite element analyses of electric field and airflow field verify that the inductive electrodes increase electric field strength near the spinneret, and the airflow assists in fiber deposition. This novel needleless electrospinning may be useful for development of high-efficiency, low energy-consumption nanofiber production systems.

RESUMEN

A novel reverse osmosis (RO) composite membrane, poly(amide-urethane-imide@Ag) (PAUI@Ag), was prepared on a polysulfone supporting film through two-step interfacial polymerization. First, in the 1st interfacial polymerization procedure, a new tri-functional crosslinking agent with -OCOCl and -COCl groups, 5-choroformyloxyisophaloyl chloride (CFIC), was reacted with 4-methyl-phenylenediamine (MMPD) without curing treatment to obtain the poly(amide-urethane) base membrane with a CFIC-MMPD precursor separation layer. And then N,N'-dimethyl-m-phenylenediamine (DMMPD) with nano-Ag particle dispersion was introduced onto the base membrane to further construct a CFIC-DMMPD modified ultrathin separation layer via the 2nd interfacial polymerization. Thus, the PAUI@Ag RO membrane with poly(amide-urethane-imide) bi-layer skin was obtained. The membrane was characterized for the chemical composition of separation layer, the membrane cross-section structure and the membrane surface morphology. Permeation experiment was employed to evaluate the PAUI@Ag membrane performance including salt rejection rate and water flux. The results revealed that the PAUI@Ag membrane composed the highly cross-linked separation layer with entire ridges and valleys, small surface roughness, and well dispersed nano-Ag particles. Upon exposure of the membranes to high concentration of free chlorine solutions, the PAUI@Ag RO membrane showed a slightly less chlorine-resistant property compared with the nascent PAUI RO membrane, but was still superior to the conventional polyamide MPD-TMC RO membrane, meanwhile it processed higher anti-biofouling property.

RESUMEN

One of the greatest challenges to the sustainability of modern society is an inadequate supply of clean water. Due to its energy-saving and cost-effective features, membrane technology has become an indispensable platform technology for water purification, including seawater and brackish water desalination as well as municipal or industrial wastewater treatment. However, membrane fouling, which arises from the nonspecific interaction between membrane surface and foulants, significantly impedes the efficient application of membrane technology. Preparing antifouling membranes is a fundamental strategy to deal with pervasive fouling problems from a variety of foulants. In recent years, major advancements have been made in membrane preparation techniques and in elucidating the antifouling mechanisms of membrane processes, including ultrafiltration, nanofiltration, reverse osmosis and forward osmosis. This review will first introduce the major foulants and the principal mechanisms of membrane fouling, and then highlight the development, current status and future prospects of antifouling membranes, including antifouling strategies, preparation techniques and practical applications. In particular, the strategies and mechanisms for antifouling membranes, including passive fouling resistance and fouling release, active off-surface and on-surface strategies, will be proposed and discussed extensively.

RESUMEN

Graphene oxide (GO) is an emerging kind of building block for advanced membranes with tunable passageway for water molecules. To synergistically manipulate the channel and surface structures/properties of GO-based membranes, the different building blocks are combined and the specific interfacial interactions are designed in this study. With vacuum-assisted filtration self-assembly, palygorskite nanorods are intercalated into adjacent GO nanosheets, and GO nanosheets are assembled into laminate structures through π-π stacking and cation cross-linking. The palygorskite nanorods in the free-standing GOP nanohybrid membranes take a 3-fold role, rendering enlarged mass transfer channels, elevating hydration capacity, and creating hierarchical nanostructures of membrane surfaces. Accordingly, the permeate fluxes from 267 L/(m(2) h) for GO membrane to 1867 L/(m(2) h) for GOP membrane. The hydration capacity and hierarchical nanostructures synergistically endow GOP membranes with underwater superoleophobic and low oil-adhesive water/membrane interfaces. Moreover, by rationally imparting chemical and physical joint defense mechanisms, the GOP membranes exhibit outstanding separation performance and antifouling properties for various oil-in-water emulsion systems (with different concentration, pH, or oil species). The high water permeability, high separation efficiency, as well as superior anti-oil-fouling properties of GOP membranes enlighten the great prospects of graphene-based nanostructured materials in water purification and wastewater treatment.
