RESUMEN

BACKGROUND@#The occurrence and development of lung cancer are closely linked to epigenetic modification. Abnormal DNA methylation in the CpG island region of genes has been found in many cancers. Protein kinase C delta binding protein (PRKCDBP) is a potential tumor suppressor and its epigenetic changes are found in many human malignancies. This study investigated the possibility of PRKCDBP methylation as a potential biomarker for non-small cell lung cancer (NSCLC).@*METHODS@#We measured the methylation levels of PRKCDBP in the three groups of NSCLC tissues. Promoter activity was measured by the dual luciferase assay, with 5'-aza-deoxycytidine to examine the effect of demethylation on the expression level of PRKCDBP.@*RESULTS@#The methylation levels of PRKCDBP in tumor tissues and 3 cm para-tumor were higher than those of distant (>10 cm) non-tumor tissues. Receiver operating characteristic (ROC) curve analysis between tumor tissues and distant non-tumor tissues showed that the area under the line (AUC) was 0.717. Dual luciferase experiment confirmed that the promoter region was able to promote gene expression. Meanwhile, in vitro methylation of the fragment (PRKCDBP_Me) could significantly reduce the promoter activity of the fragment. Demethylation of 5'-aza-deoxycytidine in lung cancer cell lines A549 and H1299 showed a significant up-regulation of PRKCDBP mRNA levels.@*CONCLUSIONS@#PRKCDBP methylation is a potential and promising candidate biomarker for non-small cell lung cancer.

RESUMEN

We report the first local transmission of the SARS-CoV-2 Delta variant in mainland China. All 167 infections could be traced back to the first index case. Daily sequential PCR testing of the quarantined subjects indicated that the viral loads of Delta infections, when they first become PCR+, were on average [~]1000 times greater compared to A/B lineage infections during initial epidemic wave in China in early 2020, suggesting potentially faster viral replication and greater infectiousness of Delta during early infection. We performed high-quality sequencing on samples from 126 individuals. Reliable epidemiological data meant that, for 111 transmission events, the donor and recipient cases were known. The estimated transmission bottleneck size was 1-3 virions with most minor intra-host single nucleotide variants (iSNVs) failing to transmit to the recipients. However, transmission heterogeneity of SARS-CoV-2 was also observed. The transmission of minor iSNVs resulted in at least 4 of the 30 substitutions identified in the outbreak, highlighting the contribution of intra-host variants to population level viral diversity during rapid spread. Disease control activities, such as the frequency of population testing, quarantine during pre-symptomatic infection, and level of virus genomic surveillance should be adjusted in order to account for the increasing prevalence of the Delta variant worldwide.

Pheochromocytoma/paraganglioma(PPGL) was a kind of neuroendocrine tumor that derived from chromaffin tissue, which seems to be an important etiology of secondary hypertension. With the development of molecular detection technology, at least 17 kinds of pathogenic genes of PPGL has been discovered, which is related to 35%-40% PPGL, and about 40% malignant PPGL is associated with SDHB gene mutation. In this study, we reported a case with a novel splicing mutation of SDHB gene induced paraganglioma.

RESUMEN

Nearly a decade of curriculum update in American medical colleges and universities are summarized in this paper. This paper introduces the situation of sampling survey of colleges and universities. The results show the diversity of medical curriculum integration, reflecting the concept of multi-disciplinary integration of basic disciplines, clinical disciplines and public health and social sciences, and giving experiences such as early clinical practice, providing personalized talent cultivation approaches, and designing competency-oriented curriculum system. Combined with the current situation of medical education curriculum reform in China, it's suggested to explore the construction of medical curriculum system with Chinese characteristics, so as to contribute to the cultivation of excellent physicians for implementing Healthy China Initiative.

OBJECTIVE@#To analyze the relationship between and gene methylation with aging in the general population.@*METHODS@#We collected peripheral blood samples from 284 male and 246 female healthy subjects for detection of methylation levels of and genes using quantitative methylation-specific PCR (qMSP). The relationship between the methylation levels of and genes and aging was analyzed using Spearman or Pearson correlation test.@*RESULTS@#We found a significant positive correlation between the methylation levels of the two genes in these subjects ( < 0.05). In the overall population as well in the female subjects, methylation was found to be inversely correlated with age ( < 0.05). The methylation levels of and genes were inversely correlated with TG, ApoE, Lp(a) and AST in the overall population ( < 0.05). In both the female and male subjects, the methylation levels of the two genes were inversely correlated with Lp(a) ( < 0.05). In the male subjects, methylation was inversely correlated with AST ( < 0.05), while methylation was inversely correlated with HDL and ApoE ( < 0.05). In the female subjects, methylation was positively correlated with LDL and inversely correlated with ApoE and AST ( < 0.05).@*CONCLUSIONS@#The methylation levels of and are closely related to age and the levels of multiple proteins in healthy subjects.

OBJECTIVE: To evaluate the potential beneficial effects of Inonotus obliquuspolysaccharides (IOP) on the alleviation of physical fatigue in mice. METHODS: Sixty-four male mice were randomly divided into four groups (n = 16 per group). Mice were orally administered IOP for a period of 14 days at 0, 100, 200 and 300 mg/kg/d, and were assigned to the control, IOP-100, IOP-200, and IOP-300 groups, respectively by the random number table method. Mice in the control group received an oral administration of sterile distilled water. A forced swimming test was performed for 8 mice per group at one hour after the last treatment. The other 8 mice in each group swam for 30 min. Blood, liver and muscle samples were taken after resting for 30 min. Levels of blood urea nitrogen and lactate, as well as glycogen contents of the liver and muscle were measured. Morphology of liver was observed by light microscopy. RESULTS: IOP extended the swimming time of mice, and increased the glycogen content of liver and muscle, but decreased blood lactic acid and serum urea nitrogen levels, IOP had no toxic effects on major organs such as the liver as assessed by histopathological examinations. CONCLUSION: IOP might be a potential anti-fatigue pharmacological agent.

Objective To study the protective effect and mechanism of Inonotus obliquus polysaccharide on liver injury induced by isoniazid( INH) combined with rifampicin( RFP) in mice. Methods Forty-eight mice were randomly divided into six groups ( control, model, huganpian, Inonotus obliquus polysaccharide at low, middle, and high dose ) . The mice were administered orally with isoniazid and rifampicin simultaneously except the control. After 2 h,the control and model groups were administered with normal saline,and others were treated with huganpian and Inonotus obliquus polysaccharide,respectively,once a day for 2 weeks. The liver index and serum levels of ALT and AST,malondialdehyde(MDA),superoxide dismutase(SOD) and glutathione peroxidase( GPx) were measured. Pathology examination of liver tissue was performed. Results The activities of ALT and AST, liver index, content of MDA in liver homogenate of mice treated with Inonotus obliquus polysaccharide and huganpian decreased obviously, while the liver SOD activity increased. Histopathological exzamination showed that Inonotus obliquus polysaccharide alleviated the degeneration and necrosis of hepatic cells. Conclusion Inonotus obliquus polysaccharide shows protective effects on mice with hepatic toxic injury induced by isoniazid and rifampicin,which maybe due to its antioxidation effect.

Objective To develop rescue device for pilots in air accidents and solve the problem that it's difficult for medical staff to carry out on-board rescue because of the height from the ground to the cockpit edge of the fighter plane. This device with hydraulic system is multi-use: it can be joined to the plane as a ladder and tied in an ambulance as a stretcher. The medical support level of air force dump and station will be improved after using it. Methods 6082 aluminium alloy for aeronautic with advanced and matured technology was utilized to weld the principal part. The fissions can be fitted together and drawn out. The cantilever with hanger was applied. The active and diagonal crutches and the separable composite structure were also used. Results With this device, two staff can board the fighter plane to carry the wounded pilot to the sliding seat which is stably controlled by hydraulic system and then after quick sliding and separating from the device, the seat become a stretcher, and thus it can avoid the second carrying to the pilot. Using this device, the rescue work on-board become faster, more steady and safer. Conclusions This device can provide on-board rescue for pilots of forced landing in air accidents and it can be configured in a new kind of outfield ambulance for both military and civilian purposes. Because of being simple in frame and light in weight, it is easy to take or transport.
