BACKGROUND: Elevated levels of reactive oxygen species may contribute to the gradual decline in muscle strength over time. Although caffeine and its metabolites have antioxidant properties that can mitigate oxidative stress, the association of caffeine and its metabolites with muscle strength remains unknown. AIM: To investigate whether caffeine metabolites in urine are associated with muscle strength in young and older adults. METHODS: A cross-sectional study was conducted with 1145 individuals aged over 20 years (n = 801 < 60 years and n = 344 ≥ 60 years) from the National Health and Nutrition Examination Survey (NHANES) 2011-2012. Muscle strength was assessed using a handgrip dynamometer, and combined grip strength was determined by summing the highest value from each hand. Caffeine and its metabolites in urine were quantified using ultra-high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (1-methyluric acid, 3-methyluric acid, 7-methyluric acid, 1,3-dimethyluric acid, 1,7-dimethyluric acid, 3,7-dimethyluric acid, 1,3,7-trimethyluric acid, 1-methylxanthine, 3-methylxanthine, 7-methylxanthine, 1,3-dimethylxanthine, 1,7-dimethylxanthine, 3,7-dimethylxanthine, 1,3,7-trimethylxanthine, 5-acetylamino-6-amino-3-methyluracil). Linear regression analyses were performed to determine the association of caffeine and its metabolites with muscle strength in young and older adults, adjusting for confounders. RESULTS: Positive associations between muscle strength and levels of 7-methyluric acid (ß = 0.029; p = 0.021), 1,3-dimethyluric acid (ß = 0.008; p = 0.004), 3,7-dimethyluric acid (ß = 0.645; p = 0.012), 3-methylxanthine (ß = 0.020; p = 0.002), 7-methylxanthine (ß = 0.020; p = 0.006), 1,3-dimethylxanthine (theophylline) (ß = 0.030; p = 0.004) and 3,7-dimethylxanthine (theobromine) (ß = 0.035; p = 0.029) were observed in older adults. In contrast, no such associations were noted in young adults. CONCLUSION: Our study indicates a positive association between certain caffeine metabolites in urine and muscle strength in older adults, but not in younger individuals. These findings indicate that specific caffeine metabolites may contribute to an antioxidant role especially in older adults.

AIM: Determining gastrointestinal parasites' frequency in non-human primates (NHPs) and handlers in different Brazilian institutions, and associate it with management information. METHODS: Different institutions in São Paulo (A), Brasília (B), Rio de Janeiro (C), Pará (D) and Santa Catarina (E) were included in the study. Fecal samples were processed by using coproparasitological techniques; information about NHP handling and professionals' routine were collected through the use of questionnaires. RESULTS: In total, 33.1% of 511 NHP samples were positive for parasites; the Entamoebidae Group and Strongyloides sp.-compatible eggs were the most detected protozoa and helminths, respectively. The Entamoebidae Group was mainly diagnosed in Alouatta from Institutions E and D, and was also the only parasite detected in handlers. Strongyloides-compatible eggs were mostly evident in fecal samples collected from Cebidae from Institutions B and D. Some risk factors associated with parasite infection were a high number of animals in the same enclosure and their use for research protocol purposes, whereas quarantine, a post-infection vacant period in enclosures and antiparasitic supply were categorized as protective factors against these agents' infection. CONCLUSIONS: Parasites showing different transmission routes but concomitantly circulating in NHP institutions located in different Brazilian regions were identified in the current study, with an emphasis on those presenting zoonotic potential.

BACKGROUND AND AIMS: Iron deficiency tendency in individuals with hyperglycemia influences the HbA1c levels' ability to estimate the stationary blood glucose levels. This study investigated the associations of iron status indicators and HbA1c levels with anthropometric, inflammatory, regulatory, metabolic, and hematologic variables in women with hyperglycemia to most widely characterize this iron deficiency tendency. METHODS: A total of 143 volunteers (68 with normoglycemia and 75 with hyperglycemia) participated in this cross-sectional study. Mann-Whitney test was used to compare groups, and the search for associations between pairs of variables used the Spearman correlation method. RESULTS: In women with hyperglycemia, decreased plasma iron level is associated directly with increased HbA1c (p < 0.001), and these changes are both related to C-reactive protein elevation (p = 0.02 and p < 0.05, respectively) and involve decreased mean hemoglobin concentration (p < 0.01 and p < 0.01), which in turn, involves enhanced osmotic stability (dX) (p < 0.05) and volume variability (RDW) (p < 0.0001) of erythrocytes, as well as decreased indirect bilirubin/total bilirubin ratio (p = 0.04). This indirect bilirubin/total bilirubin decrease, which expresses decreased hemoglobin catabolism, does not seem to be solely a process associated with diminished intracellular concentrations of this protein (p = 0.04) since it is associated with CRP elevation (p = 0.03) and reduced LDL cholesterol (p < 0.0001). CONCLUSIONS: In women with hyperglycemia, decreased plasma iron levels were associated with inflammatory status and involved increased HbA1c, osmotic stability, and volume variability of red blood cells.

Quick and reliable mass testing of infected people is an effective tool for the contingency of SARS-CoV-2. During the COVID-19 pandemic, Point-of-Care (POC) tests using Loop-Mediated Isothermal Amplification (LAMP) arose as a useful diagnostic tool. LAMP tests are a robust and fast alternative to Polymerase Chain Reaction (PCR), and their isothermal property allows easy incorporation into POC platforms. The main drawback of using colorimetric LAMP is the reported short-term stability of the pre-mixed reagents, as well as the relatively high rate of false-positive results. Also, low-magnitude amplification can produce a subtle color change, making it difficult to discern a positive reaction. This paper presents Hilab Molecular, a portable device that uses the Internet of Things and Artificial Intelligence to pre-analyze colorimetric data. In addition, we established manufacturing procedures to increase the stability of colorimetric RT-LAMP tests. We show that ready-to-use reactions can be stored for up to 120 days at -20 °C. Furthermore, we validated both the Hilab Molecular device and the Hilab RT-LAMP test for SARS-CoV-2 using 581 patient samples without any purification steps. We achieved a sensitivity of 92.93% and specificity of 99.42% (samples with CT ≤ 30) when compared to RT-qPCR.

The aim of this observational, longitudinal and retrospective study was to evaluate the fidelity of virtual surgical planning (VSP) performed on Dolphin Imaging & Management Solutions® 11.95 software on hard tissues, using the tools of the open-source software OrtogOnBlender - Blender3D. For this, linear, angular and 7-point anatomical measurements of the skeletal profile were used, and the discrepancies between the VSP and the result after bimaxillary orthognathic surgery were calculated. Pre- and postoperative cone beam CT (CBCT) scans of 43 consecutive patients with class II and III skeletal deformities were evaluated and the results of the VSP were compared to the 1-month postoperative results. All overlapping points presented values within the range considered clinically irrelevant (< 2 mm and < 4°) and differences were not significant (p > 0.05). The comparison of anatomical points showed lower values (≤ 2.11 mm) in point A of class II. For hard tissues, the comparison between VSP and 1-month postoperative tomography demonstrated the faithful results of virtual planning using this software.

Schistosomiasis, a tropical disease caused by flatworms, may affect the liver, spleen, bladder, and intestine. Casearia sylvestris Swartz, a medicinal plant, displays antiprotozoal, antimicrobial, antifungal, and antiulcer activities. We have evaluated the in vitro schistosomicidal activity of two C. sylvestris varieties against Schistosoma mansoni adult worms at concentrations between 12.5 and 200 µg/mL. At 100 and 200 µg/mL, the ethanolic C. sylvestris var. sylvestris leaf extract enriched in casearin-like diterpenes eliminated 100 % of the parasites after incubation for 72 h and 48 h, respectively, whilst the same extract at 200 µg/mL eliminated 96 %, 100 %, and 100 % of the parasites after incubation for 24, 48, and 72 h, respectively. On the other hand, the hydroalcoholic C. sylvestris var. lingua leaf extract at 200 µg/mL eliminated 60.4 and 66.7 % of the parasites after incubation for 48 and 72 h, respectively. The presence of casearin-like diterpenes and glycosylated flavonoids was confirmed based on chromatographic techniques and mass spectrometry data.

INTRODUCTION: The aim of the present study was to evaluate the presence of the median perforating canal (MPC) and its morphometric measurements in Cone Beam CT (CBCT) scans of adult patients, correlating the findings with sex, age and skeletal facial patterns. MATERIAL AND METHODS: 717 CBCT scans were selected from a Brazilian population and the presence of the MPC was recorded. MPC diameter was measured in three points: lingual, medial and buccal. To determine the correlation between MPC presence and sex, age and ANB angle classifications the Chi-square test was performed. MPC diameters were related to sex, age and skeletal discrepancies using Mann-Whitney U and Kruskal-Wallis tests. The level of significance adopted was 5%. RESULTS: Of the 717 CBCT scans analyzed, the MPC was present in 181 patients (25.24%), 279 (38.91%) men and 438 (61.09%) women. The mean age was 37.91 (±14.46) years old, ranging from 18 to 88 years old. There were no differences in the presence of MPC between women and men (p = 0.85), between skeletal discrepancies (p = 0.09) and age groups (p = 0.94). Regarding the MPC diameter, statistical difference was found only for the medial diameter between sexes (p = 0.04), with the highest values in men. CONCLUSION: The MPC was detected in 25.24% of the studied population, with significant differences only for its medial diameter in men.

This study aimed to investigate the relationship between red cell distribution width (RDW) and erythrocyte osmotic stability in non-diabetic and diabetic individuals in both sexes. The study sample (N = 122) was constituted by 53 type 2 diabetics (DM) and 69 non-diabetics (ND), being 21 and 22 men in each group, respectively. The osmotic stability of erythrocytes was obtained by the variation in saline concentration (dX) capable of determining hypoosmotic lysis. Higher RDW values and lower serum iron concentrations were found in the diabetic group when compared to the non-diabetic volunteers. In the group of diabetic women, RDW was positively correlated with the reticulocyte index, and both RDW and dX were negatively correlated with iron, haemoglobin, transferrin saturation index, mean corpuscular haemoglobin and mean corpuscular haemoglobin concentration. In all the groups studied, RDW was positively correlated with dX, especially in the diabetic group, where the correlation was the strongest. RDW elevation in both women and men with type 2 diabetes mellitus was associated with decreased serum iron indicators. Furthermore, RDW has a similar meaning to dX, as small erythrocytes have less haemoglobin, resulting in both an increase of RDW and dX.

Iron (Fe) toxicity is one of the most frequent abiotic stresses in rice, as it affects from 15% to 30% of the total production. Brassinosteroids (BRs), including 24-epibrassinolide (EBR), regulate ion homeostasis and improve the antioxidant system. The aim of this research was to determine whether EBR can contribute to the tolerance of rice plants exposed to Fe toxicity and to evaluate the possible effect on anatomical characteristics, nutrient concentrations, the antioxidant system, and gas exchange. The experiment was randomized with four treatments, two with different concentrations of Fe (250 and 6250 µM, control and toxicity, respectively) and these were either supplied with EBR or not (0 and 10 nM EBR, described as -EBR and +EBR, respectively). Treating plants grown under Fe toxic conditions with EBR caused an 70% increase in root aerenchyma area, compared to plants without steroid treatment. Our results revealed that EBR treatment could mitigate the deleterious effects of Fe toxicity in rice plants, by modulating the aerenchyma area, which contributes to the formation of an oxidative barrier and reduce the Fe mobilization at the root surface. Plants that were exposed to Fe toxic concentrations and treated with EBR showed (1) an increase in the enzyme activities of superoxide dismutase, catalase, ascorbate peroxidase and peroxidase, (2) mitigation of oxidative damage and (3) increased scavenging of reactive oxygen species. Finally, EBR alleviated the negative impacts induced by excess Fe on the net photosynthetic rate and the instantaneous carboxylation efficiency. These benefits were directly related to higher electron transport and stomatal density and indirectly linked to the protection mechanism exercised by the antioxidant enzymes on photosynthetic machinery. We conclude that EBR is able to confer tolerance to Fe toxicity in rice plants.

Cadmium (Cd2+) soil pollution is a global environmental problem caused by the high toxicity of Cd. 24-Epibrassinolide (EBR) is a biodegradable plant steroid involved in response modulation to biotic and abiotic stresses. The aim of this study was to evaluate if the leaf-application of EBR improves the gas exchange and possible repercussions on leaf anatomy in young Eucalyptus urophylla plants exposed to Cd toxicity. The experiment involved six treatments, which included three Cd concentrations (0, 450, and 900 µM) and two EBR concentrations (0 and 100 nM, described as - EBR and + EBR, respectively). Plants exposed to Cd toxicity suffered decreases in leaf anatomical and gas exchange parameters. However, the plants treated with EBR + 900 µM Cd showed an increase of 46%, 40%, and 54% in the net photosynthetic rate, water-use efficiency, and instantaneous carboxylation efficiency, respectively. The EBR application-induced improvements in gas exchange parameters, causing beneficial effects on the photosynthetic apparatus, mainly the effective quantum yield of photosystem II (PSII) photochemistry and electron transport rate. Furthermore, this steroid mitigated the effect of Cd toxicity on leaf anatomical variables, more specifically palisade and spongy parenchyma, which are intrinsically related to stomatal density, and stimulated the net photosynthetic rate of plants.

AIM: The aim of this study was to evaluate the effects of maternal exposure to a high-fat diet associated with neonatal administration of kaempferol on somatic growth, biochemical profile and feeding behavior in offspring. MATERIALS AND METHODS: Wistar rats were distributed according to diet during pregnancy and lactation into Control (C; 3.4 kcal/g; 12% kcal/lipids) or High-fat (HFD; 4.6 kcal/g; 51% kcal/lipids) groups. In the offspring, vehicle (V) or kaempferol (K, 1 mg/kg) were administered from the 8th until the 21st postnatal day (PND). Maternal body weight (BW), caloric intake and adiposity were measured. In the offspring, somatic growth parameters were evaluated on the 7th, 14th, 21st, 25th and 30th PND, except for BW, which was measured from the 8th to the 21st and from the 25th to the 30th PND. Feeding behavior was assessed by food intake and behavioral satiety sequence (BSS) on the 30th PND. The biochemical profile and relative weight of adipose tissue of offspring were also measured. KEY FINDINGS: Dams exposed to HFD showed no difference in body weight and caloric intake but exhibited increased adiposity. Neonatal administration of kaempferol increased body weight after weaning and somatic growth in the offspring of HFD dams. Neonatal kaempferol also reduced adiposity and serum creatinine levels in offspring. Neither maternal diet nor kaempferol altered offspring feeding behavior. SIGNIFICANCE: Neonatal administration of kaempferol promotes increased somatic growth post-weaning, reduces adiposity, and does not alter feeding behavior in offspring from high-fat dams.

Objective: This observational and retrospective study evaluated the prevalence and characteristics of odontogenic cysts, outlining the epidemiological profile of these lesions. Material and methods: The following data were collected from the medical records of patients with a diagnosis confirmed by microscopy: age, sex, ethnicity, anatomical location, histopathological diagnosis and treatment, between 1995 and 2018. Results: Of the 70 cysts, 75.7% were inflammatory and 24.2% development. The inflammatory periapical cyst was the most prevalent (62.8%) followed by the dentigerous cyst (71.0%). The lateral radicular cysts and odontogenic keratocysts obtained the same percentage (5.7%), followed by the residual cyst (4.3%), paradental (2.8%) and the orthokeratinized variable (1.5%). Women were slightly more affected (1.12: 1) with greater involvement of leucodermas (80%). Age and anatomical location differed according to each lesion, although the anterior maxilla region was more affected. Enucleation was the most used treatment (75.7%), except for odontogenic keratocysts, in which marsupialization was performed in 75% of the cases. Conclusion: Epidemiological data provide an important insight into the prevalence, extent and severity of these lesions, allowing early diagnosis and prevention, in order to guarantee the reduction of cases and improve the quality of life of the population.


Objetivo: Avaliar a prevalência e características dos cistos odontogênicos de forma observacional e retrospectiva, delineando o perfil epidemiológico destas lesões. Material e Métodos: Foram coletados os seguintes dados dos prontuários de pacientes com diagnóstico confirmado microscopicamente: idade, sexo, etnia, localização anatômica, diagnóstico histopatológico e tratamento, entre os anos de 1995 a 2018. Resultados: Dos 70 cistos, 75,7% eram inflamatórios e 24,2% de desenvolvimento. O cisto periapical inflamatório foi o mais prevalente (62,8%) seguido pelo cisto dentígero (71,0%). O cisto radicular lateral e queratocisto odontogênico obtiveram o mesmo percentual (5,7%), seguidos do cisto residual (4,3%), paradentário (2,8%) e a variável ortoqueratinizada (1,5%). As mulheres foram ligeiramente mais afetadas (1,12:1) com maior acometimento de leucodermas (80%). Idade e localização anatômica diferiram de acordo com cada lesão, embora a região anterior de maxila tenha sido mais afetada. A enucleação foi o tratamento mais utilizado (75,7%), exceto para o queratocisto odontogênico, no qual a marsupialização foi realizada em 75% dos casos. Conclusão: Os dados epidemiológicos fornecem uma visão importante para o entendimento da prevalência, extensão e gravidade dessas lesões, viabilizando o diagnóstico precoce e medidas preventivas, a fim de garantir a redução dos casos e melhorar a qualidade de vida da população.

BACKGROUND: Sellar region tumor growth represents an important cause of visual loss due to mechanical compression of the optic nerve apparatus. Many investigations have used non-invasive tools to evaluate the visual field consequences of this damage, and good associations have been reported between psychophysical and electrophysiological perimetries. Few reports have considered the tumor size as a predictor of visual field loss. AIMS: In the present study, we evaluated the association between the visual perimetry measured by Humphrey visual field analyzer and multifocal visual evoked cortical potential (mfVECP) and the tumor size. METHODS: Our sample was composed of 14 patients diagnosed with sellar tumors by magnetic resonance imaging. We accounted the number of sectors with negative visual responses for both methods. A simple logistic regression analysis was used to evaluate the association between the tumor dimensions and the visual field features RESULTS: Three patients had preserved visual fields, three patients showed hemianopic defects, and eight patients had generalized visual field losses at both evaluations. We observed that the three maximum diameters of the tumor and total tumor volume had different predictive abilities regarding the extent of visual field loss when using psychophysical and mfVECP data. The maximum craniocaudal diameter of the tumor was the better predictor of the psychophysical measurements, whereas for the mfVECP results, all tumor dimensions and volumes had similar values that predict visual field losses. CONCLUSION: Tumor size as a predictor of visual loss has potential to assist in the clinical intervention and to prevent the irreversible visual impairment caused by tumors of the sellar region.
