RESUMEN

Neospora caninum is an apicomplexan protozoan that causes neosporosis, which has a high economic impact on cattle herds with no available vaccine. During infection, the secretion of dense granules and the expression of surface antigens play an important role in hosting immunomodulation. However, some epitopes of those antigens are immunogenic, and using these fractions could improve the subunit antigens in vaccine design. This study evaluates the recombinant peptides rsNcGRA1 and rsNcSAG4 derived from NcGRA1 and NcSAG4 native antigens as vaccine candidates produced by a fermentative process in the yeast culture system of Komagataella phaffii strain Km71, confirmed by colony PCR, SDS-PAGE, and western blotting. The assay was conducted in BALB/c mice using the peptides at low (25 µg) and standard (50 µg) dosages in monovalent and combined administrations at three time points with saponin as an adjuvant assessing the immunogenicity by antibodies response and cytokine production. We challenge the females after pregnancy confirmation using 2 × 105 NC-1 tachyzoites previously propagated in Vero cells. We assessed the chronic infection in dams and vertical transmission in the offspring by PCR and histopathology. Mice, especially those immunised with combined peptides and monovalent rsNcGRA1 at a standard dose, controlling the chronic infection in dams with the absence of clinical manifestations, showed an immune response with induction of IgG1, a proper balance between Th1/Th2 cytokines and reduced vertical transmission in the pups. In contrast, dams inoculated with a placebo vaccine showed clinical signs, low-scored brain lesions, augmented chronic infection with 80% positivity, 31% mortality in pups, and 81% vertical transmission. These findings indicate that rsNcGRA1 peptides in monovalent and combined with rsNCSAG4 at standard dose are potential vaccine candidates and improve the protective immune response against neosporosis in mice.

RESUMEN

Brazil, a global frontrunner in pesticide consumption and sales, particularly glyphosate, appears to be at odds with other countries that increasingly ban these products in their territories. This study gathers the values of Acceptable Daily Intake and Maximum Residue Limits (MRL) in the European Union for dozens of substances and subsequently contrasts them with the corresponding benchmarks upheld in Brazil concerning its predominant crops. Furthermore, this study delves into the toxicity levels and the potential health ramifications of glyphosate on humans through the ingestion of food containing its residues. The findings from this research underscore a notable surge in glyphosate and pesticide sales and usage within Brazil over the past decade. In stark contrast to its European counterparts, Brazil not only sanctioned the sale and application of 474 new pesticides in 2019, but extended the authorization for glyphosate sales while downgrading its toxicity classification. Finally, this review not only uncovers disparities among research outcomes but also addresses the complexities of replacing glyphosate and introduces environmentally friendlier alternatives that have been subject to evaluation in the existing literature.

Goiânia, the Goiás State capital, starred in 1987, where one of the largest radiological accidents in the world happened. A teletherapy machine was subtracted from a derelict radiotherapy clinic and disassembled by scavengers who distributed fragments of the 50 TBq 137CsCl source among relatives and acquaintances, enchanted by the blue shine of the substance. During the 15 days before the accident was acknowledged, contaminated recycling materials were delivered to recycling factories in four cities in the state of São Paulo, Brazil, in the form of recycling paper bales. The contaminated bales were spotted, collected, and stored in fifty 1.6 m3 steel boxes at the interim storage facility of the Nuclear and Energy Research Institute (IPEN). In 2017, a check of the content was performed in a few boxes and the presence of high moisture content was observed even though the bales were dry when conditioned and the packages were kept sealed since then. The main objective of this work was to report the fungi found in the radioactive waste after they evolved for 30 years in isolation inside the waste boxes and their role in the decay of the waste. Examination of the microbiome showed the presence of nematodes and fungal communities. The fungi species isolated were Aspergillus quadricinctus, Fusarium oxysporum, Lecanicillium coprophilumi, Scedosporium boydii, Scytalidium lignicola, Xenoacremonium recifei, and Pleurostoma richardsiae. These microorganisms showed a significant capacity to digest cellulose in our trials, which could be one of the ways they survive in such a harsh environment, reducing the volume of radioactive paper waste. These metabolic abilities give us a future perspective of using these fungi in biotechnology to remediate radioactively contaminated materials, particularly cellulose-based waste.

INTRODUCTION: As experienced physiotherapy educators in Brazil, we observed that COVID-19 elucidated challenges in the pedagogy of entry-level education overall, and directions for their remediation. In this commentary, we describe our observations with particular attention to the opportunity for digital and distance teaching and learning in Brazil's exemplary middle-income country. BODY: First, the legislation in Brazil around health professional education, specifically entry-level physiotherapy education, is described concerning distanced learning. Then, we contrast such education before and during the COVID-19 pandemic, and in the aftermath of its peak. Our observations reinforce the need to preserve teaching and learning excellence in physiotherapy education with various approaches including distanced and digital learning; be aware of both advantages and disadvantages; and identify means of balancing these for optimal delivery and learner outcomes. Our collective experience and insights strongly support the need for change in the legislative document governing physiotherapy education in Brazil. CONCLUSION: We hope our experiences will enable other educators to evaluate their contexts, reflect on how best to deliver entry-level physiotherapy education in general and during a pandemic, and reinforce the essentiality of practical face-to-face classes in achieving physiotherapy competencies. Only in this way will global standards of practice be ensured, through quality professional education and the factors that inform and govern these.

The main goal of this study was to assess alternatives to the current challenges on environmental quality and circular economy. The former is here addressed by the treatment of radioactively contaminated solutions, and the latter by using abundant and low-cost biomass. In this paper, we examine the biosorption of hexavalent uranium (U(VI)) in a batch system using the macrophytes Limnobium laevigatum and Azolla sp. by three operational parameters: biomass dose, metal ion concentration, and contact time. Simulated solutions were firstly addressed with two biomasses, followed by studies with real liquid organic radioactive waste (LORW) with Azolla sp. The batch experiments were carried out by mixing 0.20 g biomass in 10 mL of the prepared solution or LORW. The total contact time employed for the determination of the equilibrium times was 240 min, and the initial U(VI) concentration was 0.63 mmol L-1. The equilibrium times were 15 min for L. laevigatum and 30 min for Azolla sp. respectively. A wide range of initial U(VI) concentrations (0.25-36 mmol L-1) was then used to assess the adsorption capacity of each macrophyte. Isotherm models validated the adsorption performance of the biosorption process. Azolla sp. presented a much higher U(VI) uptake (0.474 mmol g-1) compared to L. laevigatum (0.026 mmol g-1). When in contact with LORW, Azolla sp. removed much less uranium, indicating an adsorption capacity of 0.010 mmol g-1. In conclusion, both biomasses, especially Azolla sp., can be used in the treatment of uranium-contaminated solutions.

Biosorption has been examined for the treatment of aqueous solutions containing uranium, a radiotoxic pollutant. Nevertheless, the evaluation of the role of process variables by experimental design on the use of hydroxyapatite and bone meal as biosorbents for uranium has not yet been previously addressed. In this study, the effects of adsorbent dosage (M), initial uranium concentrations ([U]0), and solution pH were investigated, using a two-level factorial design and response surface analysis. The experiments were performed in batch, with [U]0 of 100 and 500 mg L-1, pH 3 and 5, and adsorbent/uranium solution ratios of 5 and 15 g L-1. Contact time was fixed at 24 h. Removal rates were higher than 88%, with a maximum of 99% in optimized conditions. [U]0 and M were found to be the most influential variables in U removal in terms of adsorption capacity (q). The experiments revealed that bone meal holds higher adsorption capacity (49.87 mg g-1) and achieved the highest uranium removal (~ 100%) when compared to hydroxyapatite (q = 49.20 mg g-1, removal = 98.5%). The highest value of q for both biomaterials was obtained for [U]0 = 500 mg L-1, pH 3, and M = 5 g L-1. Concerning the removal percentage, bone meal achieved the best performance for [U]0 = 500 mg L-1, pH 3, and M = 15 g L-1. Further experiments were made with real radioactive waste, resulting in a high uranium adsorption capacity for both materials, with 22.11 mg g-1 for hydroxyapatite and 22.08 mg g-1 for bone meal, achieving uranium removal efficiencies higher than 99%.

RHIPICEPHALUS MICROPLUS: is the most important ectoparasite of livestock in tropical and subtropical areas around the world. Research focused on developing an efficient vaccine for cattle tick control is a high priority. The aim of this study was to evaluate the rSBm7462® peptide (Bm86-B and T cell epitopes) regarding its properties of immunogenicity, protective effect in cattle and efficacy against R. microplus. This peptide was produced by a fermentative process in the yeast culture system of Komagataella (Pichia) pastoris strain Km 71. The vaccination assay was conducted in a tick-free area using non-splenectomised Holstein Friesian calves, separated into immunisation and control groups. These animals individually received the recombinant peptide and the inoculum without peptide using saponin as an adjuvant at three time points. The calves were challenged 21 days after the last immunisation using 4500 larvae per animal. An indirect ELISA was used to identify the IgG kinetics of serum samples from the calves studied. The qPCR was performed to determine the cytokine gene expression from the total RNA of the cultured PBMCs. Histomorphometry of the germinal centres (GCs) was performed measuring slides with haematoxylin-eosin staining of surgically removed lymph nodes from immunised calves. The antibody response showed a significant induction of high-affinity IgGs in calves immunised with the recombinant peptide in comparison to the controls. The kinetics of antibodies in immunised calves showed a significant increase during the experiment. This increase in high-affinity IgGs correlated with a gradual increase of the GC diameter following each peptide vaccination. Cytokine expression profiles demonstrating an adaptive immune response in calves immunised with rSBm7462® confirmed the T-dependent response. Vaccine efficacy was calculated at 72.4 % following the analysis and fecundity of collected adult female ticks, compared between control and vaccinated groups. These findings demonstrate that this new recombinant peptide is an option for control of R. microplus infestations.

Rice and coffee husks (raw and chemically activated) are examined as potential biosorption materials regarding their capacity to remove U (total), 241Am, and 137Cs. The physical parameters evaluated were the morphological characteristics of the biomass, real and apparent density, and surface area. Contact times for the batch experiments were 0.5, 1, 2, and 4 h, and the concentrations tested ranged between 10% of the total concentration and the radioactive waste itself without any dilution. The results were evaluated by experimental sorption capacity, ternary isotherm, and kinetics models. The kinetics results showed that equilibrium was reached after 2 h for all biomass. Raw coffee husk showed the best adsorption results in terms of maximum capacity (qmax) for all three radionuclides, which were 1.96, 39.4 × 10-6, and 46.6 × 10-9 mg g-1 for U, Am, and Cs, respectively. The biosorption process for the raw and activated rice husks was best represented by the Langmuir ternary isotherm model with two sites. For the coffee husk, in the raw and activated states, the biosorption process was best described by the modified Jain and Snoeyink ternary model. These results suggest that biosorption with these biomaterials can be applied in the treatment of liquid organic radioactive waste containing mainly uranium and americium.

A theoretical and experimental study of bisphenol A (BPA) degradation by the UV/H2O2 process in water is presented. The effects of the H2O2 concentration and the specific rate of photon emission (EP,0) on BPA degradation were investigated. A kinetic model derived from a reaction sequence was employed to predict BPA and hydrogen peroxide concentrations over time using an annular photochemical reactor in batch recirculation mode. The local volumetric rate of photon absorption (LVRPA) inside the photoreactor was computed using a Line Source with Parallel Plane emission model (LSPP). From the proposed kinetic model and the experimental data, the second order rate constants of the reactions between hydroxyl radicals and the main reacting species (H2O2 and BPA) were estimated applying a nonlinear regression method. A good agreement between the kinetic model and experimental data, for a wide range of operating conditions, was obtained. For BPA, H2O2, and TOC concentrations, the calculated root means square errors (RMSE) were 2.3 × 10- 2, 9.8 × 10- 1, and 9.0 × 10- 2 mmol L- 1, respectively. The simplified kinetic model presented in this work can be directly applied to scaling-up and reactor design, since the estimated kinetic constants are independent of the reactor size, shape, and configuration. Further experiments were made by employing low BPA initial concentration (100 µg L- 1) in water and real wastewater. A lower degradation rate of BPA was observed in the real wastewater, although the UV/H2O2 process has also been able to completely degrade the target pollutant in less than 1 h.

The objectives were to evaluate the effects of two non-fibrous carbohydrate sources and the rate of body weight gain on puberty status in ewe lambs. Sixty ewe lambs (½ Dorper × ½ Santa Ines) with an initial body weight of 25.1 ± 4.1 kg and 102.8 ± 1.5 days old were distributed in a randomized complete block design with 10 blocks and 3 treatments. The trial lasted 119 days, in which performance was evaluated on days 28, 56, 84, and 119, and puberty status was every week evaluated by progesterone concentration. The treatments were characterized by the inclusion of different carbohydrate sources in the diets in order for ewe lambs to reach different rates of body weight gain: CORN, total diet containing 46% of corn (% DM); PCP, total diet containing 46% of pelleted citrus pulp (% DM); and HAY, total diet containing 80% of coastcross hay (% DM). The CORN- and PCP-based diets were formulated for an average daily gain (ADG) of 0.200 kg/day and a HAY-based diet for an ADG of 0.100 kg/day. There was an interaction between treatments and experimental periods for dry matter intake (DMI) (P < 0.01), in which lambs in the CORN treatment presented DMI similar to PCP in the first and second periods, but lambs from the PCP treatment presented lower DMI than the CORN treatment in subsequent periods. In all periods, the body weight, average daily gain, and feed efficiency of CORN lambs were similar to PCP lambs, but higher than HAY lambs. The age at puberty was not affected by treatments; however, lambs fed with non-fibrous carbohydrate sources reached puberty heavier (P < 0.0001) than lambs from the HAY treatment. Despite the high increase in body weight for lambs fed with non-fibrous carbohydrate sources, it did not affect the age of puberty.

Biosorption-based technologies have been proposed for the removal of radionuclides from radioactive liquid waste containing organic compounds. Nevertheless, pytoremediation potential of uranium (U) by nonliving aquatic macrophytes Lemna sp. and Pistia stratiotes has not been previously addressed. In this study, uranium biosorption capacity by Pistia stratiotes and Lemna sp. was evaluated by equilibrium and kinetics experiments. The biomasses were added to synthetic and real waste solutions. The assays were tested in polypropylene vials containing 10 mL of uranium nitrate solution and 0.20 g of biomass. Solutions ranging from 0.25 to 84.03 mmol l-1 were employed for the assessment of uranium concentration in each macrophyte. The equilibrium time was 1 h for both macrophytes. Lemna sp. achieved the highest sorption capacity with the use of the synthetic solution, which was 0.68 mmol g-1 for the macrophyte. Since Lemna sp. exhibit a much higher adsorption capacity, only this biomass was exposed to the actual waste solution, being able to adsorb 9.24 × 10-3 mmol g-1 U (total). The results show that these materials are potentially applicable to the treatment of liquid radioactive waste.

The main effluent of oil and gas production is the discharge of produced water (PW). Despite the potential environmental impact, the structure of oil and gas platforms can act as artificial reefs. Two species of fish, Caranx crysos and Tylosurus acus, were captured under two discharging platforms at Campos Basin, P19 and P40. A nondischarging platform, P25, was taken as reference. The highest median concentration of PAH in muscle tissue was observed for C. Crysos at P40 (219.38 ng g-1) followed by P19 (68.26 ng g-1). For T. acus, the highest median concentrations were 40.07 and 42.21 ng g-1 from P19 and P40 respectively. P25 presented the smallest recorded concentrations. The results of PAH in the muscle tissue of C. crysos enabled to distinguish the platforms with PW discharge from the platform without discharge. Trace elements concentrations reflected the background levels and were not influenced by the PW release.

Twenty-eight specimens of Esophael squamous cell carcinoma (ESCC) were obtained by surgery procedures.The tissues were fixed in formalin and embedded in paraffin. In each case, all available hematoxylin and eosin stained sections were examined and a representative block was selected. The ages of these patients ranged from 40 to 93 years, with a mean age of 60 years. Results. The histological grade of tumors was 4 well-differentiated, 19 moderately differentiated and 5 poorly differentiated. Expression of Hsp27 and Hsp70 in ESCC was demonstrated in 23 (82,14%) and 26 (92,86%) cases, respectively. Adjacent normal mucosa was positive in 11 (39,29%) samples and 9 (32,15%) samples for Hsp27 and Hsp70, respectively. No relationship between the expression of Hsp27 and Hsp70 with the clinicopathological parameters, including gender, age, surgical margin, lymph node status and tumor differentiation. The median follow-up period was 60 months. Survival analysis of patients with ESCC showed no relationship with the expression of Hsp27 and Hsp70. Conclusion. Taken together, our results demonstrate that Hsp27 and Hsp70 are expressed in ESCC tissues, but they are not good prognostic factor for patients with ESCC.

OBJECTIVE: Examine the attitudes of nursing students with the suicidal behavior before and after a training course on the subject. METHODOLOGY: Performed quantitative, cross-sectional study, with 58 nursing students from a public university in Minas Gerais (Brazil) who participated in training on the theme. For data collection were used the Questionnaire of Attitudes Before Suicidal Behavior. The questionnaire was applied just before the start and the end of the training measuring attitudes toward suicidal behavior. RESULTS: Were found statistically significant differences in negative feelings factors on the patient and perception of professional competence (p <0.05). The right factor to suicide was not significantly different among nursing students. CONCLUSION: The academic training may have influenced positively the desired changes regarding the attitudes of nursing students across the suicidal behavior.

The objective of this work was to develop and evaluate agrometeorological models to simulate the production of Guineagrass. For this purpose, we used forage yield from 54 growing periods between December 2004-January 2007 and April 2010-March 2012 in irrigated and non-irrigated pastures in São Carlos, São Paulo state, Brazil (latitude 21°57'42″ S, longitude 47°50'28″ W and altitude 860 m). Initially we performed linear regressions between the agrometeorological variables and the average dry matter accumulation rate for irrigated conditions. Then we determined the effect of soil water availability on the relative forage yield considering irrigated and non-irrigated pastures, by means of segmented linear regression among water balance and relative production variables (dry matter accumulation rates with and without irrigation). The models generated were evaluated with independent data related to 21 growing periods without irrigation in the same location, from eight growing periods in 2000 and 13 growing periods between December 2004-January 2007 and April 2010-March 2012. The results obtained show the satisfactory predictive capacity of the agrometeorological models under irrigated conditions based on univariate regression (mean temperature, minimum temperature and potential evapotranspiration or degreedays) or multivariate regression. The response of irrigation on production was well correlated with the climatological water balance variables (ratio between actual and potential evapotranspiration or between actual and maximum soil water storage). The models that performed best for estimating Guineagrass yield without irrigation were based on minimum temperature corrected by relative soil water storage, determined by the ratio between the actual soil water storage and the soil water holding capacity.irrigation in the same location, in 2000, 2010 and 2011. The results obtained show the satisfactory predictive capacity of the agrometeorological models under irrigated conditions based on univariate regression (mean temperature, potential evapotranspiration or degree-days) or multivariate regression. The response of irrigation on production was well correlated with the climatological water balance variables (ratio between actual and potential evapotranspiration or between actual and maximum soil water storage). The models that performed best for estimating Guineagrass yield without irrigation were based on degree-days corrected by the water deficit factor.

RESUMEN

OBJECTIVE: To compare the efficacy of anterior transposition (AT) and graded recession (GR) in the treatment of V- pattern strabismus caused by overaction of the inferior oblique muscle. METHODS: The charts of surgically treated cases of V-pattern strabismus were analyzed retrospectively and the patients classified as AT or GR. Age, initial horizontal deviation, initial V-pattern and the amount of V-pattern correction were compared between the two groups. RESULTS: There was no significant difference in age (p = 0.066), initial horizontal deviation (p = 0.59), initial V-pattern (p = 0.15) or the amount of V-pattern correction (p = 0.78) between the two groups. CONCLUSION: AT is at least as effective as GR in the treatment of V-pattern strabismus caused by overaction of the inferior oblique muscle.
