RESUMEN

Chemoprophylaxis with dipropionate imidocarb (IMD) is a method adopted to prevent cattle tick fever (TF). Sixty weaned dairy heifers (±60 days old), without previous exposure to Rhipicephalus microplus ticks, were housed in Tifton paddocks and were subsequently exposed to R. microplus ticks and monitored up to 315 days old. Thirty animals were kept as controls (T01) and 30 received five preventive strategic treatments with IMD at 21-day intervals (T02). The heifers were monitored weekly by means of packed cell volume (PCV) and blood smears to evaluate the presence of TF agents. Salvage treatments (ST) with diminazene and enrofloxacin were administered when animals showed PCV ≤ 24%. The A. marginale prevalence was 39.3% and 37.7%, B. bovis 6.0%, and 7.3%, and B. bigemina 16.3% and 13.7% for T01 and T02, respectively. Regarding PCV values, group T01 showed lower PCV than group T02, between 119 and 161 days of life, but when animals were 196, 210, 217, and between 252 to 301 days old, an inversion occurred. The IMD treatment protocol was effective in group T02 from day 91 to 175 while treatment was being administered, but from day 182 to 315 after the IMD treatment protocol was completed, the number of salvage treatments against TF agents performed in T02 group increased significantly. The sequential application of IMD treatments with intervals less than 21 days is not recommended.

BACKGROUND: Packed cell volume (PCV) is important for assessing a patient's health status. Some blood gas analyzers measure hematocrit, and the agreement with PCV varies among different analyzers. OBJECTIVES: We aimed to determine the agreement between PCV measured by microcentrifugation and hematocrit measured by the Cobas b121 blood gas analyzer in dogs, cats, and horses. METHODS: Whole blood samples for PCV and blood gas analysis were collected in lithium-heparin syringes and analyzed within 10 min of collection. Agreement and association between the PCV and Cobas b121 generated hematocrit were assessed by the Bland-Altman method, Pearson's correlation, Deming regression analysis, and paired t tests. A total allowable error of 10% was used for the analysis. RESULTS: This study included 45 dogs, 45 cats, and 33 horses. The respective mean ± SD (minimum-maximum) of PCVs and hematocrits were: dogs, 34.9 ± 9.9% (9.0-55.0) and 32.5 ± 8.8% (10.4-50.6); cats, 29.0 ± 9.6% (11.0-51.0) and 26.9 ± 9.3% (10.2-50.9); horses, 34.2 ± 6.5% (24.0-47.0) and 34.1 ± 6.0% (22.5-46.1). There were no significant differences between the methods. The bias ± SD was: dogs, -2.4 ± 2.6%; cats, -2.2 ± 2.3%; horses, -0.1 ± 2.4%. Pearson's correlation coefficients were > 0.90 for all species (P < 0.0001). In 60%, 49%, and 85% of the samples for dogs, cats, and horses, respectively, the percentage differences between the methods were within 10%. CONCLUSIONS: The Cobas b121 blood gas analyzer provided accurate estimates of PCVs in horses. However, in dogs and cats, there was a large frequency of unacceptable differences between the methods.

Brain edema is a pathological condition with potentially fatal consequences, related to cerebral injuries such as ischemia, chronic renal failure, uremia, and diabetes, among others. Under these pathological states, the cell volume control processes are fully compromised, because brain cells are unable to regulate the movement of water, mainly regulated by osmotic gradients. The processes involved in cell volume regulation are homeostatic mechanisms that depend on the mobilization of osmolytes (ions, organic molecules, and polyols) in the necessary direction to counteract changes in osmolyte concentration in response to water movement. The expression and coordinated function of proteins related to the cell volume regulation process, such as water channels, ion channels, and other cotransport systems in the glial cells, and considering the glial cell proportion compared to neuronal cells, leads to consider the astroglial network the main regulatory unit for water homeostasis in the central nervous system (CNS). In the last decade, several studies highlighted the pivotal role of glia in the cell volume regulation process and water homeostasis in the brain, including the retina; any malfunction of this astroglial network generates a lack of the ability to regulate the osmotic changes and water movements and consequently exacerbates the pathological condition.

Data regarding parasitemia (blood smears), rectal temperature (RT), packed cell volume (PCV) and vaginal mucosa coloration (VMC) of Gyr x Holstein female calves between 3-7mo were accessed to evaluate different techniques for monitoring the bovine tick fever agents (TFA). The 1st experiment determined the correlation between the TFA parasitemia with RT and PCV. The 2nd, evaluated the associated risk of A. marginale parasitemia with RT and PCV in relation to the Gyr/Holstein genetic proportion (5/8,3/4,7/8 and 15/16) using Receiver Operating Characteristic Curve (ROC). The 3rd, two groups were performed: cattle monitored by RT (T01) and by PCV (T02), during their 80-210 days of age, data regarding TFA parasitemia, RT, PCV, VMC and weight were registered. In 1st experiment, RT showed weak correlation with TFA parasitemia, while PCV showed a strong correlation with A. marginale and B. bigemina, but not with B. bovis parasitemia. In experiment 2, the ROC curve analysis showed that when the genetic proportion of B. t. taurus increased, least reliable RT was to monitor calves infected with A. marginale. The PCV for monitoring A. marginale was the best technique, showing sensitivity of 74.2% and specificity of 97.0% than other techniques that used RT and VCM as a monitoring tool. In general, calves monitored by PCV (T02) showed higher PCV values, lower A. marginale parasitemia, less pneumonia as co-infection and less salvation treatment were performed than in animals monitored by RT (T01). Furthermore, animals from T02 gained 23.5 kg more than those from T01. The low frequency of B. bovis and B. bigemina found in this study made impossible to compare the monitoring techniques for these pathogenic agents.

The study was devised with the aim to evaluate the relationship between hemoglobin (Hb) concentration and Packed Cell Volume (PCV) in beetal goats being reared under pastoralism. It also aims to devise a hematological formula for estimation of Hb from PCV. Female (n=59) and male goats (n=41) were bled for PCV determination through microhematocrit method, and Hb estimation through Sahli's hemoglobinometer (HbD) as well as through calculation being 1/3rd of PCV (HbC). The HbD and HbC were statistically non-significant (P≥0.05) for male and female beetal goats. Overall, significantly (P≤0.01) positive correlation coefficient was noticed between HbD and PCV, and between HbD and HbC (r=0.75; adjusted r-square=0.57). As the overall model predicted that 57% variability in HbD could be deduced from PCV, hence, in order to enhance the prediction probability, the regression equation i.e. Hb concentration= 0.24(PCV) +1.5 was utilized to deduce corrected hemoglobin (CHb). The comparison of this CHb with HbD gave a non-significant (P≤0.05) difference. Similarly, linear regression of CHb with PCV gave a 99% prediction. We accordingly recommend a simplified pen-side hematological formula for deducing Hb concentration from PCV viz. Hb concentration= 0.24(PCV) +1.5 for beetal goats instead of its calculation as one-third of PCV.


O estudo foi elaborado com o objetivo de avaliar a relação entre a concentração de hemoglobina (Hb) e o volume de células embaladas (PCV) em caprinos beetal em pastoreio. O objetivo também é elaborar uma fórmula hematológica para a estimativa da hemoglobina de PCV. Cabras fêmeas (n=59) e machos (n=41) foram sangrados para determinação da PCV através do método do microhematócrito, e a estimativa de Hb através do hemoglobinômetro Sahli (HbD), bem como através do cálculo sendo 1/3 do PCV (HbC). O HbD e HbC foram estatisticamente não significativos (P≥0.05) para caprinos machos e fêmeas beetal. De modo geral, foi observado um coeficiente de correlação positiva (P≤0.01) entre HbD e PCV, e entre HbD e HbC (r=0,75; r-quadrado ajustado=0,57). Como o modelo geral previa que 57% de variabilidade em HbD poderia ser deduzida do PCV, portanto, para aumentar a probabilidade de predição, a equação de regressão i.e. concentração de Hb= 0,24(PCV) +1,5 foi utilizada para deduzir a hemoglobina corrigida (CHb). A comparação desta CHb com HbD deu uma diferença não significativa (P≤0.05). Da mesma forma, a regressão linear de CHb com PCV deu uma previsão de 99%. Assim, recomendamos uma fórmula hematológica simplificada do lado da caneta para deduzir a concentração de Hb do PCV, ou seja, concentração de Hb= 0,24(PCV) +1,5 para cabras beetal em vez de seu cálculo como um terço do PCV.

α-hemolysin (HlyA) of E. coli binds irreversibly to human erythrocytes and induces cell swelling, ultimately leading to hemolysis. We characterized the mechanism involved in water transport induced by HlyA and analyzed how swelling and hemolysis might be coupled. Osmotic water permeability (Pf) was assessed by stopped-flow light scattering. Preincubation with HlyA strongly reduced Pf in control- and aquaporin 1-null red blood cells, although the relative Pf decrease was similar in both cell types. The dynamics of cell volume and hemolysis on RBCs was assessed by electrical impedance, light dispersion and hemoglobin release. Results show that HlyA induced erythrocyte swelling, which is enhanced by purinergic signaling, and is coupled to osmotic hemolysis. We propose a mathematical model of HlyA activity where the kinetics of cell volume and hemolysis in human erythrocytes depend on the flux of osmolytes across the membrane, and on the maximum volume that these cells can tolerate. Our results provide new insights for understanding signaling and cytotoxicity mediated by HlyA in erythrocytes.

Aquaporin-4 (AQP4) is the target of the specific immunoglobulin G autoantibody (AQP4-IgG) produced in patients with neuromyelitis optica spectrum disorders (NMOSD). Previous studies demonstrated that AQP4-IgG binding to astrocytic AQP4 leads to cell-destructive lesions. However, the early physiopathological events in Müller cells in the retina are poorly understood. Here, we investigated the consequences of AQP4-IgG binding to AQP4 of Müller cells, previous to the inflammatory response, on two of AQP4's key functions, cell volume regulation response (RVD) and cell proliferation, a process closely associated with changes in cell volume. Experiments were performed in a human retinal Müller cell line (MIO-M1) exposed to complement-inactivated sera from healthy volunteers or AQP4-IgG positive NMOSD patients. We evaluated AQP4 expression (immunofluorescence and western blot), water permeability coefficient, RVD, intracellular calcium levels and membrane potential changes during hypotonic shock (fluorescence videomicroscopy) and cell proliferation (cell count and BrdU incorporation). Our results showed that AQP4-IgG binding to AQP4 induces its partial internalization, leading to the decrease of the plasma membrane water permeability, a reduction of swelling-induced increase of intracellular calcium levels and the impairment of RVD in Müller cells. The loss of AQP4 from the plasma membrane induced by AQP4-IgG positive sera delayed Müller cells' proliferation rate. We propose that Müller cell dysfunction after AQP4 removal from the plasma membrane by AQP4-IgG binding could be a non-inflammatory mechanism of retinal injury in vivo, altering cell volume homeostasis and cell proliferation and consequently, contributing to the physiopathology of NMOSD.

Saccharomyces cerevisiae has evolved diverse mechanisms to osmotic changes: the cell wall, ion and water transport systems, and signaling cascades. At the present time, little is known about the mechanisms involved in short-term responses of osmotic stress in yeast or their physiological state during this process. We conducted studies of flow cytometry, wet weight measurements, and electron microscopy to evaluate the modifications in cell volume and the cell wall induced by osmotic stress. In response to osmotic challenges, we show very fast and drastic changes in cell volume (up to 60%), which were completed in less than eight seconds. This dramatic change was completely reversible approximately 16 s after returning to an isosmotic solution. Cell volume changes were also accompanied by adaptations in yeast metabolism observed as a reduction by 50% in the respiratory rate, measured as oxygen consumption. This effect was also fully reversible upon returning to an isosmotic solution. It is noteworthy that we observed a significant recovery in oxygen consumption during the first 10 min of the osmotic shock. The rapid adjustment of the cellular volume may represent an evolutionary advantage, allowing greater flexibility for survival.

Plasma levels of hematocrit, total plasma protein, fibrinogen, creatine phosphokinase, aspartate transferase, and lactate were analyzed in blood samples of 85 Crioula breed foals, from birth to two years of age. The animals were divided into age groups: G1 (up to 15 days of age; n=70), G2 (from 16 days to one month of age; n=67), G3 (between one and three months of age; n=75), G4 (between three and six months of age; n=64), G5 (between six and nine months of age; n=59), G6 (between nine and 18 months of age; n=39), and G7 (between 18 months and two years of age; n=17). These groups were statistically analyzed by one-way variance analysis (ANOVA) and Tukey's test. Male and female means were compared by Student's t-test. Hematocrit levels were significantly higher up to 90 days of age and in G7 females. Total plasma proteins increased significantly in groups 3, 4, 6, and 7. The highest fibrinogen levels were found in G1. Yet for creatine phosphokinase, the highest concentrations were detected in G5, whereas those of aspartate aminotransferase in G7. The levels of this enzyme remained similar from 30 days to two years of age. Lactate concentrations were higher in G3. We concluded that the sex of the animal had no significant effect on laboratory test interpretations. By contrast, the age of the animal should be considered since relevant variations were observed with time. Nevertheless, specific tables for each analysis should be consulted for interpretation of results.

These groups were statistically analyzed by one-way variance analysis (ANOVA) and Tukeys test. Male and female means were compared by Students t-test. Hematocrit levels were significantly higher up to 90 days of age and in G7 females. Total plasma proteins increased significantly in groups 3, 4, 6, and 7. The highest fibrinogen levels were found in G1. Yet for creatine phosphokinase, the highest concentrations were detected in G5, whereas those of aspartate aminotransferase in G7. The levels of this enzyme remained similar from 30 days to two years of age. Lactate concentrations were higher in G3. We concluded that the sex of the animal had no significant effect on laboratory test interpretations. By contrast, the age of the animal should be considered since relevant variations were observed with time. Nevertheless, specific tables for each analysis should be consulted for interpretation of results.(AU)

The role of Cl- as an intracellular signaling ion has been increasingly recognized in recent years. One of the currently best described roles of Cl- in signaling is the modulation of the With-No-Lysine (K) (WNK) - STE20-Proline Alanine rich Kinase (SPAK)/Oxidative Stress Responsive Kinase 1 (OSR1) - Cation-Coupled Cl- Cotransporters (CCCs) cascade. Binding of a Cl- anion to the active site of WNK kinases directly modulates their activity, promoting their inhibition. WNK activation due to Cl- release from the binding site leads to phosphorylation and activation of SPAK/OSR1, which in turn phosphorylate the CCCs. Phosphorylation by WNKs-SPAK/OSR1 of the Na+-driven CCCs (mediating ions influx) promote their activation, whereas that of the K+-driven CCCs (mediating ions efflux) promote their inhibition. This results in net Cl- influx and feedback inhibition of WNK kinases. A wide variety of alterations to this pathway have been recognized as the cause of several human diseases, with manifestations in different systems. The understanding of WNK kinases as Cl- sensitive proteins has allowed us to better understand the mechanistic details of regulatory processes involved in diverse physiological phenomena that are reviewed here. These include cell volume regulation, potassium sensing and intracellular signaling in the renal distal convoluted tubule, and regulation of the neuronal response to the neurotransmitter GABA.

Cation-coupled chloride cotransporters (CCC) play a role in modulating intracellular chloride concentration ([Cl-]i) and cell volume. Cell shrinkage and cell swelling are accompanied by an increase or decrease in [Cl-]i, respectively. Cell shrinkage and a decrease in [Cl-]i increase the activity of NKCCs (Na-K-Cl cotransporters: NKCC1, NKCC2, and Na-Cl) and inhibit the activity of KCCs (K-Cl cotransporters: KCC1 to KCC4), wheras cell swelling and an increase in [Cl-]i activate KCCs and inhibit NKCCs; thus, it is unlikely that the same kinase is responsible for both effects. WNK1 and WNK4 are chloride-sensitive kinases that modulate the activity of CCC in response to changes in [Cl-]i. Here, we showed that WNK3, another member of the serine-threonine kinase WNK family with known effects on CCC, is not sensitive to [Cl-]i but can be regulated by changes in extracellular tonicity. In contrast, WNK4 is highly sensitive to [Cl-]i but is not regulated by changes in cell volume. The activity of WNK3 toward NaCl cotransporter is not affected by eliminating the chloride-binding site of WNK3, further confirming that the kinase is not sensitive to chloride. Chimeric WNK3/WNK4 proteins were produced, and analysis of the chimeras suggests that sequences within the WNK's carboxy-terminal end may modulate the chloride affinity. We propose that WNK3 is a cell volume-sensitive kinase that translates changes in cell volume into phosphorylation of CCC.

Nevertheless, the influence ofhousing systems and stabling time on glucose and PCV changes is little known. The purpose of the present study was toevaluate the patterns of cortisol, glucose and PCV in dogs housed in a kennel and normal environments, evaluating thedifferences between housing systems, by taking into account the different stabling time and sex.Materials, Methods & Results: The study comprised 98 cross-breed dogs, aged 4 ± 1.5 years, lodged in a kennel (observational group I: N=61, 29 females and 27 males), in paired household dogs (control group II: N=25, 13 females and 12males) and in unpaired household dogs (control group III: N=12, 6 females and 6 males). Females of both groups werespayed. The subjects were studied on the basis of different stabling times, ranged among <1 year, 2 years and 4 years, anddifferent sex.Discussion: This observational study showed that kennelled males lodged for 2 (P < 0.01) and 4 (P < 0.001) years showedlower cortisol concentrations than males lodged <1 year; males lodged for <1 year (P < 0.001) showed higher cortisol concentrations than females; males lodged for 4 year showed lower cortisol concentrations (P < 0.01) than females. Pre-loading handling and conditions of transport are related to welfare, disease risk and product quality of production animals. These steps continue to be one of the major animal management problems in Brazil. This study evaluated the effects of different types of pre-loading handling and road transport times on the haematological and biochemical traits of cattle. Eighteen male cattle were submitted to three travel times (24, 48 and 72 h) in a truck soon after load using different types of pre-loading handling: traditional (rough handling), training (gentle handling) and use of flags to movement cattle. Haematological traits, blood biochemical measures as well as blood and faecal cortisol were analysed in order to assess animal welfare and physiological status. The traditional management showed to be more stressful, also had animals with a greater number of neutrophils and lower numbers of lymphocytes than handling with flags, showing that animals submitted to more stressful situations can have compromised immune system. Serum aspartate aminotransferase concentrations were within the reference levels and when taken together with increased creatine kinase patterns observed indicate muscle damage in traditional management. Decrease in glucose concentrations over time from traditional management to flag management was observed, while fructosamine was increased in traditional management with 72 h of travel. When taken together, all reported factors, immune, enzymatic, energetic and hormonal, indicate that the quality of pre-loading handling and time of transport were determinant for animal welfare, its homeostatic balance and sanitary conditions.

Ion Transport across the cell membrane is required to maintain cell volume homeostasis. In response to changes in extracellular osmolarity, most cells activate specific metabolic or membrane-transport pathways to respond to cell swelling or shrinkage and return their volume to its normal resting state. This process involves the rapid adjustment of the activities of channels and transporters that mediate flux of K+, Na+, Cl-, and small organic osmolytes. Cation chloride cotransporters (CCCs) NKCCs and KCCs are a family of membrane proteins modulated by changes in cell volume and/or in the intracellular chloride concentration ([Cl-]i). Cell swelling triggers regulatory volume decrease (RVD), promoting solute and water efflux to restore normal cell volume. Swelling-activated KCCs mediate RVD in most cell types. In contrast, cell shrinkage triggers regulatory volume increase (RVI), which involves the activation of the NKCC1 cotransporter of the CCC family. Regulation of the CCCs during RVI and RVD by protein phosphorylation is a well-characterized mechanism, where WNK kinases and their downstream kinase substrates, SPAK and OSR1 constitute the essential phospho-regulators. WNKs-SPAK/OSR1-CCCs complex is required to regulate cell shrinkage-induced RVI or cell swelling-induced RVD via activating or inhibitory phosphorylation of NKCCs or KCCs, respectively. WNK1 and WNK4 kinases have been established as [Cl-]i sensors/regulators, while a role for WNK3 kinase as a cell volume-sensing kinase has emerged and is proposed in this chapter.

Neuronal activity in the retina generates osmotic gradients that lead to Müller cell swelling, followed by a regulatory volume decrease (RVD) response, partially due to the isoosmotic efflux of KCl and water. However, our previous studies in a human Müller cell line (MIO-M1) demonstrated that an important fraction of RVD may also involve the efflux of organic solutes. We also showed that RVD depends on the swelling-induced Ca2+ release from intracellular stores. Here we investigate the contribution of taurine (Tau) and glutamate (Glu), the most relevant amino acids in Müller cells, to RVD through the volume-regulated anion channel (VRAC), as well as their Ca2+ dependency in MIO-M1 cells. Swelling-induced [3H]Tau/[3H]Glu release was assessed by radiotracer assays and cell volume by fluorescence videomicroscopy. Results showed that cells exhibited an osmosensitive efflux of [3H]Tau and [3H]Glu (Tau > Glu) blunted by VRAC inhibitors 4-(2-butyl-6,7-dichloro-2-cyclopentylindan-1-on-5-yl)-oxybutyric acid and carbenoxolone reducing RVD. Only [3H]Tau efflux was mainly dependent on Ca2+ release from intracellular stores. RVD was unaffected in a Ca2+-free medium, probably due to Ca2+-independent Tau and Glu release, but was reduced by chelating intracellular Ca2+. The inhibition of phosphatidylinositol-3-kinase reduced [3H]Glu efflux but also the Ca2+-insensitive [3H]Tau fraction and decreased RVD, providing evidence of the relevance of this Ca2+-independent pathway. We propose that VRAC-mediated Tau and Glu release has a relevant role in RVD in Müller cells. The observed disparities in Ca2+ influence on amino acid release suggest the presence of VRAC isoforms that may differ in substrate selectivity and regulatory mechanisms, with important implications for retinal physiology. NEW & NOTEWORTHY The mechanisms for cell volume regulation in retinal Müller cells are still unknown. We show that swelling-induced taurine and glutamate release mediated by the volume-regulated anion channel (VRAC) largely contributes the to the regulatory volume decrease response in a human Müller cell line. Interestingly, the hypotonic-induced efflux of these amino acids exhibits disparities in Ca2+-dependent and -independent regulatory mechanisms, which strongly suggests that Müller cells may express different VRAC heteromers formed by the recently discovered leucine-rich repeat containing 8 (LRRC8) proteins.

Aparelhos de automação, cujo uso está estabelecido na hematologia dos animais domésticos, ainda não são empregados rotineiramente em felídeos selvagens. Esse estudo objetivou avaliar a técnica automatizada na contagem de células sanguíneas de jaguatiricas (Leopardus pardalis), comparando-a com a técnica manual. Foram coletadas amostras sanguíneas de oito jaguatiricas, que se submeteram à contagem de eritrócitos, leucócitos, plaquetas e à determinação do volume globular pelo método manual padrão e por meio do aparelho BC - 2800 VET® com a configuração para gatos domésticos. Os resultados foram avaliados por meio do teste t para dados pareados, e as técnicas submetidas à análise de correlação de Pearson. A técnica automatizada demonstrou resultados estatisticamente semelhantes para eritrócitos e leucócitos. Apenas para volume globular houve diferença significativa entre as técnicas manual e automatizada, mas a correlação foi alta. Apesar de não haver diferença significativa entre as técnicas para plaquetas, a correlação foi baixa. Conclui-se que o aparelho BC - 2800 VET® com a configuração para gatos domésticos é uma técnica confiável na realização do eritrograma e do leucograma para jaguatiricas. Para a determinação do parâmetro volume globular, o aparelho pode ser utilizado, desde que se faça a correção. Para a contagem de plaquetas, a técnica manual é recomendada.(AU)

The purpose of this study was to evaluate the automated technique for blood cells count of ocelots (Leopardus pardalis), comparing it with the manual technique. Blood samples were collected from eight ocelots, which were submitted to counting of erythrocytes, leukocytes, platelets and packed cell volume by the standard manual method and by the device VET ® 2800 BC with the configuration for domestic cats. The results were evaluated using the t test for paired data and the techniques submitted to Pearson correlation. The automated technique showed statistically similar results to erythrocytes and leukocytes. Significant difference was found only for packed cell volume between the manual and automated techniques, but the correlation was high. Although there was no significant difference between the techniques for platelets, the correlation was low. We concluded that the 2800 BC VET ® device with the configuration for domestic cats is a reliable technique in performing the erythrocyte and leukocyte counts for ocelots. The device may be used to determine packed cell volume, provided the correction is made. The manual technique is recommended for the platelet count.(AU)
