RESUMEN

Since 1995, national forensic DNA databases have used a maximum number of contributors, and a minimum number of loci to reduce the risk of providing false leads. DNA profiles of biological traces that do not meet these criteria cannot be loaded into these databases. In 2023, about 10 % of more than 15,000 trace DNA profiles analyzed in western Switzerland were not compared at the national level, even though they were considered to be interpretable, mainly because they contained the DNA from more than two persons. In this situation, police services can request local comparisons with DNA profiles of known persons and/or with other traces, but this occurs in only a small proportion of cases, so that DNA mixtures are rarely used to help detect potential series. The development of probabilistic genotyping software and its associated tools have made possible the efficient performance of this type of comparison, which is based on likelihood ratios (LR) rather than on the number of shared alleles. To highlight potential common contributors for investigation and intelligence purposes, the present study used the mixture-to-mixture tool of the software STRmix v2.7 to compare 235 DNA profiles that cannot be searched the Swiss DNA database. These DNA profiles originated from traces collected by six different police services in 2021 and 2022. Traces were selected by the police based on information that indicated that they were from potential series. Associations between profiles were compared with expected investigative associations to define the value of this approach. Among the 27,495 pairwise comparisons of DNA profiles, 88 pairs (0.3 %) showed at least one potential common contributor when using a LR threshold of 1000. Of these 88 pairs, 60 (68.2 %) were qualified by the police services as "expected" (60/88), 22 (25.0 %) as "possible", and six (6.8 %) as "unexpected". Although it is important to consider the limits of this approach (e.g., adventitious or missed associations, cost/benefit evaluation, integration of DNA mixture comparison in the process), these findings indicate that non CODIS loadable DNA mixtures could provide police agencies with information concerning potential series at both the local and national level.

This research highlights the underestimated significance of cigarette paper as evidence at crime scenes. The primary objective is to distinguish cigarette paper from similar-looking alternatives, addressing the first research objective. The second objective involves identifying cigarette paper brands using attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy and machine learning (ML) algorithms. Accurate differentiation of cigarette paper from normal paper is emphasized. ATR-FTIR spectroscopy, coupled with principal component analysis (PCA) for dimensionality reduction, is employed for brand identification. Among fifteen ML algorithms compared, the CatBoost classifier excels for both objectives. This research presents a non-destructive, effective method for studying cigarette paper, contributing valuable insights to crime scene investigations.

The natural occurrence of 16 inorganic ions relevant to forensic explosives investigations on human hands was studied to support the evaluation of activity-level propositions when such traces are found on the hands or in the fingerprints of a suspect. A total of 594 hand swab extracts from 297 participants throughout Europe and the United States of America were analyzed using Ion Chromatography - Mass Spectrometry. The data provides a reference framework for future covert investigations and forensic casework. The results indicate that thiocyanate, chlorate, nitrite, lithium, strontium, and barium are rarely detected on the hands of individuals who have had no direct contact with explosives (P<0.03) and in quantities below 6 µg. Perchlorate contamination sporadically occurs without deliberately handling perchlorates (P=0.03), albeit at low levels (<12 µg). It also seems that the presence of perchlorate on hands is generally related to professions that involve explosives. Detecting substantial amounts of any of these rare ions on a suspect's hands would require a specific explanation. Because legitimate activities exist that can also result in elevated levels of ions of interest on hands, the context surrounding their presence has to be carefully assessed for each individual case.

PURPOSE: Vitreous humor (VH) is used for postmortem biochemical studies because it is well protected in an uncontaminated state even after death. The goal of this research was to investigate electrolyte concentrations in the VH from human eyes with and without a history of vitrectomy surgery. METHODS: We analyzed the sodium (Na), potassium (K), chloride (Cl) and magnesium (Mg) concentrations from 34 VH samples from 34 patients. Eleven samples were from eyes with a history of vitrectomy, and the remaining 23 eyes had no history of vitrectomy. The correlations of Na, K, Cl and Mg concentrations with patient age, interval between first and second vitrectomy, and lens status (history of cataract surgery) were also evaluated. RESULTS: The Na, K, Cl and Mg concentrations in VH from vitrectomized eyes were 134.1 ± 7.9 mmol/L, 3.7 ± 0.2 mmol/L, 99.7 ± 6.7 mmol/L and 0.59 ± 0.09 mmol/L, respectively; all were significantly lower than the corresponding concentrations in VH from control eyes (lower by 5.0%, 11.0%, 11.7%, and 22.6%, respectively). Na, K, Cl and Mg concentrations in VH from vitrectomized eyes did not show significant correlations with patient ages or the interval between their first and second vitrectomies. There were no significant differences in Na, K, Cl and Mg concentrations in VH between phakic eyes and intraocular lens-implanted eyes. CONCLUSIONS: With the increasing number of vitrectomies being performed, it is necessary to consider the history of vitrectomy when using a subject's VH in forensic examination.

PURPOSE: Cannabidiol (CBD) products are widely used for pain relief, sleep improvement, management of seizures etc. Although the concentrations of Δ9-tetrahydrocannabinol (Δ9-THC) in these products are low (≤0.3% w/w), it is important to investigate if its presence and/or that of its metabolite 11-nor-carboxy-Δ9-THC, is traceable in plasma and urine samples of individuals who take CBD oil products. METHODS: A sensitive GC/MS method for the determination of Δ9-THC, 11-nor-carboxy-Δ9-THC and CBD in plasma and urine samples was developed and validated. The sample preparation procedure included protein precipitation for plasma samples and hydrolysis for urine samples, solid-phase extraction and finally derivatization with N,O-bis(trimethylsilyl)trifluoroacetamide) with 1% trimethylchlorosilane. RESULTS: For all analytes, the LOD and LOQ were 0.06 and 0.20 ng/mL, respectively. The calibration curves were linear (R2 ≥ 0.992), and absolute recoveries were ≥91.7%. Accuracy and precision were within the accepted range. From the analysis of biologic samples of 10 human participants who were taking CBD oil, it was realized that Δ9-THC was not detected in urine, while 11-nor-carboxy-Δ9-THC (0.69-23.06 ng/mL) and CBD (0.29-96.78 ng/mL) were found in all urine samples. Regarding plasma samples, Δ9-THC (0.21-0.62 ng/mL) was detected in 10, 11-nor-carboxy-Δ9-THC (0.20-2.44 ng/mL) in 35, while CBD (0.20-1.58 ng/mL) in 25 out of 38 samples, respectively. CONCLUSION: The results showed that Δ9-THC is likely to be found in plasma although at low concentrations. In addition, the detection of 11-nor-carboxy-Δ9-THC in both urine and plasma samples raises questions and concerns for the proper interpretation of toxicological results, especially considering Greece's zero tolerance law applied in DUID and workplace cases.

Single Nucleotide Polymorphisms (SNPs), as the most prevalent type of variation in the human genome, play a pivotal role in influencing human traits. They are extensively utilized in diverse fields such as population genetics, forensic science, and genetic medicine. This study focuses on the 'Rita' BeadChip, a custom SNP microarray panel developed using Illumina Infinium HTS technology. Designed for high-throughput genotyping, the panel facilitates the analysis of over 4000 markers efficiently and cost-effectively. After careful clustering performed on a set of 1000 samples, an evaluation of the Rita panel was undertaken, assessing its sensitivity, repeatability, reproducibility, precision, accuracy, and resistance to contamination. The panel's performance was evaluated in various scenarios, including sex estimation and parental relationship assessment, using GenomeStudio data analysis software. Findings show that over 95 % of the custom BeadChip assay markers were successful, with better performance of transitions over other mutations, and a considerably lower success rate for Y chromosome loci. An exceptional call rate exceeding 99 % was demonstrated for control samples, even with DNA input as low as 0.781 ng. Call rates above 80 % were still obtained with DNA quantities under 0.1 ng, indicating high sensitivity and suitability for forensic applications where DNA quantity is often limited. Repeatability, reproducibility, and precision studies revealed consistency of the panel's performance across different batches and operators, with no significant deviations in call rates or genotyping results. Accuracy assessments, involving comparison with multiple available genetic databases, including the 1000 Genome Project and HapMap, denoted over 99 % concordance, establishing the Rita panel's reliability in genotyping. The contamination study revealed insights into background noise and allowed the definition of thresholds for sample quality evaluation. Multiple metrics for differentiating between negative controls and true samples were highlighted, increasing the reliability of the obtained results. The sex estimation tool in GenomeStudio proved highly effective, correctly assigning sex in all samples with autosomal loci call rates above 97 %. The parental relationship assessment of family trios highlighted the utility of GenomeStudio in identifying genotyping errors or potential Mendelian inconsistencies, promoting the application of arrays such as Rita in kinship testing. Overall, this evaluation confirms the Rita microarray as a robust, high-throughput genotyping tool, underscoring its potential in genetic research and forensic applications. With its custom content and adaptable design, it not only meets current genotyping demands but also opens avenues for further research and application expansion in the field of genetic analysis.

This work introduces a 3D-printed portable electroplating device for the visualization of latent fingerprints (LFPs) on metallic substrates. An electroplating solution of Ag+-Cu2+ in a deep eutectic solvent (DES) is used. The electroplating is performed by two electrodes equivalent to an anode (+) and a cathode (-). The cathode is connected to the metal surface with the magnetic or alligator clip for carrying the LFP. The anode is connected to cotton dipped in the electroplating solution. The device was optimized in terms of the electroplating solution composition, and electroplating potential, current, and time. The device produced images with good resolution, revealing LFP ridges in minute detail of more than 12 points. The device also exhibited good repeatability and images were assessed against guidelines from the Centre for Applied Science and Technology (CAST) and the International Fingerprint Research Group (IFRG). The developed device could be applied to visualize LFPs in forensic investigations.

Child sexual abuse is a public health and medico-legal problem in developed and developing countries. Prompted by rare presentations of cases from this issue that change the course and outcome of events, we present the completed criminal-medico-legislative procedure as a contribution to the dynamics of the investigative process from my own many years of forensic practice. Two preschool teachers of late reproductive age from a home for neglected children reported to the State Attorney's Office a former ward of the home, now an older minor, and an auxiliary employee of the same home, for having sexual relations with a two-year-old girl. But the state attorney also ordered an examination of the reported young man, so the reported case that the teachers "saw" penetrative intercourse was rejected by the expert and the state attorney as an action impossible without injuring the girl at that age. Further investigation proved the continuous sexual abuse by two teachers of the young man since he was 13 years old, i.e. for three years, which he did not report for fear that he would be kicked out of the home. He had sexual relations with each of them several times during those incriminated years. After his statement, other employees of the home were questioned as witnesses who confirmed the suspect boy's claims. He was then released from custody, and the teachers were dismissed from their jobs and accused of years of sexual abuse of minors and abuse of their position and authority as competent persons.

The preservation of drug stability in biological evidence during the processes of collection and storage poses a substantial obstacle to the progress of forensic investigations. In conjunction with other constituents, the microorganisms present in the samples play a vital role in this investigation. The present investigation employed the high-performance liquid chromatography (HPLC) technique to assess the stability of (1R,2 S)-(-)-2-methylamino-1-phenyl-1-propanol hydrochloride in plasma and urine samples that were inoculated with Escherichia coli. These samples were subjected to storage conditions of 37 °C for 48 h and - 20 °C for a duration of 6 months. Minimal inhibitory concentration (MIC) and Minimal bactericidal concentration (MBC) of MPPH against E. coli were determined using microdilution method. The stability of MPPH in plasma and urine samples inoculated with E. coli was investigated using HPLC method. The results showed the MIC and MBC of MPPH were 87.5 ± 25 ppm and 175 ± 50 ppm, respectively. While MPPH remained stable in plasma for 48 h at 37 °C, it showed a notable decrease of about 11% in stability when stored in urine for the same period and temperature. From the beginning of the first month, a decrease in the stability of the compound appeared in all samples that were stored at - 20 °C, and the decrease reached 7% for plasma samples and about 11% for urine samples. The decrease in the stability reached its peak in the sixth month, reaching more than 30% and 70% of plasma and urine samples preserved at - 20 °C. This work concluded that E. coli can negatively affect the stability of MPPH in plasma and urine samples. This may lead to incorrect conclusions regarding the analysis of biological samples in criminal cases.

RESUMEN

Already for some decades lateral flow assays (LFAs) are 'common use' devices in our daily life. Also, for forensic use LFAs are developed, such as for the analysis of illicit drugs and DNA, but also for the detection of explosives and body fluid identification. Despite their advantages, including ease-of-use, LFAs are not yet frequently applied at a crime scene. This review describes (academic) developments of LFAs for forensic applications, focusing on biological and chemical applications, whereby the main advantages and disadvantages of LFAs for the different forensic applications are summarized. Additionally, a critical review is provided, discussing why LFAs are not frequently applied within the forensic field and highlighting the steps that are needed to bring LFAs to the forensic market.

Understanding the underlying hydrodynamics of impinging blood droplets and finding out the physical parameters determining the bloodstain characteristics are of great importance in blood related forensic investigations. In this work, the impact of non-Newtonian blood droplets on solid surfaces ranging from lyophilic to superlyophobic was systematically investigated and compared to that of Newtonian droplets with a similar dynamic shear viscosity. We show that impinging blood droplets behave as low-viscosity Newtonian droplets in the short-time spreading, which is dominated by capillary and inertial forces, but their non-Newtonian viscoelasticity would notably affect the droplet retraction and post-impact oscillation occurring in large timescales. Whereas the strong liquid-solid adhesion and the non-Newtonian elongational viscosity hinder droplet recoiling and thus alter the impact phenomena on lyophobic and superlyophobic surfaces, the shear and elongational viscosities are coupled to result in higher damping coefficients of oscillating blood droplets after deposition, in comparison to that of impinging Newtonian droplets. The size of the dried bloodstain was found to be different from both the maximum spreading radius of the droplet that can reach during impact and the final radius of the deposited droplet after oscillation, and their correlations are highly dependent on the impact velocity and surface wettability. Moreover, the morphologic characteristics of the bloodstains would also be changed by varying either the impact velocity or the surface wettability. We envision that these findings can not only find applications in the bloodstain pattern analysis, but also provide useful information for medical diagnosis based on blood droplet test.

RESUMEN

Automation is crucial for managing the increasing volume of digital evidence. However, the absence of a clear foundation comprising a definition, classification, and common terminology has led to a fragmented landscape where diverse interpretations of automation exist. This resembles the wild west: some consider keyword searches or file carving as automation while others do not. We, therefore, reviewed automation literature (in the domain of digital forensics and other domains), performed three practitioner interviews, and discussed the topic with domain experts from academia. On this basis, we propose a definition and then showcase several considerations concerning automation for digital forensics, e.g., what we classify as no/basic automation or full automation (autonomous). We conclude that it requires these foundational discussions to promote and progress the discipline through a common understanding.

The use of the drug scopolamine in drug-facilitated crimes is known. Nevertheless, given the high potency of the drug and its rapid metabolism, analysis in blood and urine may not be sufficient for drug detection in late crime declaration, especially following a single-dose administration in drug-facilitated sexual assault (DFSA) cases. Hair may constitute an essential supplemental matrix extending the drug detection window in such cases. This case report presents quantitative data on scopolamine findings in urine and hair in a DFSA case. A young female had consumed several alcoholic drinks at a party venue when her behaviour became noticeably peculiar. Later, she woke up next to an unknown man and had no recollection of the night's events. Blood and urine samples were collected 18 h after the incident. The initial toxicological target screening using UHPLC-TOF-MS detected scopolamine in the hydrolysed urine sample, and quantification yielded 41 µg/L scopolamine in urine, while blood was negative. Segmental hair analysis using multitarget UHPLC-MS/MS was performed on three washed 2-cm segments of hair collected five weeks after the incident, yielding 0.37 pg/mg scopolamine only in the relevant hair segment. This case report provides novel insight into the concentration in hair following a single exposure of scopolamine and the feasibility of detecting scopolamine in hair by comparison to published toxicological findings.

Introduction: Lip print of an individual is distinct and could be a useful form of evidence to identify the ethnicity of a terrorist. Objectives: The study analyzed the distribution of lip print patterns of two major ethnic groups in Nigeria; Ibo and Hausa, to develop a strategic plan to check the wave of ethnically driven terrorism in Nigeria, carried out by groups such as Boko Haram and Indigeneous People of Biafra (IPOB). Materials and methods: The study comprised 800 participants of Ibo and Hausa ethnic groups (400 males and 400 females). The study adopted a digital method of lip print analysis and followed the guidelines outlined by the Institute of Medicine (IOM) for anthropometric measurements. The lip was classified, using Tsuchihashi and Suzuki method of classification. Results: The predominant lip print patterns of Ibo were Type I with complete vertical groove and Type III with intersect of groove for male and Type III for female. Type I' with partial length groove was the predominant pattern for both male and female Hausa. The lip width and height of female Ibo were longer than that of the Hausa counterpart (P < 0.05), but none of the anthropometric variables could predict the lip print pattern. Conclusion: The lip size and print could aid forensic investigation, though genetic diversity and heterogeneity of ethnic groups in Nigeria, especially that of Ibo, could setback use of lip print pattern to identify the ethnicity of an unknown individual in Nigeria to help determine the terrorist group to which they may belong.

Microsoft released a new communication platform, Microsoft Teams, in 2017. Due in part to COVID-19, the popularity of communication platforms, like Microsoft Teams, increased exponentially. Given its user base and increased popularity, it seems likely that digital forensic investigators will encounter cases where Microsoft Teams is a relevant component. However, because Microsoft Teams is a relatively new application, there is limited forensic research on the application particularly focusing on mobile operating systems. To address this gap, an analysis of data stored at rest by Microsoft Teams was conducted on the Windows 10 operating system as well as on Android and Apple iOS mobile operating systems. Basic functionalities, such as messaging, sharing files, participating in video conferences, and other functionalities that Teams provides, were performed in an isolated testing environment. Cellebrite UFED Physical Analyzer and Magnet AXIOM Examine tools were used to analyze the mobile devices and the Windows device, respectively. Manual or non-automated investigation recovered, at least partially, the majority of artifacts across all three operating systems. In this study, a total of 77.6% of the populated artifacts were partially or fully recovered in the manual investigation. On the other hand, forensic tools used did not automatically recover many of the artifacts found with the manual investigation. Only 13.8% of artifacts were partially or fully recovered by the forensic tools across all three devices. These discovered artifacts and the results of the investigations are presented in order to aid digital forensic investigations.

Guidelines from the Netherlands describe that unnatural deaths should be investigated by a forensic physician and Crime Scene Technicians, but this is not always the case. In this study, we aimed to determine what predicts the non-attendance of the Crime Scene Technicians at the scene of the death of suicides in the police region in Rotterdam, the Netherlands. Data of 315 suicides (2016-2017) that have been externally examined by forensic physicians and reports from the Crime Scene Technicians were analysed. Statistical analysis was performed to determine the factors predicting the involvement of the Crime Scene Technicians at the scene of death. The Crime Scene Technicians were not attending in 23% (n = 72) cases, and over half of these cases were not found in the registration system of the Crime Scene Technicians. About some the Crime Scene Technicians was not informed. Predictors of the non-attendance of the Crime Scene Technicians were suicide by poisoning, the individual was found by an acquaintance or family, a history of suicide attempts and examination of the deceased in the hospital. In this study, we observed that the Crime Scene Technicians were sometimes not attending the investigation of apparent suicides. More research on this topic should be done to investigate the value of the presence of the Crime Scene Technicians at the scene before concluding that non-attendance of Crime Scene Technicians on-site influences the quality of the scene of death investigation and corresponding conclusions. Since 2017, several quality improvements were made, but an (inter)national conjoint protocol for uniform and structural suicide investigation could secure the quality of the scene of death investigation of suicides and would provide information useful for evaluation.

Forensic imaging is a non-invasive examination process during the forensic investigation. It is mainly used in forensic pathology as an adjunct to the traditional autopsy. In the past two decades, forensic imaging has been vigorously developed by forensic experts from computed tomography (CT) to multiple augmented techniques through CT and magnetic resonance imaging (MRI). The application field of forensic imaging has also been broadened as its advantages are recognised by more forensic practitioners. In addition to the forensic pathology, this technique has been used in other forensic disciplines, including forensic anthropology, forensic odontology, forensic ballistics and wildlife forensics, etc. This article reviews the development of forensic imaging as the practice and research development in different forensic disciplines based on the relevant literature analysis.

In forensic chemistry, when investigating seized illicit drugs, the profiling or chemical fingerprinting of drugs is considered fundamental. This involves the identification, quantitation and categorization of drug samples into groups, providing investigative leads such as a common or different origin of seized samples. Further goals of drug profiling include the elucidation of synthetic pathways, identification of adulterants and impurities, as well as identification of a drug's geographic origin, specifically for plant-derived exhibits. The aim of this state-of-art-review is to present the traditional and advanced analytical approaches commonly followed by forensic chemists worldwide for illicit drug profiling. We discussed numerous methodologies for the physical and chemical profiling of organic and inorganic impurities found in illicit drug. Applications of powerful spectroscopic and chromatographic tools for illicit drug profiling including isotope-Ratio mass spectrometry (IRMS), gas chromatography-mass spectrometry (GC-MS), gas chromatography-isotope ratio mass spectrometry (GC-IRMS), ultra-high-performance liquid chromatography (UHPLC), thin layer chromatography (TLC), liquid chromatography-mass spectrometry (LC-MS) and inductively coupled plasma-mass spectrometry (ICP-MS) were discussed. Altogether, the techniques covered in this paper to profile seized illicit drugs could aid forensic chemists in selecting and applying a suitable method to extract valuable profiling data.

BACKGROUND: We sought to evaluate the diagnostic accuracy of post-mortem cardiac magnetic resonance (PMCMR) of explanted hearts to detect the cardiac causes of sudden death. METHODS: PMCMR was performed in formalin-fixed explanted hearts of 115 cases of sudden death. Histological sampling of myocardium was performed using two different approaches: (1) guideline-based sampling; (2) guideline-based plus PMCMR-driven sampling. RESULTS: Forensic diagnosis of cardiac cause of death was ascertained in 72 (63%) patients. When the guideline-driven histological sampling was used, the PMCMR interpretation matched with final forensic diagnosis in 93 out of 115 cases (81%) with sensitivity of 88% (79-95%), specificity of 65% (47-80%), PPV of 84% (78-90%), NPV of 73% (58-84%), accuracy of 81% (72-88%), and AUC of 0.77 (0.68-0.84). When a PMCMR-driven approach was added to the guideline-based one, the matching increased to 102 (89%) cases with a PMCMR sensitivity of 89% (80-94%), a specificity of 86% (67-96%), PPV of 95% (89-98%), NPV of 73% (59-83%), accuracy of 89% (81-93%), and AUC of 0.88 (0.80-0.93). CONCLUSIONS: PMCMR has high accuracy to identify the cardiac cause of sudden death and may be considered a valid auxilium for forensic diagnosis. PMCMR could improve histological diagnosis in conditions with focal myocardial involvement or demonstrating signs of myocardial ischemia.

Background and Objectives: Aquaporins are a family of water channel proteins. In this study, the renal and intrapulmonary expression of aquaporin-5 (AQP5) was examined in forensic autopsy cases to evaluate it as a drowning marker and to differentiate between freshwater drowning and saltwater drowning. Materials and Methods: Cases were classified into three groups: freshwater drowning (FWD), saltwater drowning (SWD), and controls (CTR). Samples were obtained from forensic autopsies at less than 72 h postmortem (15 FWD cases, 15 SWD cases, and 17 other cases) and were subjected to histological and immunohistochemical investigations. Results: In FWD group, intrapulmonary AQP5 expression was significantly suppressed compared with SWD and CTR; there was no significant difference in AQP5 expression among the other two groups. The same differences in expression were also observed in the kidney. Conclusions: These observations suggest that AQP5 expression in alveolar cells was suppressed by hypotonic water to prevent hemodilution. Moreover, it is possible to hypothesize that in the kidney, with the appearance of hypo-osmotic plasma, AQP5 is hypo-expressed, as a vital reaction, to regulate the renal reabsorption of water. In conclusion, the analysis of renal and intrapulmonary AQP5 expression would be forensically useful for differentiation between FWD and SWD, or between FWD and death due to other causes.
