RESUMEN

Bacitracin Methylene Disalicylate (BMD), as a feed additive to poultry diets, enhances digestion, prevents Salmonella enteritidis (SE) colonization, and treats current infections. The objective of this study was to utilize a quantitative proteomic approach to determine the effect of BMD feed additive on broiler chickens challenged with SE in the spleen proteome. At 1 d of age, chicks were randomly allocated into four groups: control with and without SE challenge (CON, n = 60; CON-SE, n = 60), BMD with and without SE challenge (BMD, n = 60; BMD-SE, n = 60). Birds in the CON-SE and BMD-SE treatment were administered SE inoculum by oral gavage. On day three and day seven post-gavage, the spleen was collected aseptically from birds in each treatment group (CON, n = 4/day; CON-SE, n = 4/day; BMD, n = 4/day; BMD-SE, n = 4/day). Proteomic analysis by liquid chromatography-tandem mass spectrometry (LC-MS/MS) showed an increased abundance of 115 proteins and decreased of 77 due to the BMD. Proteins that decreased in abundance were enriched for fibrinogen complex and extracellular space, whereas proteins that increased in abundance were enriched for proteasome-mediated ubiquitin-dependent protein catabolic process and mitochondrion. Analysis of the interaction between BMD and the Salmonella challenge found 230 differentially abundant proteins including proteins associated with RNA binding, spliceosome, protein transport, and cell adhesion among the upregulated proteins, and those associated with protein folding, carbon metabolism, biosynthesis of nucleotide sugars, response to oxidative stress, positive regulation of NIK/NF-kappaB signaling, and inflammatory response among the downregulated proteins. The impact of BMD treatment on spleen proteome indicates an anti-apoptotic effect. BMD also modified the response of the spleen to the SE challenge with a marked decrease in proteins that prompt cytokine synthesis and an increase in proteins involved in the selective removal of unfolded proteins.

Background: The prevalence of avian coccidiosis in the poultry industry has grown, resulting in substantial financial losses from high mortality, stunted growth, reduced productivity, and expensive medical expenses. Aim: The purpose of the current study was to assess the immunological effects of neem leaf extract and toltrazuril on broilers that had contracted coccidiosis. Methods: In this investigation, 100 one-day-old Cobb broiler chicks without sexes were employed. The chicks were divided into five equal groups, with 20 birds in each. On the 14th day of life, the birds in groups 2, 3, 4, and 5 received an oral inoculation with 1 × 105 sporulated oocysts of Eimeria tenella (E. tenella) (field isolate). The first group (Gp), which consists of 20 healthy broilers, served as a negative control. Gp (2) contains experimentally infected broilers and nontreated (served as a positive control). Gp (3) contains experimentally infected broilers treated with toltrazuril (1 ml/l drinking water) for two consecutive days. Gp (4) contains experimentally infected broilers treated with neem leaf extract 4% (50 ml/l drinking water) for 5 successive days, and Gp (5) contains experimentally infected broilers treated with toltrazuril (1 ml/l drinking water) and a half dose of neem leaves extract 4% (25 ml/l drinking water) for 5 successive days. For the purpose of estimating body weight growth and feed conversion ratio, each broiler was weighed separately at the start of the trial and again on the 1st and 10th day after treatment. In addition to obtaining intestinal samples for immunohistochemistry, blood samples were also obtained for immunological examination. Results: As compared to the negative control group, the experimentally infested broilers with E. tenella showed significant decreases in serum nitric oxide, lysosome, phagocytic percent, and phagocytic index, along with significant increases in white blood cells (WBCs), lymphocyte, heterophilis, eosinophilis, basophilis, monocyte, serum total protein, γ globulin, fibrinogen, and haptoglobin. When compared to the control positive group, experimentally infested broilers treated with either neem or toltrazuril alone or in combination demonstrated significant increases in serum total protein, nitric oxide, lysozyme, phagocytic percent, and phagocytic index, but significant decreases in WBCs, lymphocytes, heterophile, eosinophile, basophile, and monocyte. The intestinal peroxidase stain of broilers infected with E. tenella exhibited a significant positive expression for CD4, but the infected broilers treated with toltrazuril and half a dosage of neem displayed a negative expression for CD4, identical to the negative control. Conclusion: The broiler chickens infested with E. tenella may have a variety of negative impacts on their immune systems and immunohistopathological findings. Nonetheless, toltrazuril and neem extract, either separately or in combination, function as anticoccidial medications that may enhance the broiler chicks' immune state.

Background: A neurological infectious viral disease, avian encephalomyelitis was initially discovered in 2-week-old commercial chicks in 1930 and classified as a neurotropic viral disease. Aim: A neurological outbreak caused by avian encephalomyelitis virus (AEV) in young chicks was first reported in Al-Ahsa in the Kingdom of Saudi Arabia (KSA) in 2010. The aim of this article is to examine the AEV in KSA, Al-Ahsa Province. Methods: Gizzard, proventriculus, cerebrum, cerebellum, and medulla oblongata tissue samples were collected from infected chicks for histopathology test and molecular identification. Results: Infected chicks showed neurological signs particularly incoordination, mild head and neck tremors, stretching of legs, and lameness. The average morbidity and mortality rates were 35% and 10%, respectively. At necropsy, no obvious identifiable macroscopic lesions were found in the infected chicks. Nonsuppurative encephalomyelitis was found histopathologically in the central nervous system, mainly in the cerebral molecular layer. Microscopic lesions in the proventriculus showed masses of heavy numbers of small lymphocytes within the muscular layer. RT-PCR followed by sequence analysis revealed that The KSA strain (KJ939252) is intimately related to chicken European strains from Poland (KC912695) and the United Kingdom (AJ225173) with identity 99.6% than Chinese strains (AY225319, AY517471, and AY275539) with identity ranged between 94.6% and 95%. The phylogenetic tree analysis showed that the KSA strain is grouped in a similar clade with chicken European strains. Conclusion: The pattern of disease findings was typical of vertically transmitted AEV. The spread of AEV in Saudi Arabia is most likely due to the trade of birds and bird products with European countries.

OBJECTIVES: Pseudomonas aeruginosa (P. aeruginosa) is one of the most serious pathogens implicated in antimicrobial resistance, and it has been identified as an ESKAPE along with other extremely significant multidrug resistance pathogens. The present study was carried out to explore prevalence, antibiotic susceptibility phenotypes, virulence-associated genes, integron (int1), colistin (mcr-1), and ß-lactamase resistance' genes (ESBls), as well as biofilm profiling of P. aeruginosa isolated from broiler chicks and dead in-shell chicks. DESIGN: A total of 300 samples from broiler chicks (n = 200) and dead in-shell chicks (n = 100) collected from different farms and hatcheries located at Mansoura, Dakahlia Governorate, Egypt were included in this study. Bacteriological examination was performed by cultivation of the samples on the surface of both Cetrimide and MacConkey's agar. Presumptive colonies were then subjected to biochemical tests and Polymerase Chain Reaction (PCR) targeting 16S rRNA. The recovered isolates were tested for the presence of three selected virulence-associated genes (lasB, toxA, and exoS). Furthermore, the retrieved isolates were subjected to phenotypic antimicrobial susceptibility testing by Kirby-Bauer disc diffusion method as well as phenotypic detection of ESBLs by both Double Disc Synergy Test (DDST) and the Phenotypic Confirmatory Disc Diffusion Test (PCDDT). P. aeruginosa isolates were then tested for the presence of antibiotic resistance genes (ARGs): int1, mcr-1, and ESBL genes (OXA-10, OXA-2, VEB-1, SHV, TEM, and CTX-M). Additionally, biofilm production was examined by the Tube Adherent method (TA) and Microtiter Plate assay (MTP). RESULTS: Fifty -five isolates were confirmed to be P. aeruginosa, including 35 isolates from broiler chicks and 20 isolates from dead in-shell chicks. The three tested virulence genes (lasB, toxA, and exoS) were detected in all isolates. Antibiogram results showed complete resistance against penicillin, amoxicillin, ceftriaxone, ceftazidime, streptomycin, erythromycin, spectinomycin, and doxycycline, while a higher sensitivity was observed against meropenem, imipenem, colistin sulfate, ciprofloxacin, and gentamicin. ESBL production was confirmed in 12 (21.8%) and 15 (27.3%) isolates by DDST and PCDDT, respectively. Antibiotic resistance genes (ARGs): int1, mcr-1, and ESBL genes (OXA-10, SHV, TEM, and CTX-M), were detected in 87.3%, 18.2%, 16.4%, 69.1%, 72.7%, and 54.5% of the examined isolates respectively, whereas no isolate harbored the OXA-2 or VEB-1 genes. Based on the results of both methods used for detection of biofilm formation, Kappa statistics [kappa 0.324] revealed a poor agreement between both methods. CONCLUSIONS: the emergence of mcr-1 and its coexistence with other resistance genes such as ß-lactamase genes, particularly blaOXA-10, for the first time in P. aeruginosa from young broiler chicks and dead in-shell chicks in Egypt pose a risk not only to the poultry industry but also to public health.

The zinc oxide nanoparticles (ZnONPs) have attracted exhilarating research interest due to their novel distinguishing characteristics such as size, shape, high surface activity, large surface area and biocompatibility. Being highly bioavailable and exerting a superior efficacy than conventional zinc sources, ZnONPs is emerging as an alternative feed supplement for poultry. The present study involves the synthesis of ZnONPs through a cost effective and eco-friendly method using planetary ball milling technique and characterized for its size, shape, optical property, functional group and elemental concentration using particle size analyzer, Transmission Electron Microscopy, X-Ray Diffraction analysis, Fourier Transform Infra-Red spectroscopy, UV-Vis spectroscopy and Inductively Coupled Plasma-Mass Spectroscopy. In vitro cytotoxicity study using Baby Hamster kidney (BHK-21) cells, Vero cells and primary chick liver culture cells revealed that ZnONPs can be safely incorporated in the broiler chick's feed up to the concentration of 100 mg/kg. To investigate the effects of ZnONPs on production performances in broiler chicks, a feeding trial was carried out using 150-day-old broiler chicks randomly allotted in five treatment groups. The dietary treatment groups were: T1 (80 mg/kg of zinc oxide), T2 (60 mg/kg of zinc methionine) and T3, T4 and T5 received 60, 40 and 20 mg/kg of ZnONPs respectively. The results showed a significant improvement (p < 0.05) in the body weight gain and feed conversion ratio of broiler chicks supplemented with 20 and 40 mg/kg of ZnONPs. The ZnONPs supplementation significantly (p < 0.05) increased the dressing percentage in addition to significant (p < 0.05) reduction in the meat pH compared to inorganic and organic zinc supplementation. Overall, an eco-friendly method for ZnONPs synthesis was demonstrated and the optimum dietary level (20 mg/kg) of ZnONPs could enhance the growth, the meat quality and Zn uptake without any negative effects on selected serum biochemical parameters in the broiler chicks.

Bioavailability is critical in ensuring bioefficacy of ginger compounds, which have not been studied in chicks. In this study, day-old chicks were treated with ginger root extract at 0.0, 0.4, 0.8, 1.5, and 3.0% for 42 days. The gingerols and shogaols in chick samples were analyzed by liquid chromatography-mass spectrometry. The primary phase-I metabolic pathway for gingerols and shogaols was the reduction of ketone groups into hydroxyl groups. Shogaols were also metabolized through thiol conjugation and hydrogenation of double-bond pathways. Within the bloodstream, gingerols and their metabolites predominantly existed as glucuronidate or sulfate conjugates. However, the levels of the free form and conjugates were comparable for shogaols. In breast meat, the quantities of both the free form and conjugates for all compounds were similar. In plasma, more than 50% of absorbed 6-gingerol (6G) and 90% of absorbed 6-shogaol underwent reduction to their respective metabolites. However, in breast meat, the percentage of reduction for absorbed 6G was less than 50%, and for absorbed 6-shogaol, it was less than 60%. Ginger compounds were absorbed into chick plasma ranging from 1.4 to 8.5 µg/mL and breast meat ranging from 7.1 to 114.6 µg/100 g across the 0.4-3.0% dose range in a dose-dependent manner.

In current study the pharmaceutically active herbs was used against coccidiosis, caused by a protozoan: Eimeria, lead to $ 3 billion loss annually. The aqueous and methanolic extracts of whole plants were applied in-vitro to assess sporulation inhibition (spi) assay and calculated the inhibitory concentration (IC50). For in-vivo study 9 groups of 14 day old broiler chicks were infected with Eimeria tenella and three groups were treated different concentrations of methanolic extracts of Verbena officinalis and Polygonum glabrum post infection. The mean weight gain, oocyst count, diarrhea, biochemical tests, hematology, and histopathology of all groups were analyzed. The herbs were characterized by antioxidant assay, phytochemical screening, Fourier transmission and infrared (FT-IR), Ultra Violet-visible (UV-Vis) spectroscopy and Gas chromatography and mass spectroscopy (GC-MS). The GC-MS identified phyto-compounds of V. officinalis were docked with S-Adenosyl methionine (SAM) synthetase. The in-vitro study revealed that V. officinalis and P. glabrum have minimum IC50 of 0.14 and 12 mg/ml respectively. The in-vivo experiment showed that V. officinalis had significantly high anticoccidial potential with significant hematological profile like drug treated controls. The histology of treated chicks also showed recovery in the studied tissues. The antioxidant assay showed that V. officinalis have 4.19U/mg Superoxide dismutase (SOD) and 33.96 µM/mg Glutathione (GSH) quantities. The chemical characterization confirmed the presence of large number of organic compounds, however Flavonoids found only in V. officinalis, which suggests the anticoccidial potential of V. officinalis because flavonoids as antagonist of thiamine (Prinzo, 1999), because it promotes the carbohydrate synthesis required. Strychane, 1-acetyl-20a-hydroxy-16-methylene has best binding of with target protein with lowest binding score (-6.4 Kcal/mol), suggests its anticoccidial potential in poultry.

This experiment was conducted to study the effect of housing systems and housing densities on the performance and digestive tract growth of broiler chicks during the first 2 wk of age. A total of 3,600 Cobb500 day-old chicks were stocked at 4 densities (30, 60, 90, and 120 chicks/m2), and reared under 2 housing systems (conventional housing system and newly developed housing system), yielding a 2 × 4 factorial arrangement. The studied traits were performance, viability, and gastrointestinal tract development. The results indicated that housing systems and housing densities significantly (P < 0.001) affected the performance and GIT development of chicks. There were no significant interactions between housing system and housing density for body weight, body weight gain, feed intake, and feed conversion. The results also showed that the effects of housing density were age-dependent. That is, the higher the density, the lower the performance and digestive tract growth with advancing age. In conclusion, birds in the conventional system outperformed birds in the newly developed housing system, and further work is needed to improve the new housing system. To achieve the highest performance, digestive tract growth, and digesta content, a density of 30 chicks/m2 is recommended for chicks up to 14-days old.

The objective was to determine the effect of rosemary essential oil (REO) as nitrite substitute on the physicochemical, microbiological and sensory quality of sausage from broilers fed different levels of thymus essential oil (TEO) and rapeseed oil (RO). Broiler chicks fed TEO (0, 300 and 500 ppm) and RO (1, 3 and 5%) in a 3 × 3 factorial design. Sausages were made from meat of chicken fed TEO (0 and 500 ppm) treated with REO (120 ppm), REO:nitrite (60:60 ppm), nitrite (120 ppm) or without REO and nitrite in a 2 × 4 factorial design. Results showed that TEO increased water holding capacity and pH value and decreased malondialdehyde concentration. RO (1 and 3%) × TEO (500 ppm) group had higher unsaturated to saturated fatty acids ratio in the meat. Results showed that treatments of REO (120 ppm) decreased lipid oxidation and increased pH value of meat. Sausage produced by nitrite (120 ppm) or REO:nitrite (60:60 ppm) had lower total microbial count, lightness but higher redness and yellowness. Based on the results, TEO in chicken feed decreased lipid oxidation of sausages. Furthermore, substitution at least 60 ppm nitrite with REO had more preserving effect on quality of produced sausages.

This experimental study was conducted to determine the ability of a novel mycotoxins detoxification agent (MR) at a concentration of 0.2% to reduce the toxicity of aflatoxin B1 (AFB1) or T-2 toxin, alone or in combination, and to examine its effect on performance, pathohistological changes (PH) and the residue of these toxins in the tissues of broiler chicks. A total of 96 broiler chicks were divided into eight equal groups: group C, which served as control (without any additives); group MR, which received the novel detoxification agent (supplemented with 0.2%); group E-I (0.1 mg AFB1/kg of diet); group E-II (0.1 mg AFB1/kg of diet + MR 0.2%); group E-III (0.5 mg T-2 toxin/kg of diet); group E-IV (0.5 mg T-2 toxin/kg of diet + 0.2% MR); group E-V (combination of 0.1 mg AFB1/kg, 0.5 mg T-2 toxin/kg of diet); and group E-VI (combination of 0.1 mg AFB1/kg, 0.5 mg T-2 toxin + 0.2% MR). Results indicate that feeds containing AFB1 and T-2 toxin, alone or in combination, adversely affected the health and performance of poultry. However, the addition of MR to diets containing AFB1 and T-2 toxin singly and in combination exerted a positive effect on body weight, feed intake, weight gain, feed efficiency and microscopic lesions in visceral organs. Residual concentration of AFB1 in liver samples was significantly (p < 0.05) decreased when chicks were fed diets supplemented with 0.2% of MR.

The present study investigated the adverse effects of arsenic and chromium in broilers and ascertained the role of vitamin E and bentonite in alleviating their harmful effects. For this purpose, we experimented on 180 one-day-old broiler chickens. The feed was administered to broiler chicks of groups 2, 6, 7, 8, and 9 chromium @ (270 mg.kg-1 BW). Groups 3, 6, 7, 8, and 9 were administered arsenic @ (50 mg.kg-1 BW). Groups 4, 7, and 9 received vitamin E (150 mg.kg-1 BW), and groups 5, 8, and 9 received bentonite (5%), respectively. Group 1 was kept in control. All the broiler chicks treated with chromium and arsenic showed a significant (p < 0.05) decline in erythrocytic parameters on experimental days 21 and 42. Total proteins decreased significantly, while ALT, AST, urea, and creatinine increased significantly (p < 0.05). TAC and CAT decreased significantly (p < 0.05), while TOC and MDA concentrations increased significantly (p < 0.05) in chromium and arsenic-treated groups on experimental days 21 and 42. Pearson correlation analysis revealed a strong positive correlation between TAC and CAT (Pearson correlation value = 0.961; p < 0.001), similarly TOC and MDA positive correlation (Pearson correlation value = 0.920; p < 0.001). However, TAC and CAT showed a negative correlation between TOC and MDA. The intensity of gross and microscopic lesions was more in chromium (270 mg.kg-1) and arsenic (50 mg.kg-1) singly or in combination-treated groups. Thus, broiler chicks treated with chromium plus arsenic exhibited higher gross and microscopic lesion intensity than other treated groups. Fatty degeneration, severe cytoplasmic vacuolar degeneration, and expansion of sinusoidal spaces were the main lesions observed in the liver. Kidneys showed renal epithelial cells necrosis, glomerular shrinkage, and severe cytoplasmic vacuolar degeneration. Co-administration of bentonite along with chromium and arsenic resulted in partial amelioration (group 8) compared to groups 7 and 9, administered arsenic + chromium + vitamin E and arsenic + chromium + vitamin E + bentonite, respectively. It was concluded that arsenic and chromium cause damage not only to haemato-biochemical parameters but also lead to oxidation stress in broilers. Vitamin E and bentonite administration can ameliorate toxicity and oxidative stress produced by arsenic and chromium.

The present study aimed to examine the impacts of supplementing hot red pepper oil (HRPO) to broiler diets. One hundred and twenty Arbor Acres chicks were divided randomly into four experimental groups as three supplementation levels of HRPO (0.25, 0.5 and 1.0 mL/kg diet) and the control group. Results showed that HRPO supplementation exhibited significantly (p < 0.001) higher red blood cells (RBCs) count, hemoglobin (Hb) and packed cells volume (PCV) percentage, while insignificant effects were shown for white blood cells (WBCs) count or its differentiation. Diets supplemented with different levels of HRPO influenced significantly (p < 0.001) the total protein (TP), albumin (Alb) and glucose (Glo) values of the studied birds. Results also indicated that different levels of HRPO supplementations significantly (p < 0.01) decreased total lipid, triglycerides (Trig), cholesterol (Cho) and low-density lipoprotein (LDL), but did not affect high density lipoprotein (HDL) values. Data revealed that supplementing broiler diets with different levels of HRPO enhanced their liver function. The bactericidal activity index was significantly increased (p < 0.02) compared with control. HRPO supplemented groups had beneficial effects (p < 0.02) on cecal microbiota count. It could be concluded that dietary HRPO supplementation could improve the general internal health status of Arbor Acres broiler chicks.

A total of 300 day-old Arbor Acres Plus broiler chicks (mixed sex) was used to evaluate the effects of dietary supplementation of Bacillus subtilis, Clostridium butyricum, and Enterococcus faecalis mixture (PB) on growth performance, ileal morphology, and cecal microbiota. All birds were randomly assigned into 3 groups based on the initial body weight. There were 5 replicate cages per group and 20 birds per cage. The experimental period was 42 days. Dietary treatments were based on a basal diet and supplemented with 0, 0.05, or 0.10% PB. The results indicated that broiler chicks fed with the diet supplemented with graded levels of PB have quadratically improved their body weight gain and feed intake; the highest value was presented in 0.05% PB-containing group. In addition, villus to crypt ratio linearly increased with the concentration of PB increased in the diet. The alpha diversity linearly increased by PB supplementation, and the highest value was presented in 0.10% PB-containing group. In terms of growth performance, the suitable dose of PB used in the diet was 0.05%. However, ternary plot showed that the harmful bacteria, Escherichia-Shigella, was enriched in 0.05% PB-containing group. In brief, we considered that dietary supplementation of graded levels of PB improved growth performance and regulated cecal microbiota in broiler chicks.

Broilers with immune stress show decline of growth performance, causing severe economic losses. However, the molecular mechanisms underlying the immune stress still need to be elucidated. One hundred and twenty broiler chicks were randomly assigned to 2 groups with 6 repeats per group, 10 birds per repeat. The model broilers were intraperitoneally injection of 250 µg/kg LPS at 12, 14, 33, and 35 d of age to induce immunological stress. Control group was injected with an equivalent amount of sterile saline. Blood samples from chickens were collected using wing vein puncture at 35 d of age and the serum was obtained for detection of CORT and ACTH. At the end of the experiment, the liver tissues were excised and collected for omics analysis. The results showed that LPS challenge significantly inhibited growth performance, increased relative weight of liver, spleen and decreased relative weight of bursa, as well as enhanced the concentration of serum ACTH and CORT, when compared with the Control. A total of 129 DEGs and a total of 109 differential metabolites were identified between Model and Control group. Transcriptomics profiles revealed that immune stress enhanced the expression of genes related to defense function while declined the expression of genes related to oxidation-reduction process. Metabolomics further suggested that immune stress changed metabolites related to amino acid metabolism, glycerophospholipid metabolism. In addition, integrated analysis suggested that the imbalance of valine, leucine and isoleucine metabolism, glycerophospholipid metabolism, and mTOR signaling pathway played an important role in immune stress of broiler chicks.

The site-specific administration of neuroactive substances or pharmacological agents is a routine practice in neurological studies. To facilitate chronic treatments intercranial cannulae are often implanted in the skull. These surgical procedures are widely described and well-documented for rodents, as the most common animal model, and even refined over the years. However, precise descriptions of proper procedure for third ventricle cannulation in young broiler chicks is not readily available. This absence is surprising, as chicken models are often used in the investigation of physiological control of the metabolism and ingestion. Furthermore, as a commercial animal, there is also a particular interest in elucidating the central regulation of feed intake and metabolism to optimize feeding and living conditions for broilers. The brain nuclei that serve as the regulatory centers of feeding and metabolism are the arcuate nucleus, the ventromedial hypothalamic nucleus, and the lateral hypothalamus, which are all located near the third ventricle. Here, we provide a full protocol for intracerebroventricular (i.c.v.) cannulation of 7-day old broiler chicks, as well as a review of some of the advances made in rodent i.c.v. cannulations and whether these advances are applicable to cannulations in chickens. Using the surgical procedure described here, we were able to achieve a success rate of proper implantation of 88%, with a mortality rate of less than 1% (n = 224).•Detailed procedure for i.c.v. canula implantation in the third ventricle of 7d-old broiler chicks.•Cement cap with anchoring screws is necessary in broilers, glue does not provide sufficient stability.•Carboxylate cement and self-adhesive resin cement were tested as an alternative for methyl methacrylate cement.

In broiler chicks, Escherichia coli lipopolysaccharide is a prominent cause for inflammatory damage and loss of immune homeostasis in broiler chicks. Ginsenosides have been shown to have anti-inflammatory and antioxidant effects. However, it has not been demonstrated that ginsenosides protect broiler chicks against stress induced by Escherichia coli lipopolysaccharide challenge. The aim of this is to investigate the protective effect of ginsenosides Rg1, Re, and Rg3 on Escherichia coli lipopolysaccharide-induced stress. Our results showed that Rg3 ameliorated growth inhibition and fever, as well as decreased the production of stress-related hormones in broilers with stress. The protective effect of Rg3 on the stressed chicks may be largely mediated by regulating inflammatory response and oxidative damage. Moreover, real-time quantitative-polymerase chain reaction (RT-qPCR) results demonstrated that Rg3 upregulated mRNA expression of mTOR, HO-1, and SOD-1. These results suggested that ginsenoside Rg3 and ginsenoside products contains Rg3 deserve further study for the control of immunological stress and inflammation in broiler chicks.

The current study aimed to discuss the impact of total sulfur amino acids (TSAA) %, photoperiod, and their interaction on growth performance, carcass and blood indices of broiler chicks. A total of 300 unsexed IR broiler chicks one-week old were used in a factorial arrangement (2 × 3), including two photoperiod systems (22 L: 2 D and 16 L: 8 D) and three experimental rations having three grades of Met + Cyst (TSAA) (70%, 85% and 100% of digestible lysine in starter and finisher diets). Results revealed that the higher LBW and BWG were noticed in birds given TSAA at grades of 1.1 or 0.90 % under 22L: 2D photoperiod at five weeks of age and the whole experimental period (1-5 weeks of age), respectively. The highest live body weight (LBW (and body weight gain (BWG) were recorded in birds received 1.1% TSAA under the long photoperiod compared to the control and the other groups. Birds fed 1.3% TSAA consumed more feed than the other groups. The opposite was found in birds fed 1.1% TSAA under the short photoperiod (16L: 8D). The best feed conversion (FCR) was detected by birds fed 1.1% and 0.90% TSAA diets during the whole experimental period. All carcass traits studied were significantly influenced by TSAA levels, except for the relative weights of abdominal fat and spleen. The interaction effect on was significant on all carcass traits except spleen %. In conclusion, the addition of TSAA at level 1.1 and 0.9 % to starter and finisher diets under a long photoperiod regime improved broiler's performance, carcass traits, and blood parameters studied.

Decabromodiphenyl ether (BDE-209) has drawn significant attention due to its suppression of immune functions in animals and even humans. In order to explore the mechanism through which BDE-209 affects the immune system, broiler chicks were fed a diet containing various concentrations of BDE-209 (0, 0.004, 0.04, 0.4, and 4 g/kg) for 42 days. Histopathological observations of immune organs found damaged and necrotic lymphocytes in the spleen and bursa, and losses of lymphoid cells in thymic gland. The activities of catalase, glutathione, glutathione peroxidase, and superoxide dismutase in both the spleen and serum were affected by BDE-209. Obvious bioaccumulation effect was found in spleen tissues (high to 1339 ± 181.9 µg/kg). Furthermore, transcriptome sequencing analyses of the spleen identified 424 upregulated and 301 downregulated DEGs, and the cytokine-cytokine receptor interaction signal pathway was most significantly enriched based on the Kyoto Encyclopedia of Genes and Genomes database. Quantitative real-time PCR affirmed the decreased expressions of interleukin IL18, IL18R1, IL18RAP, IL21, as well as interferon gamma IFNG and tumor necrosis factor superfamily members TNFSF8, indicating significant interference to immunomodulation function and possible disease progression in inflammatory effects resulting from BDE-209 exposure. The immunotoxicity of BDE-209 may cause the suppression of immune and physiological functions of spleen cells, leading to inflammation and apoptosis and ultimately spleen atrophy.

Despite restrictions on the use of antibiotics in poultry, the percentage of multidrug resistant bacteria, isolated from both adult birds and chicks, remains high. These bacteria can spread between countries via hatching eggs or chicks. Antibiotic resistant bacteria can also pose a threat to hatchery and farm workers or to consumers of poultry. The aim of the study was to perform a phenotypic and genotypic analysis of the drug resistance of E. faecalis isolates from yolk sac infections in broiler chicks from Poland and the Netherlands and to determine their genetic diversity. The tests revealed resistance to antibiotics from category D, that is, tetracycline (69.7%); category C - lincomycin (98.7%), erythromycin (51.3%), aminoglycosides (high-level streptomycin and kanamycin resistance - 10.5% and 3.95%, respectively), and chloramphenicol (7.9%); and category B - ciprofloxacin (25% with resistance or intermediate resistance). No resistance to penicillin, ampicillin, high-level gentamicin, tigecycline, or linezolid was noted. Various combinations of the erm(B), tet(M), tet(L), tet(O), ant(6)-Ia, aph(3')-IIIa, ant(4')-Ia, cat, and msr(A/B) genes were detected in all isolates (irrespective of the drug-resistance phenotype). Among isolates that carried the tet(M) and/or the tet(L) gene, 28% also had the Int-Tn gene, in contrast with isolates possessing tet(O). There were 28 sequence types and 43 PFGE restriction patterns. About 60% of isolates were of sequences types ST59, ST16, ST116, ST282, ST36, and ST82. Nine new sequence types were shown (ST836-ST844). In conclusion, broiler chicks can be a source of drug-resistant sequence types of E. faecalis that are potentially hazardous for people and animals. Restrictive programs for antibiotic use in broiler breeding flocks should be developed to decrease drug resistance in day-old chicks and reduce economic losses during rearing.

To investigate whether there are important interactions in play in broilers between thyroid hormones and the central regulation of energy homeostasis through AMP-activated protein kinase (AMPK), we induced a functional hyperthyroid and hypothyroid state in broiler chicks, and quantified systemic and hypothalamic AMPK related gene expression and related protein. Thyroid state was manipulated through dietary supplementation of triiodothyronine (T3) or methimazole (MMI) for 7 days. A hypothalamic AMPK suppressor, 0.1% α-lipoic acid (α-LA) was used to assess the effects of the T3 and MMI feed formulations on the AMPK pathways. Feed intake and body weight were reduced in both hypothyroid and hyperthyroid conditions. In hyperthyroid conditions (T3 supplementation) expression of the AMPKα1 subunit increased, while in hypothyroid conditions (MMI supplementation) active phosphorylated AMPK levels in the hypothalamus dropped, but gene expression of the AMPKα1 and α2 subunit increased. For FAS and ACC (involved in fatty acid metabolism), and CRH, TRH and CNR1 (anorexigenic neuropeptides stimulating energy expenditure) there were indications that their regulation in response to thyroid state might be modulated through AMPK pathways. Our results indicate that the expression of hypothalamic AMPK as well as that of several other genes from AMPK pathways are involved in thyroid-hormone-induced changes in appetite, albeit differently according to thyroid state.
