Previous studies indicate a positive correlation between the duration of estrus prior to ovulation and likelihood of pregnancy in embryo recipient mares. However, the mechanisms by which the duration of estrus before may affect fertility remains unclear. This study aimed to determine the effect of different durations of estradiol exposure, prior to progesterone administration, on embryo viability in anestrous recipient mares, and endometrial expression of genes thought to influence embryo survival. Three groups of anestrous recipient mares treated with different duration of estradiol were used: long (LE), short (SE) and no treatment (NE). Day 8 embryos were transferred into recipient mares four days after long-acting progesterone administration and recovered 48h later to examine embryo growth and viability. The endometrial gene expression profile of selected genes was also investigated. The likelihood of recovering an embryo 48h after transfer was 46.1% (6/13), 62.5% (5/8) and 85.7% (6/7) for recipient mares from the NE, SE and LE groups, respectively (P = .09). Embryos recovered from the different groups of recipients did not, however, differ in size, morphology or the proportion of nuclei undergoing mitosis (P > .05). Abundance of mRNA for uterocalin (P19) and insulin-like growth factor 1 (IGF1) were increased in the LE compared to the NE group, while fibroblast growth factor 2 (FGF2), progesterone receptor (PGR) and insulin-like growth factor 1 receptor (IGF1R) transcript abundances were increased (P < 0.05) in the NE group compared to both SE and LE groups. In conclusion, a longer exposure of the endometrium to estradiol before progesterone tended to improve embryo survival within 48h of transfer. However, the grade, growth rate, and proportion of mitotic cells in surviving embryos did not differ among groups. If embryos are destined to fail in a suboptimal endometrial environment, they die and disappear quickly. Moreover, a more adequately estradiol-primed uterus, before the progesterone rise, seems to create a uterine environment, in terms of P19, IGF1, FGF2 and PGR gene expression, more conducive to embryo survival and further development.

Kisspeptins are neuropeptides encoded by the Kiss1 gene that was discovered as a metastasis suppressor gene in melanoma and breast cancer. Kisspeptin has pivotal functions for gonadotropin-releasing hormone secretion and plays integrated roles in the hypothalamic-pituitary-gonadal axis. However, little is known about the peripheral expression of kisspeptin in ruminants, especially in the female reproductive tract. Here, the objectives of the current study were to investigate the spatial localization of kisspeptin and mRNA expression of Kiss1 and its receptor (Kiss1r) in the fallopian tubes (FT) and uterus of goats at varied reproductive activity (cyclic versus true anoestrous goats, n=6, each). Specimens of the uterus and FT were collected and fixed using paraformaldehyde to investigate the localizations of kisspeptin in the selected tissues by immunohistochemistry. Another set of samples was snape-frozen to identify the expressions of mRNAs encoding Kiss1 and Kiss1r using real-time PCR. Results revealed immunolocalizations of kisspeptin in the uterus and the FT. The staining of kisspeptin was found mainly in the mucosal epithelium of the uterus the FT, and the endometrial glands. Very intense staining of kisspeptin was found in the uterine and FT specimens in the true anoestrous goats compared to that in cyclic ones. The expression of mRNA encoding Kiss1 gene was significantly higher in the uterine specimen of cyclic goats (1.00±0.09) compared to that in the true anoestrous goats (0.62±0.08) (P ˂0.05), while the expression of mRNA encoding Kiss1r was significantly (P ˂0.001) higher in the uterine tissues of true anoestrous goats (1.78±0.17) compared to that in cyclic ones (1.00±0.11). In conclusion, immunohistochemical localization of kisspeptin and the expression of mRNA encoding Kiss1/Kiss1r revealed spatial changes in the uterus and FT of goats according to the reproductive potential of goats (cyclic versus true anoestrous goats). However, the definitive local role of kisspeptin in the uterus and FT need further investigation.

The objective of the present study was to investigate the different plasma metabolites between anestrus and estrus postpartum dairy cows and to provide a theoretical basis for prevention of anestrus in dairy farm cows. In the experiment, one hundred and sixty-seven Holstein dairy cows were selected with similar age and parity. According to the concentration of ß-hydroxybutyric acid, non-esterified fatty acids and glucose in plasma during 14 to 21 days in milk, all dairy cows were determined as having a status of energy balance. According to the results of clinical symptom, rectal and B ultrasound examination at 60 to 90 days postpartum, these cows were divided into twenty estrus and twenty-four anestrus group, other dairy cows were removed. 1H nuclear magnetic resonance technology was utilized to detect the plasma metabolites changes and screen different plasma metabolites between anestrus and estrus cows. Ten different metabolites including alanine, glutamic acid, asparagine, creatine, choline, phosphocholine, glycerophosphocholine, low-density lipoprotein, and very-low-density lipoprotein were significantly decreased in anestrous cows compared with estrous cows. Metabolic pathway analyses indicated that differential metabolites were primarily involved in amino acid and glycerophospholipid metabolism. These metabolites and their enrichment pathways indicate that reduced steroid hormone synthesis precursors result in lower levels of estradiol and progesterone and cause anestrus in negative energy balance. These data provide a better understanding of the changes that may affect estrus of postpartum dairy cows at NEB status and lay the ground for further research.(AU)


O objetivo do presente estudo foi investigar os diferentes metabolitos do plasma entre o cio e o cio pós-parto de vacas leiteiras e fornecer uma base teórica para a prevenção do cio de vacas em fazendas de leite. No experimento, foram selecionadas 127 vacas leiteiras Holstein com idade e paridade similares. De acordo com a concentração de ß- ácido hidroxibutírico, ácidos graxos não esterificados e glicose no plasma entre 14 e 21 dias no leite, todas as vacas leiteiras foram determinadas em estado de equilíbrio energético. De acordo com os resultados dos sintomas clínicos, do exame de ultra-som retal e B aos 60 a 90 dias pós-parto, estas vacas foram divididas em vinte cios e vinte e quatro grupos de cio, outras vacas leiteiras foram removidas. A tecnologia de ressonância magnética nuclear 1H foi utilizada para detectar as alterações dos metabólitos plasmáticos e para triar diferentes metabólitos plasmáticos entre as vacas do cio e do cio. Dez diferentes metabólitos incluindo alanina, ácido glutâmico, asparagina, creatina, colina, fosfocholina, glicerofosfocolina, lipoproteína de baixa densidade e lipoproteína de muito baixa densidade foram significativamente diminuídos nas vacas antróficas em comparação com as vacas estro. As análises da via metabólica indicaram que os metabólitos diferenciais estavam principalmente envolvidos no metabolismo de aminoácidos e glicerofosfolipídios. Estes metabólitos e suas vias de enriquecimento indicam que a redução dos precursores da síntese de hormônios esteróides resulta em níveis mais baixos de estradiol e progesterona e causa anestros no balanço energético negativo. Estes dados fornecem uma melhor compreensão das mudanças que podem afetar o cio das vacas leiteiras pós-parto no estado de NEB e preparam o terreno para mais pesquisas.(AU)

The objective of this study was to investigate the influence of plasma pathological changes before timed artificial insemination (TAI) on pregnancy of cows. The contents of estrogen (E2), progesterone (P4), glucose (Glu), selenium (Se), brain-derived neurotrophic factor (BDNF), and histamine (HIS) in plasma of 48 Holstein cows were measured before TAI. According to the estrus detection, the cows were divided into estrus (E) and anestrus (A) groups. After pregnancy testing at 28 d after TAI, two groups of E and A were divided into positive pregnancy of E group (EP+), negative pregnancy of E group (EP-), positive pregnancy of A group (AP+), and negative pregnancy of A group (AP-). The contents of E2, P4, Glu, Se, BDNF and hIS significantly differed among the four groups (P<0.01). The ROC analysis was used to determine the risk of negative pregnancy test (-) after TAI was increased when plasma E2 was less than 46.45 pmol/L in cows before TAI. The changes in E2, P4,hIS, Glu, and BDNF in the blood of natural estrus and natural anestrus cows affected the pregnancy after TAI. the level of E2 in plasma may be used to assess the risk of negative pregnancy after TAI.(AU)


O objetivo do presente estudo foi investigar a influência de mudanças patológicas de plasma antes de inseminação artificial (TAI) na gestação de vacas. O conteúdo de estrogênio (E2), progesterona (P4), glucose (Glu), selênio (Se), fator neurotrófico derivado do cérebro (BDNF), e histamina (HIS) no plasma de 48 vacas Holstein foi medido antes de TAI. De acordo com a detecção de estro, as vacas foram divididas em dois grupos: estro (E) e anestro (A). Após teste de gestação 28 d após TAI, dois grupos de E e A foram formados em gestação positiva do grupo E (EP+), gestação negativa do grupo E (EP-), gestação positiva do grupo A (AP+), e gestação negativa do grupo A (AP-). Os valores de E2, P4, Glu, Se, BDNF e hIS foram significativamente diferentes entre os quatro grupos (P<0,01). A análise ROC foi utilizada para determinar o risco de teste de gestação negativo (-) após aumento de TAI quando plasma E2 estava abaixo de 46,45 pmol/L em vacas antes de TAI. Alterações em E2, P4,hIS, Glu e BDNF no sangue de estro natural e anestro natural em vacas afetou a gestação após TAI. O nível de E2 no plasma pode ser usado para avaliar o risco de gestação negativa após TAI.(AU)

Following the induction of oestrus out of season in small ruminants, low fertility and variations in fertility rates are associated with embryonic losses. One of the main causes of embryonic loss is luteal dysfunction. Gonadotropin Releasing Hormone (GnRH) supports the luteal structure, and increasing progesterone levels may be beneficial in terms of promoting embryonic life. The main objective of the present study was to evaluate the efficacy of GnRH administration following an oestrus induction protocol in the anoestrus season for preventing embryonic loss in goats having failure to conceive during the season. In the study, 106 Damascus goats aged 3-5 years and weighing 45-60 kg were used. The oestrus of 106 goats in the anoestrous group was stimulated with progesterone and pregnant mare serum gonadotropin (PMSG) treatment. Out of breeding season, goats were divided into the 4 following groups: GnRH0 (n = 27), GnRH7 (n = 26), GnRH0+7 (n = 27) and control (n = 26). In each goat, an intravaginal sponge (IS) containing 20 mg of fluorogestone acetate (FGA) was placed into the vagina and left for 9 days. With the withdrawal of the sponge, 550IU PMSG and 125 µg of d-cloprostenol were injected intramuscularly. Oestrus detection was made via teaser bucks for 3 days starting 24 h after withdrawal of the IS. Eighteen bucks known to be fertile were used for breeding. Goats in the oestrus period were mated via natural breeding. The GnRH analogue lecirelin was injected intramuscularly at breeding in the GnRH0 group, on day 7 post-breeding in the GnRH7 group, and both at breeding and on day 7 post-breeding in the GnRH0+7 group. No injections were given to the control group. Blood samples for progesterone measurement were taken by jugular vena puncturing on days 3, 6, 7, 10, 13, 16, and 19 after breeding from 10 randomly chosen goats in all groups. The goats with a level of > 3.5 ng/mL of progesterone on day 21 post-breeding were evaluated as pregnant. Pregnancy was also viewed on day 50 after breeding by real-time ultrasonography (USG) with a 5-7.5 MHz convex probe. The oestrus rate was 96.23% (102/106) in the goats. The rates of onset of oestrus between 36-48 h, 48-60 h and 60 h and beyond were 38.7% (41/106), 21.7% (23/106) and 35.8% (38/106), respectively. The total pregnancy rate was 35.8% (38/106). There were no statistically significant differences (P > 0.05) found for the pregnancy rate, embryonic death rate or progesterone concentration of the groups. However, serum progesterone levels were statistically different in the GnRH7 group compared with the control group (P < 0.05). After synchronisation, various anti-luteolytic strategies can be used to support corpus luteum development and elevate progesterone concentration in the luteal phase to decrease embryonic loss and increase reproductive performance. Therefore, application of GnRH to support the luteal structure and to increase progesterone levels may be beneficial in terms of supporting embryonic life. The results showed that GnRH treatment on the day 7 post-breeding following oestrus induction, including FGA and PMSG, can increase serum progesterone levels in Damascus goats in the anoestrus period. However, following oestrus induction in the anoestrus period, it was seen that GnRH treatment at breeding or on day 7 after breeding did not have any positive effect on embryonic loss or reproductive performance. In conclusion, it was considered that this protocol could be implemented successfully, yielding a 35% pregnancy rate in Damascus goats in the anoestrus period, but embryonic loss must be deeply studied in detail.(AU)

Este trabalho tem por objetivo descrever um caso de carcinoma espinocelular da base do chifre, bilateral e simétrico, em uma vaca e discutir a questão hormonal, possivelmente envolvida na patogênese da doença. Tratava-se de uma fêmea bovina, 11 anos, com histórico de emagrecimento progressivo, presença de massas exofíticas na base de ambos os chifres e em anestro por muitos anos. Foi realizada biopsia incisional de ambas as lesões para exame histopatológico e dosagens hormonais. Devido à progressão do quadro clínico, optou-se pela eutanásia, seguida dos exames necroscópico e histopatológico. O exame histopatológico revelou tratar-se de um carcinoma espinocelular infiltrativo bem diferenciado, e as dosagens hormonais apresentaram alterações nos níveis do hormônio luteinizante, folículo estimulante e estrógenos totais. Apesar de existirem descrições de carcinomas espinocelulares da base do chifre, ainda não havia relatos da ocorrência do mesmo bilateral e em uma vaca com distúrbios hormonais.(AU)


The objective of this study was to describe a case of bilateral and symmetrical squamous cell carcinoma from the horn base in a cow and to discuss the hormonal question, possibly involved in its pathogenesis. A 11-year-old beef cow presenting a history of progressive thinning, presence of exophytic masses at the base of both horns and anestrous for many years was assisted. An incisional biopsy of both lesions was performed for histopathological examination and hormonal dosages. Due to the clinical progression, euthanasia followed by necroscopic and histopathological examination was carried out. Histopathological examination revealed a well differentiated infiltrative squamous cell carcinoma and the hormonal dosages presented changes in luteinizing hormone, follicle stimulating and total estrogen levels. Although there are descriptions of basal squamous cell carcinoma of the horn, there were still no reports of the occurrence of the same bilateral in a cow with hormonal disorders.(AU)

Os agonistas dopaminérgicos são utilizados para induzir estro em cadelas, pois atuam na síntese e liberação de prolactina. Objetivou-se avaliar o efeito da piridoxina como indutor de estro em cadelas por agir na neurotransmissão dopaminérgica. Foram selecionadas 40 cadelas em anestro, divididas em quatro grupos experimentais, tratadas com 10mg/kg/dia (G1) e 50mg/kg/dia (G2) de cloridrato de piridoxina, 5µg/kg/dia (G3) de cabergolina e grupo controle/placebo (G4) por até 20 dias. Foram realizadas citologias vaginais a cada 24h para acompanhamento do ciclo estral e análises hormonais (FSH, LH e PRL) no dia zero e 120h do início do tratamento. As cadelas do G3 (100%) manifestaram proestro após 12 dias de tratamento aproximadamente, tempo inferior aos demais grupos (P<0,05). Apenas uma cadela do G1 e uma do G2 ficaram gestantes contra oito fêmeas do G3 e nenhuma do G4 (P<0,05). As concentrações plasmáticas de prolactina foram reduzidas nas fêmeas do G2 e G3 (P<0,05). As demais avaliações hormonais não sofreram influência do tratamento (P>0,05). O cloridrato de piridoxina foi ineficiente para induzir estro em cadelas, mas foi capaz de suprimir a prolactina, de forma semelhante à cabergolina, quando utilizado na dose de 50mg/kg/dia.(AU)


Dopaminergic agonists are used to induce estrus in female dogs as they act in the synthesis and release of prolactin. The objective of this study was to evaluate the effect of pyridoxine as an inducer of estrus by acting on dopaminergic neurotransmission. A total of 40 female dogs in anestrous were divided into four experimental groups treated with 10mg/kg/day (G1) and 50mg/kg/day (G2) of pyridoxine hydrochloride, 5µg/kg/day (G3) of cabergoline and control group/placebo (G4) for up to 20 days. Vaginal cytologies were performed every 24h for follow-up of the estrous cycle and hormonal analyzes (FSH, LH and PRL) on day zero and 120 hours after the start of treatment. The female dogs from G3 (100%) showed proestrus after 12 days of treatment, less time than the other groups (P< 0.05). Only one female from G1 and one from G2 were pregnant against eight from G3 and none from G4 (P< 0.05). Plasma concentrations of prolactin were reduced by treatment in females from G2 and G3 (P< 0.05). The other hormonal evaluations were not influenced by the treatment (P> 0.05). Pyridoxine chloridrate was inefficient to induce estrus in female dogs but was able to suppress prolactin when used at a dose of 50mg/kg/day.(AU)

O objetivo do estudo foi comparar o efeito das técnicas hormonais e de luz artificial nas éguas receptoras de embrião acíclicas avaliando as taxas de gestação aos 14 e 28 dias durante a fase de transição de primavera. Os 48 animais foram distribuídos aleatoriamente nos grupos: controle (CONT, n=16), éguas cíclicas na fase ovulatória; luz artificial (LUZ, n=16), éguas acíclicas submetidas ao tratamento de luz artificial; e hormônio (HORM, n= 16), éguas acíclicas submetidas ao protocolo hormonal na fase de transição. As éguas do grupo LUZ foram estimuladas por 60 dias com luz artificial durante cinco horas por dia. Nos grupos CONT e LUZ, quando observada a presença de folículo ≥ 35 mm de diâmetro e edema uterino ≥ grau II, foram administrados 1,5 mg de acetato de deslorelina e 1500 UI de hCG para induzir a ovulação. As éguas do grupo HORM foram tratadas com três doses de 1,5 mg de benzoato de estradiol e seguiram os mesmos protocolos dos Grupos CONT e LUZ. Foi avaliada a taxa de gestação por ultrassonografia aos 14 dias e confirmação aos 28 em todos os grupos experimentais. Foi realizada análise descritiva e teste Qui-quadrado (significância de 5%). Taxas de gestação aos 14 e 28 dias foram semelhantes (p>0,05) entre todos os grupos. Os tratamentos HORM e LUZ durante o período de transição inverno-primavera mostraram-se eficazes para atender ao programa de transferência de embrião. Por ser um método mais natural, o protocolo LUZ tem potencial como mais uma ferramenta biotecnológica na reprodução de equinos.


The aim of this study was to compare the effect of hormonal and artificial light techniques on acyclic embryo recipient mares by assessing pregnancy rates at 14 and 28 days during the spring transition period. The 48 animals were randomly assigned to the groups: control (CONT, n = 16), cyclic mares in the ovulatory phase; artificial light (LIGHT, n = 16), acyclic mares subjected to artificial light treatment; and hormone (HORM, n = 16), acyclic mares submitted to hormonal protocol in transition phase. In the LIGHT group, mares were stimulated with artificial light for five hours a day, for 60 days. In CONT and LIGHT groups, when a follicle ≥ 35 mm in diameter and uterine edema ≥ grade II were observed, 1.5 mg of deslorelin acetate and 1500 IU hCG were administered to induce ovulation. In the HORM group, mares were treated with three doses of 1.5 mg of estradiol benzoate and followed the same protocols as the CONT and LIGHT groups. Pregnancy rate was assessed by ultrasound at 14 days and confirmation at 28 days in all experimental groups. Descriptive analysis and chi-square test (5% significance) were performed. Pregnancy rates at 14 and 28 days were similar (p> 0.05) among all groups. The HORM and LIGHT treatments during the winter-spring transition period proved to be effective during the embryo transfer programs. As it is a more natural method, the LIGHT protocol has the potential to be one more biotechnological tool in equine reproduction.

Nos grupos CONT e LUZ, quando observada a presença de folículo 35 mm de diâmetro e edema uterino grau II, foram administrados 1,5 mg de acetato de deslorelina e 1500 UI de hCG para induzir a ovulação. As éguas do grupo HORM foram tratadas com três doses de 1,5 mg de benzoato de estradiol e seguiram os mesmos protocolos dos Grupos CONT e LUZ. Foi avaliada a taxa de gestação por ultrassonografia aos 14 dias e confirmação aos 28 em todos os grupos experimentais. Foi realizada análise descritiva e teste Qui-quadrado (significância de 5%). Taxas de gestação aos 14 e 28 dias foram semelhantes (p>0,05) entre todos os grupos. The aim of this study was to compare the effect of hormonal and artificial light techniques on acyclic embryo recipient mares by assessing pregnancy rates at 14 and 28 days during the spring transition period. The 48 animals were randomly assigned to the groups: control (CONT, n = 16), cyclic mares in the ovulatory phase; artificial light (LIGHT, n = 16), acyclic mares subjected to artificial light treatment; and hormone (HORM, n = 16), acyclic mares submitted to hormonal protocol in transition phase. In the LIGHT group, mares were stimulated with artificial light for five hours a day, for 60 days. In CONT and LIGHT groups, when a follicle ≥ 35 mm in diameter and uterine edema ≥ grade II were observed, 1.5 mg of deslorelin acetate and 1500 IU hCG were administered to induce ovulation. In the HORM group, mares were treated with three doses of 1.5 mg of estradiol benzoate and followed the same protocols as the CONT and LIGHT groups. Pregnancy rate was assessed by ultrasound at 14 days and confirmation at 28 days in all experimental groups. Descriptive analysis and chi-square test (5% significance) were performed. Pregnancy rates at 14 and 28 days were similar (p> 0.05) among all groups. The HORM and LIGHT treatments during the winter-spring transition period proved to be effective during the embryo transfer programs. As it is a more natural method, the LIGHT protocol has the potential to be one more biotechnological tool in equine reproduction.

Com o objetivo de verificar a presença de VEGF e IGF-1 nos ovários de cadelas, foram realizadas análises imuno-histoquímicas do estroma cortical; teca e granulosa de folículos secundários, terciários e terciários pré-ovulatórios luteinizados; e ovócitos de folículos primários, secundários e terciários de ovários de cinco cadelas em anestro (Anest) e cinco em estro (Est). A identificação das fases do ciclo estral foi realizada por citologia vaginal associada a dosagem plasmática de progesterona. Os ovários foram submetidos a tratamento imuno-histoquímico para identificação de VEGF (anticorpo primário PU 360-UP, Biogenex, USA; diluição 1:30) e IGF-1 (anticorpo primário PabCa, Gro-Pep, Austrália; diluição 1:100). Determinou-se um índice de imunomarcação (IM), para cada tecido avaliado, pela razão entre a área positivamente marcada dividida pela área total analisada. Para os ovócitos, verificou-se imunomarcação positiva ou negativa. As comparações de IM entre tecidos foram realizadas pelo teste de Wilcoxon (diferentes tecidos em mesmo grupo) ou Mann-Whitney (mesmo tecido entre diferentes grupos), todas no nível de 5% de significância. VEGF e IGF-1 foram identificados, de forma semelhante (P>0,05), em todas as estruturas avaliadas em ambos os grupos experimentais. Conclui-se que esses fatores de crescimento estão presentes em cadelas no anestro e estro, no estroma cortical ovariano, folículos em diferentes estádios de desenvolvimento e ovócitos.(AU)


In order to verify the presence of VEGF and IGF-1 in the ovaries of bitches, immunohistochemical analyzes of the cortical stroma; theca and granulosa of secondary, tertiary and tertiary luteinized preovulatory follicles; and oocytes of primary, secondary and tertiary follicles of ovaries from five bitches in anestrous (Anest) and five in estrus (Est) was performed. The identification of the phases of the estrous cycle was performed by vaginal cytology associated with the measurement of plasma progesterone. The ovaries were treated for immunohistochemical identification of VEGF (PU 360 primary antibody-UP, Biogenex, USA, dilution 1:30) and IGF-1 (primary antibody PabCa, Gro-Pep, Australia; 1:100 dilution). The immunostaining index (MI) was determined for each tissue by the ratio of positively marked area divided by total analyzed area. For oocytes immunostaining was determined as positive or negative. Comparisons of IM between tissues were performed with the Wilcoxon test (deferent tissues in the same group) or Mann-Whitney test (same tissue between different groups), all at 5% significance level. VEGF and IGF-1 have been similarly identified (P>0.05) in all structures evaluated in both groups. It is concluded that in bitches in estrus and anestrous these growth factors are present in ovary cortical stroma, follicles at different stages of development and oocytes.(AU)

This study evaluated the influence of estrous cycle stage and transport temperature of ovaries on in vitro maturation of canine oocytes. The bitches were categorized into two groups based on stage of estrus cycle: diestrus or anestrus. One ovary of each pair collected was transported in saline solution at 4°C while the other was transported at 37°C. Thus, ovarian tissue was sliced in PBS to release cumulus oocyte complexes (COCs). A total of 345 COCs (n = 186 oocytes from ovaries of bitches in anestrus and 159 in diestrus) were cultivated in TCM 199 supplemented with HEPES, sodium pyruvate, cysteine, follicle stimulating hormone (FSH), human chorionic gonadotropin (hCG), estrogen (E2) and epidermal growth factor (EGF). After 72h of maturation, the COCs were denuded, fixed and stained to assess nuclear maturation. The Fisher test was applied to examine the differences between the groups. The significance level adopted was 0.05. The oocytes obtained from the ovaries from bitches in diestrus transported at 4°C shown increased frequency of oocytes in metaphase II stage than those maintained at 37°C (p 0.01). Similarly, there was increased frequency of oocytes in metaphase II (11.1%) stage from the ovaries of bitches in anestrus and transported at 4°C, than those maintained at 37°C (p 0.05). It was concluded that transport temperature influences the results of canine oocyte viability and in vitro maturation, regardless of reproductive stage of the female.


Foi avaliada a influencia do ciclo estral e temperatura de transporte de ovários na maturação in vitro de oócitos caninos. As cadelas foram categorizadas em dois grupos baseados no estagio do ciclo estral anestro ou diestro. Um ovário por par coletado foi transportado em solução fisiológica 0,9% a 4°C enquanto o outro foi transportado a 37°C. Então, os ovários foram seccionados em PBS para a liberação dos complexos cumulus oocito (COCs). Um total de 345 COCs (n = 186 oocitos obtidos de cadelas em anestro e 159 em diestro) foi cultivado em TCM 199 suplementado com HEPES, piruvato de sódio, cisteina, hormônio folículo estimulante (FSH), gonadotrofina coriônica humana (hCG), estrógeno (E2) e fator de crescimento epidermal (EGF). Apos 72h de maturação, os COCs foram desnudados, fixados e corados para avaliação da maturação nuclear. O teste de Fisher foi utilizado para avaliar as diferenças entre os grupos. O nível de significância adotado foi de 0,05. Os oócitos obtidos de cadelas em diestro transportados a 4°C apresentaram maior frequência de oócitos no estagio de metáfase II (21,1%) que os mantidos na temperatura de 37°C (p 0,01). De forma similar, houve maior frequência de oócitos nos estágios de metáfase II (11,2%) nos ovários obtidos de cadelas em anestro e transportados a 4°C que nos ovários mantidos a 37°C (p 0,05). Concluiu-se que a temperatura de transporte influencia os resultados de viabilidade oocitária canina e a maturação in vitro, independentemente do estagio reprodutivo da fêmea.

The bitch has reproductive peculiarities that differentiate it from other species. Several experiments have been conducted to establish efficient protocols for maturation; however, the results appear to be unsatisfactory. In this respect, the reproductive female donor should be considered, since it can be a factor of variability of findings in this species. The objective of this study was to evaluate the relationship between the estrous cycle phases phase diestrus and anestrus on canine oocyte in vitro maturation (IVM). The ovaries were transported in sodium chloride 0.9% solution, and were cut into phosphate-buffered saline (PBS) and the cumulus-oocyte complexes (COCs) selected in tissue culture medium (TCM), supplemented with 199 HEPES. A total of 469 grade 1 oocytes were collected from bitches in anestrus and diestrus. These selected oocytes were transferred to the maturation medium for a period of 72 hours, then subjected to hyaluronidase solution and stained with Hoechst 33342 to assess nuclear configuration. The comparison of anestrus and diestrus phase showed no differences (p > 0.05) between the nuclear maturation stages. Thus, the phase of the estrous cycle did not influence the in vitro maturation of canine oocytes, increasing the rates of M-II in this species.


A cadela apresenta particularidades reprodutivas que a diferencia de outras espécies. Diversos experimentos têm sido realizados visando estabelecer protocolos eficientes para a maturação, entretanto os resultados mostram-se insatisfatórios. Nesse aspecto, a fase reprodutiva da fêmea doadora deve ser considerada, já que pode ser um fator de variabilidade dos achados ate então presenciados nessa espécie. O objetivo deste estudo foi avaliar a influencia das fases do ciclo estral (anestro e diestro) na maturação in vitro (MIV) de cadelas. Os ovários foram transportados em solução de cloreto de sódio 0,9% e seccionados em solução salina fosfato tamponado (PBS) e os complexos cumulus-oócito (COCs) selecionados em meio de cultura de tecidos (TCM) 199 suplementado com HEPES. Foram obtidos 469 oócitos grau 1 de cadelas em anestro e diestro. Esses oócitos selecionados foram transferidos para o meio de maturação por um período de 72 horas, sendo posteriormente submetidos a solução de hialuronidase e corados com HOESCHT 33342 para avaliação da configuração nuclear. A comparação das fases de anestro e diestro não revelou diferença (p > 0,05) entre os estágios de maturação nuclear. Dessa maneira, a fase do ciclo estral não influenciou na maturação in vitro de oócitos caninos, incrementando os índices de M-II nesta espécie.

Em um sistema intensivo de produção de suínos, as falhas reprodutivas são uma das principais razões de descarte de matrizes e queda nos índices produtivos. A infecção urinária (cistite) e as endometrites são consideradas importantes causas de descarte em fêmeas suínas, por terem consequências reprodutivas relevantes e elevarem a taxa de reposição do plantel. O presente estudo teve o objetivo de avaliar o aparelho reprodutivo e a bexiga de fêmeas suínas de descarte normal de granjas, bem como investigar a existência de relação entre as patologias encontradas. Foram examinadas 79 matrizes suínas oriundas de 20 rebanhos localizados no Estado de Santa Catarina. De cada fêmea foram coletados os ovários, fragmentos de útero e bexiga. Dentre as fêmeas avaliadas, 32 (40,5%) tinham diferentes graduações de cistite, 24 (30,4%) tinham algum tipo de inflamação uterina, e 9 (11,4%) estavam em anestro, com ovários inativos. Contudo, não foi observada dependência significativa entre cistite e endometrite nas amostras analisadas.


Reproductive failures are the major reasons for removal of sows and decrease of production rates in an intensive swine production system. Urinary infection and endometritis are considered important causes for culling of sows, due to relevant reproductive consequences and increase of the replacement rates. The present study aimed to evaluate the reproductive and urinary system of culled sows, as well as investigate the occurrence of cystitis and endometritis in analyzed sows. Samples, such as ovaries, uterus fragments and bladder were collected from 79 sows originated from 20 farms of Santa Catarina State. Results showed that, 32 (40,5%) analyzed sows presented cystitis in different levels, 24 (30,4%) had some class of uterine inflammation, and 9 (11,4%) were in anestrous, with inactive ovaries. However, unsignificative dependence between cystitis and endometritis in analyzed samples was observed.

Este trabalho leve o objetivo de avaliar a influência do EGF na maturação in vitro (MIV) de cadelas. Realizou-se o transporte dos ovários em solução de cloreto de sódio 0,9%, que foram seccionados em solução salina fosfato temponado (PBS) e os complexos cumulus oócitos (COCs) selecionados em meio de cultura de tecidos (TCM) 199 suplementado com HEPES. Foram coletados 405 oócitos grau 1 de cadelas em anestro e diestro. Os oocitos provenientes das duas fases do ciclo estral foram submetidos a dois tratamentos: meio com adição de 10ng/mL do fator de crescimento epidermal (EGF) (T) e meio sem suplementação (C). Depois de 72 horas de maturação, os COCs foram desnudados, fixados e corados com HOESCHT 33342 para avaliação da maturação nuclear. Os oócitos obtidos dos ovários da fase de diestro do grupo T demonstraram maior porcentagem (18,8%) de oócitos no estágio de metáfase II (M-II), em relação ao grupo C (1,3%) (p < 0,01). Nos oócitos obtidos da fase de anestro houve diferença (p < 0,05) entre os grupos, observando-se menor parcela (4,1%) de oócitos no estágio de vesícula germinativa (VG) no grupo T, quando comparado ao grupo C (16,1%). Comparando-se as diferentes fases reprodutivas, em meios suplementados com EGF, foi observada diferença (p < 0,05) apenas nos oócitos obtidos da fase de diestro em relação ao estágio de M-II. Dessa maneira, a suplementação no meio de maturação na concentração de 10 ηg/mL, neste estudo, exerceu influência positiva apenas na MIV de oócitos caninos oriundos da fase de diestro, incrementando os índices de M-II nessa espécie.


This work aimed to evaluate the influence of EGF on canine oocyte in vitro maturation (IVM). We carried out the transport of the ovaries in sodium chloride 0.9% solution, which were cut into phosphate-buffered saline (PBS) and the cumulus oocyte complexes (COCs) selected in tissue culture medium (TCM), supplemented with 199 HEPES. A total of 405 grade 1 oocytes were collected from bitches in anestrus and diestrus. Oocytes from the two phases of estrous cycle were subjected to two treatments: medium with addition of 10 ηg/mL epidermal growth factor (EGF) (T) and medium without supplementation (C). After 72 h of maturation, COCs were denuded, fixed and stained with Hoechst 33342 to assess nuclear maturation. The oocytes obtained from the ovaries of the diestrus phase of the T group demonstrated higher percentage (18.8%) of oocytes at metaphase II stage (M-II) than C group (1.3%) (p < 0.01). The oocytes from anestrus phase demonstrated difference (p < 0.05) between the groups, observing smaller proportion (4.1%) of oocytes at the stage of germinal vesicle (GV) in group T compared to group C (16.1%). Comparing the different reproductive status in media supplemented with EGF difference (p < 0.05) was observed only in oocytes diestrus phase relative to the stage of M-II. Thus, supplementation on maturation medium at a concentration of 10 ηg/mL of EGF in this study had positive influence only on IVM of canine oocytes obtained from diestrus phase, increasing the levels of M-II in this species.

In canine specie, oocyte maturation rates are low and the percentage of oocytes that remain in the stage of germinal vesicle (GV) regardless of culture conditions is high. During maturation oocyte undergoes modification and the GV chromatin remodeling manifested by changes in the configuration and positioning. The objective of this work is to evaluate the configuration and positioning of chromatin of oocytes in GV stage during anestrus and diestrus bitches. The ovaries of 33 females (20 bitches in anestrous and 13 in diestrus) were isolated, sliced and only cumulus-oocyte complexes (COCs) grade 1 were subjected to solution of 0.2% hyaluronidase for release in cumulus cells. After this process, the selected oocytes were stained and evaluated. From a total of 920 oocytes, 566 were classified as grade 1 and the stages of chromatin configuration identified as GV-1, GV-2, GV-3 and GV-4. The observed changes in chromatin configuration been characterized as a transition dispersed chromatin (GV-1, GV-2) for partially condensed (GV-3) until it reaches a fully condensed stage (GV-4). The data analyzed from the chromatin configuration showed a significant difference between the stages with a higher proportion of GV-1 and GV-2 for the anoestrus and GV-3 and GV-4 during diestrus. There is need for further studies to be able to have a proper understanding of the influence of chromatin configuration of oocytes in GV stage in resumption of meiosis and consequently in oocyte meiotic competence...


Na espécie canina as taxas de maturação oocitária são baixas e a porcentagem de oócitos que permanecem em estagio de vesícula germinativa (VG), independente das condições de cultivo, e alta. Durante a maturação oocitária, a VG sofre modificação e remodelamento da cromatina, que se manifesta por alterações na sua configuração e posicionamento. Assim, o objetivo deste trabalho e avaliar a configuração e o posicionamento da cromatina de oócitos em estagio de VG durante o anestro e diestro de cadelas. Os ovários de 33 fêmeas (20 cadelas em anestro e 13 em diestro) foram isolados, fatiados e os complexos cumulus-oócitos (COCs) foram submetidos à solução de hialuronidase 0,2% para a liberação das células do cumulus. Apos esse processo, os oócitos selecionados foram corados, avaliados e apenas COCs grau 1 foram utilizados. De um total de 920 oócitos, 566 foram classificados como grau 1 e os estágios de configuração da cromatina identificados como VG-1, VG-2, VG-3 e VG-4. As alterações observadas na configuração da cromatina foram caracterizadas como transição de uma cromatina dispersa (VG-1, VG-2) para parcialmente condensada (VG-3) ate atingir um estagio totalmente condensado (VG-4). Os dados analisados da configuração da cromatina mostraram uma diferença significativa entre as fases de anestro e diestro, com maior proporção de VG-1 e VG-2 durante o anestro e de VG-3 e VG-4 durante o diestro. Ha necessidade de novos estudos para uma compreensão adequada da influencia da configuração da cromatina de oócitos no estagio de VG na retomada da meiose e na competência meiótica do oócito...

The objective of this experiment was to assess the relationship between electrical resistance of the vaginal mucosa and plasma progesterone for optimal mating time in the bitch. Eight mature beagle bitches were examined, and we observed eight times of estrus. Vaginal electric resistance was recorded weekly using a Draminski ovulation detector in anestrus, and daily in estrus. Plasma progesterone concentration was estimated by radioimmunoassay. In the bitch, incline in vaginal electric resistance (376.20 +/- 105.63 units) showed a closely association with the onset of proestrus. Ovulation day was determined as the first day when plasma progesterone concentration increased above 5.0 ng/ml (Day 0). On Day 0, vaginal mucous electric resistance was 438 +/- 132 units. Vaginal mucous electric resistance showed a slight decrease or was maintained until Day 0. However, it showed an explosive increase, and peaked on Day 1~3, which was above 600 units. Two of eight cases peaked on Day 1, three of eight cases were revealed on Day 2, and others were revealed on Day 3. After Day 4, resistance showed a rapid drop to below 600 units and reached 200 units on Day 8. The optimal mating time was determined when vaginal mucous electric resistance was above 600 units.

La melatonina es una hormona que regula los ciclos circadianos y muchos de los aspectos reproductivos de los mamíferos y es secretada por la glándula pineal en las horas de ausencia de luz. Esta hormona posee receptores de alta afinidad acoplados a proteínas de tipo G, denominados MT1. Un polimorfismo de la secuencia que codifica para estos receptores estaría involucrado en el control de la reproducción estacional de los ovinos. El propósito de este estudio busca determinar la presencia del polimorfismo del receptor MT1 en la oveja criolla Araucana, un ovino local en el que se ha registrado un corto anestro reproductivo. Para poder realizar este trabajo se utilizó la técnica denominada reacción en cadena de la polimerasa para polimorfismo en el tamaño de los fragmentos de restricción PCR-RFLP, para lo cual, se obtuvieron muestras de ADN genómico de 50 ovejas Araucanas, las cuales fueron digeridas con la endonucleasa de restricción Mnl1. Se logró identificar la presencia del polimorfismo del receptor MT1 en la oveja Araucana. Los genotipos se hallaron en una frecuencia de 68 por ciento para el genotipo +/+, 28 por ciento para el genotipo +/- y4 por ciento para el genotipo -/-. Este alto porcentaje de animales con genotipo +/+ podría explicar el corto anestro reproductivo que presenta esta raza.


Melatonin is a hormone that regulates circadian rhythms and many of the reproductive aspects of mammals and is secreted by the pineal gland during the hours of absence of light. This hormone has high affinity receptors coupled to G-like proteins, termed MT1. A polymorphism of the sequence coding for these receptors was involved in the control of seasonal reproduction in sheep. The purpose of this study was to determine the presence of MT1 receptor polymorphism in Araucana creole sheep, a local breed with a short reproductive anestrus. To carry out this work, we used a technique called polymerase chain reaction-restriction fragment lengh polymorphism PCR-RFLP, for which samples were obtained from genomic DNA of 50 Araucana sheep, which were digested with Mnl1 restriction endonuclease. It was possible to identify the presence of MT1 receptor polymorphism in Araucana sheep. The genotypes were found in a genotype frequency of 68 percent for genotype + / +, 28 percent for genotype + / - y4 percent for genotype - / -. This high percentage of animals with genotype + / + could explain the short reproductive anestrus featuring this breed.

Avaliou-se o efeito do soro de cadela em estro na maturação in vitro de ovócitos caninos, utilizando-se 92 ovócitos de cadelas, submetidas à cirurgia eletiva de ovarioisterectomia. Os ovócitos foram selecionados e distribuídos em dois tratamentos: T1 (n = 48), ovócitos cultivados in vitro durante 96 horas utilizando meio base - TCM199 + 5µg/mL de LH + 20µg/mL de FSH - mais 10 por cento de soro inativado de vaca em estro e T2 (n = 44), ovócitos cultivados em meio base mais 10 por cento de soro inativado de cadela em estro. O percentual de ovócitos observados em metáfase I não indicou diferenças (P>0,05) entre T1 (2,1 por cento) e T2 (0,0 por cento), porém a taxa de ovócitos maduros (metáfase II) foi diferente (P<0,05), sendo 27,1 por cento em T1 e 47,7 por cento em T2. O mesmo fato ocorreu com a taxa de cromatina condensada (P<0,01), com 14,6 e 0,0 por cento, respectivamente. Nos ovócitos sem configuração cromossômica, não foram observadas diferenças (P>0,05), sendo 56,3 por cento em T1 e 52,3 por cento em T2. Estes resultados indicam que a adição de soro de cadela em estro no meio de cultivo oferece melhores condições para o desenvolvimento in vitro, quando comparado à de soro de vaca em estro.


This study aimed to evaluate the effect of estrus on in vitro canine oocyte. A total of 92 oocyte from bitches under ovary-hysterectomy surgery was used. The oocytes were selected and randomly assigned to two different treatments, being T1 (n = 48) in vitro cultured for 96h using basic medium (TCM199 + 5µg/mL of LH + 20µg/mL of FSH), plus 10 percent of cow inactive serum in estrus and T2 (n = 44) basic medium plus 10 percent of bitch inactive serum in estrus. The percentage of oocyte observed on metaphase I do not indicate a difference (P>0.05) between T1 (2.1 percent) and T2 (0.0 percent). However, the rate of mature oocyte (metaphase II) was different (P<0.05), being 27.1 percent for T1 and 47.7 percent for T2. There was difference (P<0.05) in the condensed chromatin rate for T1 (14.6 percent) and T2 (0.0 percent), respectively. There was no difference (P>0.05) between T1 (56.3 percent) and T2 (52.3 percent) in oocyte with no chromosome configuration. These results indicate that supplementation with estrus bitch serum on culture media offer better conditions to in vitro development, when compared to estrus cow serum.

Dosou-se a concentração plasmática de progesterona (P4) em ovelhas Santa Inês (SI), Suffolk (SU) e Romney Marsh (RM) em anestro sazonal e submetidas ao efeito-macho, as quais receberam ou não suplementação alimentar. Machos vasectomizados foram introduzidos no grupo de fêmeas após um período prévio de isolamento de 60 dias, e amostras de sangue foram colhidas antes e após a introdução dos machos. Houve efeito (P<0,01) de período, raça, interação período x raça e interação suplementação x período x raça sobre a concentração de P4. Nas ovelhas SI ocorreu aumento (P<0,01) da concentração de P4 após a introdução do macho, indicando que houve aumento na atividade cíclica reprodutiva desse grupo. A suplementação, neste caso, potencializou este aumento. Nas ovelhas SU e RM não ocorreram modificações na concentração de P4 (P>0,01) após a introdução dos machos, nem houve efeito de suplementação. O efeito-macho foi eficaz em induzir a atividade reprodutiva durante o anestro sazonal em ovelhas SI, mas não em ovelhas SU e RM, e a associação dessa prática com a suplementação alimentar é recomendada para ovelhas da raça nativa SI.


The present study aimed to verify the plasmatic progesterone (P4) concentrations in Santa Inês (SI), Suffolk (SU) and Romney Marsh (RM) ewes submitted to male effect receiving or not food supplementation during seasonal anestrous. Vasectomized rams were introduced to the group of ewes after 60 day isolation. Blood samples were collected before and after the male's introduction. Effects (P<0.01) of period, breed, period x breed interaction and period x breed x supplementation interaction on plasmatic P 4 concentrations were observed. The plasmatic P 4 concentrations of SI ewes increased (P<0.01) after the male introduction, indicating there was an increase in the reproductive activity in this group. Supplementation increased this effect. For SU and RM ewes the plasmatic P 4 concentrations did not alter (P>0.01) after the male introduction, with no supplementation. It was concluded the male effect was efficient in inducing the reproductive activity in SI ewes, but not in SU and RM ewes. The association of the effect of the male with supplementation increased this effect.

Se evaluó el efecto de la suplementación alimenticia y el destete temporal en 46 vacas cruzadas Bos taurus x Bos indicus sobre el reinicio de la actividad ovárica postparto. Los animales fueron distribuidos al parto en tres tratamientos: T1: Control, sin suplementación y con cría al pie; T2: Suplementación + destete temporal (48 h) a los 60 días post parto; y T3: Suplementación + cría al pie. Se suplementó 3 kg de concentrado/vaca/día desde el parto hasta los 60 días post parto. Se colectó muestras de sangre dos veces por semana desde la semana 3 a la 12 postparto para determinar niveles de progesterona como indicador de reinicio de la actividad ovárica postparto. Se encontró un intervalo parto – primera ovulación de 70.6, 56.4 y 57.8 días para las vacas que ovularon dentro de las primeras 12 semanas postparto en los grupos T1, T2 y T3, respectivamente. El 31.3, 73.3 y 66.7% de las vacas de cada tratamiento ovularon en ese periodo. Se concluye que el factor nutricional y el destete temporal contribuyeron a una mejora del comportamiento reproductivo postparto del ganado bovino cruzado en el trópico peruano.


The effect of feed supplementation and temporary weaning in 46 crossbred Bos taurus x Bos indicus cows on the resumption of post partum ovarian activity was evaluated. Animals were distributed in three treatments: T1, Control, cows without supplementation + calf at foot; T2, Feed supplementation + calf at foot and temporary weaning (48 h) at 60 days post partum; and T3, Feed supplementation + calf at foot. Supplement consisted of 3 kg/cow/day from calving till 60 days post partum. Blood samples were collected twice a week since week 3 till week 16 post partum for the determination of progesterone levels as indicator of the resumption of ovarian activity. The interval calving first ovulation was 70.6, 56.4, and 57.8 days for cows that ovulate within the first 12 weeks after calving in groups T1, T2, and T3 respectively. The 31.3, 73.3, and 66.7% of cows in each treatment group ovulate within that period. It is concluded that feed supplementation and temporary weaning contribute in improving post partum reproductive performance in crossbred cattle in the Peruvian tropics.
