BACKGROUND: Cronobacter sakazakii is considered as an emerging foodborne pathogen. The aim of this study was to isolate and characterize virulent strains of Cronobacter sakazakii from food samples of Bangladesh. RESULT: Six (6) Cronobacter sakazakii was isolated and identified from 54 food samples on the basis of biochemical characteristics, sugar fermentation, SDS-PAGE of whole cell protein, plasmid profile and PCR of Cronobacter spp. specific genes (esak, gluA, zpx, ompA, ERIC, BOX-AIR) and sequencing. These strains were found to have moderately high antibiotic resistance against common antibiotics and some are ESBL producer. Most of the C. sakazakii isolates were capable of producing biofilm (strong biofilm producer), extracellular protease and siderophores, curli expression, haemolysin, haemagglutinin, mannose resistant haemagglutinin, had high cell surface hydrophobicity, significant resistance to human serum, can tolerate high concentration of salt, bile and DNase production. Most of them produced enterotoxins of different molecular weight. The isolates pose significant serological cross-reactivity with other gram negative pathogens such as serotypes of Salmonella spp., Shigella boydii, Shigella sonnei, Shigella flexneri and Vibrio cholerae. They had significant tolerance to high temperature, low pH, dryness and osmotic stress. CONCLUSION: Special attention should be given in ensuring hygiene in production and post-processing to prevent contamination of food with such stress-tolerant virulent Cronobacter sakazakii.

From January 2009 through September 2010, two cases of neonatal infections associated with Cronobacter malonaticus and one case associated with Cronobacter sakazakii, two of them fatal, were reported in the same hospital. These are the first clinical isolates of Cronobacter spp. in Argentina. The objective of this work was to characterize and subtype clinical isolates of Cronobacter spp. in neonate patients, as well as to establish the genetic relationship between these isolates and the foodborne isolates previously identified in the country. Pulsed-field gel electrophoresis analysis showed a genetic relationship between the C. malonaticus isolates from two patients. Different results were found when the pulsed-field gel electrophoresis patterns of clinical isolates were compared with those deposited in the National Database of Cronobacter spp.

Cronobacter (formerly known as Enterobacter sakazakii) is a genus comprising seven species regarded as opportunistic pathogens that can be found in a wide variety of environments and foods, including powdered infant formula (PIF). Cronobacter sakazakii, the major species of this genus, has been epidemiologically linked to cases of bacteremia, meningitis in neonates, and necrotizing enterocolitis, and contaminated PIF has been identified as an important source of infection. Robust and reproducible subtyping methods are required to aid in the detection and investigation, of foodborne outbreaks. In this study, a pulsed-field gel electrophoresis (PFGE) protocol was developed and validated for subtyping Cronobacter species. It was derived from an existing modified PulseNet protocol, wherein XbaI and SpeI were the primary and secondary restriction enzymes used, generating an average of 14.7 and 20.3 bands, respectively. The PFGE method developed was both reproducible and discriminatory for subtyping Cronobacter species.

Samples of water and barnacles Amphibalanus amphitrite were collected from Recife, Brazil, to assess if it accumulates total (TC) and thermotolerant coliforms (TTC) related with sewage pollution. The Most Probable Number (MPN) values and the standard procedures for examination of shellfish were used. Comparatively with the water samples, the highest coliform values came from the barnacles, with TC values ranging from < 3.0 × 10³ to > 2.4 × 10(6) MPN.g-1, and TTC ranging from > 2.4 × 10³ to 2.9 × 10(5) MPN.g-1. Barnacles accumulate the TC Ewingella americana, and the TTC Escherichia coli, Enterobacter gergoviae, Enterobacter aerogenes, and Enterobacter sakazakii. The results provided an indication of the level of organic contamination at the sampling locations and that this species could be a good organic pollution bioindicator.(AU)


Amostras de água e cirrípedes Amphibalanus amphitrite foram coletados em Recife, Brasil, para avaliar se estes acumulam coliformes totais (CT) e termo-tolerantes (TTC) relacionados à poluição por esgoto doméstico. Foram utilizados os valores de Números Mais Prováveis (NMP) e os procedimentos padrões para exame de bivalves. Comparativamente às amostras de água, os valores mais altos de coliformes foram dos cirrípedes, com valores de TC variando de < 3,0 × 10³ a > 2,4 × 10(6) NMP.g-1, e TTC variando de > 2,4 × 10³ a 2,9 × 10(5) NMP.g-1. Os cirrípedes acumularam TC Ewingella americana e TTC Escherichia coli, Enterobacter gergoviae, Enterobacter aerogenes e Enterobacter sakazakii. Os resultados proveram uma indicação de que há contaminação orgânica nas localidades de amostras e que esta espécie pode ser um bioindicador bom de poluição orgânica.(AU)
