Determination of the safety of agents prior to release is one of the most important research goals in biological control. In addition to concerns for the safety of non-target plants, determination of the potential toxic properties of new agents needs to be assessed. Numerous phytophagous insects are defended by chemicals against the attack of natural enemies. Some of these defensive compounds could pose an environmental risk if an agent is released. Here, larval populations of two pergid sawflies, Heteroperreyia hubrichi and H. jorgenseni, were analyzed by LC-MS/MS to investigate whether they contain alleged toxic peptides. The first species is a potential candidate for biological control of the invasive weed Brazilian peppertree in Florida and Hawaii. The chemical analyses revealed the presence of the peptides pergidin (Perg), 4-valinepergidin (VPerg), dephosphorylated pergidin (dpPerg), lophyrotomin (LGln and LGlu). The effect of sawfly population for each species was significantly influencing peptide concentration. All peptides occurred at lower concentrations compared with purportedly toxic species of this sawfly family. However, the concentrations of the peptides are of concern for the welfare of wildlife and livestock that would be exposed to these species. These results demonstrate that release of this biological control agent in the invaded range may pose an environmental threat.

Stingless bees produce geopropolis, which is popularly described for its medicinal properties, but for which few scientific studies have demonstrated pharmacological effects. The objective of this study was to investigate the chemical composition of the geopropolis of Melipona quadrifasciata anthidioides and to evaluate its antioxidant, antimutagenic, anti-inflammatory, and antimicrobial activities. The composition of the hydroethanolic extract of geopropolis (HEG) included di- and trigalloyl and phenylpropanyl heteroside derivatives, flavanones, diterpenes, and triterpenes. HEG showed antioxidant action via the direct capture of free radicals and by inhibiting the levels of oxidative hemolysis and malondialdehyde in human erythrocytes under oxidative stress. HEG also reduced the frequency of gene conversion and the number of mutant colonies of S. cerevisiae. The anti-inflammatory action of HEG was demonstrated by the inhibition of hyaluronidase enzyme activity. In addition, HEG induced cell death in all evaluated gram-positive bacteria, gram-negative bacteria, and yeasts, including clinical isolates with antimicrobial drug resistance. Collectively, these results demonstrate the potential of M. q. anthidioides geopropolis for the prevention and treatment of various diseases related to oxidative stress, mutagenesis, inflammatory processes, and microbial infections.

The chemical signature of social insects is very important for communication, and specific signs of each colony and its individuals are acquired throughout their development. This chemical signature can also be related to the composition of the materials employed during nest construction. Furthermore, the venom also contains important chemicals required for the maintenance of wasp colonies. Therefore, the objective of this study was to evaluate the chemical composition of the wasp Protopolybia exigua along its different development stages, including the nest substrate and the venom of adult worker at different ages. To achieve this, gas chromatography coupled to mass spectrometry was used. The results show that the chemical cuticular compounds present in different stages of development, and in the nest of P. exigua, are qualitatively similar, but quantitatively different, demonstrating that these compounds can be used as signals for the identification of co-specific individuals within the colonies. However, there was no significant variation in these compounds between different colonies, which may be related to the parental level between the colonies and due to them sharing the same resources. The non-polar compounds of the venom vary significantly according to the age of the workers, probably due to their different roles within the colony, and there was a clear increase in the complexity of the compounds as the wasps aged. Older wasps perform higher-risk activities, such as foraging, and therefore need to make more use of venom.

Vitellogenins are the major yolk proteins, synthesized in the fat body, released into the hemolymph and captured by the developing oocytes, but the mechanisms by which these proteins cross the follicular cell layer are still poorly understood. This study describes the actin distribution in follicular cells during vitellogenin transport to the oocyte in social Hymenoptera represented by bees Apis mellifera and Melipona quadrifasciata, the wasp Mischocyttarus cassununga and the ant Pachycondyla curvinodis. In oocytic chambers of vitellogenic follicles, vitellogenin was found within the follicular cells, perivitelline space and oocyte, indicating a transcellular route from the hemolymph to the perivitelline space. The cortical actin cytoskeleton in follicular cells underwent reorganization during transport of vitellogenin across this epithelium suggesting that in the ovary of social hymenopterans, vitellogenin delivery to oocytes requires a dynamic cytoskeletal rearrangement of actin filaments in the follicular cells.

Knowing the importance of participation by insects at cadaverous decomposition processes, and the limited use of the family Formicidae in criminal investigations, this study aims to record the foraging activity of four genera of ants in carcasses of birds and mammals. Observations occurred accidentally in two locations in the State of Minas Gerais, Brazil. In total, seven species of ants foraging in eight vertebrate carcasses were recorded. In addition, the study reported for the first time the presence of Wasmannia in carcasses in Brazil.(AU)


Sabendo da importância da participação dos insetos em processos de decomposição cadavérica e do uso limitado da família Formicidae em investigações criminais, o objetivo do presente estudo foi registrar a atividade de forrageio de quatro gêneros de formigas em carcaças de aves e mamíferos. As observações ocorreram de forma acidental em duas localidades do Estado de Minas Gerais, Brasil. No total, foram registradas sete espécies de formigas forrageando em oito carcaças de vertebrados. O trabalho registrou ainda pela primeira vez a presença do gênero Wasmannia em carcaças no Brasil.(AU)

Along with food and drug allergic reactions, a Hymenoptera insect Sting (Apoidea, Vespidae, Formicidae) is one of the most common causes of anaphylaxis worldwide. Diagnoses of Hymenoptera venom allergy (HVA) and specific immunotherapy (SIT) have been based on the use of crude venom extracts. However, the incidence of cross-reactivity and low levels of sensibility during diagnosis, as well as the occurrence of nonspecific sensitization and undesired side effects during SIT, encourage the search for novel allergenic materials. Recombinant allergens are an interesting approach to improve allergy diagnosis and SIT because they circumvent major problems associated with the use of crude venom. Production of recombinant allergens depends on the profound molecular characterization of the natural counterpart by combining some "omics" approaches with high-throughput screening techniques and the selection of an appropriate system for heterologous expression. To date, several clinically relevant allergens and novel venom toxins have been identified, cloned and characterized, enabling a better understanding of the whole allergenic and envenoming processes. Here, we review recent findings on identification, molecular characterization and recombinant expression of Hymenoptera venom allergens and on the evaluation of these heterologous proteins as valuable tools for tackling remaining pitfalls on HVA diagnosis and immunotherapy.

Melipona quadrifasciata and Melipona rufiventris are stingless bee species which present low and high heterochromatin content, respectively, on their mitotic chromosomes as assessed visually after a C-banding assay. However, these species do not show differences in the C-banding responses of their Malpighian tubule interphase nuclei. In the present study, the Feulgen-DNA response, which could inform on differences in DNA depurination due to differences in chromatin condensation, was compared in the cell nuclei of the Malpighian tubules of these species. It was hypothesized that differences in acid hydrolysis kinetics patterns, as assessed by Feulgen reaction and studied microspectrophotometrically, could discriminate M. quadrifasciata and M. rufiventris interphase nuclei not distinguishable with the C-banding method. Feulgen-DNA values corresponding to more than one ploidy class were found in both species; these values at the hydrolysis time corresponding to the maximal DNA depurination for each ploidy degree were higher in M. quadrifasciata, reflecting a higher DNA content in the Malpighian tubule cell nuclei of this species compared to those of M. rufiventris at the same larval instar. The maximal Feulgen-DNA values of M. quadrifasciata after short (50 min) and long (90 min) hydrolysis times were found to be closer to each other, while those of M. rufiventris occurred sharply at the long hydrolysis time, indicating that DNA depurination in M. quadrifasciata occurred faster. This result is probably related to the involvement of differences in chromatin condensation; it agrees with the idea that M. rufiventris contains more heterochromatin than M. quadrifasciata, which is supported by the analysis of results obtained with the image analysis parameter average absorption ratio. The depurination kinetics studied here with the Feulgen reaction were revealed to be more pertinent than the C-banding technique in establishing differences in levels of chromatin condensation for these cell nuclei.

Foraging behaviour has a remarkable significance to colonies of social insects since it is directly related to the supply of food, water, and pulp. Social wasps of the genus Apoica Lepeletier are primarily nocturnal, but studies concerning the general aspects related to this habit are still scarce. This study showed that the rates of foraging flights were higher during the full moon and last quarter moon phases. Frequencies of arrival and departure were correlated with time and temperature at night. The results suggest that the moonlight is an important component to general activities in this genus.


O comportamento de forragear é fundamental para o desenvolvimento das colônias de insetos sociais, pois permite que as colônias sejam supridas de alimento, água e materiais utilizados para a construção dos ninhos. Vespas sociais do gênero Apoica Lepeletier (Hymenoptera: Vespidae) têm comportamento de forrageamento noturno como uma de suas principais características, porém estudos sobre aspectos abióticos que influenciam esse comportamento são ainda escassos. Este estudo mostra que a freqüência de forrageamento foi maior em noites de luas cheia e crescente. Além disso, as saídas e retornos ao ninho correlacionaram-se positivamente com o período da noite e a temperatura. Os resultados sugerem que a luminosidade é um componente importante nas atividades gerais das colônias do gênero.

RESUMEN

Several stylopized specimens were found among the Hymenoptera collection of the Instituto Oswaldo Cruz (Rio de Janeiro, Brazil). A paper by Kogan & Oliveira (1966) described the parasites of Polybia represented among those specimens. I describe herein the first neotropical species of the genus Crawfordia parasitizing species of Psaenithia (Andrenidae, Panurgini). The presence of vestigial thoracic segmentation of the female cephalothorax is a peculiar trait of this genus of Strepsiptera. Triungulinids are described in detail for the first time in this genus.
