RESUMEN

A Gram stain-negative bacterial strain, designated SY4-7T, was isolated from rhizosphere mudflat of a halophyte (Zoysia sinica) collected around Seonyu Island, Republic of Korea. Cells of the organism were strictly aerobic, non-sporulating, non-motile rods and grew at 20-42 °C, pH 6-8 and 1-6% (w/v) NaCl. The 16S rRNA gene-based phylogenetic analyses revealed that strain SY4-7T formed an independent cluster separated from the recognized genera of the family Stappiaceae, which was also supported by phylogenomic analysis-based 92-core gene sequences. The type stains of the phylogenetically closest relatives were Stappia indica (95.6% sequence similarity), Stappia stellulata (95.1%) and Roseibium hamelinense (95.1%). The isoprenoid quinone was Q-10. The polar lipids consisted of phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, three unidentified aminophospholipids, an unidentified phosphoglycolipid, an unidentified aminolipid, two unidentified phospholipids and an unidentified lipid. The major cellular fatty acids are C18:1ω7c and C19:1 cyclo ω8c. The G + C content of the genomic DNA is 60.7%. Discrimination of the organism from all the recognized genera of the family Stappiaceae was apparent by the chemotaxonomic and phylogenetic features. Based on the results presented here, strain SY4-7T (= KCTC 72226T = NBRC 113902T) represents a novel species of a new genus in the family Stappiaceae, for which the name Hongsoonwoonella zoysiae sp. nov. is proposed.

A new taxon is created for the thermophilic purple nonsulfur bacterium previously designated as Rhodopseudomonas strain GI. Strain GI was isolated from a New Mexico (USA) hot spring microbial mat and grows optimally above 40 °C and to a maximum of 47 °C. Strain GI is a bacteriochlorophyll b-containing species of purple nonsulfur bacteria and displays a budding morphology, typical of species of the genus Blastochloris. Although resembling the species Blc. viridis in many respects, the absorption spectrum, carotenoid content, and lipid fatty acid profile of strain GI is distinct from that of Blc. viridis strain DSM133T and other recognized Blastochloris species. Strain GI forms its own subclade within the Blastochloris clade of purple nonsulfur bacteria based on comparative 16S rRNA gene sequences, and its genome is significantly larger than that of strain DSM133T; average nucleotide identity between the genomes of Blc. viridis and strain GI was below 85%. Moreover, concatenated sequence analyses of PufLM and DnaK clearly showed strain GI to be distinct from both Blc. viridis and Blc. sulfoviridis. Because of its unique assortment of properties, it is proposed to classify strain GI as a new species of the genus Blastochloris, as Blc. tepida, sp.n., with strain GIT designated as the type strain (= ATCC TSD-138 = DSM 106918).

Symbiosis is a diverse and complex phenomenon requiring diverse model systems. The obligate relationship between a monophyletic group of Euplotes species ("clade B") and the betaproteobacteria Polynucleobacter and "Candidatus Protistobacter" is among the best-studied in ciliates, and provides a framework to investigate symbiont replacements. Several other Euplotes-bacteria relationships exist but are less understood, such as the co-dependent symbiosis between Euplotes magnicirratus (which belongs to "clade A") and the alphaproteobacterium "Candidatus Devosia euplotis". Here we describe a new Devosia inhabiting the cytoplasm of a strain of Euplotes harpa, a clade B species that usually depends on Polynucleobacter for survival. The novel bacterial species, "Candidatus Devosia symbiotica", is closely related to the symbiont of E. magnicirratus, casting a different light on the history of bacteria colonizing ciliates of this genus. The two Devosia species may have become symbionts independently or as the result of a symbiont exchange between hosts, in either case replacing a previous essential bacterium in E. harpa. Alternatively, both may be remnants of an ancient symbiotic relationship between Euplotes and Devosia, in which case Polynucleobacter and "Ca. Protistobacter" are recent invaders. Either way, symbiont replacement between bacteria belonging to different classes must be evoked to explain this fascinating system.

A Gram-negative, strictly aerobic, yellow-pigmented and rod-shaped bacterium, designated strain NSL10(T), was isolated from the waste surface soil of a chemical factory in Hongan, China. Strain NSL10(T) was found to grow optimally at pH 7.0, 30 °C and in the absence of NaCl in modified LB medium. Cells were found to be positive for catalase and oxidase. The G+C content of the total DNA was determined to be 66.8 mol%. The 16S rRNA gene sequence of strain NSL10(T) showed the highest similarity to that of Devosia albogilva IPL15(T) (96.80 %), followed by Devosia geojensis BD-c194(T) (96.46 %) and Devosia chinhatensis IPL18(T) (96.27 %). The major cellular fatty acids of strain NSL10(T) were identified as C18:1 ω7c/C18:1 ω6c (48.2 %) and C16:0 (17.7 %). The major polar lipids were identified as diphosphatidylglycerol, phosphatidylglycerol, two unidentified glycolipids and an unidentified compound. Minor amounts of unidentified glycolipids and unidentified polar lipids were also detected. These chemotaxonomic data supported the affiliation of strain NSL10(T) to the genus Devosia. In conclusion, on the basis of biochemical, physiological characteristics and molecular properties, strain NSL10(T) represents a novel species within the genus Devosia, for which the name Devosia honganensis sp. nov., is proposed. The type strain is NSL10(T) (=KCTC 42281(T) = ACCC 19737(T)).

Strain L15(T), a Gram-negative, motile, orange colored bacterium was isolated from pond soil in the surrounding area of a hexachlorocyclohexane (HCH) dump site at Ummari village in Lucknow, India. Phylogenetic analysis based on 16S rRNA gene sequence showed that strain L15(T) belongs to the family Hyphomicrobiaceae in the order Rhizobiales. Strain L15(T) showed highest 16S rRNA gene sequence similarity to Devosia chinhatensis IPL18(T) (98.0%). Chemotaxonomic data revealed that the major fatty acids were summed feature 8 (C18:1 ω7c and/or C18:1 ω6c), C18:1 ω7c 11-methyl, C16:0 and C18:0. The major polar lipids of strain L15(T) were diphosphatidylglycerol and phosphatidylglycerol. The genomic DNA G+C content of strain L15(T) was 59.8%. Polyamine profile showed the presence of sym-homospermidine with traces of putrescine. Ubiquinone Q-10 was the major respiratory quinone present. Based on these data, strain L15(T) (=CCM 7977(T) =DSM 25398(T)) was classified as a type strain of a novel species, for which the name Devosia lucknowensis sp. nov. is proposed.

Two psychrophilic strains, Cr7-05(T) and Cr4-44(T), isolated from alpine glacier cryoconite, were characterized by using a polyphasic approach. Both strains were psychrophilic, showing good growth over a temperature range of 1-20 °C. The chemotaxonomic characteristics of these isolates included the presence of C(18:1)ω7c and summed feature 3 (C(16:1)ω7c and/or C(16:1)ω6c) as the major cellular fatty acids, Q-10 as the predominant ubiquinone and diphosphatidylglycerol, phosphatidylglycerol and unknown glycolipids as major polar lipids. The DNA G+C contents of strains Cr7-05(T) and Cr4-44(T) were 61.4 and 63.6 mol%, respectively. Phylogenetic analysis based on 16S rRNA gene sequences indicated that the two isolates belong to the genus Devosia. The 16S rRNA gene sequence similarity between the two strains was 98.6%, but DNA-DNA hybridization indicated 54% relatedness. Strains Cr7-05(T) and Cr4-44(T) exhibited 16S rRNA gene sequence similarity of 94.7-97.2 and 94.9-96.9%, respectively, to the type strains of recognized Devosia species. On the basis of phenotypic characteristics, phylogenetic analysis and DNA-DNA relatedness data, strains Cr7-05(T) and Cr4-44(T) represent two novel species within the genus Devosia, for which the names Devosia psychrophila sp. nov. (type strain Cr7-05(T) =DSM 22950(T) =CGMCC 1.10210(T) =CIP 110130(T)) and Devosia glacialis sp. nov. (type strain Cr4-44(T) =CGMCC 1.10691(T) =LMG 26051(T)) are proposed. An emended description of the genus Devosia is also provided.

Three Gram-negative, motile, aerobic bacteria were isolated from cultures of the marine red alga Porphyra yezoensis. Phylogenetic analysis based on 16S rRNA gene sequences revealed that the novel strains were closely related to Maritalea myrionectae CL-SK30(T) (97.9% 16S rRNA gene sequence similarity) and Zhangella mobilis E6(T) (96.2 %). 16S rRNA gene sequence similarity between Z. mobilis E6(T) and M. myrionectae CL-SK30(T) was 97.9%. The DNA G+C contents of the isolates (49.4-50.0 mol%) were similar to those of M. myrionectae DSM 19524(T) (52.3 mol%) and Z. mobilis JCM 15144(T) (50.3 mol%). From these results, it was difficult to differentiate the genus Zhangella from the genus Maritalea. DNA-DNA hybridization demonstrated that the isolates belonged to a single species. The isolates could also be distinguished from M. myrionectae and Z. mobilis on the basis of chemotaxonomic and phenotypic features, including fatty acid composition (particularly C(16:1)ω7c), growth with 6-9% (w/v) NaCl, carbon utilization, oxidation patterns and so on. A novel species of the genus Maritalea is proposed to accommodate the three isolates, with the name Maritalea porphyrae sp. nov. The type strain is LCM-3(T) (=LMG 25872(T)=NBRC 107169(T)). Furthermore, it is proposed that Zhangella mobilis should be transferred from the genus Zhangella to the genus Maritalea, with the name Maritalea mobilis comb. nov. (type strain E6(T)=CGMCC 1.7002(T)=JCM 15144(T)).

A Gram-negative, rod-shaped, purple non-sulfur bacterial strain, designated JA415T, was isolated from the mud of a pokkali rice field located on Vypeen Island, Ernakulam, Kerala, India. Strain JA415T was motile by means of a single polar flagellum. Photo- and chemo-organoheterotrophic growth was observed using organic compounds as carbon sources and electron donors. Photo- and chemolithoautotrophic growth using thiosulfate as electron donor did not occur. Fermentative growth could not be demonstrated. Intracellular photosynthetic membranes were lamellar stacks parallel to the cytoplasmic membrane. Bacteriochlorophyll a and carotenoids of the spirilloxanthin series were present as photosynthetic pigments. Niacin, pantothenate and p-aminobenzoate were required as growth factors. C18:1omega7c was the predominant cellular fatty acid component. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain JA415T clustered with members of the genus Rhodoplanes in the class Alphaproteobacteria. On the basis of phenotypic and molecular genetic evidence, it is proposed that strain JA415T be classified as a representative of a novel species of the genus Rhodoplanes, family Hyphomicrobiaceae, with the name Rhodoplanes pokkaliisoli sp. nov. The type strain is JA415T (=KCTC 5711T=NBRC 104972T).

A Gram-negative, pale-yellow, non-spore-forming bacterium, motile with single polar flagella, strain E6T, was isolated from coastal seawater collected from Tianjin, China, and its taxonomic position was investigated using a polyphasic approach. Strain E6T requires NaCl for growth and grows optimally at pH 8.2 and 36 degrees C and in the presence of 2.0% (w/v) NaCl. It is positive for catalase and oxidase, and reduces nitrate to nitrite. The major fatty acids (>10%) are C18:1omega7c (64.71%) and 11-methyl C18:1omega7c (12.38%), the ubiquinone system is Q-10 and the DNA G+C content is 53.1 mol%. Phylogenetic analyses based on 16S rRNA gene sequences demonstrated that strain E6T represented a new lineage in the Alphaproteobacteria and is related to genera Cucumibacter, Devosia, Ochrobactrum and Ahrensia. Strain E6T shows highest 16S rRNA gene sequence similarity (93.3%) to Cucumibacter marinus CL-GR60T and less than 92.2% similarity to other relatives. It can be differentiated from its closest phylogenetic neighbours on the basis of several phenotypic features, including nitrate reduction, assimilation of D-glucose, L-arabinose, D-mannose, mannitol and maltose and major fatty acid composition. A polyphasic analysis supported the conclusion that strain E6T represents a novel genus and species of the family Hyphomicrobiaceae, for which the name Zhangella mobilis gen. nov., sp. nov. is proposed. The type strain of Zhangella mobilis is E6T (=CGMCC 1.7002T=JCM 15144T).

A Gram-negative, rod-shaped, strictly aerobic bacterium, designated strain CL-SK30(T), was isolated from a culture of the marine ciliate Myrionecta rubra. 16S rRNA gene sequence analysis revealed that strain CL-SK30(T) was most closely related to Cucumibacter marinus (92.0 % similarity) and next to the type strains of species of the genus Devosia (89.8-91.3 % similarities) in the family Hyphomicrobiaceae. Phylogenetic analyses of the 16S rRNA gene sequences showed that strain CL-SK30(T) formed a robust clade together with C. marinus, but the sequence divergence value of 8 % between them indicated that the novel bacterium represented a distinct lineage. Strain CL-SK30(T) grew optimally in the presence of 2-5 % sea salts at 30-35 degrees C and pH 7.2-8.0. The major polar lipids were phosphatidylglycerol, diphosphatidylglycerol, two unidentified glycolipids, an unidentified phospholipid and an unidentified lipid. Ubiquinone 10 was the major quinone. The DNA G+C content was 52.7 mol%. Based on its phenotypic, chemotaxonomic and phylogenetic characteristics, strain CL-SK30(T) represents a novel genus and species of the family Hyphomicrobiaceae, for which the name Maritalea myrionectae gen. nov., sp. nov. is proposed. The type strain is CL-SK30(T) (=KCCM 90060(T)=DSM 19524(T)).

A Gram-negative, strictly aerobic, non-spore-forming bacterium, designated strain BD-c194(T), was isolated from diesel-contaminated soil in Geoje, Korea. The cells were short, motile rods with single polar flagella. Growth of strain BD-c194(T) was observed between 15 and 45 degrees C (optimally at 30-35 degrees C) and between pH 6.0 and 9.5 (optimally at pH 7.5-9.0). The predominant fatty acids were 11-methyl C(18 : 1)omega7c, C(16 : 0), C(18 : 1)omega7c, C(18 : 0) and an unknown fatty acid (equivalent chain-length 18.814); a large amount of phosphatidylglycerol and a small amount of diphosphatidylglycerol were present as polar lipids. The G+C content of the genomic DNA was 60.8 mol% and the major isoprenoid quinone was Q-10. A comparative 16S rRNA gene sequence analysis showed that strain BD-c194(T) formed a well-defined phyletic lineage within the genus Devosia (with 100 % bootstrap support). The levels of 16S rRNA gene sequence similarity with respect to the type strains of other Devosia species ranged from 95.0 to 96.1 %. On the basis of chemotaxonomic data and molecular properties, strain BD-c194(T) represents a novel species within the genus Devosia, for which the name Devosia geojensis sp. nov. is proposed. The type strain is BD-c194(T) (=KCTC 22082(T) =DSM 19414(T)).

A novel marine bacterium was isolated from a sediment sample from Hwasun Beach in Jeju, Republic of Korea. The cells were found to be Gram-negative, aerobic, oxidase-positive, catalase-positive, motile rods. The organism required natural seawater or artificial sea salts for growth. The temperature and pH ranges for growth were 20-42 degrees C and pH 5.1-12.1, respectively. Phylogenetic analyses based on 16S rRNA gene sequences showed that the organism belonged to the order Rhizobiales and formed a robust cluster with members of the genus Devosia. Its phylogenetic neighbours were the type strains of Devosia riboflavina (96.8 % 16S rRNA gene sequence similarity), Devosia neptuniae (96.7 %), Devosia soli (96.5 %) and Devosia limi (96.2 %), 'Devosia terrae' DCY11 (96.2 %) and 'Candidatus Devosia euplotis' (96.2 %). The predominant ubiquinone was Q-10, the major fatty acids were C(18 : 1), C(18 : 0) and C(16 : 0) and the G+C content of the DNA was 59.1 mol%. On the basis of phenotypic data and the results of phylogenetic analyses, strain HST3-14(T) represents a novel species of the genus Devosia, for which the name Devosia subaequoris sp. nov. is proposed. The type strain is strain HST3-14(T) (=KCTC 12772(T) =JCM 14206(T)).

RESUMEN

A Gram-negative, motile, aerobic and oval- or rod-shaped bacterial strain, DS-56(T), which is phylogenetically closely related to the genus Devosia, was isolated from soil from Dokdo, Korea. Strain DS-56(T) grew optimally at pH 6.5-7.5 and 25 degrees C in the presence of 0.5 % (w/v) NaCl. A neighbour-joining phylogenetic tree based on 16S rRNA gene sequences showed that strain DS-56(T) joins the cluster comprising Devosia species, at a bootstrap resampling value of 100 %. The levels of 16S rRNA gene sequence similarity between strain DS-56(T) and the type strains of recognized Devosia species ranged from 95.4 to 96.4 %. Strain DS-56(T) contained Q-11 as the predominant ubiquinone, unlike Devosia species, which contain Q-10 as the predominant ubiquinone. The novel strain also contained some fatty acids, including branched and hydroxy fatty acids, that are not detected in Devosia species. The DNA G+C content of strain DS-56(T) was 66.2 mol%. On the basis of the phenotypic, chemotaxonomic and phylogenetic data, strain DS-56(T) represents a novel species of the genus Devosia, for which the name Devosia insulae sp. nov. is proposed. The type strain is DS-56(T) (=KCTC 12821(T)=DSM 17955(T)).

This paper reports the identification of bacterial endosymbionts that inhabit the cytoplasm of the marine ciliated protozoon Euplotes magnicirratus. Ultrastructural and full-cycle rRNA approaches were used to reveal the identity of these bacteria. Based on analysis of 16S rRNA gene sequences, evolutionary trees were constructed; these placed the endosymbiont in the genus Devosia in the alpha-Proteobacteria. The validity of this finding was also shown by fluorescence in situ hybridization with a Devosia-specific oligonucleotide probe. Differences at the 16S rRNA gene level (which allowed the construction of a species-specific oligonucleotide probe) and the peculiar habitat indicate that the endosymbiont represents a novel species. As its cultivation has not been successful to date, the provisional name 'Candidatus Devosia euplotis' is proposed. The species- and group-specific probes designed in this study could represent convenient tools for the detection of 'Candidatus Devosia euplotis' and Devosia-like bacteria in the environment.
