In the present study, a HPLC/DAD method was set up to allow for the determination and quantification of malondialdehyde (MDA) in the brain of rodents (rats). Chromatographic separation was achieved on Supelcosil LC-18 (3 µm) SUPELCO Column 3.3 cm × 4.6 mm and Supelco Column Saver 0.5 µm filter by using a mobile phase acetonitrile (A) and phosphate buffer (20 mM, pH = 6) (B). Isocratic elution was 14% for (A) and 86% for (B). The injection volume (loop mode) was 100 µL with an analysis time of 1.5 min. Flow rate was set at 1 mL/min. The eluted compound was detected at 532 nm by a DAD detector by keeping the column oven at room temperature. The results indicated that the method has good linearity in the range of 0.2-20 µg/g. Both intra- and inter-day precision, expressed as RSD, were ≤15% and the accuracies ranged between ±15%. The lower limit of quantification (LLOQ), stability, and robustness were evaluated and satisfied the validation criteria. The method was successfully applied in a study of chronic toxicology following different treatment regimens with haloperidol and metformin.

Gliflozins and gliptins represent two different pharmacological drug classes that exert different and potentially complementary glucose-lowering effect in patients with type II diabetes mellitus. A novel, selective, and sensitive HPLC method was developed for the determination of canagliflozin, empaglifozin, linagliptin, and metformin in pure form, in laboratory prepared mixtures, and in pharmaceutical dosage form. Experimental design optimization was applied by using Plackett-Burman and face-centered composite designs to achieve the best resolution with minimum experimental trials. Three significant variables affecting optimization, namely buffer pH, percentage of methanol, and percentage of acetonitrile, were studied. Chromatographic separation was achieved using an Agilent Eclipse C8 column, and column temperature was kept at 45°C. The mobile phase was composed of dipotassium hydrogen phosphate buffer (0.05 M, adjusted to pH 6 using o-phosphoric acid):acetonitrile:methanol (50:25:25, v/v/v) at a flow rate of 1.5 mL/min. Sharp and well-resolved peaks of the cited drugs were obtained. The method was fully validated in terms of linearity, accuracy, precision, selectivity and robustness in agreement with the International Council of Harmonization (ICH) guidelines Q2 (R1). Satisfactory results were obtained by the analysis of tablets through applying the developed method. Therefore, it could be performed for the analysis of the cited drugs in quality control laboratories.

An adsorbent was developed from the residues of Moringa oleifera Lam. seed husks functionalized with iron nanoparticles for subsequent application to the removal of metformin from contaminated water. The material, MOM-Fe3O4, was characterized using TEM, SEM imaging, and EDX analysis, which revealed that iron nanoparticles were retained in the pores. The application of MOM-Fe3O4 to waste water resulted in a 93.9% reduction in the metformin content, demonstrating the efficacy of the material. Kinetic and equilibrium data were fitted to the pseudo-first-order and Langmuir models, respectively. The thermodynamic parameters indicated an exothermic and reversible process. A high adsorption capacity, 65.01 mg g-1 at 298 K, demonstrated the potential of using the new material as a low-cost adsorbent for the removal of pharmaceuticals such as metformin.

Pharmaceuticals constitute one of the most important emerging classes of environmental pollutants. A three-phase solvent system of water, water containing 0.1% of formic acid and acetonitrile was successfully used to separate, by liquid chromatography with mass spectrometry (LC-MS), polarity-matched pharmaceuticals, that is, carbamazepine, clarithromycin, and erythromycin, as well as amoxicillin and metformin. Despite of polarity similarities, these pharmaceuticals were completely resolved in the analytical run time of 15 min. The optimized three-phase solvent system based-method was validated for the simultaneous analysis of six matched-polarity pharmaceuticals in wastewater samples. Good linearity (coefficient of determination more than 0.993) and precision (relative standard deviation less than 15.66%) were achieved. Recovery of analytes from the wastewater was between 0.70 and 1.18. Limits of detections ranged from 0.0001 to 0.5114 µg/L. No significant matrix effect, evaluated by post extraction addition, was observed in the electrospray ionization (ESI) source. Then, this methodology has been successfully applied to environmental study of pharmaceutical residues occurring in influent and effluent wastewater samples, from the main wastewater treatment plant in Potenza (Basilicata, Southern Italy).

Several series of batch and continuous experiments were performed to investigate the removal of metformin and other contaminants from two wastewaters: wastewater I (WWI) containing 4 mg/L metformin and wastewater II (WWII) containing 110 g/L butanol. Biomethane potential (BMP) tests on WWII showed 77% of total chemical oxygen demand (tCOD = 110 g/L) degradability, and no apparent inhibition effects were observed. BMP tests on WWI showed an apparent inhibitory effect reflected in lower biogas production with increasing metformin concentration in the wastewater. Continuous flow hybrid vertical anaerobic biofilm (HyVAB®) experiments were consistent with the batch test findings. It was necessary to co-digest WWI (metformin) with WWII (easily degradable organics) to achieve complete metformin removal. After a period of adaptation, WWI and WWII co-digestion achieved up to 98% tCOD removal and 100% metformin removal. Most of the contaminants were removed in the anaerobic section of the HyVAB®, which implies that higher chemical oxygen demand (COD) loads than tested here are possible, given some optimization. The pilot reactor was able to manage organic loads of 11 g COD/d and above 10 mg/L metformin with a removal of 98% and 100% for tCOD and metformin, respectively.

RESUMEN

A new sample preparation method is proposed for the extraction of pharmaceutical compounds (Metformin, Phenyl biguanide, and Phenformin) of varied hydrophilicity, dissolved in an aqueous sample. When in contact with an organic phase, an interfacial potential is imposed by the presence of an ion, tetramethylammonium (TMA+), common to each phase. The interfacial potential difference drives the transfer of ionic analytes across the interface and allows it to reach up to nearly 100% extraction efficiency and a 60-fold enrichment factor in optimized extraction conditions as determined by HPLC analysis.

Organic solvents are widely used in pharmaceutical and chemical industry for chromatographic separations. In recent years, subcritical water chromatography (SBWC) has shown ability in replacing hazardous organic solvents used in traditional high-performance liquid chromatography (HPLC). In this work, a pain killer-aspirin-and an antidiabetic drug-metformin HCl-were successfully separated on an XBridge C18 column using no organic solvents in the subcritical water chromatography mobile phase. Both traditional HPLC and subcritical water chromatography were used for comparison purposes. SBWC separation of metformin HCl and aspirin were achieved at 95 °C and 125 °C, respectively. The recovery for both active pharmaceutical ingredients (APIs) obtained by SBWC is 99% in comparing with the stated content of each drug. The relative standard deviation is less than 1% for SBWC assays developed in this work. This level of accuracy and precision achieved by SBWC is the same as that resulted by the traditional HPLC analysis.

RESUMEN

A phytochemical investigation of Seidlitzia rosmarinus collected along the shoreline of the Gulf of Aqaba in the remote southern desert region of the Sinai peninsula has revealed the presence of the registered drug metformin (4). However, analysis of the 14C content revealed the drug to be an anthropogenic contaminant. Consequently, natural product researchers should be aware that compounds isolated from plants might originate from environmental contamination rather than biosynthesis. The new natural product N-(4-hydroxyphenylethyl)-α-chloroferuloylamide was isolated as a mixture of the E and Z isomers along with a number of other well-established secondary metabolites.

RESUMEN

Metformin is a widely prescribed medication that has been used to treat children with type 2 diabetes in the United States for the past 15 years. Metformin now has a variety of clinical applications in pediatrics, and its potential clinical uses continue to expand. In addition to reviewing the current understanding of its mechanisms of action including the newly discovered effects on the gastrointestinal tract, we will also discuss current clinical uses in pediatrics, including in type 1 diabetes. Finally, we examine the existing state of monitoring for metformin efficacy and side effects and discuss prospective future clinical uses.

In this study, new molecularly imprinted polymer (MIP) nanoparticles are designed for selective recognition of different drugs used for the treatment of type 2 diabetes mellitus, i.e. sitagliptin (SG) and metformin (MF). The SG- and MF-imprinted polymer nanoparticles are synthesized by free-radical initiated polymerization of the functional monomers: methacrylic acid and methyl methacrylate; and the crosslinker: ethylene glycol dimethacrylate. The surface morphology of resultant MIP nanoparticles is studied by atomic force microscopy. Fourier transform infrared spectra of MIP nanoparticles suggest the presence of reversible, non-covalent interactions between the template and the polymer. The effect of pH on the rebinding of antidiabetic drugs with SG- and MF-imprinted polymers is investigated to determine the optimal experimental conditions. The molecular recognition characteristics of SG- and MF-imprinted polymers for the respective drug targets are determined at low concentrations of SG (50-150 ppm) and MF (5-100 ppm). In both cases, the MIP nanoparticles exhibit higher binding response compared to non-imprinted polymers. Furthermore, the MIPs demonstrate high selectivity with four fold higher responses toward imprinted drugs targets, respectively. Recycled MIP nanoparticles retain 90% of their drug-binding efficiency, which makes them suitable for successive analyses with significantly preserved recognition features.

In the present study, two novel molecularly imprinted polymers (MIPs) with remarkable recognition properties for metformin and its transformation product, guanylurea, have been prepared for their selective, enrichment, isolation and removal from aqueous media. The prepared adsorbents were characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and swelling experiments. The performance of the prepared MIPs was evaluated by various parameters including the influence of pH, contact time, temperature and initial compound concentration. The effects on the adsorption behavior of the removal process parameters were studied and the equilibrium data were fitted by the Langmuir and Freundlich models. Due to the imprinting effect, adsorption performance of MIPs was always superior to its corresponding NIP (non-imprinted polymer), with maximum adsorption capacity ∼80 mg g(-1) for both MIPs. Stability and reusability of the MIPs up to the 5th cycle meant that they could be applied repeatedly without losing substantial removal ability. In the next step, the prepared MIP nanoparticles were evaluated as sorbents in a dispersive solid phase extraction (D-SPE) configuration for selective enrichment and determination of metformin and guanylurea in different aqueous matrices. Under the working extraction conditions, the D-SPE method showed good linearity in the range of 50-1000 ng L(-1), repeatability of the extractions (RSD 2.1-5.1%, n=3), and low limits of detection (1.5-3.4 ng L(-1)). The expanded uncertainty of the data obtained was estimated following a bottom-up approach. The proposed method combined the advantages of MIPs and D-SPE, and it could become an alternative tool for analyzing the residues of METF and its transformation product GUA in complex water matrices, such as wastewaters.

Antidiabetic compounds are among the most prescribed pharmaceuticals. Nevertheless, their presence in the environment has been scarcely evaluated as there is no method for their determination in environmental samples. This paper reports the development of an analytical method for the determination of traditionally used antidiabetics (metformin and glibenclamide) and novel antidiabetics (vildagliptin, sitagliptin and pioglitazone). The method is based on solid-phase extraction and determination by high-performance liquid chromatography quadrupole time-of-flight mass spectrometry. The method was applied to effluent wastewater, river water and tap water. Mean recoveries of glibenclamide, vildagliptin, sitagliptin and pioglitazone in the matrices evaluated were in the range 78-83%; limits of quantification were in the range 0.4-4.3 ng L(-1); and precision values were in the range 2.2-13%. The high hydrophilicity and polarity of metformin complicated its simultaneous extraction. Chromabond Tetracycline cartridges and sample pH 8.5 were applied to the extraction of glibenclamide, vildagliptin, sitagliptin and pioglitazone. Oasis HLB cartridges, neutral sample pH and SDS as ion-pair reagent were used for the extraction of metformin. Validation results of metformin were not as favorable as those of the other antidiabetic drugs but were comparable with others previously reported. The developed method was applied to the first-time determination of the concentrations of the five antidiabetic drugs in wastewater, river water and tap water. Metformin was the antidiabetic drug at the highest concentration in wastewater and surface water (up to 253 ng L(-1) and 104 ng L(-1), respectively). Two of the antidiabetic drugs of recent prescription, sitagliptin and vildagliptin, were found in effluent wastewater at concentrations of 117 ng L(-1) and 12 ng L(-1), respectively, and in river water at concentrations of 35 ng L(-1) and 6 ng L(-1), respectively, whereas the classic antidiabetic drug glibenclamide and the novel drug pioglitazone were not detected.

A hydride-based octadecyl stationary phase on both 4.0 and 1.8 microm silica particles is tested in both the capillary LC and the pressurized capillary electrochromatography (pCEC) modes. These two materials are compared to standard C18 stationary phase made by organosilanization and to the hydride material packed into a convention 4.6mm I.D. column. The performance of the capillary columns is evaluated in terms of analysis times for various mixtures as well as efficiency. Of particular interest are the differences between the LC mode where only laminar flow is present and pCEC operation where a flat electrodriven flow profile is superimposed on the parabolic pressurized flow. Differences in performance between columns packed with 4.0 and 1.8 microm particle silica are also evaluated.
