RESUMEN

The occurrence of hyperostotic bones is common in marine fish, especially in the Carangidae family, despite few records of this condition in fishes from Brazilian waters. The present study describes the occurrence of hyperostosis in Atlantic moonfish (Selene setapinnis) in, Rio de Janeiro State, Brazil. Radiographs of the fish specimens were taken and all of them presented hyperostosis in at least three different regions: cleithrum, pterygiophores and pleural ribs. The observed pattern is different from previously described for other species of the same genus. It is the first record for the species and a valuable contribution to the study of bone diseases in fishes from the Brazilian coast.(AU)

Extreme environmental conditions, such as temperature, can lead to meristic trait variation and skeletal deformities, which may have major impacts on individual fitness. As intertidal ecosystems experience rapid temperature and physicochemical fluctuations, intertidal fish living and reproducing in these environmental conditions may have phenotypes influenced by such variable environments. The impact of intertidal variability on fish development, however, has not been previously investigated. Skeletal deformities and meristic traits were assessed for Bathygobius cocosensis, a common intertidal fish living across the Indo-Pacific region, using a clearing and staining method on 72 individuals. Over 87% of individuals presented meristic variation and over 70% exhibited at least one type of skeletal deformity, mostly recorded in the caudal fin area. The unexpected prevalence of skeletal deformities among this intertidal fish population suggests that such deformities may be suitable markers to evaluate an individual's stress exposure during development and the subsequent fitness effects.

RESUMEN

The aim of this study was to assess histopathological alterations in the gills of Astyanax jacuhiensis from different points along the Ijuí River and to determine if these abnormalities can be used as biomarkers in biomonitoring studies. Fish specimens were collected from three sites on the Ijuí River in winter and summer and examined histologically for abnormalities of the secondary lamellae of their gills. For each fish, estimates were made of the frequencies of lamellae with edema, hyperplasia, hypertrophy, epithelium lifting, lamellar fusion or deformation, in addition to the overall frequency of abnormal lamellae and the number of abnormalities per lamella. No differences were observed between sampling points during the winter. In summer the frequency of lamellae with hypertrophy was significantly higher at site 3 (Pirapó) than at site 2 (Santo Ângelo) and the frequency of abnormal lamellae was increased at site 1 (Ijuí) in comparison to site 3. Additionally, all three sample points had a significantly higher value in the winter than in the summer for one of the histological parameters analyzed. These results indicate temporal and spatial variation in the level of contamination of the Ijuí River and also shows that fish gill histopathology can be used for in situ biomonitoring studies.

X-ray micro-computed tomography scans were used to examine the caudal-fin structure of an unusual double-tailed deformity in an adult brown surgeonfish Acanthurus nigrofuscus from the Great Barrier Reef. In both this case and in a similar double-tailed deformity in a juvenile Tomini surgeonfish Ctenochaetus tominiensis from the Philippines, the caudal fin was duplicated along the dorsoventral axis. Detailed examination of the A. nigrofuscus specimen revealed that the deformity was associated with duplication and reflection of the hypural plates and the posterior vertebrae, yet the fish survived to adulthood, indicating that the effects of duplication on survival may be limited.

RESUMEN

Different cases of vertebral ankylosis were examined in seven tripterygiid species obtained from waters around New Zealand. The skeletal deformities observed are located in the caudal region of the vertebral column. Those occurred in Forsterygion nigripenne, Matanui bathytaton and in one specimen of Ruanoho whero were severe cases. The mechanism of the formation of vertebral ankylosis and the causes behind such anomaly were discussed. Further studies are needed to relate specific pollutants with the observed types of deformities. Anat Rec, 2017. © 2017 Wiley Periodicals, Inc. RESUMEN

Laboratory breeding of annual fish is often impaired when fish fail to inflate the swim bladder after hatching. These fish, which are known as belly-sliders, cannot swim in the water column, moving mainly on the bottom of water. Therefore, we wanted to understand the factors underlying failed inflation of swim bladder in the annual fish Austrolebias nigrofasciatus and determine the implications of this condition on the growth of juveniles. To identify how the initial filling of the gaseous vesicle is compromised, and to determine the implications of this condition on juvenile growth, we analysed the histology of the swim bladder of both normal swimming fish and belly-sliders during their initial growth phase (first month of life). In addition to slower growth rate compared with fish swimming normally, we found that belly-sliders have metaplasia characterized by epithelium peeling of the swim bladder and the presence of haemorrhage. Based on our findings, it can be concluded that metaplasia of the swim bladder is recurrent in annual fish bred in the laboratory, which explains the large number of unviable larvae. However, its causes remain unknown.(AU)

The anadromous allis shad Alosa alosa has suffered dramatic population declines throughout Europe and is currently considered as endangered throughout its entire distribution range. In order to reestablish allis shad in the River Rhine, which formerly housed one of the largest and most important populations, an EU-LIFE Project 'The re-introduction of allis shad in the Rhine system' was started in 2007. In course of the LIFE+ Projects, allis shad larvae bred from genitor fish of the Gironde-Garonne-Dordogne population in France were reared in a pilot ex situ stock plant pilot facility in Aßlar, Germany. At an age of 1-2 months, about 100% of these fish developed approximately 0.5- to 0.8-cm large, fluid-filled, transparent cysts in conjunction with the upper jaw. The performed microbiological, virological, parasitological and histological examinations did not detect any infectious agents. Possible causative agents are discussed with regard to environmental factors and the nutrition of larvae. In conclusion, the observed malformations are considered a sign for a severe health problem and therefore a risk for the successful breeding of allis shad in aquaculture.

RESUMEN

We describe and compare the histology of liver and spleen ofGeophagus brasiliensis (Perciformes), Hypostomus francisci (Siluriformes) and Hoplias aff. malabaricus (Characiformes), tropical freshwater fishes. InG. brasiliensisandH. aff. malabaricusthe hepatocytes were arranged in tubular form whereas in H. franciscithey cord-like. In all species, hepatocytes presented glycogen, but in G. brasiliensis and H. aff. malabaricus they showed strong stained for hemossiderin in the cytoplasm. InG. brasiliensis and H. aff. malabaricus, melanomacrophage centres (MMCs) were associated to hepatic structures and only in G. brasiliensis was observed intrahepatic exocrine pancreas. The spleen, in all species, was characterized by red and white pulp without boundary between the two regions, but only in H. francisci was recorded nodular organization in splenic parenchyma. The G. brasiliensisandH. aff. malabaricuspresented in the white pulp MMCs linked mainly to ellipsoids. Besides, we observed large MMCs in the spleen in relation to liver of G. brasiliensis and H. aff. malabaricus. In liver, highest values of reticular fibers and collagen were observed inG. brasiliensis. In spleen, highest values of reticular fibers and collagen were recorded inH. aff. malabaricusandH. francisci, respectively. Histological differences confirm the hypothesis that the phylogenetic distance is reflected in liver and spleen.(AU)

Enrofloxacin (ENX) has been widely used in the prevention and control of bacterial diseases in sturgeon aquaculture due to its characteristics of wide antibacterial spectrum, strong antibacterial activity, less toxicity and fewer side effects, rapid action, extensive in vivo distribution, and little cross-resistance with other antibiotics. However, the spinal abnormality was found in Acipenser baerii soon after ENX administration, which resulted an "S"-shaped curvature of the spine and retarded fish growth. It was still not clear whether ENX could cause spinal abnormality in sturgeons by now. The aim of this work was to determine the accumulation rule and toxicity of ENX to A. baerii when used at a high dose and/or unusually long durations. Here, ENX was orally given to A. baerii for 3-5 d continuously at the dosage of 0, 20, 40, and 80 mg/kg once daily, respectively. The accumulation of ENX in blood, liver, kidney, and cartilage was detected after withdrawal, and the tissues were made into sections for morphological examination. The results showed that the levels of ENX increased in the four tissues with the increase of dose and duration, and the ENX level in serum was far lower than that in other tissues. At 240 h, ENX levels in the four tissues decreased significantly. The histology indicated that the liver, kidney, and cartilage began to show structural damages at 5 d after withdrawal of 40 mg/kg ENX. The damage was aggravated at 3-5 d after withdrawal of 80 mg/kg ENX. At 240 h, the damaged tissues showed signs of recovery. These results suggested that ENX should be no more than 40 mg/kg and that exposure time should not be greater than 5 d to prevent liver, kidney, and cartilage damage. More attention should be paid to the impact of ENX on the occurrence and development of chondrocytes in juvenile A. baerii and the potential damage to the cartilage.

RESUMEN

Effects of environmental variables at different spatial scales on freshwater fish assemblages are relatively unexplored in Neotropical ecosystems. However, those influences are important for developing management strategies to conserve fish diversity and water resources. We evaluated the influences of site- (in-stream) and catchment-scale (land use and cover) environmental variables on the abundance and occurrence of fish species in streams of the Upper Araguari River basin through use of variance partitioning with partial CCA. We sampled 38 1st to 3rd order stream sites in September 2009. We quantified site variables to calculate 11 physical habitat metrics and mapped catchment land use/cover. Site and catchment variables explained > 50% of the total variation in fish species. Site variables (fish abundance: 25.31%; occurrence: 24.51%) explained slightly more variation in fish species than catchment land use/cover (abundance: 22.69%; occurrence: 18.90%), indicating that factors at both scales are important. Because anthropogenic pressures at site and catchment scales both affect stream fish in the Upper Araguari River basin, both must be considered jointly to apply conservation strategies in an efficient manner.(AU)

Seahorses, pipefish and seadragons are fish of the Family Syngnathidae. From 1998 to 2010, 172 syngnathid cases from the Toronto Zoo were submitted for post-mortem diagnostics and retrospectively examined. Among the submitted species were yellow seahorses Hippocampus kuda Bleeker (n=133), pot-bellied seahorses Hippocampus abdominalis Lesson (n=35) and weedy seadragons Phyllopteryx taeniolatus (Lacépède; n=4). The three most common causes of morbidity and mortality in this population were bacterial dermatitis, bilaterally symmetrical myopathy and mycobacteriosis, accounting for 24%, 17% and 15% of cases, respectively. Inflammatory processes were the most common diagnoses, present in 117 cases. Seven neoplasms were diagnosed, environmental aetiologies were identified in 46 cases, and two congenital defects were identified.

RESUMEN

The Wilms tumor suppressor gene Wt1 encodes a zinc finger transcription factor that is essential for development of multiple organs including kidneys, gonads, spleen and heart. In mammals Wt1 comprises 10 exons with two characteristic splicing events: inclusion or skipping of exon 5 and alternative usage of two splice donor sites between exons 9 and 10. Most fish including zebrafish and medaka possess two wt1 paralogs, wt1a and wt1b, both lacking exon 5. Here we have characterized wt1 in guppy, platyfish and the short-lived African killifish Nothobranchius furzeri. All fish except zebrafish show alternative splicing of exon 4 of wt1a but not of wt1b with the wt1a(-exon 4) isoform being the predominant splice variant. With regard to function, Wt1a(+exon 4) showed less dimerization but stimulated transcription more effectively than the Wt1a(-exon 4) isoform. A specific knockdown of wt1a exon 4 in zebrafish was associated with anomalies in kidney development demonstrating a physiological function for Wt1a exon 4. Interestingly, alternative splicing of exon 4 seems to be an early evolutionary event as it is observed in the single wt1 gene of the sturgeon, a species that has not gone through teleost-specific genome duplication.

RESUMEN

Selenium pollution from coal ash wastewater was investigated in Lake Sutton, NC. This lake has been continuously used as a cooling pond for a coal-fired power plant since 1972. Historic and recent levels of contamination in fish tissues (14-105µg Se/g dry weight in liver, 24-127 in eggs, 4-23 in muscle, 7-38 in whole-body) exceeded toxic thresholds and teratogenic effects were observed in fish collected in 2013. A high proportion (28.9 percent) of juvenile Lepomis spp. exhibited spinal and craniofacial malformations that were consistent with selenium poisoning. Teratogenic Deformity Index values indicated population-level impacts on the fishery. The partially monetized cost of resultant fishery losses was calculated at over $US 8.6 million annually, and over $US 217 million for the entire period of damage, which dates back to 1987 when chemical and biological monitoring began.

This paper aims to standardize the use of eugenol as an anesthetic for the patinga (Piaractus brachypomus), measuring the time for anesthesia induction and recovery. Induction and recovery times to anesthesia were measured in different concentrations of eugenol and plasma glucose. The results were analyzed according to a completely randomized design (CRD) with six treatments (control, 25, 35, 50, 75 and 150 L-1 of eugenol) and ten repetitions each. The analysis of variance (ANOVA) and averages were compared by Tukey test. Concentrations above 150 mg.L-¹ presented 100% mortality of the population. The deep anesthesia induction times recorded for concentrations of 50 and 75 mg L-1 were the ones presenting the shortest sedation times. It can be observed that the patinga recovery time for 50mg.L-1 doses was 93 seconds. The 25mg.L-1 dose of eugenol was the only dose obtaining a decrease in plasma glucose levels which can be an indication to the use of small doses for the management of patinga. It can also be concluded that eugenol is a suitable anesthetic for patinga weighing 256.6 ± 73.18 g, and it can be used at concentrations of 50mg. L-1 eugenol...

Pejerrey (Odontesthes bonariensis) is a native species from Rio Grande do Sul, Uruguay and Argentina where it is of great economic importance for artisanal fishing. One difficulty in laboratory research with pejerrey is related to its sensitivity, as it presents higher basal cortisol levels than other freshwater species. For this reason, the aim of this work was to evaluate the efficiency of benzocaine and clove oil as anesthetics for pejerrey fingerlings. Two experiments were done where fingerlings (57±7.8mm and 1.1±0.44g) were exposed to benzocaine with concentrations between 40mgL-1 and 120mgL-1 and to clove oil with concentrations between 12mgL-1 and 75mgL-1. Survival, anesthesia induction time and recovery time for each pharmaceutics were evaluated. Both benzocaine and clove oil pharmaceutics showed efficiency as anesthetics for pejerrey fingerlings, with negative correlation between the dose of anesthetics and the anesthesia induction time. For benzocaine, the concentrations between 80mgL-1 and 100mgL-1 showed better results, as for clove oil the optimal concentrations were between 25mgL-1 and 50mgL-1. On the other hand, the anesthesia recovery time did not present significant variation on the different concentrations of the tested products. The tested products are highly metabolizable by pejerrey.

This study aimed to evaluate the influence of cryoprotectants, thawing times and temperatures on pacu (Piaractus mesopotamicus) sperm abnormalities. Semen samples from four pacu were collected and diluted in 2 solutions, cryoprotectant dimethylsulfoxide (DMSO) 10% and methanol 10%. After thawing, semen was collected for morphological analysis, evaluating primary and secondary abnormalities. There was no interaction (P>0.05) of secondary abnormalities, abnormalities in total and normal sperm in the different treatments. The primary abnormalities showed interaction between temperature and thawing time, where the higher temperature was associated with the shortest time (60°C for 8s) or the lowest temperature associated with longer thawing (40°C for 12s) provided under (P<0.05) percentages of spermatozoa with primary abnormalities. In the process of semen cryopreservation of P. mesopotamicus using the cryoprotectants DMSO and methanol, it is recommended that thawing be performed at 40° C for 12 seconds or 60°C for 8 seconds.

Aquatic ecosystems near urban areas suffer a steady deterioration caused by physical and chemical factors related to human activity, representing a danger to organisms that inhabit them, especially the fsh. These effects may be biomonitored by widely used techniques, such as the Micronucleus Test (Mn) and Nuclear Abnormalities (NA), among others. The aim of this study was to verify and compare the frequency of micronuclei and nuclear abnormalities in three fsh species collected in an urban lake, Cyprinus carpio, Astyanax eigenmanniorum and Cheirodon interruptus. After the capture and anesthesia of the fsh, weight and length were registered, and a blood sample was obtained from each individual. The samples were fxed and stained with 10% Giemsa, subsequently they were analyzed with the microscope (1000X) and photographed with a digital camera. The body con-dition index (K) was calculated for each fsh caught. A correlation analysis Spearman rank (rs) between frequency of Mn and NA vs. K was performed for each species. The Kruskal-Wallis test was applied to assess differences in Mn and NA between species. The results showed no statistically signifcant correlation (P> 0.05) between the frequencies of Mn and NA vs K. The frequencies of Mn between different species yielded no signifcant differences (P> 0.05), whereas NA (P <0.05) did. A. eigenmanniorum was the most sensitive species. NA preliminary differences between the species used, convert A. eigenmanniorum in a sort of "sentinel specie" and a possible biomonitor of xenobiotic agents.

RESUMEN

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous environmental contaminants, which are known carcinogens and teratogens. However, the toxicity of PAHs during skeletal development and the mechanism involved are not completely clear. In the present study, rockfish (Sebastiscus marmoratus) embryos were exposed to pyrene (Pyr) for 7 days at 0.5, 5 and 50 nM which resulted in craniofacial skeleton deformities. Pyr exposure for 6 days reduced the expression of PCNA, Col2a1 and Sox9 in the craniofacial skeleton revealed using in situ hybridization. These results suggest that Pyr exposure impairs skeleton development via disrupting the proliferation of the chondrocytes. At the same time, Pyr exposure reduced the expression of lox1 and inhibited the activity of lysyl oxidase, which is the key enzyme controlling the collagen cross-linking, and which might therefore have been one of the reasons for the deformative Meckel's cartilage (lower jaw).
