RESUMEN

The molecular targets and mechanisms of propolis ameliorating metabolic syndrome are not fully understood. Here, we report that Brazilian green propolis reduces fasting blood glucose levels in obese mice by disrupting the formation of CREB/CRTC2 transcriptional complex, a key regulator of hepatic gluconeogenesis. Using a mammalian two-hybrid system based on CREB-CRTC2, we identify artepillin C (APC) from propolis as an inhibitor of CREB-CRTC2 interaction. Without apparent toxicity, APC protects mice from high fat diet-induced obesity, decreases fasting glucose levels, enhances insulin sensitivity and reduces lipid levels in the serum and liver by suppressing CREB/CRTC2-mediated both gluconeogenic and SREBP transcriptions. To develop more potential drugs from APC, we designed and found a novel compound, A57 that exhibits higher inhibitory activity on CREB-CRTC2 association and better capability of improving insulin sensitivity in obese animals, as compared with APC. In this work, our results indicate that CREB/CRTC2 is a suitable target for developing anti-metabolic syndrome drugs.

Propolis is a non-toxic natural substance with multiple pharmacological properties including anti-cancer, antioxidant, fungicidal, antibacterial, antiviral, and anti-inflammatory among others. The aim of this study was to determine the chemical and botanical characterization of Chilean propolis samples and to evaluate their biological activity against the cariogenic bacteria Streptococcus mutans and Streptococcus sobrinus. Twenty propolis samples were obtained from beekeeping producers from the central and southern regions of Chile. The botanical profile was determined by palynological analysis. Total phenolic contents were determined using colorimetric assays. Reverse phase HPLC and HPLC-MS were used to determine the chemical composition. The minimum inhibitory concentration (MIC) was determined on S. mutans and S. sobrinus. All propolis samples were dominated by structures from native plant species. The characterization by HPLC/MS, evidenced the presence of quercetin, myricetin, kaempferol, rutine, pinocembrin, coumaric acid, caffeic acid and caffeic acid phenethyl ester, that have already been described in these propolis with conventional HPLC. Although all propolis samples inhibited the mutans streptococci growth, it was observed a wide spectrum of action (MIC 0.90 to 8.22 µg mL(-1)). Given that results it becomes increasingly evident the need of standardization procedures, where we combine both the determination of botanical and the chemical characterization of the extracts. Research conducted to date, describes a promising effectiveness of propolis in the prevention of caries and other diseases of the oral cavity, making it necessary to develop studies to identify and understand the therapeutic targets or mechanisms of molecular action of the various compounds present on them.

Honey bees use propolis to defend against invaders and disease organisms. As some colonies produce much more propolis than others, we investigated whether propolis collecting is associated with disease resistance traits, including hygienic behavior and resistance to the parasitic bee mite, Varroa destructor. The three highest (HP) and three lowest propolis-producing (LP) colonies among 36 Africanized honey bee colonies were initially selected. Queens and drones from these colonies were crossed through artificial insemination to produce five colonies of each of the following crosses: HP♀ X HP♂, LP♀ X HP♂, HP♀ X LP♂, and LP♀ X LP♂. Colonies headed by HP♀ X HP♂ queens produced significantly more propolis than those with HP♀ X LP♂ and LP♀ X HP♂ queens and these in turn produced significantly more propolis than those headed by LP♀ X LP♂ queens. The brood cell uncapping rate of the high-propolis-producing colonies in the hygienic behavior test was significantly superior to that of the other groups. The LP X LP group was significantly less hygienic than the two HP X LP crosses, based on the evaluation of the rate of removal of pin-killed pupae. The HP X HP colonies were significantly more hygienic than the other crosses. No significant differences were found in mite infestation rates among the groups of colonies; although overall, colony infestation rates were quite low (1.0 to 3.2 mites per 100 brood cells), which could have masked such effects. Honey and pollen stores were significantly and positively correlated with propolis production.
