RESUMEN
PREMISE: Bracken (Pteridium, Dennstaedtiaceae) is a cosmopolitan genus of aggressive disturbance colonizers that are toxic to agricultural livestock. The taxonomy of Pteridium has been treated in multiple schemes, ranging from one to six species worldwide, with numerous subspecies and varieties. Recent work has focused on the worldwide distribution and systematics of the bracken fern, but South America has been poorly represented. We present the first continent-wide sampling and analysis of Pteridium esculentum, a Southern Hemisphere diploid species. METHODS: Within South America, P. esculentum has several morphotypes, distinguished into subspecies by variation in indument and lamina architecture. We used double digest restriction site-associated DNA sequencing (ddRADSeq) to assess the phylogenetic relationships of P. esculentum subspecies. RESULTS: We found a striking genetic homogeneity in the species, being able to support only two morphotypes from molecular data: P. e. arachnoideum and P. e. campestre. We had high confidence for shallow and deep phylogenetic relationships, but less support for relationships among crown groups. CONCLUSIONS: We describe an east-west geographic pattern that would explain the relationships between populations; and, in contrast to previous studies, we detected differences with P. esculentum from Australia. These results will lay the foundations for studying variations in this species' behavior as a weed, as well as its impact on the production of agricultural livestock in South America.

Bovine enzootic hematuria (BEH) is a chronic disease caused by ingestion of Pteridium esculentum subsp. arachnoideum (Kaulf.) Thomson. This disease has great importance because it affects the animal wellbeing, causes economic losses, and hinders public health due to the toxic principle that is present in the milk of cows that ingest this plant. Early diagnosis assists in the decision making for disposal and replacement of animals in endemic regions. Despite ultrasonography is a useful tool for diagnosis of this disease, there is a lack of studies evaluating BEH in the state of Tocantins, Brazil, using ultrasound and epidemiological data and clinical signs; however, the histopathological examination is the method that provides the definitive diagnosis. The objective of this study was to describe the occurrence of BEH in the state of Tocantins and evaluate the efficiency of bladder ultrasound examination for diagnosis and prognosis of BEH. Ten animals that presented intermittent hematuria and hypo-colored mucous membranes were subjected to clinical and complementary exams. The urine physical analysis presented light red color, high turbidity, and positive sedimentation. The ultrasonography showed a thick, irregular mass in the bladder wall, containing an echogenic liquid of high cellularity and pedunculated papillary lesions. Macro and microscopic analyses of the animals' bladders showed areas with vascular proliferation in the sub-mucous membranes associated with mononuclear inflammatory cell infiltrate and proliferation of the epithelium. Ultrasonography is an excellent option and, combined with clinical and laboratory exams of blood and urine, enables the diagnosis and prognosis of BEH. (AU)
Hematúria enzoótica bovina (HEB) doença crônica causada pela ingestão de Pteridium esculentum subsp. arach-noideum (Kaulf.) Thomson.

Tropical montane forests are threatened by uncontrolled fire events because of agricultural expansion. Consequently, deforested areas frequently are dominated by the bracken fern, Pteridium spp., for long periods, and forest regeneration is limited. Despite considerable research on bracken-dominated ecosystems, little is known about the relationship between bracken mycorrhizal fungi and tree seedlings. Arbuscular mycorrhizal fungi (AMF) form symbiotic relationships with terrestrial plants, providing nutrients and protection against pathogens and promoting seedling growth and establishment. Therefore, AMF inoculum have high potential for forest restoration programs. Here, we compare the species diversity of AMF spores, root colonization, and seedling growth of Clusia trochiformis 1 year after the addition of different liquefied root inocula: forest conspecific, forest heterospecific, and from Pteridium rhizomes. Thirteen morphospecies of arbuscular mycorrhizal fungi were identified on the roots of C. trochiformis, and Glomus spp. were the most abundant in all treatments. No differences were observed in spore species richness and diversity among treatments, but spore density was the highest subsequent to the Pteridium inoculum. There was no significant difference in mycorrhizal root colonization and seedling growth of C. trochiformis among inoculated treatments. We found a positive relation between root colonization and total biomass. This study shows that the AMF communities in bracken areas and forests present similar characteristics and that the bracken fern does not limit AMF inoculum potential, favouring seedling growth of Clusia.

Pteridium aquilinum (Dennstaedtiaceae) colonization affects ecological and restoration processes. The knowledge of the impacts on the ecological succession by this species allows the use of restoration strategies in invaded environments. This work aimed to evaluate the floristic composition, diversity, structure, density, basal area, height, and diameter of natural regeneration in three areas of the Atlantic Forest in the Serra do Espinhaço Biosphere Reserve in an area invaded by P. aquilinum after a fire. Three environments with different coverage intensities by P. aquilinum were studied, and the plants over 10 cm in height or 5 cm in canopy diameter were measured. The floristic composition and diversity were analyzed using indices presented by Chao, Fisher, Margalef, Pielou, Shannon-Weaver, and Simpson, and similarity was evaluated by the Jaccard index. Species density, basal area, height, and canopy diameter classes were also evaluated. The floristic composition, diversity, structure of natural regeneration, density, and basal area were higher in post-fire areas with a lower coverage by P. aquilinum. The topsoil coverage with plant litter and the possible effect of P. aquilinum allelopathy probably reduced the species richness and diversity. The proportion of plants from the lowest height and canopy diameter classes was higher under moderate coverage by P. aquilinum. The reduction in the floristic composition, diversity, number of species, and basal area in post-fire areas colonized by P. aquilinum is probably due to this species aggressiveness. The population of this plant is high, accumulating large quantities of plant litter as a physical barrier preventing light and propagules from reaching the soil, reducing the germination of the seed bank and, consequently, the natural regeneration. The floristic composition, diversity, structure of natural regeneration, density, and basal area were lower in areas with higher coverage by P. aquilinum. The proportion of plants in the most significant height and canopy diameter classes was higher with reduced coverage by P. aquilinum. The P. aquilinum reduced forest succession in areas after a fire.

It is reported the occurrence of enzootic hematuria (EH) in buffaloes in Brazil after performing an epidemiological survey and clinicopathological analises. To date, EH caused by ingestion of Pteridium esculentum subsp. arachnoideum, a radiomimetic plant popularly known as "bracken fern", has not been described in this species in Brazil. Bovine EH is responsible for high economic losses in Brazil's Southeast Region not only because of the deaths it causes, but also owing to its negative effect on productivity. In São José do Barreiro County, São Paulo, some farmers in areas with a high incidence of bovine EH have been replacing cattle with buffaloes, based on the premise that the latter would be more resistant to poisoning by ingestion of Pteridium spp. However, even though initial observations indicated that buffaloes are indeed less sensitive than cattle to the toxic principle of Pteridium spp., cases of hematuria in this species have been reported. According to preliminary date, EH only occurs in buffaloes over six years of age. Macroscopic examination revealed a thickened urinary vesicle mucosa, along with multiple foci of ulcerated, exophytic, verrucous, and pedunculated lesions. In one of the buffaloes studied, the bladder wall was ruptured and exhibited marked secondary inflammation. Histologically, neoplastic and non-neoplastic changes similar to those described in cattle poisoned by Pteridium spp. were observed. The neoplasms found included papilloma, carcinoma in situ, urothelial carcinoma (low and high grade), inverted, microcystic, and trabecular variants, urothelial carcinoma with divergent differentiation (squamous and glandular), squamous cell carcinoma, lymphangioma, hemangioma, and hemangiosarcoma. There was also coexistence of epithelial and mesenchymal neoplasms. Bovine papillomavirus particles were not detected by polymerase chain reaction in the bladder samples analyzed.

Pteridium arachnoideum, a fern of the Pteridium aquilinum species complex found in South America, is responsible for several different syndromes of poisoning. Cases of bovine enzootic hematuria and upper alimentary squamous cell carcinoma are both frequent occurrences in Brazil, whereas only bovine enzootic hematuria is noted with any frequency around the world. The reason for the high frequency of upper alimentary squamous cell carcinoma in Brazil is not currently known. One possible explanation may be the higher levels of ptaquiloside and pterosin B in Brazilian Pteridium than those present in the plant in other countries. However, these levels have not yet been determined in P. arachnoideum. Thus, the present study aimed to measure and compare ptaquiloside and pterosin B levels in mature green fronds and sprouts of P. arachnoideum collected from different locations in Brazil. Samples of P. arachnoideum were collected from the states of Minas Gerais and Rio Grande do Sul. A total of 28 mature leaf samples and 23 sprout samples were used. The mean concentrations of ptaquiloside and pterosin B present in the mature green fronds of P. arachnoideum ranged from 2.49 to 2.75 mg/g and 0.68 to 0.88 mg/g, respectively; in P. arachnoideum sprouts, mean concentrations of ptaquiloside and pterosin B ranged from 12.47 to 18.81 mg/g, and 4.03 to 10.42 mg/g for ptaquiloside and pterosin B, respectively. Thus, ptaquiloside and pterosin B levels in P. arachnoideum samples collected in Brazil were higher in sprouts than in mature green fronds, as observed in other countries. However, there was no variation in ptaquiloside levels among plants collected from different cities in Brazil. The high frequency of upper alimentary squamous cell carcinoma in Brazilian cattle may not be attributed to greater levels of ptaquiloside and pterosin B in P. arachnoideum than in other Pteridium species in other countries.

Ferns of the genus Pteridium are known to be responsible for cases of poisoning in cattle. The commonly found species of Pteridium in Brazil are P. arachnoideum and P. caudatum. The toxic principle of these ferns is ptaquiloside, a norsesquiterpene glucoside, which is a known carcinogen. Ptaquiloside can be secreted in the milk of cows grazing on bracken ferns, thereby posing a potential health risk for the milk consumers. The objective of the present study was to develop an analytical methodology using high performance liquid chromatography (HPLC) with ultraviolet (UV) detection to determine the concentrations of ptaquiloside in cow´s milk. HPLC-UV method, based on the conversion of ptaquiloside into pterosin B and its subsequent extraction using dichloromethane, was successfully developed for estimating concentrations of ptaquiloside in milk samples. However, the presence of ptaquiloside was not detected in milk samples obtained from cows with a history of bovine enzootic hematuria resulting from grazing on P. arachnoideum infested pastures in Brazil.(AU)
The diet of shredder chironomid larvae depends on the local and temporal conditions of the food resources. We analysed the gut content of shredder chironomid larvae that colonised the leaf litter of three riparian species: Hedychium coronarium, Pteridium arachnoideum and Magnolia ovata. We hypothesised that the differences in the decomposition rates of leaf litter species influence the consumption of plant tissue by shredder chironomid taxa over time. We incubated perforated bottles with each leaf species within four low-order streams during 1st, 3rd, 7th, 22nd, 36th, 55th and 85th day of exposure. We used an analysis of covariance (ANCOVA) to compare differences in the percentage of AFDM (ash-free dry mass) and AOM (amorphous organic matter) among leaf litter species. To verify differences in the larvae abundance, we used a general linear model, and to test if there were feeding preferences for AFDM and AOM, we used the adapted Paloheimo selectivity index. Magnolia ovata presented a higher quantity of AOM followed by H. coronarium and P. arachnoideum. Pteridium arachnoideum showed a higher AFDM followed by H. coronarium and M. ovata. The larvae abundance was different among plant species and varied significantly with AFDM and AOM quantities. The consumption of plant tissue by shredder chironomid differed temporarily and among riparian species, where facultative or strict shredders showed strong association with different leaf litter species. The amount of AFDM and AOM in plant tissues explained these differences. We highlighted that shredder chironomids displayed an important role as co-participants in the decomposition process.

Upper digestive tract (UDT) cancer is rare in cattle, however in Southern Brazil, the UDT squamous cell carcinomas (SCCs) are relatively common and have been associated with bracken fern consumption and the presence of papillomas. Although a theory of pathogenesis considers bovine papillomavirus type 4 (BPV-4) as a cofactor in the development of these SCCs, some aspects of the etiopathogenesis of this disease need to be more investigated. In fact, detection of BPV-4 in UDT papillomas is scarce in other regions of the world and has not been performed in Brazil. Therefore, this study had two aims: 1) to analyze the epidemiological, clinical and pathological aspects of 100 natural cases of SCCs in the UDT of cattle grazing on bracken fern (Pteridium arachnoideum) highly contaminated areas, investigating the associations between these parameters; and 2) to investigate the presence of papillomavirus DNA by polymerase chain reaction (PCR) in the UDT papillomas (n = 47) from 30 cattle that also had UDT SCCs. There were statistically significant associations between clinical signs and tumor localization in the UDT; between histological grade of differentiation and tumor localization; and a trend towards significant association between histological grade of differentiation and presence of metastases. The average age of cattle with oropharyngeal SCCs was 7.39 years, with statistically significant difference comparing to cattle with esophageal SCCs (8.6 years). No statistical association was observed among other clinical-pathological parameters (growth pattern and primary site of the tumor) analyzed. No BPV DNA was detected in papillomas by PCR. Therefore, these results suggest the possibility that papillomas of the UDT are not necessarily associated with BPV infection.

In tropical agriculture, the vigorously growing Bracken fern causes severe problems by invading pastures and out-competing the common pasture grasses. Due to infestation by that weed, pastures are abandoned after a few years, and as a fatal consequence, the biodiversity-rich tropical forest is progressively cleared for new grazing areas. Here we present a broad physiological comparison of the two plant species that are the main competitors on the pastures in the tropical Ecuadorian Andes, the planted forage grass Setaria sphacelata and the weed Bracken (Pteridium arachnoideum). With increasing elevation, the competitive power of Bracken increases as shown by satellite data of the study region. Using data obtained from field measurements, the annual biomass production of both plant species, as a measure of their competitive strength, was modeled over an elevational gradient from 1800 to 2800 m. The model shows that with increasing elevation, biomass production of the two species shifts in favor of Bracken which, above 1800 m, is capable of outgrowing the grass. In greenhouse experiments, the effects on plant growth of the presumed key variables of the elevational gradient, temperature and UV radiation, were separately analyzed. Low temperature, as well as UV irradiation, inhibited carbon uptake of the C4-grass more than that of the C3-plant Bracken. The less temperature-sensitive photosynthesis of Bracken and its effective protection from UV radiation contribute to the success of the weed on the highland pastures. In field samples of Bracken but not of Setaria, the content of flavonoids as UV-scavengers increased with the elevation. Combining modeling with measurements in greenhouse and field allowed to explain the invasive growth of a common weed in upland pastures. The performance of Setaria decreases with elevation due to suboptimal photosynthesis at lower temperatures and the inability to adapt its cellular UV screen.

The infestation of pastures by populations of Pteridium arachnoideum has been a considerable and global problem to the livestock production, due to animal mortality and pasture coverage reduction. Given the impact of P. arachnoideum on cattle production, it has been proposed to monitor a beef cattle farm in Rio Grande do Sul, to assess some of the losses associated with the plant consumption and the methods employed for the plant control. A survey of cattle deaths associated with fern poisoning on the farm was carried out from January 2007 to January 2015. In addition, the viability of sheep as a biological control tool was tested. Attempts to control the plant through the use of the herbicides methylsulfuron-methyl and picloram associated or not with previous mowing were monitored. Cases of natural poisoning observed in the farm included the acute form (22), known as hemorrhagic diathesis as well as the chronic form, consisting in digestive carcinomas (6). Five cases of hemorrhagic diathesis in young cattle went along with marked laryngeal edema, which was clinically manifested as dyspnea and roaring, in addition to the classic pathological changes of widespread hemorrhages and infarcts. Even though less frequent, upper digestive tract carcinomas caused significant losses, due to annual mortality of mature cows. In occasions, when cattle were moved to newly mowed infested areas, outbreaks of acute poisoning were observed. The attempted control by sheep introduction showed to be unpractical, due to the little consumption seen at a stocking rate of four sheep per hectare. However, sheep mortality due to P. arachnoideum consumption was not recorded. The plant control method applied at the farm is based on both, the mowing of densely populated areas and the use of herbicides, namely metylsulfuron-methyl and picloram, in all paddocks. The reduction of P. arachnoideum coverage showed to be varied in different paddocks, however allowed the recovery of some grazing areas. This practice needs further studies, as some areas have not had satisfactory pasture recovery, and its indiscriminate use could lead to environmental impacts and resistance of P. arachnoideum to herbicides.(AU)

Oesophageal squamous cell carcinomas (SCCs) may be observed as exophytic masses or ulcerative or infiltrative endophytic neoplasms. However, in cattle, there is also an uncommon gross presentation as an annular stenotic thickening of the oesophageal wall. Thirteen cases of annular stenotic oesophageal SCC in cattle grazing in bracken fern (Pteridium arachnoideum) areas are reported. The lesions consisted of endophytic masses, focally extensive, firm and circumferential (annular) in the oesophageal wall. Pronounced wrinkling of the mucosa, with retracted uneven areas and subsequent luminal narrowing (stenosis), was observed in all cases. Papillomas and squamous intraepithelial lesions also were observed in these cases. The SCCs were graded as well differentiated (n = 7), moderately differentiated (n = 5) or poorly differentiated (n = 1). The neoplastic keratinocytes were surrounded by moderate to abundant fibrous connective tissue (a desmoplastic reaction), that was better demonstrated by Masson's trichrome stain. Picrosirius red-stained sections showed abundant collagen type I fibres, which contributed to the stenosing characteristics of this tumour. Although it might be easily misdiagnosed as oesophageal scar tissue, the oesophageal SCCs of cattle grazing bracken fern may have a distinctive gross appearance that should be included in the differential diagnosis of oesophageal stenosis.

The aim of this study was evaluate the effects of Bracken fern (BF) (Pteridium aquilinum (L.) Kuhn.) on biological systems. When consumed by animals can cause acute intoxication, hematuria, biochemistry alterations and cancer. To humans the toxicity is associated with its intake on contaminated ground water or milk and inhalation of its spores. In order to check the BF aqueous extract (AEB) deleterious effects on animals blood vessels system, chick embryo chorioallantoic membrane (CAM) was used. It were applying on CAM 0.1, 0.5, 1, 5, 10 e 15 µg/mL of AEB and saline as control. The angiogenesis was analyzed and the vascular density index (VDI) calculated. The CAM samples were prepared and stained with H&E to evaluation of microvessels, Massons trichrome to characterize collagen and fibrin deposition and Picro-sirius used to evaluate collagen using polarized light. Also the morphological aspects of embryos were analysed. We observe on the results of neovascularization that AEB did not change significantly the number of vessels/mm², however, membranes treated with AEB (5 or 10 µg/ mL) exhibit opacity and tissue fibrosis, both signs of inflammation. Histological analysis with Massons trichrome and picro-sirius on tissues exposed to AEB respectively has shown increased collagen fibers and presence of fibrilar collagen. The embryos exposed to concentrations of 5 or 10 μg/mL AEB, showed changes as poor face formation and poor closing of abdominal wall. The highest concentration of AEB (15 μg/mL) was lethal to embryos. Although significant effects on the CAMs vasculature has not observed, tissue aggression was detected, a desmoplasia (an extensive inflammatory signal triggered by tissue injury), changes caused on embryos as well as the presence of toxic substances in the AEB show us an important and deleterious pathway of this bracken fern extract on its intoxicants effects on humans and animals, and even cancer or the death of animals.(AU)
