RESUMEN

A high-performance fluorescent "turn-on" aptasensor (Eu-MOFs@SMZ-Apt) for sulfamethazine (SMZ) determination was designed using dual-emitting europium metal-organic frameworks (Eu-MOFs) as a signal transducer and an amplifier. Eu-MOFs featuring dual emission peaks (430 nm and 620 nm) were first prepared via a facile self-assembly strategy employing Eu (III) ions and 2-aminoterephthalic acid as precursors. The high-affinity aptamer was bonded with Eu-MOFs to form Eu-MOFs@SMZ-Apt through the amidation reaction. Benefiting from the integration of inherent virtues from Eu-MOFs and aptamer, the Eu-MOFs@SMZ-Apt-based sensor allowed sensitive and selective determination of SMZ with good linear relationships in a range of 1.4-40 ng mL-1 and a low detection line (0.379 ng mL-1). This sensor was successfully applied to the determination of trace SMZ in real samples with satisfactory recoveries (86.47-113.52%) and a relative standard deviation (<6.51). Consequently, the Eu-MOFs@SMZ-Apt ratiometric fluorescence sensor furnishes new possibilities for the accurate detection of various pollutants in food.

RATIONALE: Retroperitoneal dedifferentiated liposarcoma (RPDDL) is an uncommon malignancy, which often remains undetected for many years due to having adequate space in the retroperitoneal cavity and lacking clinical manifestations in the early stage of the disease. Surgical procedure is usually used as the first choice for treatment. However, it is prone to local recurrence after the operation, resulting in an unfavorable prognosis. Our aim is to draw useful lessons from the new case and provide some experience for management of the disease. PATIENT CONCERNS: We describe a 55-year-old male patient who was admitted for a 3-week history of persistent dull ache of the left waist. A large mass of the left upper abdomen was palpated in physical examination. Moreover, the imaging examination revealed that the diameter of the mass was about 21 cm, and some adjacent vital organs were invaded, which brought great challenges to complete surgical resection. DIAGNOSIS: The postoperative pathological results confirmed that the mass was RPDDL with invasion of the surrounding vital structures including pancreas, spleen, left adrenal gland, left kidney, and vasculature with tumor emboli. INTERVENTIONS: Surgical resection of the mass was performed by our multidisciplinary team. The patient received chemotherapy 1 month after surgery. OUTCOMES: The effect of chemotherapy seemed to be unsatisfactory. Local multifocal recurrence of the tumor was considered about 2 months after surgery. Finally, he gave up any treatments and died of the disease. LESSONS: Regular physical examination and ultrasound screening may detect the disease as early as possible, especially for high-risk group aged 60 to 70, which should be popularized. Incomplete resection, vascular invasion, and interruption of postoperative treatment may lead to an unfavorable prognosis. Therefore, we think that patients with the disease may benefit from complete surgical resection and uninterrupted adjuvant therapy.

Self-assembled Eu-dipeptide (tryptophan-phenylalanine) microparticles with multi-emission fluorescence was prepared and modified with a single-stranded DNA corresponding to the sulfamethazine (SMZ) adapter (Eu-PMPs@cDNA). Aptamer-functionalized magnetic Fe3O4 (MNPs@aptamer) was used to specifically bind the target SMZ. Using Eu-PMPs@cDNA as fluorescent signal probe and MNPs@aptamer as catcher, a noncompetitive fluorescence sensing strategy was developed for determination of SMZ with good sensitivity, accuracy, selectivity, and stability. Under the optimized conditions, fluorescence increases linearly in the 0-20 ng/mL SMZ concentration range, and the detection limit is 0.014 ng/mL. The fluorescence sensing method was applied to analysis of water and fish muscle samples, and recoveries ranged from 81.78 to 119.46 % with relative standard deviations below 4.2 %. This study offered a reliable and sensitive fluorescence sensing strategy for SMZ determination in food samples, which owns great potential for wide-ranging application in harmful compounds assay by simply changing the type of aptamer and its complementary single-stranded DNA.

Mycobacterium farcinogenes (M. farcinogenes) is rapidly growing mycobacterium, belonging to non-tuberculous mycobacterial (NTM). M. farcinogenes is an exceedingly rare causative agent of human infection. Only seven cases with M. farcinogenes infections in humans were reported. This is a case of soft tissue infection and osteomyelitis caused by M. farcinogenes after heart surgery. Microbial identification was achieved by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). The clinical outcome was favorable after surgical debridement and 4-month antibiotics treatment. We also provide a comprehensive literature review on this disease.

Crystalline carbon nitride (CCN), derived from amorphous polymeric CN, is considered as a new generation of metal-free photocatalyst because of its high crystallinity. In order to further promote the photocatalytic performance of CCN, p-type MnO nanoparticles are in situ synthesized and merged with n-type CCN through a one-pot process to form p-n heterojunction. The formed interfacial electric field between the semiconductors with different work functions efficiently breaks the coulomb interaction between MnO and CCN. The prepared catalysts exhibit drastically increased photocatalytic hydrogen evolution (PHE) activity integrated with oxidation of alkyl and aryl alcohols under irradiation of visible light. In the aqueous solution of benzyl alcohol (BzOH), the hydrogen generation rate over MnO/CCN (39.58 µmol h-1) is nearly 7 times and 37 times that of pure CCN (5.76 µmol h-1) and CN (1.06 µmol h-1), respectively, combining with oxidation of BzOH to benzaldehyde. This work proposes an avenue for in situ construction of a novel 2D material-based S-scheme heterojunction and extends its application in solar energy conservation and utilization.

Photocatalytic performance of polymeric carbon nitride (CN) is primarily restricted by limited light utilization and poor charge separation efficiency. To this end, skeleton modification strategy was adopted by attaching thiophene ring and polar nickel complex (NiL) onto CN. The obtained bifunctionalized carbon nitride (TCN-NiL) displayed obviously elevated optical absorption and photoexcited charge separation efficiency. The NiL, with polar structure, plays as active sites like cocatalyst thus exhibited platinum-like H2 evolution activity from water splitting under visible light. The optimized photocatalytic H2 generation rate over TCN-NiL reached 136.7 µmol·h-1 without any cocatalyst, the highest rate reported so far in noble-metal-free CN-based catalysts, which is 5 times of that of CN loaded with 3 wt% Pt. Additionally, the maximum wavelength of performing H2 production capacity over TCN-NiL extends to 550 nm from 450 nm of CN, suggesting an excellent visible light absorption ability. This work provides a way for modifying CN to enhance the photocatalytic activities in a noble metal free system.

Background: Infection is the most important cause of non-relapse mortality in hematologic malignancy patients, leading to increased costs and prolonged hospitalization times. However, comprehensive and comparable reports on infection-specific mortality (ISM) trends in hematologic malignancy patients are lacking. Objectives: We aimed to provide updated ISM trends and factors associated with ISM among hematologic malignancy patients. Design: This is a retrospective study. Methods: Patients diagnosed with the five most common hematologic malignancies from 1983 to 2016 from the Surveillance, Epidemiology, and End Results database were included. Joinpoint regression was used to analyze mortality trends. Results: ISM decreased beginning in 1983, 1988, and 1994, with yearly decreases of -2.1% for acute leukemia (AL), -1.3% for Hodgkin lymphoma (HL), and -14.3% for non-Hodgkin lymphoma (NHL). In contrast, ISM in patients with chronic leukemia (CL) and multiple myeloma (MM) increased dramatically beginning in 2000, with yearly increases of 2.8% and 3.3%, respectively. ISM rates were higher in males than in females across all hematologic malignancy subtypes. The mortality trends significantly differed according to race, age, sex, and stage, which could help in further etiological investigations. Moreover, male sex, older age at diagnosis, black race, and unmarried status were poor prognostic factors for ISM across all hematologic malignancy subtypes. Conclusion: A promising downward trend in ISM in recent years occurred in patients with AL, HL, and NHL; however, ISM increased dramatically in patients with CL and MM. Our data suggest that risk assessment and careful infection monitoring are recommended for hematologic malignancy patients, particularly those with CL and MM.

Introduction: Diffuse large B-cell lymphoma (DLBCL) is the most common subtypes of lymphoma. Clinical biomarkers are still required for DLBCL patients to identify high-risk patients. Therefore, we developed and validated the platelet-to-albumin (PTA) ratio as a predictor for DLBCL patients. Methods: A group of 749 patients was randomly divided into a training set (600 patients) and an internal validation set (149 cases). The independent cohort of 110 patients was enrolled from the other hospital as an external validation set. Penalized smoothing spline (PS) Cox regression models were used to explore the non-linear relationship between the PTA ratio and overall survival (OS) as well as progression-free survival (PFS), respectively. Results: A U-shaped relation between the PTA ratio and PFS was identified in the training set. The PTA ratio less than 2.7 or greater than 8.6 was associated with the shorter PFS. Additionally, the PTA ratio had an additional prognostic value to the well-established predictors. What's more, the U-shaped pattern of the PTA ratio and PFS was respectively validated in the two validation sets. Discussion: A U-shaped association between the PTA ratio and PFS was found in patients with DLBCLs. The PTA ratio can be used as a biomarker, and may suggest abnormalities of both host nutritional aspect and systemic inflammation in DLBCL.

Mycoplasma hominis, Ureaplasma parvum, and Ureaplasma urealyticum commonly colonize the human urogenital tract, which may cause urogenital infections. However, infection by M. hominis, U. parvum, or U. urealyticum is rarely reported in patients with peritoneal dialysis (PD)-associated peritonitis. Herein, we reported four cases of PD-associated peritonitis caused by these pathogens, along with a review of the literature. The four cases were female patients with recurrent "culture-negative" PD-associated peritonitis and were related to menstruation. M. hominis, U. parvum, or U. urealyticum was detected in the PD fluid of the patients by metagenomic next-generation sequencing. All four patients were cured by intraperitoneal tigecycline combined with oral azithromycin or minocycline. M. hominis, U. parvum, and U. urealyticum should be paid more attention in female patients with recurrent culture-negative PD-associated peritonitis, especially when the peritonitis is related to menstruation, sexual intercourse, or urogenital tract operation. Moreover, metagenomic next-generation sequencing can provide a reasonable method to identify the pathogen for culture-negative PD-associated peritonitis.

An innovative core-shell magnetic molecularly imprinted polymers, Fe3O4@SiO2@MIPs, was elaborately tailored for specific separation and enrichment of olaquindox (OLA) and its metabolic marker methyl-3-quinoxaline-2carboxylic acid (MQCA). Herein, benefiting from the combination of functionalized magnetic nanoparticles (Fe3O4@SiO2) and MIPs, Fe3O4@SiO2@MIPs not only possessed favorable magnetic properties and stability, avoiding tedious sample pretreatment process, but also demonstrated exceptional selective recognition ability and adsorption capacity, suppressing influence of coexisting interfering substances. Encouraged by prominent merits, Fe3O4@SiO2@MIPs-based magnetic solid phase extraction with HPLC method was developed, realizing simultaneous measurement of OLA and MQCA. Under optimal conditions, excellent linear ranges of 0-100 µg/L with detection limit of 0.175-0.271 µg/L were obtained. The proposed method was finally utilized for determination of OLA and MQCA in fish muscle and milk samples with satisfactory recoveries (80.56-95.26%) and relative standard deviation below 8.1%, furnishing a reliable and sensitive strategy for enrichment and detection residual veterinary drugs in food samples.

Tetracycline (TC) residues not only endanger human health, but also are detrimental to the sustainable development of aquaculture and animal husbandry. Herein, a novel fluorescence sensor with high sensitivity and selectivity was developed based on nitrogen-doped carbon dots embedded in zinc-based metal-organic frameworks and incorporating molecularly imprinted polymer (ZIF-8&N-CDs@MIP). The physical and chemical properties of the ZIF-8&N-CDs@MIP had been characterized by SEM, TEM, FTIR, XRD, BET, TGA, etc. Under optimal conditions, the limit of detection (LOD) of the novel sensor was 0.045 µg mL-1 with the concentration of TC in the range of 0.1-4.0 µg mL-1. In addition, the prepared imprinted polymers showed superior adsorption selectivity to tetracycline compared with non-imprinted polymers, and the quenching mechanism of ZIF-8&N-CDs@MIP was demonstrated to be attributed to the inner filter effect (IFE). This work provided an effective and reliable method for the specific detection of tetracycline and was successfully applied in milk and egg samples with satisfactory recoveries (80.67-95.22%).

Olaquindox residues in food from its illegal use has received great attention. Here, an immunoassay strategy integrating an upconversion nanoparticle (UCNP)-based immunochromatographic strip with a fluorescence reader was proposed for the highly selective and sensitive detection of olaquindox. Polyacrylic acid-functionalized UCNPs were synthesized using a simple ligand exchange process and combined with an olaquindox polyclonal antibody to form a fluorescent probe. This approach achieved a sensitive response and specific recognition of olaquindox. A convenient upconversion fluorescence reader was introduced to implement accurate and sensitive quantitative analysis of olaquindox based on the fluorescence intensity of control and test lines on a strip. Under optimal conditions, the method demonstrated a favorable linear range (0-50 ng/mL) and sensitive detection (1.42 ng/mL, S/N = 3). This method was applied successfully to determine olaquindox in fish muscle and water samples, and results were consistent with an HPLC approach, and considered a promising strategy for monitoring olaquindox residuals.

Various types of fluorescent nanoparticles have been proposed for the detection of veterinary drug residues in food. However, structure-induced fluorescence nanoparticles with biodegradability and the capacity to conjugate with molecular recognition elements are lacking. Here, a biodegradable tryptophan-phenylalanine dipeptide-based nanomaterial was assembled and modified with a sulfadimethoxine aptamer (TPNPs@aptamer). Then, a novel and efficient fluorescence sensing strategy, based on TPNPs@aptamer and BHQ1-labeled single-stranded DNA (BHQ1@cDNA), was developed for sulfadimethoxine determination. Under optimal conditions, the fluorescence was linear in the range of 0-200 ng/mL sulfadimethoxine with a detection limit of 1.47 ng/mL (S/N = 3). Satisfactory recoveries of standard additions were found between 92.88 % and 114.15 % for water and milk samples with a relative standard deviation less than 5.0 %, suggesting that the proposed fluorescence sensing strategy can be applied reliably in sulfadimethoxine analysis of liquid foods.

Ultrasensitive and onsite detection of oxytetracycline (OTC) is of vital significance for ensuring public health. Herein, a novel and versatile fluorescence biomimetic nanosensor, Mg,N-CDs@MIL-101@MIP, was elaborately tailored for the assay of OTC. MIL-101 with extraordinarily high surface area and porosity, as a favorable supporter, suppressed self-quenching of Mg,N-CDs and boosted mass transfer rate, realizing signal amplification. As an ultrasensitive signal transducer, high luminescent Mg,N-CDs yielded conspicuous fluorescence responses for OTC, enhancing the sensitivity of Mg,N-CDs@MIL-101@MIP. High-affinity imprinting sites further endowed Mg,N-CDs@MIL-101@MIP with superior anti-interference ability and reusability. Given prominent merits, Mg,N-CDs@MIL-101@MIP demonstrated a good linear range (0.05-40 µg mL-1) with a lower limit of detection (16.8 ng mL-1), supplying high accessibility to realize ultrasensitive and highly selective measurement of OTC in samples. Additionally, to attain precise onsite profiling of OTC, an intelligent sensing platform was developed by integrating Mg,N-CDs@MIL-101@MIP with a portable smartphone-assisted optical device. As both signal reader and analyzer, smartphone can instantly capture concentration-dependent fluorescent images and accurately digitize them, accomplishing quantitative analysis of OTC. More delightfully, the portable platform was utilized for visual determination of OTC in milk samples with satisfactory results, offering a promising tool for the high-performance onsite evaluation of food safety and environmental health.

Astrocytes (ASTs) and oligodendroglial lineage cells (OLGs) are major macroglial cells in the central nervous system. ASTs communicate with each other through connexin (Cx) and Cx-based network structures, both of which allow for quick transport of nutrients and signals. Moreover, ASTs interact with OLGs through connexin (Cx)-mediated networks to modulate various physiological processes in the brain. In this article, following a brief description of the infrastructural basis of the glial networks and exocrine factors by which ASTs and OLGs may crosstalk, we focus on recapitulating how the interactions between these two types of glial cells modulate myelination, and how the AST-OLG interactions are involved in protecting the integrity of the blood-brain barrier (BBB) and regulating synaptogenesis and neural activity. Recent studies further suggest that AST-OLG interactions are associated with myelin-related diseases, such as multiple sclerosis. A better understanding of the regulatory mechanisms underlying AST-OLG interactions may inspire the development of novel therapeutic strategies for related brain diseases.

BACKGROUND: Despite its efficacy, emerging concerns exist regarding radiation therapy (RT)-associated toxicity in adolescent and young adult (AYA) lymphoma patients. Few long-term follow-up studies have examined the association between RT and outcomes. METHODS: Lymphoma patients aged 15-39 years were identified in the Surveillance, Epidemiology and End Results (SEER) database from 1992 to 2016. Mortality was assessed by comparing those with and without RT using the Fine-Gray competing risk model. Standardized mortality ratios (SMRs) were used to assess the relative risk of death compared with the general U.S. RESULTS: In total, 29,686 patients were included; 10,708 (36.07%) received RT. Cause-specific mortality was compared between patients with and without RT while considering other competing events, including death due to index cancer, second malignant neoplasms (SMNs), and noncancer causes. Patients with RT had a lower probability of death and crude 5-year cumulative incidence of death. Moreover, there were significantly lower SMRs in patients with RT than in patients without RT. Differences between the two groups were greatest for mortality due to hematological malignancies and infections. Additionally, in the RT cohort, the SMR for index-cancer-related death was highest in the first year after diagnosis and gradually decreased. Hematological malignancies and infections were the most common specific SMN and noncancer causes of death, respectively. CONCLUSIONS: RT did not increase mortality from index cancer, SMNs, or noncancer causes in AYA patients with lymphoid malignancies. The current analysis may serve as a reference for healthcare providers monitoring RT application for AYA lymphoid malignancy survivors.

Although the link of white matter to pathophysiology of schizophrenia is documented, loss of myelin is not detected in patients at the early stages of the disease, suggesting that pathological evolution of schizophrenia may occur before significant myelin loss. Disrupted-in-schizophrenia-1 (DISC1) protein is highly expressed in oligodendrocyte precursor cells (OPCs) and regulates their maturation. Recently, DISC1-Δ3, a major DISC1 variant that lacks exon 3, has been identified in schizophrenia patients, although its pathological significance remains unknown. In this study, we detected in schizophrenia patients a previously unidentified pathological phenotype of OPCs exhibiting excessive branching. We replicated this phenotype by generating a mouse strain expressing DISC1-Δ3 gene in OPCs. We further demonstrated that pathological OPCs, rather than myelin defects, drive the onset of schizophrenic phenotype by hyperactivating OPCs' Wnt/ß-catenin pathway, which consequently upregulates Wnt Inhibitory Factor 1 (Wif1), leading to the aberrant synaptic formation and neuronal activity. Suppressing Wif1 in OPCs rescues synaptic loss and behavioral disorders in DISC1-Δ3 mice. Our findings reveal the pathogenetic role of OPC-specific DISC1-Δ3 variant in the onset of schizophrenia and highlight the therapeutic potential of Wif1 as an alternative target for the treatment of this disease.

In this work, a suppressive piezoelectric immunosensor based on the ultra-sensitive quartz crystal microbalance and highly specific immunosorbent assay was constructed for the detection of ribavirin for the first time. The carboxyl group was modified on the surface of the gold electrode by electropolymerization, and then the carboxyl group was combined with the amino-modified antigen, and then the corresponding antibody was able to bind to it according to the principle of immune recognition, which interlocked to cause the frequency change of the piezoelectric sensor. The preparation conditions of the sensor and the antibody concentration were optimized, and the performance of the sensor was evaluated. Under the optimal conditions, a wide linear range of 1-750 µg L-1, a low detection limit (IC15) of 2.64 µg L-1, and a good sensitivity (IC50) of 31.49 µg L-1 were obtained. The sensor was used for the detection of ribavirin in chicken and milk samples, and the recovery rate ranged from 88.01 to 94.42%, which shows satisfactory consistency with the detection results of high performance liquid chromatography. It was suggested that the proposed piezoelectric immunosensor has good sensitivity and selectivity for ribavirin, and can be applied to the detection of actual food samples.

In this work, an Eu3+-MOF-253@Au electrochemiluminescence sensor was successfully constructed for the first time by encapsulating nanogold in the metal-organic frameworks (MOFs) backbone and pore channels, and assembling Eu3+ on the MOF backbone. Firstly, the introduction of nanogold overcomes the weakness of MOFs, which was difficult to achieve, and enhances its catalytic performance, followed by the modification of Eu3+ to confer the electrochemiluminescence performance and the function of target detection on the sensor. Moreover, carbaryl was placed in an alkaline working solution to enhance the intensity of electrochemiluminescence signal, as well as to promote the hydrolysis of carbaryl into 1-naphthol, which caused the burst of Eu3+-MOF-253@Au electrochemiluminescence sensor, thereby achieving the sensitive detection of carbaryl. On this basis, the electrochemiluminescence detection conditions were optimized, the performance was analyzed, and finally it was successfully used for the detection of carbaryl with good linearity in the range of 0.2-200 µg L-1 and a low detection limit (0.14 µg L-1).

In this study, a novel fluorescent molecularly imprinted nanosensor (N, S-GQDs@ZIF-8@MIP) based on the nitrogen and sulfur co-doped graphene quantum dots decorated zeolitic imidazolate framework-8 was constructed for the detection of octopamine (OA). Herein, ZIF-8 with a large surface area was introduced as a supporter of the sensing system, which effectively shortened the response time of the sensor. Meanwhile, high green luminescent N, S-GQDs and a maximum emission wavelength of 520 nm under 460 nm excitation and a 12.5% quantum yield were modified on the surface of ZIF-8 as a signal tag that can convert the interactions between the sensor and OA into detectable fluorescent signals. Finally, N, S-GQDs@ZIF-8@MIP was acquired through the surface molecular imprinting method. Due to the synergy of N, S-GQDs, ZIF-8, and MIP, the obtained sensor not only demonstrated higher selectivity and sensitivity than N, S-GQDs@ZIF-8@NIP, but also displayed faster fluorescence response than N, S-GQDs@MIP. Under optimal conditions, the developed sensor presented a favorable linear relationship in the range of 0.1-10 mg L-1 with a detection limit of 0.062 mg L-1. Additionally, the proposed N, S-GQDs@ZIF-8@MIP strategy was effectively applied to the detection of OA in fermented samples, and the obtained results had a satisfactory correlation with those of HPLC.
