ABSTRACT

The expression of the gene of pattern recognition receptor TLR2, proinflammatory cytokine IL-1ß, and anti-inflammatory cytokine IL-10 was analyzed in the peripheral blood of nonagenarians (n=219; mean age 92.1 years, 77 men and 142 women) in comparison with healthy young donors (n=24; mean age 22.5 years, 16 women and 8 men). Nonagenarians were interviewed, medical records were analyzed, and a comprehensive geriatric assessment was performed according to the Clinical Guidelines on Frailty. The level of gene expression was determined by real-time PCR. The participation of inflammatory mechanisms in the immunosenescence was revealed. It was shown that increased expression of IL1B and TLR2 genes is associated with the development of frailty in nonagenarians and can be a factor of pathological aging. Increased expression of IL10 gene can be considered as a factor of successful aging in nonagenarians.

ABSTRACT

For evaluation of effects of release-active antibodies to CD4 on cultured lymphocytes from human peripheral blood, we measured intracellular content of lck-kinase cell-based ELISA. In cells treated with release-active antibodies to CD4, the content of intracellular lck-kinase significantly (p<0.01) decreased in comparison with the control (purified water processed in a similar way). Phytohemagglutinin had no effect on the concentration of lck-kinase in cells. The decrease in the content of CD4-associated lck protein suggests that the preparation enhanced intracellular coupling of lck-kinase with T-cell receptor and potentiated T-cell immune response.

ABSTRACT

AIM: Study of the role of innate immunity factors, namely, expression of TLR2 and HBD-2 genes, as well as TNF-α arid TGF-ß in pathogenesis of periodontium tissue inflammation. MATERIALS AND METHODS: 59 individuals with periodontitis illnesses were included into the study; 15 healthy donors represented the comparison group. Study of the TLR2 and HBD-2 gene expression levels was carried out by rRT-PCR, cytokine production evaluation--by EIA. RESULTS: The results of the study have shown the presence of TLR-mediated disbalance in the innate immunity system in the periodontium tissue during chronic generalized periodontitis. TLR2 gene hyperexpression was accompanied by the reduction of expression of anti-microbial peptide HBD-2, as well as an increase of production of TNF-αand TGF-P by epithelial cells of periodontium mucosa. Conclusion.-The study carried out has shown that disturbance of molecular mechanisms of innate immunity system has an important place in pathogenesis of periodontitis. -

ABSTRACT

Molecular study of congenital immune factors was conducted in 45 patients with inflammatory diseases of maxillofacial area. The study focused on expression of cationic low-molecular peptides α- and Β-defensines in oral mucosa. These peptides are involved in antibacterial activity and regulation of immune reactions. The results showed 20-fold increase in defensines expression in oral mucosa of patients with inflammatory diseases of maxillofacial area when compared to control group (12 healthy individuals) thus proving an important role they play in the development of the disease. Therapy with cytokines may be indicated in these cases.

ABSTRACT

AIM: To study the influence of the COX inhibitor--lornoxicam (LX)--on Toll-like receptor (TLR)-mediated production of proinflammatory and anti-inflammatory cytokines by peripheral blood mononuclear cells (PBMC) from healthy subjects and patients with acute pancreatitis (AP) in vitro. MATERIALS AND METHODS: Cytokine production by PBMC of healthy donors was stimulated by TLR1/2 ligand peptidoglycan (PG) and TLR4 ligand lypopolysaccharide (LPS) in presence of LX. Levels of cyotokines (IL-1beta, IL-6, IL-8, IL-10, IL-12, and TNFalpha) were measured by ELISA. Group of patients with acute pancreatitis of toxic etiology included 11 subjects: patients from main group received combined therapy supplemented with NSAID from the oxicam class--LX; patients who received only standard basic treatment formed comparison group. RESULTS: It was found that in vitro LX inhibits production of both proinflammatory and anti-inflammatory cytokines by PBMC of healthy subjects mediated by ligands of TLR1/2 and TLR4. Maximal inhibitory effect of LX was observed when cytokine production was induced through TLR1/2. Patients with AP demonstrated increased production of TNFalpha induced by TLR1/2 and TLR4 ligands. CONCLUSION: LX inhibits TLR-mediated production of both proinflammatory (IL-1, IL-6, IL-8, IL-12, TNFalpha) and anti-inflammatory (IL-10) cytokinesby PBMC of healthy subjects in vitro. Treatment with LX in patients with AP results in diminished effector function of TLR1/2 and TLR4 already during 1st day of the illness and normalization of these indices by 6th day.
