ABSTRACT

BACKGROUND: Psoriasis is a chronic inflammatory skin disease with manifestations that go beyond the visual manifestation, and include psychological aspects. Some mental disorders or personality traits in psoriasis patients have also been highlighted, such as a negative or problematic attitude towards life, impulsive or avoidant behavior, and lower satisfaction with life. The aim of our cross-sectional study was to explore the associations between adult attachment, temperament, and quality of life of patients with psoriasis. METHODS: A sample of 75 patients with psoriasis was evaluated with the Attachment Style Questionnaire (ASQ) to study adult attachment, the Temperament Evaluation of Memphis, Pisa, and San Diego Auto-questionnaire (TEMPS-A) to study temperament traits, and the Dermatology Life Quality Index (DLQI) to study the impact of dermatological diseases on patients' lives. RESULTS: Depressive, cyclothymic, and irritable temperaments were found to be significantly positively associated with a need for approval and preoccupation with relationships subscales of the ASQ. The severity of skin disease effect on the patient's life was higher in women than in men. Moreover, a statistically significant effect of the need for approval subscale of the ASQ was found. The positive correlation between the severity of skin disease effect on the patient's life with a need for approval was statistically significant and stronger in women than in men. CONCLUSIONS: A better understanding of the impact of mental comorbidities on psoriasis and vice versa places an ever-greater responsibility on dermatologists involved in the management of psoriasis to recognize these problems and collaborate with psychologists and psychiatrists to help these patients.

ABSTRACT

Immunometabolism has been demonstrated to control immune tolerance and the pathogenic events leading to autoimmunity. Compelling experimental evidence also suggests that intracellular metabolic programs influence differentiation, phenotype, proliferation, and effector functions of anti-inflammatory CD4+CD25+Foxp3+ regulatory T (Treg) cells. Indeed, alterations in intracellular metabolism associate with quantitative and qualitative impairments of Treg cells in several pathological conditions. In this review, we summarize the most recent advances linking how metabolic pathways control Treg cell homeostasis and their alterations occurring in autoimmunity. Also, we analyze how metabolic manipulations could be employed to restore Treg cell frequency and function with the aim to create novel therapeutic opportunities to halt immune-mediated disorders.

ABSTRACT

INTRODUCTION AND OBJECTIVES: Ischemic heart disease is the single most common cause of death in Europe. Mortality in patients presenting with ST-elevation myocardial infarction (STEMI) is associated with many factors, one of which is the time delay to treatment. The purpose of this work is to analyze the coronary pathway in our region in terms of timing, taking into consideration the place of first medical contact (FMC). METHODS: Consecutive patients admitted to our center with STEMI to undergo percutaneous coronary intervention (PCI) between 2013 and 2022 were analyzed. Age, gender, and time delays were collected. Analysis was performed with IBM SPSS version 28 for a significance level of 0.05. RESULTS: We found that non-PCI centers had a significantly greater FMC to diagnosis delay and diagnosis to wire delay compared to other places of origin. Only 2.2% of patients met the 10-min FMC to diagnosis target; 44.8% met the target of 90 min from diagnosis to wire in transferred patients, while 40.6% met the 60-min target for patients admitted to a PCI center. Median patient, electrocardiogram (ECG) and logistic delays are 92.0±146.0 min, 19.0±146.0 min and 15.5±46.3 min, respectively. CONCLUSION: A significant difference between state-of-the-art targets and reality was found, depending on the place of FMC, with the worst delays in non-PCI centers. Patient delay, ECG delay, FMC to diagnosis and logistic delay are identified as key areas in which to intervene.

ABSTRACT

The binding of a cognate antigen to T cell receptor (TCR) complex triggers a series of intracellular events controlling T cell activation, proliferation, and differentiation. Upon TCR engagement, different negative regulatory feedback mechanisms are rapidly activated to counterbalance T cell activation, thus preventing excessive signal propagation and promoting the induction of immunological self-tolerance. Both positive and negative regulatory processes are tightly controlled to ensure the effective elimination of foreign antigens while limiting surrounding tissue damage and autoimmunity. In this context, signals deriving from co-stimulatory molecules (i.e., CD80, CD86), co-inhibitory receptors (PD-1, CTLA-4), the tyrosine phosphatase CD45 and cytokines such as IL-2 synergize with TCR-derived signals to guide T cell fate and differentiation. The balance of these mechanisms is also crucial for the generation of CD4+ Foxp3+ regulatory T cells, a cellular subset involved in the control of immunological self-tolerance. This review provides an overview of the most relevant pathways induced by TCR activation combined with those derived from co-stimulatory and co-inhibitory molecules implicated in the cell-intrinsic modulation of T cell activation. In addition to the latter, we dissected mechanisms responsible for T cell-mediated suppression of immune cell activation through regulatory T cell generation, homeostasis, and effector functions. We also discuss how imbalanced signaling derived from TCR and accessory molecules can contribute to autoimmune disease pathogenesis.

ABSTRACT

Introduction: Urbanization processes are constantly increasing, and most of the European population currently live in urban areas. Nevertheless, evidence is consistent in highlighting the positive association between nature exposure and human wellbeing, although individual differences might affect this association. Methods: The present study aimed to investigate the association among nature connectedness, conceptualized as Love and Care for Nature, place identity, and physical wellbeing, via restorativeness and positive and negative affect. A total of 312 visitors of an urban green area (i.e., Milan's Parco Nord) participated in the study. They completed an anonymous questionnaire. Results: Findings showed that nature connectedness and place identity positively affect physical wellbeing, via restorativeness and positive affect, but not through the negative ones. Discussion: Results highlight the importance of the joint role of exposure to nature and individual differences in promoting wellbeing. This study offers implications for interventions aimed at enhancing individuals' health through exposure to nature. Limitations of the study and future research developments are discussed.

ABSTRACT

Climate change is a reality that can no longer be ignored, so much so that combating climate change and its impact is one of the main goals of the UN Agenda 2030. Youths, albeit the main victims of climate change, are often excluded from decision-making processes on sustainable actions. More and more young people are joining collective pro-environmental movements, raising their voices against the current inadequate sustainable policies and claiming to be the main actors of change. However, pro-environmental collective actions are often judged negatively by public opinion, diminishing their effectiveness and potentially impacting youth participation. In light of this, it is critical to understand the individual, contextual and relational aspects that lead young people to engage with these movements. The present study aimed to systematically review the existing literature on factors that might promote youth participation in pro-environmental movements. According to the PRISMA guidelines, we conducted a literature search of three databases (PsycINFO, ProQuest, and SCOPUS). Moreover, we deepened our research by focusing on two relevant theoretical models on collective actions, the Social Identity Model of Collective Action and the Social Identity Model of Pro-Environmental Action. After the screening and the eligibility phases, 11 articles (12 studies) were included. Most of the selected studies adopted a cross-sectional quantitative design. The results revealed individual and relational factors involved in promoting youths' involvement in pro-environmental movements. To the aim of deepening young people's pro-environmental activism, findings highlighted the need to consider personal and social drivers together. Limitations of the study, future directions, and practical implications are discussed.

ABSTRACT

We are launching a series to celebrate the 40th anniversary of the first issue of Molecular Biology and Evolution. In 2024, we will publish virtual issues containing selected papers published in the Society for Molecular Biology and Evolution journals, Molecular Biology and Evolution and Genome Biology and Evolution. Each virtual issue will be accompanied by a perspective that highlights the historic and contemporary contributions of our journals to a specific topic in molecular evolution. This perspective, the first in the series, presents an account of the broad array of methods that have been published in the Society for Molecular Biology and Evolution journals, including methods to infer phylogenies, to test hypotheses in a phylogenetic framework, and to infer population genetic processes. We also mention many of the software implementations that make methods tractable for empiricists. In short, the Society for Molecular Biology and Evolution community has much to celebrate after four decades of publishing high-quality science including numerous important inferential methods.

ABSTRACT

Recently, many studies have addressed the performance of phylogenetic tree-building methods (maximum parsimony, maximum likelihood, and Bayesian inference), focusing primarily on simulated data. However, for discrete morphological data, there is no consensus yet on which methods recover the phylogeny with better performance. To address this lack of consensus, we investigate the performance of different methods using an empirical dataset for hexapods as a model. As an empirical test of performance, we applied normalized indices to effectively measure accuracy (normalized Robinson-Foulds metric, nRF) and precision, which are measured via resolution, one minus Colless' consensus fork index (1-CFI). Additionally, to further explore phylogenetic accuracy and support measures, we calculated other statistics, such as the true positive rate (statistical power) and the false positive rate (type I error), and constructed receiver operating characteristic plots to visualize the relationship between these statistics. We applied the normalized indices to the reconstructed trees from the reanalyses of an empirical discrete morphological dataset from extant Hexapoda using a well-supported phylogenomic tree as a reference. Maximum likelihood and Bayesian inference applying the k-state Markov (Mk) model (without or with a discrete gamma distribution) performed better, showing higher precision (resolution). Additionally, our results suggest that most available tree topology tests are reliable estimators of the performance measures applied in this study. Thus, we suggest that likelihood-based methods and tree topology tests should be used more often in phylogenetic tree studies based on discrete morphological characters. Our study provides a fair indication that morphological datasets have robust phylogenetic signal.

ABSTRACT

Nutrition affects all physiological processes occurring in our body, including those related to the function of the immune system; indeed, metabolism has been closely associated with the differentiation and activity of both innate and adaptive immune cells. While excessive energy intake and adiposity have been demonstrated to cause systemic inflammation, several clinical and experimental evidence show that calorie restriction (CR), not leading to malnutrition, is able to delay aging and exert potent anti-inflammatory effects in different pathological conditions. This review provides an overview of the ability of different CR-related nutritional strategies to control autoimmune, cardiovascular, and infectious diseases, as tested by preclinical studies and human clinical trials, with a specific focus on the immunological aspects of these interventions. In particular, we recapitulate the state of the art on the cellular and molecular mechanisms pertaining to immune cell metabolic rewiring, regulatory T cell expansion, and gut microbiota composition, which possibly underline the beneficial effects of CR. Although studies are still needed to fully evaluate the feasibility and efficacy of the nutritional intervention in clinical practice, the experimental observations discussed here suggest a relevant role of CR in lowering the inflammatory state in a plethora of different pathologies, thus representing a promising therapeutic strategy for the control of human health.

ABSTRACT

Work-family conflict is a prominent issue, especially in our society, where people are expected to fulfil many roles simultaneously. Work and family life demands significantly impact an individual's overall well-being, especially for women, since they typically balance caregiving for children and elderly relatives with careers. Therefore, highlighting which factors might protect women from experiencing work-family conflict is essential to enhance women's and their family's well-being. Thus, the main aim of the present study was to systematically review previous research on women's coping strategies and protective factors which can reduce the negative effects of work-family conflict. Following the PRISMA guidelines, we conducted a literature search of three databases (PubMed, PsycINFO, and Scopus). After the screening and the eligibility phases, we included a final set of 13 studies. Most of these studies adopted a cross-sectional design (N = 10), and a few adopted a longitudinal one (N = 3). Results highlighted the role of different personal (e.g., hardiness, self-esteem, locus of control) and relational factors (e.g., family and work support) that significantly reduce the negative effects of work-family conflict in women's lives. Findings, practical implications, and future research directions are discussed.

BACKGROUND: Personal values have been extensively found to be relevant variables linked to prosociality; they are desirable and trans-situational goals that serve as guiding principles in people's lives to select modes, means and actions, these reflecting what people consider relevant and worthy. Research has investigated how cultural background influences people's personal values and prosociality, but little is known about the influence of the perception of the values endorsed by significant others, namely the people belonging to the micro-relational context with whom daily interactions and exchanges are possible. Based on Schwartz's theory of basic human values, we analyzed the moderating role of the perceptions of significant others' values in the relationship between personal values and self-reported prosociality. PARTICIPANTS AND PROCEDURE: Two hundred and forty-five Italian young adults (66.9% women) aged between 18 and 30 years (M = 22.58, SD = 2.53) completed a self-report questionnaire. RESULTS: Specifically, openness to change values were a significant positive predictor of self-reported prosociality when respondents perceived low importance assigned both to openness to change and self-transcendence by significant others, whereas conservation values were a significant positive predictor of self-reported prosociality when respondents perceived low importance assigned to self-enhancement by significant others. CONCLUSIONS: Our findings show a complex interplay between personal values and perceived significant others' values in shaping young adults' self-reported prosociality.

BACKGROUND AND AIM: Parental self-efficacy is recognized as a core element of the transition to parenthood process. It affects parental mental health, as well as children's psychosocial, neurodevelopmental and health outcomes since early infancy. Parents with higher parenting self-efficacy have a higher likelihood of engaging in quality parenting practices that support optimal self-regulation development in children. Our study evaluated the psychometric properties of the Italian TOPSE-Short Form (I-TOPSE-SF) questionnaire. METHODS: 673 Italian mothers (Mage=37.5; SD=5.7) of children aging 0 to 6 years (Mage=3.9; SD=1.7) were involved in this study. RESULTS: Initial results of Confirmatory Factor Analysis highlighted that the fit indices of the hypothesized 6-factor structure of the TOPSE weren't satisfactory. Acceptable internal consistencies for the total score and the six dimensions of the measure were observed. Evidence for convergent and divergent validity were provided. CONCLUSIONS: The I-TOPSE-SF could be a valuable tool to assess parental self-efficacy up to school age, but it is in need of further considerations about its measurement properties.

ABSTRACT

Although a consensus exists that all living turtles fall within either Pleurodira or Cryptodira clades, estimating when these lineages split is still under debate. Most molecular studies date the split in the Triassic Period, whereas a Jurassic age is unanimous among morphological studies. Each hypothesis implies different paleobiogeographical scenarios to explain early turtle evolution. Here we explored the rich turtle fossil record with the Fossilized Birth-Death (FBD) and the traditional node dating (ND) methods using complete mitochondrial genomes (147 taxa) and a set of nuclear orthologs with over 10 million bp (25 taxa) to date the major splits in Testudines. Our results support an Early Jurassic split (191-182 Ma) for the crown Testudines with great consistency across different dating methods and datasets, with a narrow confidence interval. This result is independently supported by the oldest fossils of Testudines that postdate the Middle Jurassic (174 Ma), which were not used for calibration in this study. This age coincides with the Pangaea fragmentation and the formation of saltwater barriers such as the Atlantic Ocean and the Turgai Strait, supporting that diversification in Testudines was triggered by vicariance. Our ages of the splits in Pleurodira coincide with the geologic events of the Late Jurassic and Early Cretaceous. Conversely, the early Cryptodira radiation remained in Laurasia, and its diversification ensued as all its major lineages expanded their distribution into every continent during the Cenozoic. We provide the first detailed hypothesis of the evolution of Cryptodira in the Southern Hemisphere, in which our time estimates are correlated with each contact between landmasses derived from Gondwana and Laurasia. Although most South American Cryptodira arrived through the Great American Biotic Interchange, our results indicate that the Chelonoidis ancestor probably arrived from Africa through the chain islands of the South Atlantic during the Paleogene. Together, the presence of ancient turtle diversity and the vital role that turtles occupy in marine and terrestrial ecosystems underline South America as a chief area for conservation.

ABSTRACT

The aim of the study was to assess which kill site characteristics were selected by a lone wolf living in a protected Mediterranean coastal area near the city of Pisa, Italy, where both wild and domestic ungulates were available as potential prey. Between 2017 and 2019, we monitored the wolf's predatory behaviour through a combination of camera trapping and active search for kill sites and prey carcasses. The main prey found was the fallow deer (n = 82); only two wild boars and no domestic ungulates were found preyed upon. The features and habitat of kill sites were modelled to test for selection by the wolf. The habitat type of kill site was composed of meadows and pastures (89.3%), woods (7.3%), degraded coastal areas (1.9%), roads and rivers (1.1%), and marshes (0.5%). We calculated their distance from landscape features and ran a binomial generalised linear model to test the influence of such landscape variables. The distance of kill sites from landscape elements was significantly different from random control sites, and a positive selection for fences was found. In fact, the wolf pushed fallow deer towards a fence to constrain them and prevent them from escaping. We also analysed the body condition of predated fallow deer as a percentage of fat content in the bone marrow of the hind legs. Our results revealed the selection of the lone wolf for deer in good body condition. This is a possible outcome of the habitat selection shown by fallow deer in the study area, where fenced open pastures are the richest in trophic resources; therefore, our findings suggest a high efficacy for the lone wolf hunting strategy, but also the adoption of a high risk feeding strategy by deer. This study suggests that a lone predator can take advantage of human infrastructures to maximise its predatory effectiveness.

Prejudice against immigrants is a relevant research topic within social psychology. Researchers identified several individual variables affecting anti-immigrant prejudice, such as morality and personality. However, until now, prejudice has never been studied in relation to kindness, which might be a significant protective factor against prejudice. Based on Kohlberg's theory of moral judgement, four stage dimensions of kindness were identified, from egocentric to authentic kindness (i.e., a means for social progress and improvement). This study aims to explore the relationship between the four kindness dimensions and blatant and subtle prejudice against immigrants in adolescence, by also considering the moderating role of adolescents' sex. It involved 215 Italian participants (77% girls), who were asked to fill in a self-report questionnaire. Results showed that boys scored higher on egocentric kindness than girls, but no sex differences emerged for prejudice. Egocentric and extrinsically motivated kindness appeared to be risk factors for prejudice, whereas the most authentic form of kindness was a protective factor. In addition, adolescents' sex moderated the relationship between egocentric kindness and blatant prejudice, whereby this association was stronger for boys. The implications of these findings, the study's limitations, and suggestions for future research are discussed.

ABSTRACT

Anthropic areas play a pivot role for main wolf conservation challenges. Wolf presence in the higher Pisan hills has been well documented while wolf settlement in the lower Pisan hills is still uncertain. In this study, long-term information on wolf presence in a highly anthropic area of the lower Pisan hills was collected by using non-invasive monitoring techniques. Furthermore, both the relationship of this predator with human activity and the impact of hunting on wolf presence have been investigated. The results obtained indicate the presence of a stable and reproductive wolf pack composed by both Italian wolf and hybrids individuals in the municipalities of Crespina Lorenzana and Casciana Terme Lari. A high impact of wolf on livestock was recorded in this area since no prevention systems were adopted by farmers. Wolf appears not to have a negative impact on wild boar population. Similarly, wild boar drive hunting does not appear to affect the wolf pack presence in the area. Thereby wolf may play a key role as controller of wild population. Prevention strategies improvement becomes instrumental to promote wolf-human coexistence. Further investigation to monitor pack hybridization level and turnover and to assess the impact of packs on wild population and livestock in anthropic areas is desirable.

ABSTRACT

Compelling experimental evidence links immunity and metabolism. In this perspective, we propose forkhead-box-P3 (FoxP3)+CD4+CD25+ regulatory T (Treg) cells as key metabolic sensors controlling the immunological state in response to their intrinsic capacity to perceive nutritional changes. Treg cell high anabolic state in vivo, residency in metabolically crucial districts, and recirculation between lymphoid and non-lymphoid sites enable them to recognize the metabolic cues and adapt their intracellular metabolism and anti-inflammatory function at the paracrine and systemic levels. As privileged regulators at the interface between neuroendocrine and immune systems, the role of Treg cells in maintaining metabolic homeostasis makes these cells promising targets of therapeutic strategies aimed at restoring organismal homeostasis not only in autoimmune but also metabolic disorders.

BACKGROUND: Children with Attention-Deficit/Hyperactivity (ADHD) and/or Developmental Coordination Disorder (DCD) show high rates of motor difficulties in daily activities. This study aimed to examine the validity and reliability of the Developmental Coordination Disorder Questionnaire (DCDQ) in identifying motor characteristics in daily activities, differentiating children with ADHD-only, with DCD-only and with ADHD and DCD comorbidity. METHODS: Thirty-three children with ADHD-only, 30 with DCD-only, 33 with ADHD/DCD, and 35 controls participated to the study. Diagnosis satisfied DSM-5 criteria for ADHD and DCD. The DCDQ was administered to all children; moreover, the association between DCDQ scores and ADHD symptoms, measured by SNAP-IV, and motor coordination severity, measured by M-ABC subscales, was examined. RESULTS: The DCDQ subscale scores were significantly lower in all clinical groups than in controls, but only minimal differences were found between the clinical groups. Principal Component Analysis (PCA) of DCDQ identified five components showing the best adaptability and accounting for 71% of the variance. Both ADHD-only and ADHD/DCD achieved a better performance than DCD-only in the "Motor control in running/jumping". Conversely, children with DCD-only performed better in "General coordination". ADHD-only reached better mean scores than ADHD/DCD in "Motor control with the ball" and, than DCD, in "Planning". Inattention, hyperactivity-impulsivity at SNAP-IV, and balance at M-ABC, represented the main predictors for ADHD-only, ADHD/DCD and DCD-only, respectively. CONCLUSIONS: DCDQ may represent a useful measure to detect the different areas of difficulties in children with neurodevelopmental disorders and provides suggestions for identifying distinct altered processes underlying ADHD and DCD, alone or in comorbidity.
