ABSTRACT
Eimeria are ubiquitous parasites and eimeriosis treatment is based on coccidiostats or coccidicides used prophylactically, metaphylactically, or therapeutically. The long-term efficacy of toltrazuril (TZR, 15 mg/kg) against experimentally infected naïve calves was investigated. Seven groups (six treated and one control) of six animals each were formed. Animals of each group received a single TZR prophylactic oral treatment on days - 42, - 35, - 28, - 21, - 14, and - 7 before the challenge with infectious sporulated oocysts of Eimeria spp. (100,000 oocysts: 59.5% E. zuernii, 38.1% E. bovis, 1.2%, E. alabamensis, and 1.2% E. ellipsoidalis). The long-term efficacy was assessed based on Eimeria spp. oocyst excretion by fecal oocyst counts. Three calves from the control group presented diarrhea with blood, which was not observed in animals belonging to the treatment groups. The TZR achieved efficacy greater than 95.0% up to 14 and 7 days. This formulation showed efficacy above 95% for 7 to 14 days, between 82 and 84% for 21 to 28 days and between 50 and 64% for 35 to 42 days.

Avian coccidiosis continues to be one of the costliest diseases of commercial poultry. Understanding the epidemiology of Eimeria species in poultry flocks and the resistance profile to common anticoccidials is important to design effective disease prevention and control strategies. This study examined litter samples to estimate the prevalence and distribution of Eimeria species among broiler farms in 4 geographic regions of Colombia. A total of 245 litter samples were collected from 194 broiler farms across representative regions of poultry production between March and August 2019. The litter samples were processed for oocysts enumeration and speciation after sporulation. End-point polymerase chain reaction (PCR) analysis was conducted to confirm the presence of Eimeria species. Anticoccidial sensitivity was determined with 160 Ross AP males in 5 treatment groups: noninfected, nonmedicated control (NNC), infected, nonmedicated control (INC), infected salinomycin treated (SAL, dose: 66 ppm), infected diclazuril treated (DIC, dose: 1 ppm), and infected methylbenzocuate-Clopidol treated (MET.CLO, dose: 100 ppm), All birds were orally inoculated with 1 × 106 sporulated oocysts using a 1 mL syringe, except for the NNC- group who received 1ml of water.Eimeria spp. were found in 236 (96.3%) out of 245 individual houses, representing 180 (92.8%) out of 194 farms. Eimeria acervulina was the most prevalent species (35.0%) followed by Eimeria tenella (30.9%), Eimeria maxima (20.4%), and other Eimeria spp. (13.6%). However, mixed species infections were common, with the most prevalent combination being mixtures of E. acervulina, E. maxima, E. tenella, and other species in 31.4% of the Eimeria-positive samples. PCR analysis identified E. acervulina, E. maxima, E. tenella, Eimeria necatrix, Eimeria mitis, and Eimeria praecox with variable prevalence across farms and regions. Anticoccidial sensitivity testing of strains of Eimeria isolated from 1 region, no treatment difference (P > 0.05) was observed in final weight (BW), weight gain (BWG) or feed conversion (FCR). For the global resistance index (GI) classified SAL and MET.CLO as good efficacy (85.79 and 85.49, respectively) and DIC as limited efficacy (74.52%). These results demonstrate the ubiquitous nature of Eimeria spp. and identifies the current state of sensitivity to commonly used anticoccidials in a region of poultry importance for Colombia.

Rotation with different active ingredients is among the most effective and recommended strategies to preserve the efficacy of anticoccidial drugs and reduce the emergence of resistance. Tools such as anticoccidial sensitivity tests (ASTs) are ideally used to make rational rotation programs and bring benefits to production. The objective of this study was to evaluate the sensitivity of E. acervulina (EA) and E. maxima (EM) from 3 different regions in Brazil, by using four ASTs. Feces samples weighing 6 to 7 kg were collected in the regions of São Paulo, Paraná, and Minas Gerais. Prevalent oocysts from feces were filtered, identified, and quantified to conduct 2 ASTs with EA and 2 with EM. The same experimental design was used in every AST (4 replicates per treatment, with 6 birds each, for a total of 240 birds). Treatment groups were a nonchallenged and nonmedicated control group (T1), a challenged and nonmedicated control group (T2), and the other groups challenged and treated with the following compounds: lasalocid (90 ppm - T3), maduramycin (6 ppm - T4), decoquinate (30 ppm - T5), nicarbazin+semduramicin (66 ppm - T6), monensin (110 ppm - T7), salinomycin (66 ppm - T8), narasin+nicarbazin (100 ppm - T9), and nicarbazin (125 ppm - T10). At the end of each AST (20 d), the percent change (delta value) between the treated group (T3 to T10) and the control group (T2) was calculated for the following variables: body weight gain, feed conversion ratio, lesion score, and an indicator of percentage of optimal anticoccidial activity (POAA) that included T2. Different sensitivity levels of EA and EM isolates could be identified. As a whole, drugs from T5 and T3 groups showed higher delta values when compared to other compounds, whereas the lowest sensitivity levels of these isolates were observed in groups T4 and T7. Despite some limiting factors, ASTs can be a good tool for strategic selection of anticoccidial drugs in order to maintain efficacy and extend the lifespan of these molecules.

The incidence of coccidiosis was determined by oocysts count in feces (OoPG), testing the interactions between treatment, age of birds and the number of flocks housed. Samples were selected to identify, through the Multiplex PCR, species of the genus Eimeria. circulating in the shed. The results showed that in litter where fermentation was adopted, the efficiency to control the sporulation of Eimeria spp. was significantly (p <0.05) higher than in the other treatments. The use of the Multiplex PCR technique requires improvements in the preparation of samples for the extraction of genetic material.

Eimeria spp. infection is one of the main diseases that affect young cattle in beef and dairy herds, leading to economic losses and compromising the performance and growth of animals. This study aimed to evaluate the effectiveness of different protocols of metaphylactic treatment with 5% toltrazuril (15 mg/kg) in naturally infected, extensively reared beef calves from birth to 1 year of age. We used 92 calves aged up to 2 weeks of life, initially divided into two experimental groups: treated and control. The animals were evaluated fortnightly with individual count of oocysts per gram of feces (OPG) tests until weaning; thereafter, they were again divided into four subgroups: I-treated at both birth and weaning; II-treated at birth but not treated at weaning; III-not treated at birth but treated at weaning; and IV-neither treated at birth nor at weaning (control). During the experimental period, the animals were evaluated for weight gain, excretion of oocysts, and presence of pathogenic species of Eimeria spp. In the first stage of the study, less oocysts were excreted in the treated group for up to 75 days and at 150 days of age (p < 0.05). In the final stage of the study, significant differences occur between groups I and II, I and IV, II and III, and III and IV (p = 0.3512), in the collection 20 days after treatment. Eimeria bovis was the most frequently identified species during the experimental period. Moreover, there was a positive correlation between the presence of diarrhea in the calves and the number of oocysts excreted, treated group (p = 0.0092) and untreated group (p = 0.2). There was no significant difference in weight gain of animals in any of the study periods (p > 0.05).

Resumo O objetivo deste trabalho foi avaliar a eficiência dos manejos sanitários aplicados à cama de aviários comerciais para o controle da salmonelose nas criações de frango de corte, na região Oeste do Paraná, e verificar seu efeito no controle da coccidiose. Foram selecionados três tratamentos de cama: a fermentação associada à calagem (FL); calagem (L) e fermentação (F). As amostras de fezes foram coletadas de 18 aviários, com um delineamento experimental inteiramente casualizado, testando-se diferentes áreas de coleta. As coletas foram realizadas a partir do 5° dia do alojamento e se repetiram semanalmente até o 40° dia, durante cinco lotes sucessivos. A incidência da parasitose foi determinada pela contagem de oocistos por grama de fezes (OoPG), testando as interações entre tratamento, idade das aves e número de lotes alojados. Amostras de fezes foram selecionadas para identificar os oocistos esperulados das espécies do gênero Eimeria circulantes no galpão, por meio do Multiplex PCR. Observou-se que a fermentação foi significativamente (p<0,05) mais eficiente, comparando-se o mesmo lote ao longo de todos os observados. A utilização da técnica de PCR Multiplex, precisa ser padronizada para amostras de campo.

Toxoplasma gondii is a protozoan that has great genetic diversity and is prevalent worldwide. In 2018, an outbreak of toxoplasmosis occurred in Santa Maria, Brazil, which was considered the largest outbreak ever described in the world. This paper describes the isolation and molecular characterization of Toxoplasma gondii from the placenta of two pregnant women with acute toxoplasmosis who had live births and were receiving treatment for toxoplasmosis during the outbreak. For this, placental tissue samples from two patients underwent isolation by mice bioassay, conventional PCR and genotyping using PCR-RFLP with twelve markers. Both samples were positive in isolation in mice. The isolate was lethal to mice, suggesting high virulence. In addition, the samples were positive in conventional PCR and isolates submitted to PCR-RFLP genotyping presented an atypical genotype, which had never been described before. This research contributes to the elucidation of this great outbreak in Brazil.

The current study aimed to determine the anti-Eimeria efficacy of an extract of grapefruit peels (GF) and commercial naringenin (NAR) in naturally-infected lambs, as well as the influence of these flavonoids on the oxidative status during ovine coccidiosis. Pharmacokinetic profiles were also determined. Extracts were administered per os to Eimeria naturally infected growing lambs during 90 consecutive days. The commercial anticoccidial drug toltrazuril (TTZ) was included in this trial as a standard. Twenty-four lambs were divided into four groups: NAR, lambs given a daily dose of 5mg of a commercial naringenin extract of 98% higher purity per kg body weight; GF, lambs that recived a daily dose of 5mg of ethanolic extract of grapefruit peels per kg body weight; TTZ, lambs treated with 20mg of toltrazuril/kg body weight on days 0 and 15 of the experiment; and CTRL, untreated lambs that received daily dose of 30ml of water. Daily doses of GF and NAR were dissolved in 30ml of water and orally given to animals; whereas toltrazuril was administered as a single dose of an undiluted suspension to lambs of the TTZ group. The CTRL group received 30ml of water; as well as the TTZ group for the period after the single dose administration. Fecal and serum samples were collected from all lambs. Anticoccidial efficacy was estimated by coprological techniques. Generation of nitric oxide levels and the antioxidant capacity of the experimental compounds were determined by the Griess and ABTS assays, respectively. The pharmacokinetic parameters of NAR and the GF extract were obtained. On day 30 post-ingestion, anticoccidial efficacy was 91.76% (NAR) and 89.65% (GF); whereas 99.63% of efficacy was achieved with TTZ 15days after treatment. NAR, GF and TTZ significantly reduced oxidative stress in infected animals. The mean daily weight gain for each group was 122g (NAR), 122g (GF), 143g (TTZ) and 98g (CTRL). Following the oral administration of NAR and GF, values in plasma approached maximum concentrations within 2.1 to 2.5h. In conclusion, the administration of NAR and the GF extract reduced Eimeria oocyst output, oxidative stress and promoted higher mean daily weight gains in infected lambs.

Resumo O objetivo deste estudo foi avaliar a eficácia de uma formulação experimental de toltrazuril 7,5% + Trimix™ em cordeiros mantidos em pastagem com infecção naturalmente adquirida por Eimeria spp. e, em outro teste, a eficácia comparativa entre lasalocida sódica e toltrazuril 7,5% + Trimix™ em ovinos recém-desmamados, naturalmente infectados com Eimeria spp. em condições de confinamento. No primeiro experimento, 30 cordeiros lactantes foram divididos em dois grupos: A - tratados com toltrazuril 7,5% + Trimix™; e B - controle. No experimento 2, 30 ovinos desmamados foram divididos em três grupos: I - tratados com toltrazuril 7,5% + Trimix™; II - tratados com lasalocida sódica; e III - controle. O grupo A (tratado) obteve uma eficácia de 90, 99,4 e 87,3% nos dias 5, 10 e 20, respectivamente. O grupo I teve eficácia de 98,2, 92,6 e 94,5%, enquanto o grupo II teve uma eficácia de 72,7, 81,6 e 95.9% nos dias 7, 21 e 42, respectivamente. Foram identificadas oito espécies de Eimeria sendo E. ovinoidalis a mais comum. O tratamento com a formulação de toltrazuril 7,5% + Trimix™ foi eficaz contra Eimeria spp. em cordeiros em lactação em condições de campo e em ovinos desmamados em confinamento.

Effectiveness of an oral combination of toltrazuril and iron dextran (Baycox(®) Iron) to maintain weaning weight by preventing coccidiosis caused by Isospora suis and iron-deficiency anaemia in neonatal piglets was investigated on three commercial pig farms with a history of coccidiosis: two in Mexico and one in Brazil. On day (SD) 2 of life, piglets were randomised within litter by bodyweight to treatment or control group. On SD 3 piglets allocated to the control group (CG) each received 1 mL Baycox(®), containing 50 mg/mL toltrazuril orally and commercially available iron (200 mg/piglet) by intramuscular injection. Piglets allocated to the treatment group (TG) each received 1 mL toltrazuril and iron combination orally (Baycox(®) Iron) containing 50 mg/mL toltrazuril and 228 mg iron as iron dextran. All piglets had access to creep feed. 6493 piglets completed the study. Bodyweight at weaning on SD 21 of piglets treated with the oral toltrazuril and iron combination was confirmed to be non-inferior to the control treatment with <1 % difference between group mean body weights. Faecal samples from at least 10 % of litters on SD 14 demonstrated control of coccidiosis. Haemoglobin levels on SD 21 were lower in the oral toltrazuril and iron combination treated piglets compared to control levels but above minimum haemoglobin levels to maintain health. There was no difference in mortality between the two groups. This large scale field evaluation clearly demonstrated the effectiveness of a combination of oral toltrazuril and iron (Baycox(®) Iron) in maintaining body weight at weaning compared to conventional treatment. The combination was effective in preventing coccidiosis and anaemia and thus provides a valuable alternative that reduces stressful events in neonatal piglets. There were no product related adverse events.

Blood sample and placenta were taken from a 37-week pregnant woman; serologic results indicated acute toxoplasmosis. Placenta was inoculated into mice. Seropositive mice were sacrificed and tissue cysts from brain were inoculated into new mice. Specific DNA was detected by PCR, and the isolate was characterized as Type II by nPCR-RFLP for nSAG2, SAG3, BTUB, GRA6, c29-2, c22-8, L358, PK1 and Apico markers. This is the first isolation and molecular characterization of Toxoplasma gondii from humans in Argentina.

BACKGROUND: Control programs have been executed in an attempt to reduce vertical transmission and the severity of congenital infection in regions with a high incidence of toxoplasmosis in pregnant women. We aimed to evaluate whether treatment of pregnant women with spiramycin associated with a lack of monitoring for toxoplasmosis seroconversion affects the prognosis of patients. METHODS: We performed a prospective cohort study with 246 newborns (NB) at risk for congenital toxoplasmosis in Goiânia (Brazil) between October 2003 and October 2011. We analyzed the efficacy of maternal treatment with spiramycin. RESULTS: A total of 40.7% (66/162) of the neonates were born seriously infected. Vertical transmission associated with reactivation during pregnancy occurred in 5.5% (9/162) of the NB, with one showing severe infection (systemic). The presence of specific immunoglobulins (fetal IgM and NB IgA) suggested the worst prognosis. Treatment of pregnant women by spiramycin resulted in reduced vertical transmission. When infected pregnant women did not undergo proper treatment, the risk of severe infection (neural-optical) in NB was significantly increased. Fetal IgM was associated with ocular impairment in 48.0% (12/25) of the fetuses and neonatal IgA-specific was related to the neuro-ophthalmologic and systemic forms of the disease. When acute toxoplasmosis was identified in the postpartum period, a lack of monitoring of seronegative pregnant women resulted in a higher risk of severe congenital infection. CONCLUSION: Treatment of pregnant women with spiramycin reduces the possibility of transmission of infection to the fetus. However, a lack of proper treatment is associated with the onset of the neural-optical form of congenital infection. Primary preventive measures should be increased for all pregnant women during the prenatal period and secondary prophylaxis through surveillance of seroconversion in seronegative pregnant woman should be introduced to reduce the severity of congenital infection in the environment.

Toxoplasma gondii is an intracellular parasite widely distributed in nature. Infection is asymptomatic in immunocompetent individuals; however, various clinical manifestations may occur in immunocompromised individuals. Although there are medications for the treatment of toxoplasmosis, such as pyrimethamine and sulfonamide, they cannot always be used due to adverse reactions or to therapeutic failures related to intolerance or malabsorption of drugs and to parasite drug resistance. In recent years, the search for new antimicrobial agents derived from plants has intensified because a quarter of synthetic drugs that are currently prescribed have been isolated from a plant source, demonstrating that natural products are important in the development of new drugs. A systematic literature search was conducted to evaluate the use of natural products as an alternative for the treatment of T. gondii infection. The search was conducted for the 2000-2014 period in Science Direct, Scopus, PubMed, EMBASE, and SciELO databases, using the following MeSH terms: anti-Toxoplasma activity, toxoplasma AND natural products, toxoplasma AND plant extracts. Ethnobotanical and experimental evidence (in vitro/in vivo) was found supporting the use of natural products as a source for the discovery of new therapies against T. gondii.

The weight gain performance and oocysts reduction, as response to the metaphylactic treatment with Diclazuril 0.25% at the start of a coccidiosis outbreak, was studied by a cases-controls transverse study. Fifty-eight suckling calves of approximately 90 days old were randomly selected from an infected beef herd on extensive farming. The calves were weighted and individual faecal samples were taken for oocyst per gram count (OPG). Out of those, 29 were drenched with 1 mg kg(-1) of Diclazuril in one oral dose (group T), since the other 29 remained as control group (group C). Samples for OPG and weights were measured again at days 7 and 21 after treatment, respectively. Later, the groups were divided (by the median) in higher or lower OPG counts, and were compared in the same way, in order to remove those without apparent infection (lower OPG counts). The faecal oocysts reduction reached 99.5% (p<0.0001), for the treated group. Along the three weeks of study, an increment of 2.65 kg in 21 days (125 g day(-1), p=0.036), was seen in treated group respect to controls, but this difference increased to 3.94 kg in 21 days (p<0.0001), when only the calves with higher OPG were taken into account. These results highlight the magnitude of the subclinical impact of coccidiosis, biased by the individual susceptibility to infection, which leads to get heavier infections and express higher oocysts output.

However, there are few studies on this parasite in the State of Rio Grande do Norte. The aim of this study was to evaluate the action of decoquinate, added to mineral salt, for controlling Eimeria infection in lambs, and to identify which species are infecting sheep in the eastern region of the state. This study was carried out from August 2009 to January 2010, and used 76 animals. These were divided into two treatment groups: one with common mineral salt, and the other with mineral salt enriched with 6% micronized decoquinate. Fecal samples and body weight measurements were taken every 14 days for parasitological diagnosis, weight gain follow-up and quantitative analysis. The study showed that there was a significant difference in OPG only at the 7th collection, but no significant difference in weight gain. The Eimeria species found were E. ahsata. E. crandallis. E. granulosa. E. intrincata. E. ovina. E. faurei. E. ovinoidalis. E. pallida and E. parva. It was concluded that addition of decoquinate to mineral salt gave rise to lower oocyst elimination, thus favoring eimeriosis control in sheep.

Porém, poucos são os estudos com esse parasito no Estado do Rio Grande do Norte. O objetivo deste trabalho foi avaliar a ação do decoquinato, adicionado ao sal mineral, no controle da infecção causada por parasitas do gênero Eimeria em cordeiros, e identificar quais as espécies infectam ovinos na região leste Potiguar. O trabalho foi desenvolvido entre agosto de 2009 e janeiro de 2010, e foram usados 76 animais, distribuídos em dois tratamentos, um com sal mineral comum e o outro com sal mineral enriquecido com decoquinato a 6% micronizado. Amostras fecais e pesagens dos animais foram feitas a cada 14 dias para o diagnóstico parasitológico, acompanhamento do ganho de peso ponderal e análise quantitativa. O estudo evidenciou que houve diferença significativa na redução do OoPG apenas na 7º semana de experimento, mas não houve diferenças significativas para ganho de peso dos animais. As espécies encontradas foram E. ahsata. E. crandallis. E. granulosa. E. intrincata. E. ovina. E. faurei. E. ovinoidalis. E. pallida . E. parva. Conclui-se que a adição de decoquinato ao sal mineral propiciou uma menor eliminação de oocistos favorecendo o controle da eimeriose ovina.

This study aimed to investigate the association of salinomycin and semduramicin, in different doses, against controlled mixed infection of Eimeria acervulina, E. maxima and E. tenella in broiler chickens. Eight hundred birds were divided into 5 groups (T1: not medicated feed; T2: 30 ppm of salinomycin and 12.5 ppm of semduramicin; T3: 30 ppm of salinomycin and 15 ppm of semduramicin; T4: 40 ppm of salinomycin and 12,5 ppm of semduramicin and T5: 40 ppm of salinomycin and 15 ppm of semduramicin) and inoculated at 15 days of age with sporulated oocysts of E. acervulina, E. maxima and E. tenella in a mixed suspension, through the feed. Performance data and lesion scores were recorded. All treated groups showed statistically better cumulative weight gain at 21 days old. At 35 days old only the T3 group showed significant difference. Cumulative feed conversion showed statistical difference in the groups T4 and T5. The treatment T5 was more effective in the coccidiosis control of E. tenella. T3 and T5 achieved statistical differences in the average lesion scores of the three analyzed species. The association of salinomycin and semduramicin used in lower doses than the usual, showed to be an option in the coccidiosis control in this experiment.

O presente estudo teve por objetivo investigar a associação de salinomicina e semduramicina, em diferentes doses, frente à infecção mista controlada de Eimeria acervulina, E. maxima e E. tenella em frangos de corte. Oitocentas aves foram divididas em 5 grupos (T1: ração não medicada; T2: 30 ppm de salinomicina e 12,5 ppm de semduramicina; T3: 30 ppm de salinomicina e 15 ppm de semduramicina; T4: 40 ppm de salinomicina e 12,5 ppm de semduramicina e T5: 40 ppm de salinomicina e 15 ppm de semduramicina) e inoculadas aos 15 dias de idade com oocistos esporulados de E. acervulina, E. maxima e E. tenella, em inóculo misto, via ração. Parâmetros produtivos e escore de lesões foram registrados. Todos os grupos tratados apresentaram estatisticamente melhores ganhos de peso cumulativo aos 21 dias de vida. Aos 35 dias de vida, somente o grupo T3 apresentou diferença significativa. A conversão alimentar cumulativa apresentou diferença estatística nos grupos T4 e T5. O tratamento T5 foi mais eficaz no controle de E. tenella. T3 e T5 obtiveram diferenças estatísticas no escore médio de lesão das três espécies. O uso de salinomicina, associada a semduramicina, em baixas doses demonstrou uma opção viável no controle da coccidiose neste experimento.
