ABSTRACT
Selenium is an essential micronutrient for ruminants, which participates in the optimal functioning of proteins and enzymes that can combat oxidative stress in the body; however, its toxicity is documented in different species. The objective of this work was to describe macroscopic and microscopic lesions in lambs intoxicated with selenium administered through intraruminal boluses. The main lesions at necropsy were pulmonary oedema; the myocardial surface presented multifocal pale areas; the thyroid and thymus glands were decreased in size, and areas of necrosis, haemorrhage and hyperkeratosis were observed in the reticulum and rumen. At the microscopic level, congestion, haemorrhage, oedema and hyaline membranes were observed in the lung; hepatic congestion, haemorrhage, degeneration and necrosis; degeneration and necrosis of the reticulum mucosa, as well as areas of hyperplasia and hyperkeratosis; myocardial degeneration, necrosis and fibrosis; congestion, haemorrhage, degeneration and renal tubular necrosis; thyroid follicular atrophy and thymic cortical atrophy. This study evidenced the main lesions related to selenium poisoning in lambs supplemented with the mineral through intraruminal boluses.

This study aimed to investigate the effects of meat biofortified with antioxidants and canola oil on the health of older adults through blood parameters. Eighty institutionalized older persons were divided into four groups who received the following treatments: C-control meat with 46 µg/kg of meat with selenium, 3.80 g/kg of meat with vitamin E and 0.78 g/100 g of meat with conjugated linoleic acid (CLA); A-antioxidant meat with 422 µg/kg of meat with selenium, 7.65 g/kg of meat with vitamin E and 0.85 g/100 g of meat with CLA; O-oil meat with 57 µg/kg of meat with selenium, 3.98 g/kg of meat with vitamin E and 1.27 g/100 g of meat with CLA; OA-oil and antioxidant meat with 367 µg/kg of meat with selenium, 7.78 g/kg of meat with vitamin E and 1.08 g/100 g of meat with CLA. Blood samples were collected at 0, 45 and 90 days after the start of meat intake. Older adults who consumed ANT (A and AO) meat had higher concentrations of selenium (p = 0.039), vitamin E and HDL (higher concentrations of high-density lipoprotein, p = 0.048) in their blood. This study demonstrates that the consumption of Se- and vitamin E-biofortified meat increases the concentration of these metabolites in blood from older adults.

The aim of this study was to evaluate the effects of different selenium compounds on the sperm quality of cryopreserved ram semen. Ejaculates from four rams, collected using an artificial vagina heated to 38 °C, were individually evaluated. The approved ejaculates were pooled and diluted (1:1 v:v) in Tris-egg yolk extender (20%, v/v) and separated into two control groups, one cooled for 2 h and the other for 4 h. The pooled ejaculates at the two cooling periods were supplemented with two doses (0.5 and 1 µg/mL) of organic selenium (ORG), and inorganic selenium (SeNa), each. The samples were packed in 0.25 ml straws, at a concentration of 400 × 106 sperms/mL and stored in liquid nitrogen. The straws were thawed in a water bath at 37 °C for 20 s, and the samples were subjected to sperm kinetics evaluation by Computer Assisted Semen Analysis software. Sperm membrane integrity, acrosome morphology, and mitochondrial potential were assessed. In addition, oxidative stress markers reactive oxygen species (ROS), ferric reducing antioxidant power (FRAP), thiobarbituric acid reactive species (TBARS), and glutathione peroxidase (GPx) enzyme activity) were also evaluated. No significant improvement was observed in the ram semen quality at the two cooling times. Supplementation of the freezing extender with 0.5 µg/mL ORG, subjected to 4 h cooling period, increased the sperm motility when compared with the control group at the same cooling time. In addition, the 0.5 µg/mL SeNa group, under the 2 h cooling period, showed an increase in sperm motility when compared to the control group at the same cooling period. Considering the importance of sperm motility as a fertility parameter, our study indicates that supplementation with ORG and SeNa can help improve the total motility of the cryopreserved ram semen.

INTRODUCTION: Diet-based interventions may be a culturally acceptable option to decrease mercury levels and thus prevent the adverse effects of this metal on population health. Selenium is an element present in Colombian geology that can act as a chelator, decreasing mercury concentrations in the human body. OBJECTIVE: To identify potentially useful selenium-rich foods to control the effects of mercury exposure among Afro-Colombians. MATERIALS AND METHODS: A cross-sectional study was carried out with 320 individuals from five municipalities of Chocó. They were asked about the frequency of consumption of selenium-rich foods, and their association with mercury concentrations in hair was estimated with multiple robust regression. RESULTS: Guava, whole wheat flour, strawberries, cow liver, spinach and yeast extract were the foods with higher consumption. Walnuts, whole wheat flour, and yeast extract were identified in multiple robust regression as foods to consider in future interventions. CONCLUSION: It is proposed that the banana juice, the pineapple colada, the borojó (Borojoa patinoi) sorbet, the cucas, and the enyucado are basic elements for a culturally acceptable intervention.
Introducción: Las intervenciones basadas en la dieta pueden ser una opción culturalmente aceptable para disminuir los niveles de mercurio y prevenir los efectos adversos de este metal en la salud de la población. El selenio es un elemento presente en la geología colombiana que puede actuar como quelante, disminuyendo las concentraciones de mercurio en el cuerpo humano. Objetivo: Identificar los alimentos ricos en selenio potencialmente útiles para controlar los efectos de la exposición a mercurio entre afrocolombianos. Materiales y métodos: Se realizó un estudio transversal con 320 individuos de cinco municipios del departamento del Chocó. Se les preguntó sobre la frecuencia de consumo de alimentos ricos en selenio y su asociación con las concentraciones de mercurio en cabello fue estimada con regresión múltiple robusta. Resultados: La guayaba, la harina de trigo integral, las fresas, el hígado de vaca, la espinaca y el extracto de levadura fueron los alimentos de mayor consumo. Las nueces, la harina de trigo integral y el extracto de levadura fueron identificados en regresión múltiple robusta como alimentos por considerar en futuras intervenciones. Conclusiones: Se propone que el jugo de plátano, la piña colada, el sorbete de borojó (Borojoa patinoi), las cucas, el enyucado y la sábida de plátano primitivo maduro son elementos básicos para una intervención culturalmente aceptable.

The objective of this study was to evaluate the effects of dietary supplementation with different levels of nano-selenium (NS) on growth performance, antioxidant activity, biochemical parameters, and selenium content in Landes geese. A total of 120 80-week-old healthy Landes geese (4.44±0.03 kg) were randomly assigned to three groups, each with four replicates of 10 birds. The control birds were fed a basal diet without further dietary supplementation (0.0 mg/kg of NS) and the two experimental groups were fed the basal diet supplemented with dietary NS at 0.2 or 0.4 mg/kg of feed. The results demonstrated that NS dietary supplementation had no significant effect on growth performance. Increased total superoxide dismutase activity in serum, breast muscle and liver, glutathione peroxidase level in serum and liver, and catalase in breast muscle and liver were observed for both NS supplemented groups. Additionally, reduced malondialdehyde in serum, breast muscle, and liver was detected in both NS-supplemented groups. Compared with the control, the birds fed diets supplemented with NS had lower concentrations of alanine aminotransferase, triglyceride, aspartate aminotransferase, and low-density lipoprotein cholesterol in serum, while high-density lipoprotein cholesterol was increased. Furthermore, increased selenium, especially in the liver, was found in groups with dietary supplementation of NS. These findings suggest the supplementation of NS in diets can improve antioxidant status, biochemical parameters, and tissue selenium content, although it has no significant effect on growth performance of Landes geese.

An experimental study was conducted to perform Comparative analysis of Selenium yeast with inorganic Se in broilers under heat stress. A total of 120 broilers chicks of one day were assigned to 4 groups each consisting 30 chicks fed on same basal diet but different selenium sources. The basal diet of group D1 was not supplemented with Se source (Negative control), group D2 basal diet was supplemented with inorganic selenium (Sodium selenite 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase), group D3 basal diet was supplemented with commercially available organic selenium (Seleno-methionine 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase) and group D4 basal diet was supplemented with self-developed organic selenium (Se-enriched yeast 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase). The performance parameters i.e. feed intake (FI), live body weight (BW) and FCR were not significantly (p>0.05) effected by selenium supplementation in the starter phase but were significantly (p<0.05) effected in the finisher phase. Selenium supplementation significantly (p<0.05) effected serum Se level in different supplemented groups. Higher serum Se value (58.20±0.06) was recorded in D4 group. Similarly significantly lower selenium value was recorded for D4 and higher was recorded for D1 (11.36±0.08). However lower serum Paraoxonase (PON) value was recorded for D4 (13.24±0.01) and higher for D1 (13.33±0.03). Comparatively self-developed Se enriched yeast increased the Se accumulation and improved antioxidant system. A suplementação de probióticos enriquecidos com selênio é eficaz na redução do estresse oxidativo e na manutenção da estabilidade da qualidade da carne em frangos de corte, especialmente sob estresse por calor. Um estudo experimental foi conduzido para realizar uma análise comparativa da levedura selênio com o Se inorgânico em frangos de corte sob estresse térmico. Um total de 120 pintos de um dia foi dividido em 4 grupos, cada um consistindo de 30 pintos alimentados com a mesma dieta basal, mas com diferentes fontes de selênio. The aim of the study was to evaluate whether supplementation with different levels of selenium (Se) can change the biochemical and electrolytic components of semen, causing an improvement in seminal quality in rams. Thirty animals were kept in an intensive pen system, fed with hay and commercial ration, allocated into five groups (six animals/pen) and supplemented with a mineral mixture containing Omg (G1), 5mg (G2), 10mg (G3), 15mg (G4) and 20mg (G5) of Se/kg. Each group received a different treatment every 56 days and treatments were rotated between groups following a dynamic sequence. Semen samples were collected by electroejaculation after the end of each treatment to evaluate the levels of fructose, citric acid, potassium (K), sodium (Na), calcium (Ca), Se, zinc (Zn), manganese (Mn), sulfur (S) and lead (Pb). The statistical design was a 5x5 Latin square. The different levels of Se supplementation evaluated maintained the concentrations of electrolytes and minerals in the semen at the required levels and did not change the sperm quality, concluding that higher intakes of Se do not cause antagonistic effects on the absorption and subsequent action of other essential minerals supplied to the animals and still maintains electrolyte balance.
O objetivo do estudo foi avaliar se a suplementação com diferentes níveis de selênio (Se) pode alterar os componentes bioquímicos e eletrolíticos do sêmen, ocasionando, por conseguinte, uma melhoria na qualidade seminal em carneiros. Foram utilizados 30 animais, mantidos em sistema intensivo de baias, alimentados com feno e ração comercial, sendo alocados em cinco grupos (seis animais/baia) e suplementados com uma mistura mineral contendo 0mg (G1), 5mg (G2), 10mg (G3), 15mg (G4) e 20mg (G5) de Se/kg. Cada grupo recebeu um tratamento diferente a cada 56 dias, e foi realizado um rodízio de tratamentos entre os grupos seguindo uma sequência dinâmica. Amostras de sêmen foram colhidas por eletroejaculação após o fim de cada tratamento, a fim de se avaliarem os níveis de frutose, ácido cítrico, potássio (K), sódio (Na), cálcio (Ca), Se, zinco (Zn), manganês (Mn), enxofre (S) e chumbo (Pb). O desenho estatístico foi um quadrado latino 5x5. Os diferentes níveis de suplementação de Se avaliados mantiveram as concentrações de eletrólitos e de minerais do sêmen nos níveis exigidos e não modificaram a qualidade espermática, concluindo-se que maiores ingestões de Se não causam efeitos antagônicos na absorção e subsequente ação de outros minerais essenciais fornecidos aos animais e ainda mantêm o equilíbrio eletrolítico.

Salmonid Rickettsial Septicaemia (SRS), caused by Piscirickettsia salmonis, is the most important infectious disease in the Chilean salmon farming industry. An opportunity to control this disease is to use functional micronutrients to modulate host mechanisms of response to the infection. Since P. salmonis may affect the host antioxidant system in salmonids, particularly that dependent on selenium (Se), we hypothesized that fish's dietary selenium supplementation could improve the response to the bacterial infection. To address this, we defined a non-antibiotic, non-cytotoxic concentration of selenium to evaluate its effect on the response to in vitro infections of SHK-1 cells with P. salmonis. The results indicated that selenium supplementation reduced the cytopathic effect, intracellular bacterial load, and cellular mortality of SHK-1 by increasing the abundance and activity of host glutathione peroxidase. We then prepared diets supplemented with selenium up to 1, 5, and 10 mg/kg to feed juvenile trout for 8 weeks. At the end of this feeding period, we obtained their blood plasma and evaluated its ability to protect SHK-1 cells from infection with P. salmonis in ex vivo assays. These results recapitulated the observed ability of selenium to protect against infection with P. salmonis by increasing the concentration of selenium and the antioxidant capacity in fish's plasma. To the best of our knowledge, this is the first report of the protective capacity of selenium against P. salmonis infection in salmonids, becoming a potential effective host-directed dietary therapy for SRS and other infectious diseases in animals at a non-antibiotic concentration.

BACKGROUND: Selenium (Se) has been recognized as an essential micronutrient for nearly all forms of life. In recent decades, broiler responses to dietary Se supplemental levels and sources have received considerable attention. On environmental grounds, organic trace mineral utilization in practical broiler feeds has been defended due to its higher bioavailability. In such feeds, trace minerals are provided simultaneously in the same supplement as inorganic salts or organic chelates, a fact commonly ignored in assays conducted to validate organic trace mineral sources. The current assay aimed to investigate growth and biochemical responses, as well as Se retention of growing chicks fed diets supplemented with organic and inorganic Se levels and where the trace minerals (zinc, copper, manganese, and iron) were provided as organic chelates or inorganic salts according to Se source assessed. In so doing, a 2 × 5 factorial arrangement was used to investigate the effects of sodium selenite (SS) and selenium-yeast (SY) supplemented in feeds to provide the levels of 0, 0.08, 0.16, 0.24, and 0.32 mg Se/kg. RESULTS: Chicks fed selenium-yeast diets had body weight (BW), and average daily gain (ADG) maximized at 0.133 and 0.130 mg Se/kg, respectively. Both Se sources linearly increased (P < 0.05) the glutathione peroxidase (GSH-Px) activity in chick blood but higher values were observed in sodium selenite fed chicks (P < 0.05). Both Se sources influenced thyroid hormone serum concentrations (P < 0.05). Chicks fed SY exhibited greater retention of Se in the feathers (P < 0.05). Relative bioavailability of selenium yeast compared with SS for the Se content in carcass, feathers, total and Se retention were, 126, 116, 125 and 125%, respectively. SY supplementation resulted in lower liver Se concentration as Se supplementation increased (P < 0.05). CONCLUSIONS: Based on performance traits, the supplemental level of organic Se as SY in organic trace minerals supplement to support the maximal growth of broiler chicks is 0.133 mg Se/kg.

Objetivou-se avaliar a influência da suplementação com minerais biocomplexados sobre a qualidade da carne de frangos da linhagem Label Rouge, de ambos os sexos, criados em sistema alternativo. O delineamento experimental foi inteiramente ao acaso (DIC), disposto em esquema fatorial (3x2), sendo três dietas (controle; 0,50ppm de selênio; 0,40ppm de cromo) e dois sexos. Para a estabilidade lipídica, foi utilizado o DIC, disposto em esquema fatorial (3x2x2), sendo três dietas, dois sexos e dois tempos de armazenamento (zero e 12 meses). Os parâmetros avaliados no peito e na coxa foram: pH final, cor (L* - luminosidade, a* - índice de vermelho, b* - índice de amarelo, C* - índice de saturação e h* - ângulo de tonalidade), perda de peso por cozimento (PPC), força de cisalhamento (FC) e estabilidade lipídica. Além disso, foram avaliadas a proporção das formas químicas da mioglobina da coxa e a quantificação do conteúdo de selênio no peito. Não houve efeito isolado da dieta sobre os parâmetros físicos e químicos do peito e da coxa e sobre a concentração de selênio no peito. As fêmeas apresentaram médias superiores de L* (57,57), b* (10,55) e C* (10,60) do peito; e os machos de L* (55,09) da coxa. Entre os machos, as aves alimentadas com a dieta controle e com cromo expressaram valores superiores de FC da coxa em relação às suplementadas com selênio; para o tratamento com cromo, os machos apresentaram média superior de FC da coxa e, em contrapartida, para o tratamento com selênio, as fêmeas manifestaram a maior média. As amostras submetidas ao período de 12 meses de armazenamento mostraram maiores valores de substâncias reativas ao ácido tiobarbitúrico (TBARS) em ambos os cortes. A suplementação com cromo e selênio biocomplexados não provocou alterações na qualidade da carne; as fêmeas demonstraram melhores atributos de qualidade da carne.(AU)
The objective of the present study was to evaluate the influence of the supplementation with biocomplexed minerals on the quality of the meat of label Rouge lineage chicken, of both genders, reared in an alternative system. The experimental design was completely randomized (CRD) in a factorial scheme (3x2), with three diets (control; 0.50ppm of selenium; 0.40ppm of chromium) and two genders. For lipid stability, the CRD arranged in a factorial scheme (3x2x2) was used, being three diets, two genders and two storage times (0 and 12 months). The parameters evaluated in the breast and in the thigh were: final pH, color (L* - luminosity, a* - red index, b* - yellow index, C* - saturation index and h* - tonality angle), weight loss per cooking (WLC), shear force (SF) and lipid stability. In addition, we evaluated the proportion of chemical forms of myoglobin of the thigh and the quantification of selenium content in the breast. There was no isolated effect of diet on the physical and chemical parameters of the breast and thigh and selenium concentration in the breast. Females had higher mean values of L* (57.57), b* (10.55) and C* (10.60) of the breast; and males of L* (55.09) of the thigh. Among males, poultry fed with the control diet and chromium showed higher SF values of the thigh than those supplemented with selenium; for the treatment with chromium, the males presented superior average of SF of the thigh and, in contrast, for the treatment with selenium, the females had the highest average. Samples submitted to the period of 12-month of storage showed higher values of substances reactive to thiobarbituric acid (TBARS) in both cuts. Supplementation with biocomplexed chromium and selenium did not cause changes in meat quality; females presented better attributes of meat quality.(AU)

This study was designed to examine the protective effects of nano-selenium and nano-zinc oxide on queen and workers performance under heat stress condition and gene expression of heat shock protein 70 (hsp70) as an index of heat tolerance. Sixty colonies were randomly assigned to five treatments with 12 replicates from June until early September. Sugar syrup (50%) containing no supplement or nano-selenium at levels of 50 and 100 µg L-1 or nano-zinc at levels of 100 and 200 µg L-1 was fed to colonies. Nano-selenium supplementations had no effect, but nano-zinc at level of 100 µg L-1 significantly decreased body malondialdehyde concentration. The highest bee population was seen in nano-zinc at level of 100 µg L-1 and the lowest one in control group. The lowest and the highest body weight, fat and protein deposition was found in group received nano-zinc at level of 100 µg L-1 and control, respectively. The highest gene expression was for group received nano-zinc at level of 100 µg L-1 In group received nano-zinc at level of 100 µg L-1, an increase in hsp70 gene expression was found. In conclusion, nano-zinc oxide at level of 100 µg L-1 could increase queen and worker performance and heat resistance of bees in the hot climate condition.

In the present study, we aimed to evaluate the effects of different concentrations of selenium (Se) ovine nutritional supplementation on spermatozoa DNA integrity. Thirty male ovines (age: 10 months) were used. They were fed with hay and ram food in an intensive system, which was divided into stalls (5 m long and 3 m wide) with feeding troughs, and had ad libitum access to food and water. Ovines in group 1 (G1, the negative control) received mineral salt supplementation without Se; ovines in G2 received the same mineral salt mixed with 5 mg Se (as sodium selenite)/kg mineral supplement;ovines in G3 received 10 mg Se/kg mineral supplement; ovines in G4 received 15 mg Se/kg mineral supplement; and ovines in G5 received 20 mg Se/kg mineral supplement. Ovines in all groups remained untreated for 14 days, followed by a treatment period of 56 days. Semen samples were obtained by electroejaculation. The DNA damage in semen samples was evaluated using the comet assay. The experimental design was implemented using a 5 × 5 Latin Square, i.e., five treatments and five experimental periods. The mean differences were compared using Tukeys test at a significance level of 5%. The control group (G1) showed a high percentage of DNA damage compared to the Se-treated groups (G2-G5). Therefore, Se supplementation could decrease the basal level of DNA damage in sperm cells, suggesting that Se might exert protective effects on sperm DNA.(AU)
O presente estudo teve por objetivo avaliar os efeitos da suplementação mineral com diferentes concentrações de selênio (Se) sobre a integridade de DNA espermático de ovinos. Utilizaram-se 30 machos, com 10 meses de idade. Eles foram mantidos em sistema intensivo, sendo alimentados com feno e ração própria para ovinos, divididos em baias (5 m x 3 m), com cochos e água ad libitum. Os ovinos do grupo 1 (G1=controle negativo) receberam suplementação de sal mineral sem a adição de Se, os animais do G2 receberam a mesma mistura mineral, porém com 5 mg de Se (selenito de sódio)/kg mistura mineral, os ovinos do G3 receberam 10 mg Se/kg mistura, os animais do G4 receberam 15 mg Se/kg mistura, os do G5 receberam 20 mg Se/kg mistura. Os ovinos de todos os grupos passaram por um período de adaptação de 14 dias, seguido por um período de tratamento de 56 dias. O sêmen foi colhido por meio de eletroejaculação. A integridade do DNA espermático foi avaliada por meio do teste de cometa. O modelo experimental utilizado foi Quadrado Latino 5 x 5, com cinco grupos e cinco períodos experimentais. A diferença entre as médias foi analisada pelo teste de Tukey, com 5% de nível de significância. O grupo controle (G1) apresentou elevada porcentagem de danos quando comparada aos demais grupos de tratamentos (G2 a G5). Portanto, a suplementação de Se diminui o nível de danos ao DNA espermáticos, sugerindo que o Se pode exercer efeitos protetores sobre o DNA dos espermatozoides de ovinos.(AU)

SUMMARY: Over dose or long-term clinical use of therapeutic doses of acetaminophen (APAP) causes hepatotoxicity. Various strategies attempted to ameliorate APAP-hepatotoxicity have been found to be unsuitable for clinical practice. This study was aimed to illustrate the histopathological changes induced by therapeutic dose of APAP and investigate the hepatoprotective role of oral co-administration of selenium/ Tribulus terrestris (TT) extract concurrently against hepatotoxicity induced by APAP in rats. Fifty-four healthy male albino Wistar rats were randomized into nine groups (G1-G9) of six rats each, and administered with APAP and TT orally for 30 days as follows: Control (2ml normal saline), APAP (470 mg/kg), APAP (470 mg/kg) + selenium (2 mg/kg), APAP (470 mg/kg) + TT (98 mg/kg), APAP (470 mg/kg) + selenium (2mg/kg) + TT (98 mg/kg), APAP (470 mg/kg) + silymarin (200 mg/kg), selenium (2 mg/ kg), TT (98 mg/kg) and silymarin (200 mg/kg) groups. The results demonstrated that exposure of rats to therapeutic dose of APAP for 30 days caused significant histopathological changes parallel to elevated blood chemistry parameters. Co-administration of selenium/TT extract showed significantly reduced histopathological lesions and, restored or decreased levels of the examined blood chemistry parameters. Liver histology in selenium/TT extract showed normal hepatic architecture with mild changes and silymarin treated rats showed no histopathological changes. Histochemically PAS staining, showed that APAP-induced hepatotoxicity was characterized by hepatocytes glycogen depletion. Selenium/TT co-supplementation plays a potential role in preventing APAP-induced glycogen depletion by increasing detoxification and scavenging the reactive metabolites. Selenium/TT extract oral co-administration possesses a significant hepatoprotective property and mitigates APAP-induced hepatotoxicity by enhancing its antioxidant role and improving tissue integrity. Selenium/TT supplementation could represent an effective treatment against APAP-induced hepatotoxicity. Further studies are needed to elucidate the exact mechanism underlying the protective role of TT extract.
RESUMEN: La dosis excesiva o el uso clínico a largo plazo de dosis terapéuticas de acetaminofeno (APAP) causa hepatotoxicidad. Se ha descubierto que varias estrategias que intentaron mejorar la hepatotoxicidad por APAP no son adecuadas para la práctica clínica. Este estudio tuvo como objetivo ilustrar los cambios histopatológicos inducidos por la dosis terapéutica de APAP e investigar el papel hepatoprotector de la administración conjunta de extracto de selenio / Tribulus terrestris (TT) simultá- neamente contra la hepatotoxicidad inducida por APAP en ratas. Cincuenta y cuatro ratas Wistar albino machos sanas se aleatorizaron en nueve grupos (G1 - G9) de seis ratas cada una, y se administraron con APAP y TT por vía oral durante 30 días de la siguiente manera: Control (2 ml de solución salina normal), APAP (470 mg / kg), APAP (470 mg / kg) + selenio (2 mg / kg), APAP (470 mg / kg) + TT (98 mg / kg), APAP (470 mg / kg) + selenio (2 mg / kg) + TT (98 mg / kg), APAP (470 mg / kg) + silimarina (200 mg / kg), selenio (2 mg / kg), TT (98 mg / kg) y silimarina (200 mg / kg). Los resultados demostraron que la exposición de las ratas a la dosis terapéutica de APAP durante 30 días causó cambios histopatológicos significativos paralelos a parámetros elevados de química sanguínea. La administración conjunta de extracto de selenio / TT mostró lesiones histopatológicas significativamente reducidas y niveles restaurados o disminuidos de los parámetros de química sanguínea. La histología hepática en el extracto de selenio / TT mostró una arquitectura hepática normal con cambios leves y las ratas tratadas con silimarina no mostraron cambios histopatológicos. La tinción histoquímica de PAS mostró que la hepatotoxicidad inducida por APAP se caracterizó por la pérdida de glucógeno de los hepatocitos. La suplementación con selenio / TT juega un papel potencial en la prevención de la pérdida de glucógeno inducido por APAP al aumentar la desintoxicación y eliminar los metabolitos reactivos. La administración conjunta de extracto de selenio / TT posee una propiedad hepatoprotectora significativa y mitiga la hepatotoxicidad inducida por APAP al mejorar su papel antioxidante y la integridad del tejido. La suplementación con selenio / TT podría representar un tratamiento efectivo contra la hepatotoxicidad inducida por APAP. Se necesitan más estudios para dilucidar el mecanismo exacto que subyace a la función protectora del extracto TT.

Recently, we demonstrated the promising anxiolytic action of 7-chloro-4-(phenylselanyl) quinoline (4-PSQ) in mice. For this reason, the objective of this study was to expand our previous findings by investigating the contribution of serotoninergic and GABAergic systems to the anxiolytic action of this compound. Pretreatment with different serotoninergic antagonists (pindolol, WAY100635 and ketanserin) blocked the anxiolytic effect caused by 4-PSQ (50 mg/kg, per oral) in the elevated plus maze (EPM) test. The contribution of the GABAergic system was investigated by pretreatment with pentylenetetrazole (a GABAA receptor antagonist) (PTZ). 4-PSQ diminished the PTZ-induced anxiety, and did not modify the locomotor, exploratory and motor activities of mice. Later, this group of animals was euthanized and the blood was removed to determine the levels of corticosterone, and cerebral cortex and hippocampus to determine the mRNA expression levels of cAMP response element binding protein (CREB), brain derived neurotrophic factor (BDNF) and nuclear factor kappa B (NF-κB), as well as the Na+, K+ ATPase activity and reactive species (RS) levels. 4-PSQ was able to significantly reverse the increase in RS and corticosterone levels, as well as the decrease of CREB and BDNF expression in the cerebral structures and increase of NF-κB expression in the hippocampus. Finally, 4-PSQ restored the Na+, K+ ATPase activity in the cerebral structures evaluated. Here, we showed that the modulation of serotonergic and GABAergic systems, factors related to neurogenesis, oxidative status and Na+, K+ ATPase activity contributes to the anxiolytic effect of 4-PSQ and reinforces the therapeutical potential of this compound for the treatment of anxiety.

Eimeriosis is caused by a protozoan parasite of the genus Eimeria and infection affecting most domestic animal species. The aim of this research was to comprehend the impact of selenium nanoparticles (SeNPs) on eimeriosis induced by Eimeria papillata in mouse jejunum, and how they work as antioxidants and anti-apoptotic agents against eimeriosis. The numbers of meronts, gamonts, and developing oocysts of E. papillata reduced after the infected mice were treated with the SeNPs. The levels of malondialdehyde (MDA), nitric oxide (NO), and other oxidative stress-related molecules, such as glutathione (GSH), catalase (CAT), and superoxide dismutase (SOD), were assayed. E. papillata was able to change the redox status of the jejunal cells; this was confirmed by the elevation of the MDA and NO levels, and the decrease of the GSH levels and the activities of the antioxidant enzymes CAT and SOD. SeNP treatment significantly reversed this disturbance of the redox status. The expression levels of the apoptotic markers Bax and caspase-3 in the jejunal samples were evaluated using qRT-PCR. The SeNPs decreased the Bax and caspase-3 expression after being administered to the E. papillata-infected mice. Collectively, the SeNPs demonstrated antioxidant and anti-apoptotic activities against murine eimeriosis.

Bearing in mind that pain and major depressive disorder (MDD) often share biological pathways, this condition is classified as depression-pain syndrome. Mounting evidence suggests that oxidative stress is implicated in the pathophysiology of this syndrome. The development of effective pharmacological interventions for the depression-pain syndrome is of particular importance as clinical treatments for this comorbidity have shown limited efficacy. Therefore, the present study aimed to evaluate whether the 3,5-dimethyl-1-phenyl-4-(phenylselanyl)-1H-pyrazole (SePy) was able to reverse the depression-pain syndrome induced by intracerebroventricular (i.c.v) streptozotocin (STZ) in mice and the possible modulation of oxidative and nitrergic pathways in its effect. The treatment with SePy (1 and 10 mg/kg) administered intragastrically (i.g.) reversed the increased immobility time in the tail suspension test, decreased grooming time in the splash test, latency time to nociceptive response in the hot plate test, and the response frequency of Von Frey hair (VFH) stimulation induced by STZ (0.2 mg/4 µl/per mouse). Additionally, SePy (10 mg/kg, i.g.) reversed STZ-induced alterations in the levels of reactive oxygen species, nitric oxide, and lipid peroxidation and the superoxide dismutase and catalase activities in the prefrontal cortices (PFC) and hippocampi (HC) of mice. Treatment with SePy (10 mg/kg, i.g.) also reversed the STZ-induced increased expression of inducible nitric oxide synthase (iNOS) and glycogen synthase kinase 3 beta (GSK3ß) in the PFC and HC. An additional molecular docking investigation found that SePy binds to the active site of iNOS and GSK3ß. Altogether, these results indicate that the antidepressant-like effect of SePy is accompanied by decreased hyperalgesia and mechanical allodynia, which were associated with its antioxidant effect.

The enrichment of meat with selenium is important to improve the intake of selenium by humans. The effects of supranutritional doses of sodium selenite or selenium-enriched yeast on performance, carcass characteristics and meat quality were evaluated using 63 Nellore cattle in a completely randomized design with two sources (sodium selenite and selenium-enriched yeast), three levels (0.3, 0.9 and 2.7 mg Se/kg DM) and control treatment (without addition of selenium). Final body weight (BW), average daily gain, dry matter intake and gain to feed ratio (G : F) at the end of 84 days of supplementation were not influenced by treatments (P>0.05). Values of pH, ribeye area, back fat thickness and marbling score were also not influenced by treatments ( P>0.05). Dressing percentage was greater (P=0.02) in Nellore cattle supplemented with organic Se (58.70%) compared to animals supplemented with inorganic Se (57.94%). Hot carcass weight increased ( P=0.05) with the increasing of Se levels in the diet. Colour, shear force (SF), cooking and drip loss remained unchanged ( P>0.05); however thiobarbituric acid reactive substances was 15.51% higher with inorganic Se compared with organic Se. The selenium concentration in the meat of animals receiving organic selenium was higher ( P<0.001) than that of animals receiving sodium selenite, at all levels (0.3; 0.9 and 2.7 mg/kg DM). The meat of animals receiving 2.7 mg of organic Se/kg of DM presented concentration of 372.7 µg Se/kg in the L.dorsi muscle, and the intake of 150 g of this meat by humans provides approximately 100% of the recommended Se intake (55 µg Se/day for adults). Therefore, the use of supranutritional doses of 2.7 mg Se/kg of DM, regardless of source, is a way of naturally producing selenium-enriched meat without compromising performance, carcass characteristics and quality of Nellore bovine meat.

The present study was performed to evaluate the association of chromium-yeast and selenium-yeast as nutritional additives to enhance performance and quantitative characteristics in the carcass of finishing barrows. A total of 48 barrows, commercial hybrid, with initial weight of 68.3±3.5 kg and final weight of 99.6±3.6 kg, were utilized for the study. The animals were distributed in a randomized complete block design, consisting of three levels of chromium-yeast and selenium-yeast (i.e., without supplementation of chromium-yeast and selenium-yeast, supplementation of 0.4 mg kg-1 chromium-yeast and 0.3 mg kg-1 selenium-yeast, and supplementation of 0.8 mg kg-1 chromium-yeast and 0.6 mg kg-1 selenium-yeast) with eight replicates and two animals per experimental unit. The supplementation of chromium and selenium-yeast did not significantly affect (P>0.05) the average daily feed intake, digestible lysine, crude protein and metabolizable energy intake, average daily weight gain, feed conversion, weight and carcass length, back fat thickness, muscle depth, percentage and amount of lean meat, and carcass allowance. These findings suggest that supplementation of chromium and selenium-yeast does not affect the performance or quantitative characteristics in the carcass of finishing barrows.(AU)
Realizou-se este estudo com o objetivo de avaliar a associação de cromo-levedura e selênio-levedura como aditivos nutricionais melhoradores de desempenho e das características quantitativas de carcaça de suínos machos castrados em terminação. Foram utilizados 48 suínos machos castrados, híbridos comerciais, com peso inicial de 68,3±3,5 kg e peso final de 99,6±3,6 kg. Os animais foram distribuídos em delineamento experimental de blocos casualizados, composto por três níveis de cromo-levedura e selênio-levedura (controle sem suplementação de cromo-levedura e selênio-levedura; suplementação de 0,4 mg kg-1 de cromo-levedura e 0,3 mg kg-1 de selênio-levedura; e 0,8 mg kg-1 de cromo-levedura e 0,6 mg kg-1 de selênio-levedura), com oito repetições e dois animais por unidade experimental. A suplementação de cromo-levedura e selênio-levedura não influenciou (P>0,05) o consumo de ração diário, os consumos de lisina digestível, de proteína bruta e de energia metabolizável, o ganho de peso diário, a conversão alimentar, o peso e comprimento de carcaça, espessura de toucinho, profundidade de músculo, percentagem e quantidade de carne magra e índice de bonificação. A suplementação associada de cromo e selênio-levedura não influencia o desempenho e as características quantitativas de carcaça de suínos machos castrados em terminação.(AU)
