RÉSUMÉ

Defeitos ósseos constituem um problema de saúde global. O sistema Rigenera permite a extração de microenxertos ricos em células-tronco mesenquimais (CTMs). Objetivou-se avaliar o processo de regeneração óssea por enxertos obtidos pelo sistema Rigenera em defeitos críticos na calvária de ratos. Foram utilizados 18 ratos Wistar, machos, pesando 285±29g, distribuídos em três grupos (n=6), sendo cada animal controle de si mesmo, denominados G15-Controle e G15-Tratado (15 dias); G30-Controle e G30-Tratado (30 dias) e G60-Controle e G60-Tratado (60 dias). Foram realizadas duas lesões de 5mm de diâmetro em cada antímero da calvária. Nos grupos tratados, foram utilizados microenxertos autólogos de cartilagem xifoide, obtidos pelo sistema Rigenera. O defeito contralateral serviu como controle em todos os animais. Os animais foram eutanasiados aos 15, 30 e 60 dias após a cirurgia, e as amostras foram processadas para a histoquímica. Nos grupos controle, não foram observados sinais de regeneração óssea, enquanto nos grupos tratamento foram verificadas áreas de formação óssea e tecido mesenquimal ativado. O sistema Rigenera foi eficiente na obtenção de microenxertos autólogos, para terapia celular em defeito crítico de calvária de ratos. Com o aprimoramento do protocolo, o sistema Rigenera poderá ser amplamente utilizado no tratamento de lesões ósseas.(AU)


Bone defects are a global health problem. The Rigenera system allows the extraction of micro grafts rich in mesenchymal stem cells (MSCs). The objective of this study was to evaluate the bone regeneration process by grafts obtained by the Rigenera system in defects in the rats calvarian. Eighteen male Wistar rats were used, weighing 285 ± 29g, distributed in three groups (n = 6), where each animal was treatment and control, called G15-Control and G15-Treated (15 days); G30-Control and G30-Treated (30 days) and G60-Control and G60-Treated (60 days). Two 5mm diameter lesions were performed on each calvaria side. In the treated groups, autologous micrograft from xiphoid cartilage, obtained by the Rigenera system, were used. The other defect served as a control in all animals. The animals were euthanized at 15, 30 and 60 days after the surgery and the samples were processed for histochemistry. In the control groups, no signs of bone regeneration were observed, while in the treatment groups, areas of bone formation and activated mesenchymal tissue were verified. The Rigenera system was efficient in obtaining autologous micrograft for cell therapy in a critical calvaria defect in rats. Rigenera system can be widely used in the treatment of bone injuries.(AU)

Cheeses are able to serve as suitable matrices for supplying probiotics to consumers, enabling appropriate conditions for bacteria to survive gastric transit and reach the gut, where they are assumed to promote beneficial processes. The present study aimed to evaluate the microbiological, immunological, and histological changes in the gut of Salmonella Enteritidis-challenged rats fed goat cheese supplemented with the probiotic strain Lactobacillus rhamnosus EM1107. Thirty male albino Wistar rats were randomly distributed into 5 experimental groups with 6 animals each: negative (NC) and positive (PtC) control groups, control goat cheese (CCh), goat cheese added with L. rhamnosus EM1107 (LrCh), and L. rhamnosus EM1107 only (EM1107). All animals, except NC group were challenged with Salmonella Enteritidis (109 cfu in 1 mL of saline through oral gavage). Microbial composition was assessed with high-throughput 16S rRNA sequencing by means of Illumina MiSeq (Illumina, San Diego, CA). Nuclear factor kappa B (NF-κB) from the animal cecum tissue was determined by real-time PCR and interleukins (TNF-α, IL-1ß, IL-10, and IFN-γ) by means of ELISA. Myeloperoxidase and malondialdehyde levels were determined biochemically. The administration of the L. rhamnosus EM1107 probiotic strain, either as a pure culture or added to a cheese matrix, was able to reduce Salmonella colonization in the intestinal lumen and lessen tissue damage compared with rats from PtC group. In addition, the use of cheese for the probiotic strain delivery (LrCh) was associated with a marked shift in the gut microbiota composition toward the increase of beneficial organisms such as Blautia and Lactobacillus and a reduction in NF-κB expression. These findings support our hypothesis that cheeses might be explored as functional matrices for the efficacious delivery of probiotic strains to consumers.

O presente trabalho objetivou investigar a prevalência da bactéria Helicobacter spp. em gatos da Cidade do Recife, Pernambuco. As amostras de estômagos analisadas foram colhidas de 119 gatos sem raça e idade definida, sendo 56 machos e 63 fêmeas. Para pesquisa de Helicobacter spp., foram realizados os métodos de teste rápido da urease e exames histopatológicos. As análises pelo teste rápido da urease mostraram que em 82,35% das amostras foi detectada reação positiva para Helicobacter. Na análise histopatológica, verificou-se que em 59,66% das amostras (71/119) apresentaram lesões. Essas lesões consistiram em infiltrado linfoplasmocitário com folículo linfoide, microabcessos, necrose, hipotrofia; em 40,34% das amostras (48/119) não foram observadas alterações histológicas. Conclui-se que gatos oriundos da Cidade de Recife, Pernambuco< apresentam alta incidência de infecção por Helicobacter spp. Infiltrado difuso de células linfoplasmocitárias foram as alterações histopatológicas mais frequentes observadas na mucosa gástrica de gatos infectados por Helicobacter spp. (AU)


INCIDENCE AND PATHOLOGICAL ASPECTS OF INFECTION BY HELICOBACTER SPP. IN CATS FROM THE CITY OF RECIFE, PERNAMBUCO, BRAZIL. The present study aimed to investigate the incidence of the bacterium Helicobacter spp. in cats from Recife, Pernambuco State, Brazil. Stomach samples were collected from 119 cats, without distinction between race or age, the breakdown by sex being 56 males and 63 females. The urease quick test and histopathological examination using both hematoxilin-eosin (HE) and Giemsa stain were performed. The urease quick test showed positive results for Helicobacter in 82.35% of the samples. The histopathology revealed an alteration in 59.66% (71/119) of the samples, such as lymphoplasmocitic infiltration with a lymphoid foliculum, microabscess, necrosis, and hypotrophy, while in 40.34% of the samples (48/119) no alterations were observed. It was concluded that the cats from the city of Recife, State of Pernambuco, Brazil, presented a high incidence of infection with Helicobacter spp. Diffuse infiltrate of lymphocytes cells was the most frequent histopathological change observed in the gastric mucosa of cats infected with Helicobacter spp. (AU)
