The objective of this work was to discover a biochemical pathway to explain the transfer of cadmium, a toxic element, from seawater to cultured mussels. Understanding the intricacies of this transfer is crucial for global mussel crops, as it has the potential to mitigate risks to human health and prevent economic losses in the industry. We focused our investigation on Yal Bay, a typical area with intense mussel aquaculture activity (16,000 t y-1) in the inland sea of southern Chile. Seasonal samples of blue mussels (Mytilus chilensis) were collected and analyzed from September 2014 to December 2015 at two integrated depths (0-5 m and 5-10 m). Diurnal and nocturnal seston, seawater, benthic sediments and decanted suspensions from the water column were recorded. Our findings indicate that nocturnal seston satisfactorily explains the presence of cadmium in Mytilus chilensis aquaculture throughout its annual temporal distribution (Spearman rs = 0.63, p = 0.002).

Globally, Chile is the second largest producer of mussels, with 99% of production concentrated in the inland sea of the Los Lagos Region, Southern Chile. This study reveals that seasons produce a drastic difference in the cadmium concentration (Cd) in marine mussels in bay, channel, and fjord ecosystems in this area. As the global mussel industry continues its rapid expansion, a complete understanding of cadmium pathways is critical in order to minimize the cadmium content in harvests. In this study, biweekly sampling was conducted in Chiloé (Southern Chile), during five consecutive seasons from June 2014 to November 2015. Cadmium in the soft tissues (ST) and in the content of the digestive gland (CDG) of Mytilus chilensis were investigated, in addition to resuspensions and seston to determine the effect of the seasons on metal bioassimilation capacity. In spring, the (Cd) between CDG and ST varied by approximately 2 mg Cd kg-1 dry mass (DM). In summer and autumn, the (Cd) in CDG increased from 3 to 6 mg Cd kg-1 while the (Cd) in ST decreased from 2.5 to 1.5 mg Cd kg-1 DM. The three ecosystems showed the same cadmium bioconcentration trends in all seasons, revealing coherent global trends. These findings should caution the industry and coastal populations about the seasonal variability and intensity of cadmium metal transfer to biofilters, especially because of the adverse effects of cadmium consumption on human health. Additionally, this study found that mussels in natural beds concentrate more Cd (> 1 mg Cd kg-1 DM) than in industrial facilities. Multiregression analysis showed and explained the cadmium in CDG for three ecosystems: channel (R2 0.9537), bay (R2 0.5962), and fjord (R2 0.4009). The independent variable nocturnal seston was able to explain the increase in cadmium.

Cadmium is present in agricultural soil composition and is assimilated by plants. The mussel industry generates large volumes of calcareous valves as a byproduct of processing in factories. This solid waste is ground, stored and disposed of in the form of agricultural supplies near production areas in almost all regions of the world. Cd content was recorded in these calcareous substrate by-products of industrial processes. In this study, mussel shells were investigated in three areas in the sea of Chile; two areas with high aquaculture activity and one in a non-production area. Mass Spectrometry was used for [Cd] readings. There are no significant differences (p=0.92) in [Cd] between the studied areas. Additionally, the [Cd] observed in valves was also similar relative to different depths. The low [Cd] observed in valves (~0.014±0.0049mgCdkg-1 dry) suggests the potential use of this industrial byproduct in agricultural applications.

OBJECTIVE: To evaluate the effectiveness of a reproductive management program consisting of combinations of Ovsynch/TAI and prostaglandin (PG) F(2alpha) treatments in Holstein dairy cows under a pasture-based dairying system. DESIGN: Field trial. PROCEDURE: A total of 1177 cows in 8 commercial dairy farms were randomly allocated to control and treatment groups. Treatment group cows received one of two interventions depending upon the number of days postpartum (DPP) before the planned start of breeding. Cows more than 50 DPP by the planned start of breeding received the Ovsynch/TAI treatment, consisting of gonadotrophin releasing hormone (GnRH) - PGF(2alpha)- GnRH plus timed artificial insemination. Cows between 40 and 50 DPP received a PGF(2alpha) treatment followed by oestrus detection and, if the cow was not seen in oestrus, the cow received a second PGF(2alpha) 14 days later. Control cows were submitted to twice a day heat detection followed by artificial insemination. The experimental period was the start of the breeding season plus 21 days for cows over 50 DPP at the start of breeding, and was 40-61 DPP for cows that calved later and passed their voluntary waiting period after the start of the breeding season. RESULTS: Submission rate was higher for the treated group than for the control group (84.9% vs. 55.1%; P < 0.0001), as was the conception rate (51.0% vs. 46.1%; P < 0.03). Due to farm variations, pregnancy rate was similar in both groups (38.5% vs. 28.2%; P > 0.1). Within the treated group, conception rate and pregnancy rate of the cows inseminated after a PGF(2alpha) were higher than for timed artificial inseminated cows (51.4% vs. 32.6%; P < 0.001), and (37.8% vs. 32.6%; P < 0.001). CONCLUSION: A programmed reproductive management protocol may improve reproductive efficiency in dairy farms with seasonal breeding, by increasing submission and conception rates at the beginning of the breeding season and/or at the end of the voluntary waiting period. Fertility of cows bred after a PGF(2alpha) synchronised heat was greater than after an Ovsynch/TAI protocol.

This study describes the effect of parity (multiparous versus primiparous) and body condition score (BCS) at calving (<3 or > or =3; scale 1-5) on variations of BCS, body weight (BW) and metabolic profiles in Holstein cows grazing on improved pastures. Forty-two cows were studied (21 multiparous and 21 primiparous) from 2 months before to 3 months after calving. BCS, BW and milk production were measured every 2 weeks. Blood samples were taken every 2 weeks to determine total protein, albumin, urea, non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHB), cholesterol, aspartate aminotransferase (AST), calcium, phosphorus and magnesium. Primiparous cows had lower BCS during the early postpartum (PP) period and produced less milk than multiparous. In primiparous cows NEFA concentrations were higher during the early postpartum period; BHB levels were similar in both categories during this period. Primiparous cows showed a more unbalanced metabolic profile than multiparous cows, reflecting that they are recovering from the loss of BCS after calving with less success.

RÉSUMÉ

This study characterized endometrial expression of mRNAs of oestrogen and progesterone receptors (ER, PR) and insulin-like growth factor-I (IGF-I) during the oestrous cycle. Seven Holstein heifers that showed standing oestrus on the same day (day 0) were selected and blood samples for oestradiol (E2) and progesterone (P4) determinations by RIA were taken daily until day 23. Endometrial samples were taken by transcervical biopsies on days 0, 5, 12 and 19 for mRNA determination by solution hybridization. The highest endometrial mRNA levels of ERalpha and PR were observed at oestrus and a decline was observed already at day 5, which then decreased progressively at the end of the luteal phase. IGF-I mRNA levels were higher at day 0 and 5 than at day 12. At day 19, mRNA levels of ERalpha, PR and IGF-I were the lowest in heifers that were at the end of their luteal phase (n=4), but were high again in heifers which P4 levels were basal (n=3). The temporal changes in mRNA endometrial expression of ERalpha, PR and IGF-I and their relation to the changes in steroid concentrations during the bovine oestrus cycle are described.
