RÉSUMÉ

The osseous vascular endothelium encompasses a vast intricate framework that regulates bone remodeling. Osteoporosis, an age-associated systemic bone disease, is characterized by the degeneration of the vascular architecture. Nevertheless, the precise mechanisms underpinning the metamorphosis of endothelial cells (ECs) with advancing age remain predominantly enigmatic. In this study, we conducted a systematic analysis of differentially expressed genes (DEGs) and the associated pathways in juvenile and mature femoral ECs, utilizing data sourced from the Gene Expression Omnibus (GEO) repositories (GSE148804) and employing bioinformatics tools. Through this approach, we successfully discerned six pivotal genes, namely Adamts1, Adamts2, Adamts4, Adamts14, Col5a1, and Col5a2. Subsequently, we constructed a miRNA-mRNA network based on miRNAs displaying differential expression between CD31hiEMCNhi and CD31lowEMCNlow ECs, utilizing online repositories for prediction. The expression of miR-466i-3p and miR-466i-5p in bone marrow ECs exhibited an inverse correlation with age. Our in vivo experiments additionally unveiled miR-466i-5p as a pivotal regulator in osseous ECs and a promising therapeutic target for age-related osteoporosis.

Although research has been conducted on various insect species that induce galls on various plant tissues, information is particularly scarce when it comes to insects that form galls on the tough trunk of their host plants. This contribution describes the gall-inducing aphid Nipponaphis hubeiensis sp. nov. from the Zhushan County, Shiyan City, Hubei Province of China. This aphid induces enclosed galls with woody external layer on the trunk of Sycopsis sinensis (Saxifragales: Hamamelidaceae), an uncommon ecological niche in the aphid-plant interaction system. Morphological features for the identification of new species are provided. In addition, a partial sequence of the nuclear gene EF1 was amplified and sequenced to construct a cluster graph. Based on the clustering graph combined with morphology traits, the gall-forming aphid was classified into Nipponaphis. The unique ecological habits of this new aphid will bring innovative perspectives to the study of the evolution and diversity in aphid-host interaction.

We want to construct a yeast expression system for thymosin a1 (Ta1) to make the orally administered Ta1 preparation possible. The whole Ta1 DNA fragment was obtained by PCR. After being digested with restriction enzymes, it was cloned into pYES2 vector. Sequencing was performed to identify the recombinant. The sequence of Ta1 in recombinant coincided with the original one reported in Genbank. When pYES2-Ta1 plasmid was transformed into yeast, galactose instead of glucose was used to induce Ta1 expression. Western blot was performed to identify the quality of the expressed Ta1. Dried yeast containing pYEST2-Ta1 was fed to Balb/c mice whose immunities were inhibited by cyclophosphamide in advance. Synthesized Ta1 peptide was used as positive control and empty yeast was used as negative control. Compared with the negative control group, both dried yeast containing pYEST2-Ta1 and synthesized Ta1 peptide can significantly increase the CD8+ level (22.74±1.09 and 18.77±4.72 vs 7.49±2.14, p<0.01), while both of them had little effect on the CD4+ lymphocytes (61.86±6.94 and 65.91±4.78 vs 57.93±10.40, p>0.05). We concluded that a high effective yeast expression system for Ta1 was constructed successfully and the Ta1 protein expressed by this system can improve CD8+ level in immune inhibited mice.

Two premeditated anionic rosette-layer architectures have been constructed using guanidinium and ubiquitous C3-symmetric oxoanions that carry unequal charges, employing bulky organic cations as interlayer templates. The undulate guanidinium-carbonate network occurs in 4[(C2H5)4N+].8[C(NH2)3+].3(CO3)2-.3(C2O4)2-.2H2O, in which the disordered (C2H5)4N+ guests are accommodated in pouches and channels within a complex three-dimensional hydrogen-bonded host framework. In [(C2H5)4N+]2.[C(NH2)3+].[1,3,5-C3H3(COO-)3].6H2O, ordered (C2H5)4N+ cations are sandwiched between planar guanidinium-trimesate host layers whose honeycomb cavities are tightly fitted with flattened-chair (H2O)6 clusters of symmetry 2.
