The health of calves has a significant impact on the production of cows and livestock. Some desert plants have pharmacological importance, as they can be used to reduce antibiotic resistance. Our hypothesis is designed to detect Virulent- Multidrug-Resistant and Extended- spectrum Beta- lactamase Enterobacteriaceae (Virulent-MDR-ESBL Enterobacteriaceae and to determine whether Moringa oleifera has antibacterial activity against the detected isolates. A total of 39 Enterobacteriaceae isolates from 28 diarrheic samples were collected from calves aged between 20 days and 20 months from 3 different flocks in North Sinai, Sahl-Eltina region, Egypt. E.coli 46% (18/39), O157 13% (5/39), Klebsiella pneumoniae 41% (16/39). MDR members accounted for 87%, while ESBL isolates accounted for 43%. The antibacterial activity is represented by microdilution. Minimum inhibition concentration (MIC) for the methanol extract of Moringa oleifera ranged from 2.5,5,10, and 25mg/ ml among E.coli isolates, and O157 was susceptible to (2.5mg/ ml), Klebsiella pneumoniae isolates were susceptible to (5-50mg/ ml). Analysis of the methanol extract revealed that ferulic acid was the dominant phenolic compound with a concentration of 29,832 parts per million (ppm). In silico docking study expected the active site of ferulic acid to act on the tyrosine bacterial enzyme through Pi-alkyl, Pi-anion, Carbon hydrogen bonds, and extra ionic attractive interactions with copper ions which can stabilize ferulic acid inside the targeted pocket Diverse virulent gene profiles were observed in E. coli. The Shiga toxin-producing Escherichia coli (STEC) was reported in 83% of the isolated E. coli, while the DNA gyrase (gyrA) was harbored in 100% of Klebsiella pneumoniae isolates. Various profiles of antibiotic resistance genes for both E. coli and Klebsiella pneumoniae isolates were distinguished. blaTEM genes were detected in 99% of E. coli and 100% of Klebsiella pneumoniae. Sequence analysis for E. coli strain DRC-North Sinai-Eg was placed in accession numbers (OP955786) for the Shiga toxin 2 gene (Stx2A), (OP997748) and (OP997749) for the Adhesion to host cell gene (Eae). For the hemolysine gene (hylA), the accession number was (OP946183). Klebsiella pneumoniae strain DRC-North Sinai-Eg was placed in (OP946180) for (gyrA). This study has proven the broad range of Moringa oliefera's antibacterial effects in vitro against the virulent-MDR- ESBL E. coli and Klebsiella pneumoniae isolated from North Sinai calves diarrhea. These are congruent with the disability effect on bacterial tyrosinase enzyme through docking study therefore, we recommend the usage of this desert plant as a prospective feed additive, we endorse this as an antibacterial new insight natural source and for the medication of considered pathogens with zoonotic impacts.

Infectious diseases are the most important cause of death worldwide. Many of these diseases show great resistance to drugs and antibiotics with long-term use. Extracts of some medicinal plants have antimicrobial properties, which can treat and overcome these diseases. Meliaceae is a family of timber trees used extensively in treating many bacterial and fungal diseases, especially Swietenia mahagoni (L.) Jacq. In this study, phytochemical screening, gas chromatography/mass spectrometry (GC/MS) analysis, and antimicrobial, antioxidant, and antitumor activities of the methanolic extract of S. mahagoni (L.) leaves were performed. Phytochemical screening exhibited the presence of alkaloids, flavonoids, saponins, phenols, triterpenoids, glycosides, and tannins. GC/MS analysis exhibited 40 compounds, mainly 7-hexadecene, (Z)-, imidazole-4,5-d2, and 1-acetyl-4,4-bis[4-(3-bromopropoxy)-3,5-dimethoxyphenyl] piperidine. The antibacterial and antifungal potentials of the methanolic extract of S. mahagoni (L.) leaves was investigated using the agar well diffusion technique. Potent antibacterial activity against Staphylococcus aureus, Escherichia coli, Salmonella enterica, Enterobacter aerogenes, and Proteus vulgaris and antifungal activity against Aspergillus flavus, Aspergillus niger, and Candida albicans were found. The minimum inhibitory concentration and minimum bactericidal and fungicidal concentrations ranged from 12.5 to 25 mg/mL. Antioxidant activity was studied using the free radical scavenging assay, and the IC50 value of the leaf extract was 69.9 µg/mL. Cytotoxic activity was screened using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, and the IC50 value was 44.2 µg/mL. These findings suggested the importance of this plant in treating some bacterial and fungal infections and cancer.

The present work has been designed to investigate the hepatoprotective and renoprotective efficiency of alcoholic extract of Allium porrum and Bauhinia variegata leaves in thioacetamide-induced toxicity in adult Wistar rats. Allium porrum leaf extract, Bauhinia variegata leaf extract and their combinations were orally administered for 14 days then TAA (300 mg/kg) i.p. was injected once and the rats were sacrificed 2 days later. Plasma AST, ALT, GGT, total bilirubin, creatinine, urea, uric acid, triglyceride, cholesterol, HDL and LDL were measured. Liver MDA, GSH, CAT, SOD and TNF-α were evaluated. Histological examination was performed. The rats treated with TAA showed a significant increase in AST, ALT, GGT, total bilirubin, creatinine, urea, uric acid, total, triglyceride, cholesterol and HDL while it led to a significant decrease in protein and HDL. The treatment of rats with TAA resulted in a significant decrease of the hepatic GSH, SOD and CAT and a significant elevation of MDA and TNF-α. Allium porrum and Bauhinia variegata extracts alleviated the toxic effects of TAA on the liver and the kidney. In conclusion, treatment with Allium porrum and Bauhinia variegata extracts and their combination reduced deleterious effects of TAA on liver through antioxidant and anti-inflammatory properties.

CONTEXT: Cajanus cajan L. (Fabaceae), a food crop, is widely used in traditional medicine. OBJECTIVES: The phytochemical composition of C. cajan seeds and evaluation of the anti-inflammatory, immunomodulatory, antinociceptive, and antioxidant activities were studied. MATERIALS AND METHODS: Unsaponifiable matter and fatty acids were analyzed by GC and GC/MS. The n-butanol fraction was chromatographed on polyamide column. The anti-inflammatory activity of hexane extract (200 and 400 mg/kg, p.o.) was evaluated using the carrageenan-induced rat paw edema at 1, 2, and 3 h. The serum tumor necrosis factor-α, interleukin-6, and immunoglobulin G levels were detected by ELISA. The hexane extract antinociceptive activity was determined by adopting the writhing test in mice. DPPH radical scavenging, total reduction capability, and inhibition of lipid peroxidation of butanol fraction were evaluated. RESULTS AND CONCLUSION: Twenty-one unsaponifiable compounds (mainly phytol, 2,6-di-(t-butyl)-4-hydroxy-4-methyl-2,5-cyclohexadiene-1-one, ß-sitosterol, stigmasterol, and campesterol), as well as 12 fatty acids (primarily 9,12-octadecadienoic and palmitic acids) were identified in hexane extract of C. cajan seeds. n-BuOH fraction contains quercetin-3-O-ß-d-glucopyranoside, orientin, vitexin, quercetin, luteolin, apigenin, and isorhamnetin. For the first time, quercetin-3-O-ß-d-glucopyranoside is isolated from C. cajan plant. The hexane extract (200 and 400 mg/kg) inhibited carrageenan-induced inflammation by 85 and 95%, respectively, 3 h post-carrageenan challenge. This was accompanied by an 11 and 20%, 8 and 13%, respectively, decrease of TNF-α and IL-6, as well as significant decrease in IgG serum levels. Moreover, hexane extract (200 and 400 mg/kg) decreased the number of writhings by 61 and 83%, respectively. The butanol fraction showed DPPH radical scavenging (inhibitory concentration (IC50) value: 9.07 µg/ml).
