RÉSUMÉ

A sensitive chemiluminescence method, based on the enhancive effect of phenobarbital on the chemiluminescence reaction between luminol and dissolved oxygen in a flow injection system, was proposed for the determination of phenobarbital. The chemiluminescence intensity responded to the concentration of phenobarbital linearly ranging from 0.05 to 10 ng x ml(-1) with the detection limit of 0.02 ng x ml(-1) (3 sigma). At a flow rate of 2.0 ml x min(-1), a complete determination of phenobarbital, including sampling and washing, could be accomplished in 0.5 min, offering the sampling efficiency of 120 h(-1) accordingly. The method was applied successfully in an assay of PB for pharmaceutical preparations, human urine and serum without any pretreatment with recovery from 95.7 to 106.7% and RSDs of less than 3.0%.

RÉSUMÉ

A 3-year study of spotted fever group rickettsial ecology in Inner Mongolia revealed that nearly half of the human population tested had antibodies to Rickettsia sibirica detected by complement fixation test. Infected persons, ticks and a high proportion of seropositive livestock and wild rodents were found in all five vegetation zones (desert, steppe, forest, forest-grassland and grassland).

In earlier publications, we have recorded evidence that micro-injection of globin mRNA from rabbit into goldfish eggs leads to the production of rabbit globin in mature red blood cells of goldfish. This paper is concerned with the mechanism of that apparent transfer of genetic information from the cytoplasm to the nucleus. We investigate the possibility that the injected mRNA is reverse transcribed to create a corresponding cDNA in goldfish eggs. By using purified mRNA from rabbit reticulocytes and micro-injection into enucleated and nucleated goldfish eggs, we show that the production of a DNA sequence hybridizes to cloned cDNA of rabbit globin mRNA and appears to become incorporated into the chromosomes of the developing eggs.

RÉSUMÉ

DNAs were isolated from goldfish, and the balancer bearing species of newt. They were injected separately into fertilized goldfish eggs. A balancer developed in some newt-DNA injected goldfish. DNA injected goldfish eggs occasionally developed a non-balancer process. These results demonstrated the newt DNA-mediated transfer of the species character from newt to goldfish. The injected DNA was localized by the method of autoradiography using H3-cDNA. Silver grains were found in areas over the colchicine induced metaphase. The incorporation of foreign DNA into chromosomes provides a material basis for genetic transformation.
