This study aimed to isolate and identify pathogenic bacteria in the intestinal tract, skin, and muscles of Sciades herzbergii; detect histopathological changes in the gill and liver; and use these biomarkers for the assessment of potential risks to human health. Fish were sampled during the rainy and dry seasons at two points in São Marcos Bay, Maranhão, Brazil: Ilha dos Caranguejos (IC) and Porto Grande (PG). Isolation and quantification were carried out using COLItest®. Colonies were subjected to identification and phenotypic investigation of antimicrobial resistance using Vitek®. Gill and liver samples were subjected to routine histological examination. The results indicated the presence of Klebsiella pneumoniae and Escherichia coli, the latter of which showed phenotypic resistance to norfloxacin and gentamicin. Fish caught at PG exhibited more extensive gill and liver damage than fish caught at IC. The findings suggest that histological changes in target organs of S. herzbergii may be influenced by infection with pathogenic bacteria.

In the present study, the objective was to evaluate in situ interaction between Benzo[a]anthracene 7,2-dione 7-oxime (BZA) and calf thymus dsDNA (ct-dsDNA) using electroanalytical genosensor. Analytical techniques based on Ultraviolet/Visible (UV-Vis) spectroscopy and electroanalytical were used to investigate the interaction processes in solution and immobilized on carbon screen-printed electrodes modified with electrochemical mediator Meldola blue. In addition, was possible to evaluate the degree of damage caused to the genetic material by the analyte through of toxicity estimate (S%). The interaction evaluated by genosensor showed processes of intercalation, degradation, and breaks of the double strand of ct-dsDNA, suggesting that the interaction simulates highly toxic (values varying from 0.6 to 0.8 µA in 48 h of interaction), such as 8-oxoguanine (+0.48 V), which is a by-product of guanine oxidation. Furthermore, monitoring A (+1.10 V) after 1 h showed an S% value between 50 and 90%, indicative of high toxicity, and monitoring G (+0.85 V), which showed S>90%, indicated no toxicity after 10 min. Overall, the electroanalytical genosensor developed in a miniaturized system displayed good reproducibility and stability over time being a quick alternative for assesses the degree of toxicity between toxic xenobiotics and biologically electroactive molecules, such as DNA.

In this study, we analyzed biomarkers and heavy metals in Ucides cordatus specimens caught in mangrove areas [A1 = reference (low impact area); A2, A3 and A4 = port complex (potentially impacted area)] of the Amazonian northern coast of Brazil. Sediments and crab muscles sampled from the potentially impacted mangroves showed high levels of metals. Additionally, catalase and glutathione S-transferase enzymes in the gills and hepatopancreas of the collected crabs were elevated in crabs from A2, A3 and A4 relative to those from A1. The histological characteristics of the gills and hepatopancreas differed in crabs collected from A2, A3 and A4 compared with those collected from A1. The crabs were larger and heavier in the rainy season in A1 and A3 and heavier in the dry season in A4. Heavy metal analyses of sediments, crab muscles and biological responses indicated that the animals are subjected to stress levels differentiated in the mangroves due to a gradient (A4>A3>A2>A1) of anthropogenic pressure in São Marcos Bay. In conclusion, the analyses of metals in the sediment and in the muscles of crabs, as well as the biochemical biomarker and histological analyses, suggest that crabs from mangroves in the port complex region are subject to pollutants that compromise their health.

The current study combined chemical data on trace elements and polycyclic aromatic hydrocarbons (PAH) from sediment and used a multibiomarker approach in the catfish Sciades herzbergii to evaluate two different sites in São Marcos Bay, Brazil. Higher diffuse contaminations by trace elements and PAH were detected in the sediment of Porto Grande (PG) harbor than in the Ilha dos Caranguejos (IC) reference area. A multibiomarker was used in catfish to evaluate the bioavailability of PAH in bile and the effects of pollutants in target tissues. The parameters considered were oxidative stress biomarkers (SOD, CAT, GSH, GST and LPO) and histopathological alterations and were compared between two seasons. The biological responses revealed adverse effects on the population, as indicated by the presence of trace elements and PAH as stressors. Principal component analysis (PCA) of the biomarkers corroborated these results and indicated that fish from the PG site during the rainy season in 2019 exhibited many biological effects compared to 2018. Overall, the present study showed that environmental contamination increased over the years and provides information on the contamination of sediments in the São Marcos Bay, Brazil. The results showed that the presence of contaminants was correlated with the health status of the catfish S. herzbergii.

Nowadays biomonitoring programs can benefit with mathematical models able to correlate biomarkers to monitor water pollution. The aim of this study was to develop a screening test based on hematological parameters and histological lesions in tambaqui (Colossoma macropomum), to allow the assessment of environmental impacts on fish inhabiting a protected area in Maranhão inside of Brazilian Amazon. Samples collected during three years (2012, 2013 and 2014) were grouped by season (dry and rainy) Water samples were also collected for physical chemistry analysis. Blood samples were stained with Acridine Orange to detect micronuclei and erythrocyte abnormalities. Gill tissues were stained with hematoxylin and counterstained with alcoholic eosin, and histopathological lesions were scored on a scale of 1-3, being 1 = minimal pathological importance, 2 = moderate pathological importance and 3 = marked pathological importance. A screening test for evaluating environmental impact was developed by fitting the measured data (necrosis, erythrocyte abnormalities, number of micronuclei) from tambaqui. A three-dimensional surface was fit to the empirical data. Our proposed model predicted the probability of necrosis (observed in euthanized animals) based on the numbers of micronuclei and abnormal erythrocytes (observed in blood samples from live animals) (correlation coefficient R = 0.89). The methodology could be applied for predicting contamination histories (chronic pollution that induces branchial lesions) in rivers using the micronucleus and erythrocyte abnormalities of the fishes (with a simple blood sample).

Catfish have been used as a model system for studying biochemical mechanisms of biotransformation. The main goal of this study was to identify alterations in hepatic glutathione S-transferase (GST) activity and changes in the parental care pattern of a mouth-brooding catfish, Sciades herzbergii, as a biomarker of anthropogenic impact in a port area on the northeastern coast of Brazil. The fish were sampled from a natural reserve (A1 = reference site) and from an industrialized port area (A2 = impacted site). Two analyses were carried out: hepatic GST activity and mouth-brooding behavior of males. Catfish collected from the A1 site displayed all stages of gonadal maturation, and some of the adult males were mouth brooding 12-30 embryos. Not all gonadal maturation stages of the catfish were represented at the A2 site, and no mouth-brooding males were observed. GST activity in the liver of S. herzbergii was significantly higher in fish from the impacted site compared with fish from the reference site. Values for the enzymatic activity increased progressively in fish sampled from the reserve area as they became more reproductively mature (immature ≤ maturing ≤ mature ≤ spent). However, the greatest values for GST activity (2.84 ± 0.31 µmol min-1 mg protein-1) among fish sampled from the impacted area were found in (immature) juveniles. These data suggest that changes in hepatic GST activity and mouth-brooding behavior of S. herzbergii can be used as biomarkers of anthropogenic impact.

Genotoxic and hematological parameters in tambaqui (Colossoma macropomum), a native freshwater fish, were used as biomarkers to assess exposure to environmental stressors within the Maracanã Protected Area of Maranhão State, Brazil. Fish were sampled at two sites-Serena Lagoon (control) and Ambude River-on four occasions (dry and rainy season), and biometric data (length and weight) recorded and blood collected from all fish for analysis. Erythrocyte indices-mean corpuscular volume, mean corpuscular hemoglobin, and mean corpuscular hemoglobin concentration-were calculated. Blood samples were stained with Giemsa and acridine orange, and microscopically examined for micronuclei and morphological nuclear abnormalities. Micronuclei were observed in fish from both sites, although the frequency was significantly higher in fish from the Ambude River and morphological nuclear abnormalities were only observed in fish from the Ambude River. More morphological nuclear abnormalities and a larger number of micronuclei were observed in erythrocytes stained with acridine orange compared with those stained with Giemsa. On average, erythrocyte indices were lower in fish from the Ambude River than from the Serena Lagoon. The results confirm that genotoxic and hematological parameters in C. macropomum can be used as indicators of environmental health and could be valuable tools for monitoring environmental conditions within protected areas.
