RÉSUMÉ

Background/Aims@#This study aimed to investigate the biological function and regulatory mechanism of TCN1 in colorectal cancer (CRC). @*Methods@#We studied the biological function of TCN1 by performing gain-of-function and loss-offunction analyses in HCT116 cell lines; examined the effects of TCN1 on the proliferation, apoptosis, and invasion of CRC cells; and determined potential molecular mechanisms using HCT116 and SW480 CRC lines and mouse xenotransplantation models. Tumor xenograft and colonization assays were performed to detect the tumorigenicity and metastatic foci of cells in vivo. @*Results@#TCN1 knockdown attenuated CRC cell proliferation and invasion and promoted cell apoptosis. Overexpression of TCN1 yielded the opposite effects. In addition, TCN1-knockdown HCT116 cells failed to form metastatic foci in the peritoneum after intravenous injection. Molecular mechanism analyses showed that TCN1 interacted with integrin subunit β4 (ITGB4) to positively regulate the expression of ITGB4. TCN1 knockdown promoted the degradation of ITGB4 and increased the instability of ITGB4 and filamin A. Downregulation of ITGB4 at the protein level resulted in the disassociation of the ITGB4/plectin complex, leading to cytoskeletal damage. @*Conclusions@#TCN1 might play an oncogenic role in CRC by regulating the ITGB4 signaling pathway.

RÉSUMÉ

In this study, we investigated the inhibition of spontaneous canine benign prostatic hyperplasia by a crude polysaccharide fraction extracted from Urtica fissa roots and stems. After oral administration of U. fissa polysaccharide fraction for 3 months, the dog prostatic volume reduced significantly when compared to that before treatment using CT examination. The high-dosage U. fissa polysaccharide fraction (120 mg/kg body weight/day) and finasteride (0.5 mg/kg body weight/day) treatments showed both almost 30 % reduction of the initial prostatic volume. At the end of the administration of U. fissa polysaccharide fraction, the prostates were excised, and the volumes were measured by water displacement. The prostatic volume showed significant decrease by 11 %, 15 %, and 21 % for the 30, 60, and 120 mg/kg/day U. fissa polysaccharide fraction treatment groups, respectively, compared to the control group. Histological observation found that U. fissa polysaccharide fraction inhibited the dog prostatic epithelial cells proliferation and enlarged glandular lumen diameter. The crude polysaccharide fraction of U. fissa is a possible new candidate for the treatment of benign prostatic hyperplasia.

RÉSUMÉ

When confronted with a real-world problem, heuristic knowledge and experience can guide the solution of a specific technical problem as the key step toward innovation. In particular, a heuristic prototype must be used correctly to cue the technical problem that exists in a particular situation. The present study selected an innovative paradigm and scientific innovation materials to investigate the neural basis of insight induced by heuristic prototypes using event-related functional magnetic resonance imaging (fMRI). The day prior to undergoing fMRI scanning, participants were asked to solve 42 difficult technical problems that scientists might have already encountered but were unknown to the participants. In the subsequent fMRI experiment, the same participants were randomly presented with 84 prototypes classified into two types: related prototypes (RPs), which were useful for solving previously encountered problems, and unrelated prototypes (UPs), which sometimes did not contribute to problem solving. While being scanned, participants were asked to assess whether a prototype is relevant to any of the technical problems. This study comprised two conditions: solving technical problems when presented with a related heuristic prototype and failing to solve technical problems using unrelated heuristic prototypes. The authors assumed that the regions significantly activated by the RP condition, compared with the UP condition, reflected brain activity related to the role of heuristic prototypes in scientific insight. fMRI data showed that the left dorsolateral prefrontal gyrus (left DLFPC, BA9) and the left angular gyrus (left AG, BA39) were more significantly activated when presented with RPs than with UPs. The results suggest that the DLPFC may be involved in the automatic retrieval of technical problems and breaking of mental sets. Moreover, the left AG may be involved in forming novel associations between technical problems and related prototypes.

RÉSUMÉ

Heuristics through the application of heuristic knowledge to the creation of imitation devices may be one of the most common processes in scientific innovation. In particular, heuristics suggests that innovation includes the automatic activation of heuristic knowledge and formation of novel associations between heuristic knowledge and problem situations. In this study, 76 scientific innovation problem situations were selected as materials. Among these, 36 contain related heuristic knowledge and 40 have no such information. Through functional magnetic resonance imaging, the learning-testing paradigm was used to explore the brain mechanisms of scientific problem finding inspired by heuristic knowledge. Participants were asked to find a problem on the basis of a given innovation problem situation. Two scenarios were presented: finding scientific problems with related heuristic knowledge and finding conventional problems without related heuristic knowledge. The authors assumed that the regions in the brain significantly activated by the finding scientific problems with related heuristic knowledge condition compared with the finding normal problems without related heuristic knowledge condition are relevant to the brain mechanisms of scientific problem finding inspired by heuristic knowledge. The first scenario more significantly activated the left precuneus and left angular gyrus than did the second scenario. These findings suggest that the precuneus is relevant to the successful storage and retrieval of heuristic knowledge and that the left angular gyrus is involved in the formation of novel associations between heuristic knowledge and problem situations for finding scientific problems.

RÉSUMÉ

Multidrug resistance (MDR) of tumor cells is one of the major problems encountered during cancer chemotherapy. In this paper, we isolated eight triterpenoids from Poria cocos and evaluated their effects on reversing MDR of KBV200 cells. Eight triterpenoids increase significantly vincristine-induced cytotoxicity in drug-resistant KBV200 cells at the concentrations of 12.5 µg/mL and 25 µg/mL. Dehydrotumulosic acid showed the best reversal effect: it increased KBV200 apoptosis induced by vincristine and inhibited P-gp function through enhancing the accumulation and retention of fluorescent P-gp substrate rhodamine 123 in KBV200 cells but had no effect on P-gp expression.

Objective To gain more pathophysiolgic knowledge about acute obstructive hydrocephalus and to explore its rapid and effective treatment by establishing canine acute obstructive hydrocephalus model. Methods Acute obstructive hydrocephalus model was established by injecting cyan-acrylic gel glue into the fourth ventricle via posterior fosse craniotomy in 9 male adult mongrel dogs.At the same time, lateral ventricle catheterization were performed and were fixed on the scalp to connect reservoir bag so that the changes of intracranial pressure (ICP) could be measured dynamically, and the changes of neurological function were observed. Results Acute obstructive hydrocephalus model was successfully established in 6 of the total 9dogs.ICP was (48.2 ± 6.1 ) cm H2 O at 48 hours after the injection and was (56.4 ± 5.7 ) cm H2 O at 72 hours after the injection,it increased 392% and 459 % respectively. And the ICP after injection was significantly different(P < 0.01 )compared with that before injection (12.3 ± 3.1 )cm H2O.Conclusion The establishment of acute obstructive hydrocephalus model has high success rate, and is easy to reduplicate; ICP could be measured dynamically and also could be reduced by releasing CSF;Thus, ventricular drainage is the most rapid and effective treatment for acute obstructive hydrocephalus.

RÉSUMÉ

Plasmid DNA for biopharmaceutical applications is produced easily in Escherichia coli bacteria. The cell lysis is the most crucial step for purification of plasmid DNA. In this paper, we describe a continuous cell alkaline lysis, neutralization, and clarification combination process for production of plasmid pUDK-HGF using hollow fiber ultrafiltration column as a lysis chamber and compare the plasmid DNA yield and homogeneity with the T-connector and manual processes, respectively. The results show that the plasmid pUDK-HGF yield of the combination process is 13% higher than manual lysis, twice higher than using T-connector. When the proportion of lysed cells and neutralization solution is 3:1, the plasmid pUDK-HGF yield can improve by 70%. This process could be easily scaled up to meet the industrial scale for cell lysis.
