In the present study, we have evaluated the cytotoxic activity of 282 extracts from 72 native plant species of the Brazilian Atlantic Forest biome. As a result, Casearia arborea and Sorocea hilarii leaves extracts showed cytotoxic activity against three tumour cell lines tested (B16F10, SW480 and Jurkat). After bioassay-guided fractionation, the bioactive fractions were submitted to the dereplication study via High-performance Liquid Chromatography, connected to High-resolution Mass Spectrometry (HPLC-ESI-QTOF/MS) analysis, combined with a Global Natural Products Social Molecular Networking (GNPS) tool. A combination of bioactivity-guided and dereplication approaches resulted in the putative annotation of 27 clerodane diterpenes and 9 flavonoids as main compounds present in the cytotoxic fractions of C. arborea. Regarding the active fraction of S. hilarii, 10 megastigmans, 17 spirostane steroids derivatives and 2 lignans were putatively identified. In conclusion, Casearia arborea and Sorocea hilarii are potential sources of antitumor compounds.

Donkeys are a public health concern in the Northeast region of Brazil, with thousands of stray animals. Orchiectomy is an important population control measure; however, the long postoperative period with daily treatment of open wounds in the scrotum makes it difficult to perform a large number of castrations in sheltering centers. We evaluate a novel surgical procedure for orchiectomy in donkeys using parascrotal access. Twelve donkeys were used, divided into two groups: I - submitted to orchiectomy through parascrotal surgical access (novel procedure), and II - submitted to orchiectomy through scrotal access (conventional). Postoperative evaluations consisted of a macroscopic evaluation of the surgical wound (bleeding and intensity of edema), hematological parameters, and peritoneal fluid, which occurred in both groups at the moments (M): M0 - before the surgical procedure. The others moments occurred after surgery: M12 (twelve hours); M24 (twenty-four hours); M48 (forty-eight hours); M72 (seventy-two hours); M8D (eight days); and M16D (sixteen days). The surgical techniques did not generate an important systemic inflammatory response to the point detected by the leukogram, fibrinogen dosage, and peritoneal fluid. The parascrotal technique required long surgery but promoted less bleeding, less edema, and faster healing. The techniques used did not promote sufficient systemic inflammation to alter the number of leukocytes and the fibrinogen concentration; however, evaluation of the peritoneal fluid proved to be important for evaluating inflammatory processes involving the scrotum and inguinal canal. We describe a novel surgical procedure for orchiectomy in Donkeys using a parascrotal access that promoted less risk of bleeding, shorter period of edema, and healing time, but required longer surgery time.

Marek's disease is an important neoplastic disease in birds caused by a serotype 1 specific herpesvirus; it is controlled by vaccination. In commercial breeders and layers in Brazil, current vaccination programs use the combination of attenuated or non-pathogenic strains of the HVT virus (turkey herpesvirus - serotype 3) and CVI 988 (Rispens - serotype 1). The combination of serotype 3 and 1 it has been an important and effective control strategy through the vaccination of long-lived birds. In addition, more recently the recombinant rHVT strain (vectorized vaccines) has been used in some vaccine programs. This study’s main objective was to compare CVI and HVT components’ replication in feather tips in three different Marek's disease (MD) immunoprophylactic programs (T01 – program A, T02 – program B and T03 – program C). Quantification of these two vaccine strains was performed by real-time PCR in samples collected at the ages of 14, 21, and 28 days. At 14 days, mean of log[cvi] in program B was significantly higher than C (p<0.05). For mean of log[hvt], at 28 days, program B was significantly higher than A (p<0.05). For proportion of positives, at 14 days, program B had 2.7 times more risk to be positive in CVI than program C (p<0.001). At 21 days, program B had 1.7 times more risk to be positive in CVI than program C (p=0.005). For HVT, at 28 days, program B had 3.2 times more risk to be positive than program A (p=0,009). Results showed significant differences between the treatments evaluated. In general the conventional combination Marek’s vaccine containing CVI+HVT (program B) showed higher replication rate and percentage of vaccine coverage than the programs with rHVT vector vaccines (program A and C).


A doença de Marek é uma importante doença neoplásica das aves causada por um herpesvírus específico do sorotipo 1 e seu controle se faz por vacinação. Em reprodutoras e poedeiras comerciais do Brasil, os programas de vacinação utilizam a combinação de estirpes atenuadas ou não patogênicas do vírus HVT (turkey herpesvirus - sorotipo 3) e CVI 988 (Rispens - sorotipo 1). A combinação do sorotipo 3 e 1 tem sido uma importante e efetiva estratégia de controle para aves de vida longa. Além disso, mais recentemente a fração rHVT recombinante (vacinas vetorizadas) vem sendo utilizada em alguns programas vacinais. O objetivo deste estudo foi comparar a replicação das frações CVI e HVT no folículo da pena em três programas imunoprofiláticos distintos (T01 – programa A, T02 – programa B e T03 – programa C). A quantificação das duas estirpes vacinais foi realizada por PCR em tempo real nas amostras colhidas nas idades de 14, 21 e 28 dias. Aos 14 dias, em média, log[cvi] do programa B foi significativamente maior do que C (p<0.05). Aos 21 e 28 dias, a média do log[cvi] do programa C foi significativamente menor do que A e B (p<0.05). Para log[hvt], aos 28 dias, a média do programa B foi significativamente maior do que A (p<0.05). Para proporção de positivos, aos 14 dias, o programa B teve 2,7 vezes mais risco de ter positivos no CVI do que C (p<0.001). The objective of this study was to analyze the diversity and discrimination of high-performance Brazilian rice cultivars using microsatellite markers. Twenty-nine rice cultivars belonging to EMBRAPA Arroz e Feijão germplasm bank in Brazil were genotyped by 24 SSR markers to establish their structure and genetic discrimination. It was demonstrated that the analyzed germplasm of rice presents an expressive and significant genetic diversity with low heterogeneity among the cultivars. All 29 cultivars were differentiated genetically, and were organized into two groups related to their upland and irrigated cultivation systems. These groups showed a high genetic differentiation, with greater diversity within the group that includes the cultivars for irrigated system. The genotyping data of these cultivars, with the morphological e phenotypical data, are valuable information to be used by rice breeding programs to develop new improved cultivars.

RESUMO Este trabalho objetivou verificar se os profissionais de nível superior, ligados a Estratégia de Saúde da Família (ESF) do Município de Petrolina-PE percebem a importância, utilização e indicações de Plantas Medicinais e Fitoterápicos. Trata-se de um estudo transversal de caráter exploratório e descritivo, no qual participaram 96 profissionais de nível superior locados em Unidades da ESF. Os dados foram obtidos através de entrevista individual utilizando questionário semiestruturado. Para análise estatística, foi utilizado o teste Pearson Qui quadrado. As plantas medicinais mais citadas foram o Boldo (Plectranthus barbatus A.) e a Camomila (Matricaria recutita L.). Os fitoterápicos foram indicados apenas por 5 (5,2%) médicos e os mais citados foram Guaco® e Maracugina®. 36 (37,5%) dos profissionais acham que não estão preparados para repassar as informações sobre o uso de plantas medicinais e apenas 35 (36,5%) se sentem preparados para prescrever fitoterápicos. Observa-se a necessidade de capacitação e motivação desses profissionais para a utilização correta e segura das Plantas Medicinais e Fitoterapia.


ABSTRACT This study aimed to determine if the top-level professionals of the Strategy of Health of the Family, perceive the importance, use and indications of phytotherapy and Medicinal Plants. This is a cross-sectional, exploratory and descriptive study, with the participation 96 professionals serving in the family health strategy units. Data were collected through individual interviews in a semi-structured questionnaire. For statistical analysis, we used the Pearson Chi-square test. The most cited medicinal plants were the Boldo (Plectranthus barbatus A.) and Chamomile (Matricaria recutita L.). The phytotherapics were indicated only for 5 (5.2%) physicians and the most cited were Guaco® and Maracugina®. 36 (37.5%) of the professionals think are not prepared to pass on the information about the use of medicinal plants and only 35 (36.5%) feel prepared to prescribe phytotherapy. One observes the need for training and motivation of these professionals for the correct and safe use of Medicinal Plants and phytotherapy.

Phyla scaberrima (Verbenaceae) is a herbaceous perennial species that is distributed from Mexico (center of origin) to Colombia, growing in forest and swamp edges or grasslands from sea level up to an altitude of 1800 m. The chemical properties and uses in popular medicine have drastically affected the population size of this species. In this study, we investigated genetic variability in populations of P. scaberrima using AFLP markers. Three AFLP primer combinations rendered a total of 997 markers in a sample of 131 individuals from five populations, including two populations from Mexico and three from Colombia. The average percentage of polymorphic loci, gene diversity and Shannon-Wiener index were 46.62, 0.0695, and 0.119, respectively. Analysis of molecular variance showed that the distribution of the genetic variability within populations (85.41%) was higher than between groups (8.11%) and between populations (6.48%). Principal coordinate analysis and Bayesian analysis for the K number of clusters showed that the individuals were dispersed in five (K= 5) clusters. The low levels of genetic diversity observed in these populations demonstrated that the populations from Mexico and Colombia need urgent management to recover their genetic variability.

Chrysophyllum gonocarpum is a tropical tree species that is very important in the recovery of heterogeneous forests and of degraded areas of permanent preservation. We identified microsatellite loci for C. gonocarpum to assess the genetic variability and the patterns of the population structure of the species. We isolated 8 microsatellite primers by using CT- and GT-enriched genomic libraries. We detected 2-4 alleles with 2.9 alleles per locus on average, by polymerase chain reaction. Test for cross-amplification showed that some loci were successfully amplified in 2 other Chrysophyllum species. The microsatellites can be used to assess the genetic diversity and population structure of C. gonocarpum. Some primer pairs can be amplified in C. marginatum and C. splendens.

Parapiptadenia rigida, locally known as angico, is a tropical tree common in the semideciduous Brazilian forest. Its wood is naturally resistant to insect attack and is useful for construction. Extracts from the tree have medicinal properties. We characterized nine microsatellite loci for P. rigida. Thirty-five alleles were detected in a sample of 45 individuals from 3 different populations, with an average of 3.9 alleles per locus. The average polymorphic information content ranged from 0.099 to 0.640. Observed and expected heterozygosities varied from 0.111 to 0.489 and from 0.106 to 0.707, respectively. One locus exhibited significant deviation from Hardy-Weinberg equilibrium and four pairs of loci showed significant linkage disequilibrium. All nine primers were tested for cross-amplification in species from the Fabaceae-Mimosoidea family, yielding a transferability success rate of 7 loci in Stryphnodendron adstringens to 0 transferred loci in Pithecellobium incuriale and Inga marginata. These microsatellites will be valuable to study population genetics of this and other species where primer transferability was detected.

Macaúba (Acrocomia aculeata) is a palm of economic importance, widely distributed in natural forests from Mexico to Uruguay. We analyzed the genetic diversity of populations of macaúba (A. aculeata) in the northern region of the State of Minas Gerais, Brazil. Young leaves from 10 macaúba individuals encompassing 49 genotypes of macaúba were collected from Montes Claros, Itacambira, Brasília de Minas, Mirabela, and Grão Mogol. After extraction and amplification of samples, the amplified fragments were separated by electrophoresis. We found high levels of genetic diversity within the populations. Genetic diversity indices were high, except in the Itacambira and Mirabela populations. Results show that Mirabela and Itacambira populations can require conservation strategies because they present lower values of genetic diversity.

O objetivo deste estudo foi avaliar os efeitos da utilização de diferentes ingredientes fibrosos nas dietas sobre o desempenho, a composição corporal e a morfometria intestinal de juvenis de pacu. Foram avaliadas cinco dietas isoproteicas (23 por cento de proteína digestível), isoenergéticas (3250kcal de energia digestível/kg) e isofibrosas (9 por cento de fibra bruta), sendo a principal fonte fibrosa de cada constituída por farelo de soja, casca de soja, farelo de girassol e polpa cítrica; esta última em dois níveis de inclusão (30 e 45 por cento). Foram utilizados 300 juvenis de pacu (25,12±0,78 gramas), alojados em 25 aquários (200 litros). Os melhores resultados de crescimento e conversão alimentar foram obtidos com as dietas contendo farelo de soja e farelo de girassol. As dietas contendo casca de soja e polpa cítrica prejudicaram o desempenho dos juvenis de pacu, e o efeito negativo foi acentuado com o aumento da inclusão de polpa cítrica. Além disto, somente nos peixes alimentados com a dieta com 45 por cento de polpa cítrica foi observada diminuição na densidade de vilosidades por área no epitélio intestinal. A partir dos resultados obtidos, pode-se concluir que alguns ingredientes fibrosos podem afetar negativamente o desempenho e as características do epitélio intestinal de juvenis de pacu.


The aim of this study was to evaluate the effects of diets containing different fiber sources on the performance, corporal composition and intestinal morphometry of pacu juveniles (Piaractus mesopotamicus). This study evaluated five isoproteic (23 percent digestible protein), isoenergetic (3250kcal digestible energy/kg) and isofibrous (9 percent crude fiber) diets, each one containing a fiber source such as soybean meal, soybean hulls, sunflower meal and citric pulp in two levels of inclusion (30 and 45 percent). A total of 300 pacu juveniles (25,12±0,78g) were stocked in 25 experimental aquariums (200 liters) equipped with aeration and heating system. The best growth and feed conversion results were obtained with diets containing soybean meal and sunflower meal. Diets containing soybean hulls and citrus pulp worsened the juvenile pacu performance. Moreover, only the diet containing 45 percent citrus pulp decreased the villous number per area in the intestinal epithelium. From the results we can conclude that some fibrous ingredient may affect juvenile pacu performance, corporal composition and intestinal epithelium characteristics.

Studies on intraspecific variation can contribute to the development of conservation strategies by identifying units of conservation for threatened species. Aspidosperma polyneuron is a tropical tree of seasonal semideciduous forests that is currently endangered and protected because it has been heavily logged for lumber, although it was once common in Brazil and neighboring countries. We investigated genetic structure in two samples of A. polyneuron collected from steep hillsides and from flat areas of a natural forest fragment in northern Paraná State, Brazil. Seven AFLP primer combinations yielded 200 markers, with a polymorphic rate of 88.5% for samples from the flat area and 99% for samples from the high declivity area. Total genetic diversity (H(T)) was 0.387, while the genetic diversity within the populations (H(S)) was 0.307 and 0.372, for samples from the flat and the high declivity areas, respectively. Genetic differentiation between samples was high, with a mean F(ST) of 0.265 and a genetic distance of 0.148, indicative of a high degree of genetic structure over a short distance. Principal coordinate analysis separated the samples into three groups of individuals; the first group included individuals from the high declivity area, the second group consisted of individuals only from the flat area, and the third group had individuals from both areas. Bayesian analysis also showed K = 3 clusters. The unexpected high level of intraspecific variation of A. polyneuron in this small forest fragment should be taken into account when evaluating the genetic impact of forest degradation on this species in other semideciduous forest fragments.

Knowledge of the effects of flooding on plant survival is relevant for the efficiency of management and conservation programs. Schinus terebinthifolius is a tree of economic and ecological importance that is common in northeast Brazil. Flooding tolerance and genetic variation were investigated in two riparian populations of S. terebinthifolius distributed along two different ecological regions of the Tibagi River basin. Flooding tolerance was evaluated through the investigation of young plants, submitted to different flooding intensities to examine the morphological and anatomical responses to this stress. The growth rate of S. terebinthifolius was not affected by flooding, but total submersion proved to be lethal for 100% of the plants. Morphological alterations such as hypertrophied lenticels were observed in both populations and lenticel openings were significantly higher in plants from one population. Genetic analysis using DNA samples obtained from both populations showed a moderate degree of genetic variation between populations (13.7%); most of the variation was found within populations (86.3%). These results show that for conservation purposes and management of degraded areas, both populations should be preserved and could be used in programs that intend to recompose riparian forests.

The aim of this study was to evaluate the effects of diets containing different fiber sources on the performance, corporal composition and intestinal morphometry of pacu juveniles (Piaractus mesopotamicus). This study evaluated five isoproteic (23% digestible protein), isoenergetic (3250kcal digestible energy/kg) and isofibrous (9% crude fiber) diets, each one containing a fiber source such as soybean meal, soybean hulls, sunflower meal and citric pulp in two levels of inclusion (30 and 45%). A total of 300 pacu juveniles (25,12±0,78g) were stocked in 25 experimental aquariums (200 liters) equipped with aeration and heating system. The best growth and feed conversion results were obtained with diets containing soybean meal and sunflower meal. Diets containing soybean hulls and citrus pulp worsened the juvenile pacu performance. Moreover, only the diet containing 45% citrus pulp decreased the villous number per area in the intestinal epithelium. From the results we can conclude that some fibrous ingredient may affect juvenile pacu performance, corporal composition and intestinal epithelium characteristics.


O objetivo deste estudo foi avaliar os efeitos da utilização de diferentes ingredientes fibrosos nas dietas sobre o desempenho, a composição corporal e a morfometria intestinal de juvenis de pacu. Foram avaliadas cinco dietas isoproteicas (23% de proteína digestível), isoenergéticas (3250kcal de energia digestível/kg) e isofibrosas (9% de fibra bruta), sendo a principal fonte fibrosa de cada constituída por farelo de soja, casca de soja, farelo de girassol e polpa cítrica; esta última em dois níveis de inclusão (30 e 45%). Foram utilizados 300 juvenis de pacu (25,12±0,78 gramas), alojados em 25 aquários (200 litros). Os melhores resultados de crescimento e conversão alimentar foram obtidos com as dietas contendo farelo de soja e farelo de girassol. As dietas contendo casca de soja e polpa cítrica prejudicaram o desempenho dos juvenis de pacu, e o efeito negativo foi acentuado com o aumento da inclusão de polpa cítrica. Além disto, somente nos peixes alimentados com a dieta com 45% de polpa cítrica foi observada diminuição na densidade de vilosidades por área no epitélio intestinal. A partir dos resultados obtidos, pode-se concluir que alguns ingredientes fibrosos podem afetar negativamente o desempenho e as características do epitélio intestinal de juvenis de pacu.

Investigou-se os efeitos de algumas vitaminas B associadas ao diclofenaco ou nimesulida, na contorção abdominal em camundongos e edema de pata em ratos apenas com a nimesulida ou sua associação com algumas vitaminas B.Verificou-se que o pré-tratamento com as vitaminas tiamina[B1], piridoxina[B6] e cianocobalamina [B12], isoladasou administradas conjuntamente, não inibiu o edema de pata produzido pela carragenina em ratos, nem as contorções abdominais produzidas pelo ácido acético em camundongos. Por outro lado, a nimesulida [5mg/kg]reduziu o edema de pata em ratos, e a associação das três vitaminas à este fármaco, mas não as vitaminas administradas isoladamente, potencializou seus efeitos nos tempos de duas, três e quatro horas após a carragenina. As contorções abdominais em camundongos foram inibidas pelas doses de 25 ou 50mg/kg de diclofenaco. A associação das três vitaminas ou apenas da cianocobalamina, potencializou as duas doses do diclofenaco utilizadas. As contorções abdominais foram reduzidas também pelo nimesulida [5mg/kg] e a associação das três vitaminas, oucada uma das vitaminas administradas isoladamente, foram capazes de potencializar os efeitos do nimesulida. É provável que a diferença no mecanismo de ação destes fármacos sejaresponsável pela diferença dos efeitos das vitaminas. O presente trabalho aponta para a realização de novos estudos com o uso destes antiinflamatórios, combinados com as vitaminas tiamina[B1], piridoxina[B6] e cianocobalamina [B12], em doenças inflamatórias crônicas, na tentativa dereduzir a dose destes fármacos e consequentemente diminuindo seus efeitos colaterais.


The effects of a combination of some B vitamins and diclofenac or nimesulide on chemical nociception in mice or paw edema in rats were investigated. While the vitamins alone had no effect, combination of thiamine (B1), pyridoxine (B6) and cyanocobalamin (B12), given i.p. in doses of 100mg and 5mg/kg, respectively, potentiated the inhibition by nimesulide (5mg/kg) of paw edema induced by carrageenin in rats. Antinociceptive effects of diclofenac and nimesulide (inhibition of abdominal writhing induced by acetic acid in mice) were also potentiated by the combination of the vitamins B1, B6 and B12. Thiamine, pyridoxine and cyanocobalamin given singly were effective in potentiating antinociceptive effects of nimesulide, but only cyanocobalamin potentiated these effects of diclofenac, probably reflecting the differing mechanisms of action of the two drugs. The results document the positive influence of B vitamins on the antinociceptive effects of diclofenac or nimesulide and support the use of B vitamins to shorten the treatment time and reduce the daily dose of anti-inflammatories.

Os "timbós verdadeiros" (plantas do gênero Derris), originários da Amazônia Brasileira, tem demonstrado importância crescente por produzirem uma classe de compostos flavonoídicos relacionados à rotenona, que possuem atividade tóxica para peixes e mamíferos. Neste estudo foi determinado a dose letal 50% (DL50) do extrato alcoólico do pó de Derris spp para três espécies de peixes filogeneticamente diferentes e um mamífero roedor (rato). As DL50 de 2,6 microgramas/ml para Collosoma macropomum (tambaqui), 4,8 microgramas/ml para Oreochromis niloticus (tilápia), 14,2 microgramas/ml para Plecostomus sp (cascudo) e DL50 de 100,0 mg/kg para Rattus norvegicus (rato) denotam acentuadas diferenças entre os valores de DL50, principalmente entre os peixes e o rato. Isto possivelmente é devido a fatores farmaco-cinéticos que se relacionam com as diferentes barreiras teciduais encontradas pelos rotenóides quando administrados pela via oral em mamíferos

True "timbós" from the Brazilian Amazon region, plants belonging to the genus Derris of the Leguminosae family, have there considerable importance, because they contain rotenoids which are toxic to fish and mammals. The aim of the study was to establish the lethal doses (LD50) for three phylogenetical different species of fish and also for the rat. The lethal dose for Collosoma macropomum (tambaqui) was 2.6 mg/ml, for Oreochromis niloticus (tilápia) 4.8 mg/ml, for Plecostomus sp (cascudo) 14.2 mg/ml and for Rattus norvegicus 100 mg/kg. The large differences between the lethal doses for the fish and the one for the rat are possibly due to different barrier tissues against the action of rotenoids in the mammal when given orally.
