RÉSUMÉ

This research communication addresses the hypothesis that double premilking teat disinfection (DD) is more effective in reducing soiling and bacterial count on the cows' teat skin and milkers' gloves than conventional disinfection (CONV). The design was a 3 × 3 Latin square (three groups of cows and three treatments) with conventional teat disinfection (CONV, lactic acid application after forestripping), double teat disinfection using the same disinfectant (DD1D, lactic acid application before and after forestripping), and double teat disinfection using two different disinfectants (DD2D, application of lactic acid before and chlorine-based disinfectant after forestripping). All groups were assigned for six days for each treatment, and the evaluations and samples were collected on the last day, before and after treatment. We evaluated the teat cleanliness score (TCS), count of Gram-negative bacteria (coliforms and noncoliforms), Staphylococcus spp., Streptococcus spp., and total bacterial count (TBC) on the cows' teats and TBC on the milkers' gloves. TCS after premilking was lower in DD than CONV treatment. The reduction of Staphylococcus spp. count was greater in the DD treatment and tended to be higher in the DD2D. The TBC reduction on the cows' teats was greater in the DD treatments. The TBC on the milkers' gloves was lower for DD before and after premilking. In conclusion, DD can be an alternative for reducing some bacterial populations on cow teats and preventing the transmission of microorganisms between cows via the milkers' hands.

RÉSUMÉ

This study aimed to compare the feed efficiency (FE) and physiological parameters of Holstein and crossbred Holstein × Simmental cows in a confinement system during winter and summer. The study was conducted in a dairy farm in southern Brazil by including a total of 48 multiparous cows. The cows were studied for 21 days in two periods, summer and winter, and their daily dry matter intake (DMI), milk yield (MY), rectal temperature (RT), respiratory rate (RR), body weight, and body condition score were recorded. An analysis of variance was conducted using the SAS statistical package. The results showed that crossbred Holstein × Simmental cows have a similar FE as Holstein cows in a high-production system (1.83 × 1.81 kg DMI/kg MY, respectively), and they can achieve the same production levels as purebred Holstein cows (43.8 vs. 44.5 milk/cow/day). Our findings indicated a difference for the period as both genetic groups achieved higher FE in winter than in summer (1.98 vs. 1.67 DMI/kg MY, respectively). In addition, we found evidence that crossbred cows are better at dissipating body heat during heat-stress situations, as they have higher RR in summer compared to purebred cows, while Holstein cows have higher RT in summer afternoons than crossbred cows. Therefore, using crossbred Holstein × Simmental cows is an alternative for high-production systems.

RÉSUMÉ

Technical regulations that determine milk quality standards in Brazil establish the minimum criteria that the processing industry's raw material must present. These rules can challenge the logistics sector (transport) to maintain adequate milk conservation levels. We aimed to evaluate the influence of bulk transport and storage conditions on refrigerated raw milk quality. The experiment was carried out in a dairy industry in Santa Catarina State. Information and samples of refrigerated raw milk were collected from dairy farms, from isothermal compartments of milk trucks, and from the industry's storage silo. These samples were submitted to microbiological analysis, somatic cell count(SCC), and physicochemical analysis. The results were evaluated by ANOVA and multivariate analysis (factor analysis). The transport of raw milk in bulk and the transfer of raw material to the industrial silo worsened the milk's microbiological quality (p < 0.0001) for standard plate count (SPC) and psychrotrophic microorganism count(PMC), without affecting, the physicochemical characteristics of the milk (p > 0.05). Poor hygienic conditions in milk storage rooms, in the refrigeration bulk tanks of the dairy farms, and during the transport, as well as the transport on longer routes, were related to higher SPC and psychrotrophic count while receiving milk by the dairy industry with higher temperatures was only associated with higher SPC.(AU)

RÉSUMÉ

The study aimed to evaluate the use of the azidiol® preservative for psychrotrophic microorganism count (PMC) in cooled raw milk. Two studies were carried out, one under controlled conditions (experiment 1) and the other under field conditions (experiment 2), in which samples of raw milk were taken with and without the use of the azidiol® preservative and analyzed at predefined times (0, 6, 12 and 24 hours -experiment 1) and at varying times (experiment 2). Analysis of variance and regression analysis using SAS were applied for data statistical analysis. Milk samples without azidiol®showed higher PMC with increasing time between sampling and analysis, while in samples preserved with azidiol®, this count remained constant. Samples of cooled raw milk intended for PMC should be collected in flasks containing the azidiol®preservative.(AU)

RÉSUMÉ

Leptospirosis is a zoonotic infectious disease caused by the bacterium Leptospira spp. The consequences of infection in horses are unclear, and the serogroups involved vary depending on the region. This study aimed to evaluate the seropositivity of unvaccinated horses in the Serra Catarinense region and the possible risk factors associated with the infection. We used 207 horses from 26 properties, with no clinical history of infection and unvaccinated for leptospirosis. We submitted serum samples to the microscopic agglutination serum (SAM) to obtain the main reference serogroups. We considered animals with titre 100 as infected. The results of the serology revealed seropositivity in 45.4% (94/207) of the animals studied. The most frequent antibodies reportedwere those against the serogroups Australis 16.9% (35/207), Ichterhaemorrhagiae 14.4% (30/207), and Grippotyphosa 5.31% (11/207). Among the primary risk factors associated with the infection we reported extensive rearing system (OR = 1.27; P 0.05) and the presence of other animal species such as cattle (OR = 3.85; P 0.01) and capybaras (OR = 2.07; P=0.06). The findings presented in this study showed that leptospirosis is endemic in horses in the Serra Catarinense region, and revealed the need for emergency surveillance and control measures specific to this important zoonosis.(AU)

Our objective was the characterization and staging of histological lesions in different anatomical sites of the central nervous system (CNS) of rabid cattle. The severity of the lesions was compared with the clinical stages of the disease, the variants of viral isolates, and with the load of virus. Thirty-one spontaneously affected rabid cattle the state of Santa Catarina underwent clinical follow-up and were eventually necropsied. CNS tissues were sampled and submitted to direct fluorescent antibody technique (DFAT), immunohistochemistry (IHC), routine histopathology with hematoxylin and eosin stain (HE), reverse transcriptase polymerase chain reaction (RT-PCR), and polymerase chain reaction in quantitative reverse transcriptase in real time (qRT-PCR). Affected cattle were allotted in four groups according to their clinical stage when euthanized: G1, euthanized while standing; G2, euthanized when in sternal recumbence; G3, euthanized when in lateral recumbence; and G4, affected cattle with natural death. In order to evaluate the degree of severity of the lesions and the presence of Negri bodies (NBs), the brain was sectioned at 9 sites. Additionally, spinal cord and trigeminal ganglion sections were examined. The intensity of the lesions was graded as either absent, mild, moderate, or marked, and the presence or absence of the NBs was noted. Histological lesions were characterized by lymphocytic and monocytic meningoencephalitis with NBs in 28 cases. In all analyzed groups, intensities of histological lesions ranging from mild to severe were observed. Brain regions with the highest inflammatory lesion intensity were the medulla at the level of obex, followed by the colliculus and thalamus. NBs were observed in a higher percentage in the cerebellum, followed by medulla at the obex level, striatum complex, and frontal telencephalon. The duration of the clinical course of the disease did not influence the intensity of the inflammatory lesion, but it did influence the presence of NBs, with a higher percentage of these inclusions in cattle that died naturally than in euthanized cattle. All isolated rhabdovirus included in this study were genetically compatible with samples from hematophagous bats Desmodus rotundus. The evaluation by qRT-PCR did not demonstrate a correlation between lesion intensity and the amount of virus.(AU)

The objective of this study was to compare the reproductive performance, milk yield and composition, and udder health and conformation traits of Holstein (Ho), F1, and R1 crossbred Ho × Simmental (Sim) cows. Three commercial dairy farms in south Brazil were used as the research units. All farms held Ho, F1, and R1 crossbred Ho × Sim (¾ Ho × » Sim and ¾ Sim × » Ho) cows. The collection of milk samples and evaluation of udder conformation traits occurred during four visits to each farm. In addition to the actively collected data, retrospective reproduction records of the farms served as the basis for the statistical analysis using analysis of variance models using SAS. The F1 crossbred Ho × Sim cows and ¾ Sim (first rotational crossbreeding generation = R1 using Sim semen) cows had a shorter calving interval and calving to first service interval compared to the Ho cows (P < 0.0001). Milk yield did not differ among the genetic groups except for R1 (¾ Sim) that produced approximately 10% less milk than the other groups (P = 0.0245). Fat plus protein yield and somatic cell score did not differ among the genetic groups. Ho cows had shallower udders (P < 0.0001) and a higher udder clearance (P < 0.0001) than the other groups. F1 and R1 crossbred Ho × Sim cows had shorter reproduction intervals than purebred Ho cows. Although udder conformation traits lacked high-quality scores in crossbred cows, somatic cell scores reached the same level as in purebred Ho cows.

RÉSUMÉ

To reduce costs, lactating cows are often fed a higher proportion of roughage and alternative feeds; however, such protocols may not lead to optimal milk production. Using a meta-analysis, we evaluated the effects of dietary roughage sources on the composition, voluntary intake, and digestibility of diets fed to crossbred Holstein x Zebu cows under experimental feeding conditions in Brazil. Finally, we assessed the effects of dietary roughage sources on milk yield and composition. The database was composed of 43 experiments with 183 treatments, including 943 lactating cows. The data were obtained from studies published between January 2000 and December 2015. The data were evaluated by analysis of variance, considering the distribution of variables. The diets differed in dry matter and crude protein (CP) contents according to the roughage sources. Diets based on corn silage demonstrated higher intakes of neutral detergent fiber (NDF), CP, and total digestible nutrients, which improved the digestibility of NDF and CP, and resulted in a higher milk yield compared to the diets with one roughage source or those with a combination of two roughage sources. The present study demonstrated that feeding crossbred Holstein x Zebu lactating cows alternative roughage sources compromised milk production and that lactating cows produced higher milk yields when fed corn silage.

RÉSUMÉ

We aimed to determine the relationship of milk yield, milk composition, and somatic cell score (SCS) with functional traits (lameness score, udder cleanliness score, udder depth, and teat end hyperkeratosis score) and physiological status (parity and days in milk (DIM) of cows housed in compost bedded pack barn system (CBP) using multivariate analysis. Data were collected in September and October of 2017 and evaluated using factor and cluster analysis. Four factors were formed, with the first factor showing the traits responsible for altering the lactose content in milk, the second factor comprising the effect of DIM, the third factor representing the teat end hyperkeratosis score, and the fourth factor demonstrating the udder cleanliness. The cluster analysis formed three clusters. Clusters 2 and 3 differed for protein and lactose content, SCS, parity, DIM, lameness score, and udder depth. Cluster 1 differed from the others, especially due to the high milk yield. Cow conformation traits and physiological status affected milk yield and composition in CBP. There is an association between udder depth, parity, and SCS with negative impact on lactose content. Parity is also associated with higher milk yield and teat end hyperkeratosis score. Cows with lower lameness score presented dirtier udders due to increased movements on the feed parlor and water troughs areas. Special attention to the improvement of the udder conformation traits, as well as management practices that reduce hyperkeratosis, are necessary to keep high yielding, healthy, and cows with higher longevity. The cleanliness in the feed parlor and water troughs area is very important to preserve milk quality and healthy cows.(AU)

RÉSUMÉ

Artisanal mountain cheese (queijo artesanal serrano; AMS) is produced from raw milk, which makes it a suitable medium for the multiplication of pathogenic microorganisms. The ripening process involves physical, chemical and microbiological factors unfavorable to pathogen development. For this reason, the Brazilian legislation requires a minimum ripening time of 60 days. However, according to Normative Instruction (NI) no 30, of August 7, 2013, and Decree no. 9.013, of March 29, 2017, this time can be reduced provided that scientific studies show that there is no alteration in the safety of the product. This study aimed to evaluate the microbiological quality of AMS in 19 cheese factories in the Serra Catarinense region, at 14, 21, 28 and 35 days of ripening, and to determine which period would meet the microbiological limits. A total of 76 samples of AMS, 19 samples of water and 19 samples of raw milk were studied. The microorganisms were investigated following Ordinance no. 146, of March 7, 1996. The investigation and count of coliforms at 30 °C, E. coli and coagulase-positive Staphylococcus followed protocols by Silva et al. (2010), using Petrifilm™. The investigation of Salmonella spp. followed Silva et al. (2010), whereas the investigation of Listeria monocytogenese and count of mesophilic aerobes followed NI no. 30, of June 26, 2018. Water potability was evaluated by the Most Probable Number (MPN) technique, in accordance with NI no. 30/2018 and Consolidation Ordinance no. 5, of September 28, 2017. Results were tested by Analysis of Variance and Linear Regression using the SAS statistical package with significance determined at p < 0.05. No safe ripening period for sale could be determined. At 35 days of ripening, 14 farms still showed counts above the limit for the microorganisms described in the legislation. On one of the farms, the presence of L. monocytogenes was detected in the four evaluated periods, demonstrating that ripening was not efficient to eliminate the pathogen. The microbiological quality of the milk was not satisfactory, with 63.16% of the samples not conforming to the standards. As regards the potability of the water used in the cheese factories, 52.63% of the analyzed samples showed unsatisfactory results. The research revealed heterogeneity in the hygienic-sanitary standards adopted by the cheese factories, indicating that there is a need for training in Good Agricultural Practices and Good Manufacturing Practices, support from institutions, periodic inspection and commitment of producers to standardize the process.(AU)

The aim of this study was to assess the metaphylactic effect of subcutaneous doses of trace minerals (copper, zinc, selenium, and manganese) and vitamins A and E on biochemical (NEFA, BHBA, fructosamine, urea, triglycerides, HDL, and calcium) and hormonal parameters (cortisol, insulin, and IGF-1) of Holstein cows in the transition period. Sixty animals kept in a semi-confinement system, with an average production of 22 liters of milk per day (Farm 1) and 24 liters of milk per day (Farm 2) and a diet according to NRC (2001) with a low metabolic challenge, were divided into two groups: a treatment group, supplemented with trace minerals and vitamins (MTVG) (n=30), and a control group (CG) (n=30). The collection of blood samples was performed at the moments M1 (21 days prepartum), M2 (14 days prepartum), M3 (7 days prepartum), M4 (parturition day), M5 (7 days postpartum), M6 (14 days postpartum), and M7 (21 days postpartum). The experimental design was a randomized block design and the data were submitted to analysis of variance with repeated measures in time within the random variable cow. Differences were considered significant at 5% significance level and trend at 10% level. No difference was observed between treatment and the interaction treatment × day in the assessed parameters. However, some variables showed a significant difference of day. NEFA levels (P < 0.0001) were higher on the parturition day. BHBA concentrations were higher after parturition (P=0.0031), with a peak at the third week of lactation. Fructosamine showed increasing values until the parturition day (P=0.0373), in which higher concentrations were observed. Average values of HDL were higher at 21 days postpartum (P < 0.0001). Triglycerides levels remained high during the prepartum period but showed a sudden drop on the parturition day (P < 0.0001), with lower values postpartum.[...]

This study compared the superovulatory response of embryos donor mares of Quarter Horse (QH) and Crioula using a protocol with low dose of equine pituitary extract (EPE). Eight QH mares and six Crioula mares were monitored during 3 consecutive estrous cycles, with each cycle corresponds to a group. Control Group - monitoring of follicle growth up to the ovulation; EPE group - when the follicles reached 20 mm in diameter were administered 7 mg EPE twice daily. When the follicles achieved a diameter 35 mm, the ovulation was induced with 2500UI hCG; Post-EPE Group - idem to the control group. Data were analyzed using GLM procedure of the SAS statistical package. The variables were submitted to the Tukeys test and least square means adjusted for multiple comparisons using the Tukey-Kramers test, with significance level of 5%. Values are presented as mean ± SD. In the Crioula mares treated with EPE, there was an increase in the number of ovulations (p =0.0534) with a mean of 3.33 ± 2.06 ovulations when compared to the control and post-EPE groups, and mares QM. (2.00±0.53). Treatment with EPE in the recommended dose allowed better superovulatory response and rate of embryo recovery in Crioula mares when compared to the QM mares.(AU)

RÉSUMÉ

The defatted dry extract (DDE) comprises of the solid constituents of the milk, except for the fat, and must have a minimum established content of 8.4% for raw milk in Brazil. The objective was to evaluate the effects of commercialized milk volume, somatic cell count (SCC) and total bacterial count (TBC) on the DDE content of dairy farm milk samples. The sampling included data from monthly milk analyzes of dairy farms, which supply four dairy industries located in the state of Santa Catarina, Paraná and Rio Grande do Sul, and comprised of 106.470 observations sampled in 2015.The data were submitted to variance and regression analysis. To evaluate the influence of the volume of milk commercialized by the farmers, the SCC and TBC levels, and the levels of DDE, the values of these explanatory variables were stratified in quintiles. The mean DDE was 8.55%, with 25.6% of the samples being below the established minimum. In regard to the volume of milk marketed by the farmers, seasonal variations were observed in all quintiles of dairy farms analyzed, but with lower levels of DDE in milk samples from small farms throughout the year. Samples of milk with high TBC and, especially high SCC, presented lower percentage of DDE during the year. It is concluded that the DDE content of milk is related to the season of the year, the production volume per dairy, SCC and TBC.(AU)

The objective was to investigate the effects of available shaded area on behavioural characteristics of lactating cows during the warm season in the subtropics. Twelve Holstein cows were divided into groups, with 2 (SH2) or 10 m2 (SH10) of available shade per cow (six cows per treatment). The behaviour was recorded when animals were grazing from 09.00 to 16.00 h (GMT −02.00 h) throughout the trial. Data considered in the statistical analysis were restricted to the five days when the maximum temperature was ≥ 25 °C. Cows in the SH10 group had lower panting score, but higher proportions of time in shade and lying down and stayed more time per visit in the shaded area than cows in the SH2 group. Cows in the SH10 group had less events of competition near the water trough and in the shaded area than cows in the SH2 group. The solar radiation threshold that triggered the use of the shade was lower in SH10 than in SH2 group. Hourly proportion of time spent grazing or ruminating differed between shaded areas at specific times. The 2 m2 of available shaded area per cow is sufficient for all cows to enter the shaded area at the same time. In spite of that, 2 m2 of available shaded area per cow negatively affects the proportion of time spent lying and in the shaded area; in addition, it enhances agonistic behaviours between cows when compared with 10 m2 of shade per cow.(AU)

RÉSUMÉ

The aim of this study was to evaluate the composition, voluntary intake, and digestibility of the diet of Holstein cows, as well as milk yield and milk composition as a function of different roughage sources used under experimental feeding conditions in Brazil, through meta-analysis. The database of this study consisted of 109 experiments with 424 treatments of 3,903 lactating Holstein cows. The data were obtained from studies published between January 2000 and December 2015, and were evaluated by variance analysis. Lactating dairy cow diets with a single source of roughage other than corn silage resulted in lower voluntary intake, milk yield, and differences in milk composition, compared to diets based on corn silage or the combination of two roughages, probably due to reduced nutrient digestibility. Diets based on corn silage are distinguished by feed efficiency, milk yield, and composition. Diets based on the combination of two roughage sources are similar with respect to milk yield and composition, compared to diets based on corn silage; whereas, diets with a single source of roughage (other than corn silage), even when containing a higher proportion of concentrate, led to reduced voluntary intake, milk yield, and food efficiency due to the lower utilization of the nutrients. Diets based on corn silage in general allow a higher proportion of roughage in the diet due to energy density.(AU)
