RÉSUMÉ

Advanced mRNA vaccines play vital roles against SARS-CoV-2. However, most current mRNA delivery platforms need to be stored at -20 °C or -70 °C due to their poor stability, which severely restricts their availability. Herein, we develop a lyophilization technique to prepare SARS-CoV-2 mRNA-lipid nanoparticle vaccines with long-term thermostability. The physiochemical properties and bioactivities of lyophilized vaccines showed no change at 25 °C over 6 months, and the lyophilized SARS-CoV-2 mRNA vaccines could elicit potent humoral and cellular immunity whether in mice, rabbits, or rhesus macaques. Furthermore, in the human trial, administration of lyophilized Omicron mRNA vaccine as a booster shot also engendered strong immunity without severe adverse events, where the titers of neutralizing antibodies against Omicron BA.1/BA.2/BA.4 were increased by at least 253-fold after a booster shot following two doses of the commercial inactivated vaccine, CoronaVac. This lyophilization platform overcomes the instability of mRNA vaccines without affecting their bioactivity and significantly improves their accessibility, particularly in remote regions.

RÉSUMÉ

In the face of resource and environmental problems caused by extensive economic development, China has put forward a green development strategy. Scientific measurement and analysis of green total factor productivity (GTFP) is of great significance for achieving high-quality economic development. By introducing the human capital composition, including education, health, scientific research, and training, this paper study adopts the Slack Based Measure-Global Malmquist-Luenberger (SBM-GML) index to re-measure the GTFP and its decomposition of 30 provinces, municipalities, and autonomous regions in China from 2000 to 2019. The results show that: (1) China's GTFP has a fluctuating growth trend, with an average annual growth rate of 2.31%. (2) In terms of its decomposition, technical progress is the main force driving GTFP growth, with a contribution rate of 1.59%; the improvement of technical efficiency is a secondary driving force, with a contribution rate of 0.71%. (3) The measurement results of GTFP, considering the human capital composition, are generally higher than those without consideration, and the GTFP growth under the two modes shows a trend of "high in the east and low in the west". The conclusions have enlightening significance for improving GTFP and the growth potential of the economy in China.

RÉSUMÉ

This paper makes a new attempt to identify the effectiveness of innovation factor allocation with a random forest method. This method avoids the evaluation bias of the relative effectiveness caused by the noneffective selection of production frontier in the nonparametric DEA method. It does not refer to other optimal subjects but shifts the focus to the judgment of its own effectiveness. In addition, it also gets rid of the constraints of the model and variables in the parameter SFA method, ensuring the reliability of the measurement results by resampling thousands of times. The data is collected from 30 provinces in China from 2009 to 2018. The findings show the innovation factor allocation in more than half of the provinces is not fully effective. It indicates that how to make use of innovation factor inputs to achieve the actual innovation output higher than own optimal levels is currently still in a period of exploration in China. To further improve innovation factor allocation efficiency, it deeply analyzes the impacts of innovation factor inputs and finds out the important innovation factor inputs. Furthermore, this study presents the nonlinear characteristics and optimal combination of important innovation factor inputs. According to this, it offers the detailed suggestions about how to adjust current important innovation factor inputs for each province in order to greatly enhance the effectiveness of innovation factor allocation in the future.
